RESUMEN

The emergence of next-generation sequencing (NGS) technologies has revolutionized the way to investigate the microbial diversity in traditional fermentations. In the field of food microbial ecology, different NGS platforms have been used for community analysis, including 454 pyrosequencing from Roche, Illumina's instruments and Thermo Fisher's SOLiD/Ion Torrent sequencers. These recent platforms generate information about millions of rDNA amplicons in a single running, enabling accurate phylogenetic resolution of microbial taxa. This review provides a comprehensive overview of the application of NGS for microbiome analysis of traditional fermented milk products worldwide. Fermented milk products covered in this review include kefir, buttermilk, koumiss, dahi, kurut, airag, tarag, khoormog, lait caillé, and suero costeño. Lactobacillus-mainly represented by Lb. helveticus, Lb. kefiranofaciens, and Lb. delbrueckii-is the most important and frequent genus with 51 reported species. In general, dominant species detected by culturing were also identified by NGS. However, NGS studies have revealed a more complex bacterial diversity, with estimated 400-600 operational taxonomic units, comprising uncultivable microorganisms, sub-dominant populations, and late-growing species. This review explores the importance of these discoveries and address related topics on workflow, NGS platforms, and knowledge bioinformatics devoted to fermented milk products. The knowledge that has been gained is vital in improving the monitoring, manipulation, and safety of these traditional fermented foods.

OBJECTIVE: To determine whether tocilizumab improves clinical outcomes for patients with severe or critical coronavirus disease 2019 (covid-19). DESIGN: Randomised, open label trial. SETTING: Nine hospitals in Brazil, 8 May to 17 July 2020. PARTICIPANTS: Adults with confirmed covid-19 who were receiving supplemental oxygen or mechanical ventilation and had abnormal levels of at least two serum biomarkers (C reactive protein, D dimer, lactate dehydrogenase, or ferritin). The data monitoring committee recommended stopping the trial early, after 129 patients had been enrolled, because of an increased number of deaths at 15 days in the tocilizumab group. INTERVENTIONS: Tocilizumab (single intravenous infusion of 8 mg/kg) plus standard care (n=65) versus standard care alone (n=64). MAIN OUTCOME MEASURE: The primary outcome, clinical status measured at 15 days using a seven level ordinal scale, was analysed as a composite of death or mechanical ventilation because the assumption of odds proportionality was not met. RESULTS: A total of 129 patients were enrolled (mean age 57 (SD 14) years; 68% men) and all completed follow-up. All patients in the tocilizumab group and two in the standard care group received tocilizumab. 18 of 65 (28%) patients in the tocilizumab group and 13 of 64 (20%) in the standard care group were receiving mechanical ventilation or died at day 15 (odds ratio 1.54, 95% confidence interval 0.66 to 3.66; P=0.32). Death at 15 days occurred in 11 (17%) patients in the tocilizumab group compared with 2 (3%) in the standard care group (odds ratio 6.42, 95% confidence interval 1.59 to 43.2). Adverse events were reported in 29 of 67 (43%) patients who received tocilizumab and 21 of 62 (34%) who did not receive tocilizumab. CONCLUSIONS: In patients with severe or critical covid-19, tocilizumab plus standard care was not superior to standard care alone in improving clinical outcomes at 15 days, and it might increase mortality. A digestibility experiment was conducted to evaluate the effects of dietary exogenous monocomponent protease on the coefficient of apparent total tract digestibility (ATTD) and coefficient apparent ileal digestibility (AID) of amino acids in meat and bones meal (MBM), poultry byproducts meal (PBPM), and feather meal (FM). A total of 512 Cobb-500 male broiler chickens (aged 14 d) were randomly placed into 64 metabolism cages (8 birds per pen) and were allocated to eight treatments with eight replicates in a semi-controlled environmental room. The experimental diets consisted of the basal diet (corn/SBM) and the replacement of 300 g/kg on a weight basis with MBM, PBPM, or FM. The excreta were collected during 3 d (19 to 21 d), and the ileal digesta (using Celite as an indigestible marker) only 1 d (21 d). The protease contained 75,000 PROT units/g. The use of the enzyme increased (P < 0.05) ATTD for alanine, cysteine, glycine, and threonine in the basal diet and AID for the amino acids alanine, cysteine, glycine, lysine, threonine, and valine for the basal diet (vegetable). Regarding meals, there was an increase (P < 0.05) in the amino acid digestibility in large part due to the amino acids of MBM (14) and PBPM (9), with only five amino acids for FM. The amino acids glycine and threonine showed increases (P < 0.05) in both total and ileal digestibility for all animal meals and for the diet based on corn and soybean meal, which indicates a high specificity of the enzyme for these amino acids. The overall results obtained in this study found satisfactory effectiveness of this exogenous protease. The total collection method was lower by 1.83% of amino acids digestibility than the ileal method.

