The aim of this study was to evaluate the effect of grazing methods and intensities on the metabolic profile of lambs grazing ryegrass. This study was performed in Eldorado do Sul (RS). The treatments consisted of two grazing intensities with rotational and continuous stocking methods: moderate and low. The experimental design was a randomized block arranged in a factorial scheme with four replicates. To evaluate the metabolic and nutritional conditions of the lambs, blood samples were collected to evaluate phosphorus, albumin, glucose, magnesium, globulin, cortisol, urea and total proteins. The highest blood urea value was observed in the month of September in lambs kept in continuous method grazing. There was no statistical difference between the grazing intensities and grazing methods for albumin, glucose, magnesium, globulin, cortisol and total protein levels in profile lambs. The cortisol values suggested that the animals were not subjected to high stress levels. In conclusion, rearing female lambs on ryegrass, irrespective of grazing methods or intensities, allowed the maintenance of body homeostasis and did not cause any metabolic unbalances, nutritional unbalances or stress.(AU)


O objetivo deste estudo foi avaliar o efeito dos métodos e das intensidades de pastejo no perfil metabólico de cordeiros. Este estudo foi realizado em Eldorado do Sul (RS). Os tratamentos consistiram em duas intensidades de pastejo (moderada e baixa), com métodos lotação rotativa e lotação contínua. O delineamento experimental foi em blocos ao acaso, em esquema fatorial com quatro repetições. Para avaliar as condições metabólicas e nutricionais dos cordeiros, foram coletadas amostras de sangue para avaliar fósforo, albumina, glicose, magnésio, globulina, cortisol, ureia e proteínas totais. O maior valor de ureia no sangue foi observado no mês de setembro nos cordeiros mantidos em pastejo contínuo. Não houve diferença estatística entre as intensidades de pastejo e os métodos de pastejo para albumina, glicose, magnésio, globulina, cortisol e níveis de proteína total nos cordeiros perfilados. Os valores de cortisol sugeriram que os animais não foram submetidos a altos níveis de estresse. Concluindo, a criação de cordeiros em azevém, independentemente dos métodos ou das intensidades de pastejo, permitiu a manutenção da homeostase corporal e não causou desequilíbrios metabólicos, desequilíbrios nutricionais ou estresse.(AU)

An understanding of the processes involved in grazing behaviour is a prerequisite for the design of efficient grassland management systems. The purpose of managing the grazing process is to identify sward structures that can maximize animal forage daily intake and optimize grazing time. Our aim was to evaluate the effect of different grazing management strategies on foraging behaviour and herbage intake by sheep grazing Italian ryegrass under rotational stocking. The experiment was carried out in 2015 in southern Brazil. The experimental design was a randomized complete block with two grazing management strategies and four replicates. The grazing management treatments were a traditional rotational stocking (RT), with pre- and post-grazing sward heights of 25 and 5 cm, respectively, and a 'Rotatinuous' stocking (RN) with pre- and post-grazing sward heights of 18 and 11 cm, respectively. Male sheep with an average live weight of 32 ± 2.3 kg were used. As intended, the pre- and post-grazing sward heights were according to the treatments. The pre-grazing leaf/stem ratio of the Italian ryegrass pasture did not differ between treatments (P > 0.05) (~2.87), but the post-grazing leaf/stem ratio was greater (P < 0.001) in the RN than in the RT treatment (1.59 and 0.76, respectively). The percentage of the non-grazed area was greater (P < 0.01) in post-grazing for RN compared with RT treatment, with an average of 29.7% and 3.49%, respectively. Herbage nutritive value was greater for the RN than for the RT treatment, with greater CP and lower ADF and NDF contents. The total time spent grazing, ruminating and resting did not differ between treatments (P > 0.05), with averages of 439, 167 and 85 min, respectively. The bite rate, feeding stations per min and steps per min by sheep were greater (P < 0.05) in the RN than in the RT treatment. The grazing time per hour and the bite rate were greater (P < 0.05) in the afternoon than in the morning in both treatments. The daily herbage intake by sheep grazing Italian ryegrass was greater (P < 0.05) in the RN than in the RT treatment (843.7 and 707.8 g organic matter/sheep, respectively). Our study supports the idea that even though the grazing time was not affected by the grazing management strategies when the animal behaviour responses drive management targets, such as in 'Rotatinuous' stocking, the sheep herbage intake is maximized, and the grazing time is optimized.