Microalgae are sources of nutritional products and biofuels. However, their economical processing is challenging, because of (i) the inherently low concentration of biomass in algal cultures, below 0.5%, (ii) the high-water content in the harvested biomass, above 70%; and (iii) the variable intracellular content and composition. Cell wall structure and strength vary enormously among microalgae, from naked Dunaliella cells to robust Haematococcus cysts. High-value products justify using fast and energy-intensive processes, ranging from 0.23 kWh/kg dry biomass in high-pressure homogenization, to 6 kWh/kg dry biomass in sonication. However, in biofuels production, the energy input must be minimized, requiring slower, thermal or chemical pretreatments. Whichever the primary fraction of interest, the spent biomass can be processed into valuable by-products. This review discusses microalgal cell structure and composition, how it affects pretreatment, focusing on technologies tested for large scale or promising for industrial processes, and how these can be integrated into algal biorefineries.

This experiment was carried out to study the effect of dietary exogenous monocomponent protease on the coefficient apparent total tract (ATTD) and apparent ileal (AID) digestibility of amino acids of corn, soybean meal (SBM), and full fat soybean meal (FFSM) in broilers. A total of 400 males Cobb-500 (14 d of age) were equally allocated in 80 metabolic cages (50 cm × 50 cm × 45 cm) in a completely randomized design and a semi-controlled environment. Eight treatments (basal diet with or without a protease and three ingredients replacing the basal diets in 40% to corn and 30% to SBM and FFSM, with and without protease), with 10 replicates each were evaluated. The protease was added at 200 mg/kg resulting in 15,000 unit of PROT/kg. The total collection of excreta was held during 3 d, after 5 d for adaptation of broilers at the diets. The ileal content was collected on d 21, after slaughter of birds. The enzyme increased (P < 0.05) the ATTD and AID of most amino acids contained in SBM and FFSM. The digestibility of cysteine, glycine, proline, and threonine had higher (P < 0.05) ATTD and AID in all tested ingredients with the use of protease. On average, the dietary protease increased in 5.19% and 3.86% the total and ileal digestibility of amino acids, respectively. It was concluded that the dietary protease exerts major effects on toasted full-fat soybean, followed by soybean meal and corn to broilers.

Here, we report the draft genome sequence of Pediococcus acidilactici strain LPBC161, a lactic acid bacterium isolated from mature coffee cherries in Brazil. The genome sequence of P. acidilactici LPBC161 provides valuable information on the mechanisms of adaptation and metabolism of lactic acid bacteria (LAB) in the environment and stressor factors of coffee processing.

An environmental friendly process was developed to produce Arthrospira maxima's biomass from sugarcane vinasse, which was generated in a bioethanol production chain, at laboratory and pilot scale. Peptides fractions were than obtained from enzymatically hydrolyzed biomass. High microalgae biomass productivities were reached (0.150 g L-1 day-1) coupled with a significant reduction of BOD and COD (89.2 and 81%, respectively). Three peptide fractions were obtained from microalgae biomass through single or sequential enzymatic hydrolysis. Antioxidant, antimicrobial, anti-inflammatory, and/or anti-collagenase activities of biopetides' fractions were observed. The PHS showed multi-biological activities. The three peptides fractions could be potential candidates for different applications in pharmaceutical, cosmetic and food industry.

Lactic acid is a product that finds several applications in food, cosmetic, pharmaceutical and chemical industries. The main objective of this work was the development of a bioprocess to produce L(+)-lactic acid using soybean vinasse as substrate. Among ten strains, Lactobacillus agilis LPB 56 was selected for fermentation, due to its ability to metabolize the complex oligosaccharides. Fermentation was conducted without need for supplementary inorganic nitrogen sources or yeast extract. Kinetic and yield parameters determined at laboratory scale were 0.864 and 0.0162 for YP/S and YX/S, 0.0145 g/L h (rx), 1.32 g/L h (rs) and 1.13 g/L h (rp). The use of vinasse enriched with soybean molasses provided higher lactic acid concentration (138 g/L), the best proportion of inoculum being 25% (v/v). After scale-up to a pilot plant, kinetic and yield parameters were 0.849 and 0.0353 for YP/S and YX/S, 0.0278 g/L h (rx), 0.915 g/L h (rs) and 0.863 g/L h (rp).

RESUMEN

The aim of this work was to develop an economical bioprocess to produce the bio-ethanol from soybean molasses at laboratory, pilot and industrial scales. A strain of Saccharomyces cerevisiae (LPB-SC) was selected and fermentation conditions were defined at the laboratory scale, which included the medium with soluble solids concentration of 30% (w/v), without pH adjustment or supplementation with the mineral sources. The kinetic parameters - ethanol productivity of 8.08g/Lh, YP/S 45.4%, YX/S 0.815%, m 0.27h(-1) and microX 0.0189h(-1) - were determined in a bench scale bioreactor. Ethanol production yields after the scale-up were satisfactory, with small decreases from 169.8L at the laboratory scale to 163.6 and 162.7L of absolute ethanol per ton of dry molasses, obtained at pilot and industrial scales, respectively.