Diante da escassez de dados sobre a topografia e a sintopia das vísceras abdominopélvicas do tamanduá-bandeira (Myrmecophage tridactyla - Linnaeus, 1758), o presente estudo teve como objetivo elucidar essas características e compará-las com as demais espécies animais, mormente as domésticas. Utilizaram-se três espécimes, dois machos e uma fêmea, provenientes de doação da Polícia Militar Ambiental de Franca ao Laboratório de Anatomia Veterinária da Universidade de Franca, após óbitos por atropelamentos. Os animais foram fixados e mantidos em solução aquosa de formaldeído a 10%, seguidos de dissecação convencional das cavidades abdominopélvicas para posterior inspeção direta e descrição topográfica das vísceras, visando a análises comparativas com outras espécies, cujo posicionamento e cujas particularidades já são bem estabelecidos na literatura. Observou-se que a maioria das vísceras dessas cavidades possuem localização e sintopia similares aos animais domésticos, exceto os rins e os testículos. Diante da metodologia estabelecida e dos resultados obtidos, admite-se que mais espécimes de tamanduás-bandeiras, de ambos os gêneros, devam ser avaliados e registrados cientificamente, visando à confirmação dos dados da atual pesquisa e à preconização anatômica da cavidade abdominopélvica, visto que variações anatômicas individuais são passíveis entre animais da mesma espécie.(AU)


Objetivou-se avaliar a fauna vetorial e os aspectos ambientais e climáticos relacionados à transmissão das leishmanioses. Foi realizado um estudo eco-epidemiológico prospectivo de coleta sistemática de flebotomíneos e inquérito censitário sorológico canino em áreas de um município do Brasil. Para determinar a taxa de prevalência de LVC, foram examinadas amostras de sangue de 1752 cães. Na avaliação entomológica, foram instaladas 24 armadilhas luminosas em 12 residências distribuídas, instaladas no ambiente de peridomicílio e intradomicílio durante 12 meses. Para análise dos aspectos climáticos, utilizou-se a correlação simples de Spearman e para análise espacial foram utilizadas a Lógica Fuzzy e a Função K. A taxa de prevalência em cães foi de 4,1% e 7,1%. No estudo entomológico, foram capturados 431 flebotomíneos. A maior parte (74%) dos espécimes foi capturada no peridomicílio. Em relação à infecção natural, 5,6 % das amostras analisadas por biologia molecular apresentaram positividade à infecção por Leishmania spp.. Em 100% das amostras positivas, encontrou-se infecção por Leishmania infantum. Na análise espacial uma Área apresentou maior concentração de pontos de sobreposição de alta densidade de Lutzomyia longipalpis e cães sororreagentes, indicando maior risco na ocorrência concomitante dos dois eventos. Os resultados mostram que a interface parasito-reservatório-vetor está ativa nas áreas estudadas.(AU)

Chromobacterium violaceum is a free-living bacillus with several genes that enables it survival under different harsh environments such as oxidative and temperature stresses. Here we performed a label-free quantitative proteomic study to unravel the molecular mechanisms that enable C. violaceum to survive oxidative stress. To achieve this, total proteins extracted from control and C. violaceum cultures exposed during two hours with 8 mM hydrogen peroxide were analyzed using GeLC-MS proteomics. Analysis revealed that under the stress condition, the bacterium expressed proteins that protected it from the damage caused by reactive oxygen condition and decreasing the abundance of proteins responsible for bacterial growth and catabolism. GeLC-MS proteomics analysis provided an overview of the metabolic pathways involved in the response of C. violaceum to oxidative stress ultimately aggregating knowledge of the response of this organism to environmental stress. This study identified approximately 1500 proteins, generating the largest proteomic coverage of C. violaceum so far. We also detected proteins with unknown function that we hypothesize to be part of new mechanisms related to oxidative stress defense. Finally, we identified the mechanism of clustered regularly interspaced short palindromic repeats (CRISPR), which has not yet been reported for this organism.

Campomanesia adamantium (Cambess.) O. Berg. is originally from Brazil. Its leaves and fruits have medicinal properties such as anti-inflammatory, antidiarrheal and antiseptic properties. However, the mutagenic potential of this species has been reported in few studies. This study describes the mutagenic/antimutagenic, splenic phagocytic, and apoptotic activities of C. adamantium hydroethanolic extract with or without cyclophosphamide in Swiss mice. The animals orally received the hydroethanolic extract at doses of 30, 100, or 300 mg/kg with or without 100 mg/kg cyclophosphamide. Mutagenesis was evaluated by performing the micronucleus assay after treatment for 24, 48, and 72 h, while splenic phagocytic and apoptotic effects were investigated after 72 h. Short-term exposure of 30 and 100 mg/kg extract induced mild clastogenic/aneugenic effects and increased splenic phagocytosis and apoptosis in the liver, spleen, and kidneys. When the extract was administered in combination with cyclophosphamide, micronucleus frequency and apoptosis reduced. Extract components might affect cyclophosphamide metabolism, which possibly leads to increased clearance of this chemotherapeutic agent. C. adamantium showed mutagenic activity and it may decrease the effectiveness of drugs with metabolic pathways similar to those associated with cyclophosphamide. Thus, caution should be exercised while consuming these extracts, especially when received in combination with other drugs.

Acrocomia aculeata is a plant rich in antioxidant compounds. Studies suggest that this plant has anti-inflammatory, antidiabetic, and diuretic potential. We assessed the antigenotoxic, antimutagenic, immunomodulation, and apoptotic potentials of A. aculeata alone and in combination with an antitumor agent, cyclophosphamide. Swiss male mice (N = 140) were used. The animals were divided into 14 experimental groups as follows: a negative group, a positive group (100 mg/kg cyclophosphamide), groups that only received the oil extracted from the almond (AO) and from the pulp (PO) of A. aculeata at doses of 3, 15, and 30 mg/kg, and the associated treatment groups (oils combined with cyclophosphamide) involving pretreatment, simultaneous, and post-treatment protocols. Data suggest that both oils were chemopreventive at all doses, based on the tested protocols. The highest damage reduction percentages, observed for AO and PO were 88.19 and 90.03%, respectively, for the comet assay and 69.73 and 70.93%, respectively, for the micronucleus assay. Both AO and PO demonstrated immunomodulatory activity. The oils reduced the capacity of cyclophosphamide to trigger apoptosis in the liver, spleen, and kidney cells. These results suggest that A. aculeate AO and PO can be classified as a functional food and also enrich other functional foods and nutraceuticals with chemopreventive features. However, they are not appropriate sources for chemotherapeutic adjuvants, in particular for those used in combination with cyclophosphamide.

Management strategies for increasing ruminant legume consumption and mitigating methane emissions from tropical livestock production systems require further study. The aim of this work was to evaluate the herbage intake, animal performance and enteric methane emissions of cattle grazing dwarf elephant grass (DEG) (Pennisetum purpureum cv. BRS Kurumi) alone or DEG with peanut (Arachis pintoi cv. Amarillo). The experimental treatments were the following: DEG pastures receiving nitrogen fertilization (150 kg N/ha as ammonium nitrate) and DEG intercropped with peanut plus an adjacent area of peanut that was accessible to grazing animals for 5 h/day (from 0700 to 1200 h). The animals grazing legume pastures showed greater average daily gain and herbage intake, and shorter morning and total grazing times. Daily methane emissions were greater from the animals grazing legume pastures, whereas methane emissions per unit of herbage intake did not differ between treatments. Allowing animals access to an exclusive area of legumes in a tropical grass-pasture-based system can improve animal performance without increasing methane production per kg of dry matter intake.

To maximize herbage DMI, pregrazing sward surface height (SSH) and level of herbage depletion (HD) must be such that variables determining short-term herbage intake such as bite mass (BM) and bite rate (BR) are optimized. The objective of this study was to determine a SSH target and the level of HD as a proportion of the SSH that optimizes BM and BR of beef heifers grazing Sorghum bicolor swards. Two experiments were conducted using 2 S. bicolor swards and 4 beef heifers (25 mo old; 322 kg BW). Experiment 1 compared the effect of 6 pregrazing SSH, 30, 40, 50, 60, 70, and 80 cm, on BM, BR, and jaw movements. Experiment 2 assessed the effect of HD level as a proportion of SSH (0.17, 0.34, 0.50, 0.67 and 0.84) on BM, BR, and jaw movements using the optimal pregrazing SSH defined in Exp. 1. Short-term herbage DMI was estimated using a double-weighing technique and corrected for insensible BW loss. Herbage DMI was subsequently used to calculate the BM. Net eating time and jaw movements for apprehension and manipulation + mastication during grazing as well as total jaw movements were determined using the IGER (Institute of Grassland and Environmental Research) behavior recorders. Bite rate and the number of total jaw movements per gram herbage DMI were derived from jaw movement count and measurements of herbage DMI. The results of Exp. 1 showed low and high SSH constraint the ease of herbage harvesting. Greater BM are maintained until a SSH of 50 cm is reached (P < 0.05) and then decline at greater SSH due to herbage dispersion. The nonbiting jaw movement rate increased at greater SSH whereas BR decreased (P < 0.05). For both variables, the turning point was close to a SSH of 50 cm. Experiment 2 showed that such an optimization of BM and BR was maintained until an HD level of 0.34 was reached (P < 0.05). There was a linear increase in both the total jaw movements per unit herbage DMI and the nonbiting jaw movements rate (manipulation + mastication) subsequent to levels of HD greater than 0.34 (P < 0.05). These studies provide, for the first time, sward feature targets to manage grazing and optimize BM and BR, aiming to maximize the short-term herbage DMI of cattle grazing S. bicolor swards.

Mass-spectrometry-based shotgun proteomics has become a widespread technology for analyzing complex protein mixtures. Here we describe a new module integrated into PatternLab for Proteomics that allows the pinpointing of differentially expressed domains. This is accomplished by inferring functional domains through our cloud service, using HMMER3 and Pfam remotely, and then mapping the quantitation values into domains for downstream analysis. In all, spotting which functional domains are changing when comparing biological states serves as a complementary approach to facilitate the understanding of a system's biology. We exemplify the new module's use by reanalyzing a previously published MudPIT dataset of Cryptococcus gattii cultivated under iron-depleted and replete conditions. We show how the differential analysis of functional domains can facilitate the interpretation of proteomic data by providing further valuable insight.

Avaliaram-se os efeitos da estratégia de pastoreio rotacionado sobre a dinâmica do rebaixamento do milheto (Pennisetum glaucum (L.) R. Br.) pastejado por vacas leiteiras. Os tratamentos consistiram de um fatorial entre duas alturas pré-pastejo, 60 e 40cm, e duas pós-pastejo, 20 e 10cm, alocados em um delineamento em blocos ao acaso com quatro repetições. A estrutura do pasto foi avaliada durante o processo de pastejo a cada 10 minutos, por meio de medições de altura. A taxa de rebaixamento da altura do pasto no tratamento 40-20 apresentou-se relativamente constante ao longo de todo o rebaixamento. O tratamento 60-10 apresentou o maior declínio na taxa de rebaixamento. A taxa de rebaixamento no tratamento 40-10 diminuiu em ritmo mais lento em relação à dos tratamentos com 60cm de altura pré-pastejo. As maiores taxas de ingestão foram observadas nos tratamentos de altura pré-pastejo de 60cm, e considerou-se como a melhor estratégia de manejo a combinação 60-20cm de altura do pasto.


This study aimed to evaluate the effects of rotational grazing strategies on the dynamics of the grazing pastures of millet (Pennisetum glaucum (L.) R. Br.) grazed by dairy cows. Treatments consisted of two pre-grazing heights (60 and 40cm) and two post-grazing heights (20 and 10cm) allocated in a randomized block design with four replications. The sward height was evaluated during grazing every 10 minutes. The rate of grazing down of the sward height of the pasture on the 40-20 grazing strategy was relatively constant throughout the grazing down. Treatment 60-10 showed the largest decline in the rate of grazing down. The rate of grazing down in the 40-10 treatment decreased with a slower intensity when compared to the treatments with 60cm of pre-grazing sward height. The major intake rates were observed in treatments with pre-grazing sward height of 60cm. The best management strategy was considered the combination of 60-20cm of sward height.

Os tratamentos consistiram de um fatorial entre duas alturas pré-pastejo, 60 e 40cm, e duas pós-pastejo, 20 e 10cm, alocados em um delineamento em blocos ao acaso com quatro repetições. A estrutura do pasto foi avaliada durante o processo de pastejo a cada 10 minutos, por meio de medições de altura. A taxa de rebaixamento da altura do pasto no tratamento 40-20 apresentou-se relativamente constante ao longo de todo o rebaixamento. O tratamento 60-10 apresentou o maior declínio na taxa de rebaixamento. A taxa de rebaixamento no tratamento 40-10 diminuiu em ritmo mais lento em relação à dos tratamentos com 60cm de altura pré-pastejo. As maiores taxas de ingestão foram observadas nos tratamentos de altura pré-pastejo de 60cm, e considerou-se como a melhor estratégia de manejo a combinação 60-20cm de altura do pasto.(AU)


This study aimed to evaluate the effects of rotational grazing strategies on the dynamics of the grazing pastures of millet (Pennisetum glaucum (L.) R. Br.) grazed by dairy cows. Treatments consisted of two pre-grazing heights (60 and 40cm) and two post-grazing heights (20 and 10cm) allocated in a randomized block design with four replications. The sward height was evaluated during grazing every 10 minutes. The rate of grazing down of the sward height of the pasture on the 40-20 grazing strategy was relatively constant throughout the grazing down. Treatment 60-10 showed the largest decline in the rate of grazing down. The rate of grazing down in the 40-10 treatment decreased with a slower intensity when compared to the treatments with 60cm of pre-grazing sward height. The major intake rates were observed in treatments with pre-grazing sward height of 60cm. The best management strategy was considered the combination of 60-20cm of sward height.(AU)

RESUMEN

The treatment of a transverse maxillary deficiency in skeletally mature individuals should include surgically assisted rapid palatal expansion. This study evaluated the distribution of stresses that affect the expander's anchor teeth using finite element analysis when the osteotomy is varied. Five virtual models were built and the surgically assisted rapid palatal expansion was simulated. Results showed tension on the lingual face of the teeth and alveolar bone, and compression on the buccal side of the alveolar bone. The subtotal Le Fort I osteotomy combined with intermaxillary suture osteotomy seemed to reduce the dissipation of tensions. Therefore, subtotal Le Fort I osteotomy without a step in the zygomaticomaxillary buttress, combined with intermaxillary suture osteotomy and pterygomaxillary disjunction may be the osteotomy of choice to reduce tensions on anchor teeth, which tend to move mesiobuccally (premolar) and distobuccally (molar).

Gastric cancer is one of most frequent causes of death in Brazil. The city of Manaus has one of the highest incidences of this disease in Brazil. The Epstein-Barr virus (EBV) is a ubiquitous herpesvirus that is classified as a group 1 carcinogen by the International Agency for Research on Cancer. We obtained biopsies from 6 control subjects and 10 patients with gastric carcinomas living in Manaus. In the patients, the samples were taken from tumors and from adjacent non-cancerous mucosa. These samples were screened for EBV DNA by PCR to amplify the 288-bp fragments from the Bam M region. The EBV DNA was detected in 8 of the 10 tumor cases and in none of the 6 control subjects. In the positively identified samples, EBV DNA was detected in five corresponding resection margins. Previous research indicated only a weak association between EBV and gastric cancer. We suggest that EBV should be considered as a risk factor for gastric adenocarcinomas in Manaus.

Spectral counting is a strategy to quantify relative protein concentrations in pre-digested protein mixtures analyzed by liquid chromatography online with tandem mass spectrometry. In the present study, we used combinations of normalization and statistical (feature selection) methods on spectral counting data to verify whether we could pinpoint which and how many proteins were differentially expressed when comparing complex protein mixtures. These combinations were evaluated on real, but controlled, experiments (yeast lysates were spiked with protein markers at different concentrations to simulate differences), which were therefore verifiable. The following normalization methods were applied: total signal, Z-normalization, hybrid normalization, and log preprocessing. The feature selection methods were: the Golub index, the Student t-test, a strategy based on the weighting used in a forward-support vector machine (SVM-F) model, and SVM recursive feature elimination. The results showed that Z-normalization combined with SVM-F correctly identified which and how many protein markers were added to the yeast lysates for all different concentrations. Statistical modeling of links between genetic profiles with environmental and clinical data to aid in medical diagnosis is a challenge. Here, we present a computational approach for rapidly selecting important clinical data to assist in medical decisions based on personalized genetic profiles. What could take hours or days of computing is available on-the-fly, making this strategy feasible to implement as a routine without demanding great computing power. The key to rapidly obtaining an optimal/nearly optimal mathematical function that can evaluate the "disease stage" by combining information of genetic profiles with personal clinical data is done by querying a precomputed solution database. The database is previously generated by a new hybrid feature selection method that makes use of support vector machines, recursive feature elimination and random sub-space search. Here, to evaluate the method, data from polymorphisms in the renin-angiotensin-aldosterone system genes together with clinical data were obtained from patients with hypertension and control subjects. The disease "risk" was determined by classifying the patients' data with a support vector machine model based on the optimized feature; then measuring the Euclidean distance to the hyperplane decision function. Our results showed the association of renin-angiotensin-aldosterone system gene haplotypes with hypertension. The association of polymorphism patterns with different ethnic groups was also tracked by the feature selection process. A demonstration of this method is also available online on the project's web site.

Objetivo: descrever características espirométricas de crianças e adolescentes asmáticos acompanhados em ambulatório especializado. Métodos: cem asmáticos maiores de seis anos foram aleatoriamente convidados para o estudo. Sete recusaram-se a participar. Testes de função pulmonar (FP) foram realizados pela manhã (espirômetro Vitalograph®), quando os pacientes estavam livres de infecções respiratórias há pelo menos duas semanas e após suspensão de broncodilatadores de curta elonga ação. Foram obtidas curvas de fluxo-volume antes e 15 minutos após inalação de 400g de salbutamol. Doze pacientes não foram capazes de realizar curvas reprodutíveis. Resultados: a mediana de idade, foi 11,5 anos. Dezenove pacientes apresentavam asma intermitente (AI), 24 asma persistente leve (APL), 22 asma persistente moderada (APM) e 16 asma persistente grave (APG). As medianas dos valores basais de FP para AI, APL, APM e APG foram respectivamente: capacidadevital forçada (CVF): 96 per cent, 88 per cent, 84 per cent e 90 per cent; volume expiratório forçado no primeiro segundo (VEF1): 84 per cent, 82 per cent, 73 per cent e 65 per cent; fluxo expiratório forçado entre 25 per cent e 75 per cent da CVF (FEF25-75 per cent): 78 per cent, 66 per cent, 60 per cent e 43 per cent. Houve aumento significante nos valores de FP após salbutamol para todos os parâmetros avaliados. Não houve correlação entre a gravidade da asma e o incremento de FP após salbutamol. O FEF25-75 per cent foi o parâmetro que melhor discriminou a gravidade da asma e que indicou alterações funcionais em maior número de casos. Nãohouve correlação entre o tempo de doença e os valores de FP. Conclusões: a espirometria é útil na avaliação de crianças e adolescentes asmáticos. O FEF25-75 per cent é o parâmetro mais sensível para a avaliação da gravidade da doença.

RESUMEN

A 67 year-old normotensive woman had a syncope followed by shock and remained anuric after hemodynamic stabilization. Paraplegia and paresis of the right upper limb, as well as signs of ischemia of the distal lower limbs were noted. The possibility of acute aortic dissection was raised and confirmed by computed tomography. The paraplegia was attributed to an ischemic infarction of the spinal cord. The patient died on the fourth hospital day due to a pericardial temponade. This rare and not well recognized complication of aortic dissection is briefly reviewed.
