RESUMEN

The Coral Triangle encompasses nearly 30% of the world's coral reefs and is widely considered the epicenter of marine biodiversity. Destructive fishing practices and natural disturbances common to this region damage reefs leaving behind fields of coral rubble. While the impacts of disturbances in these ecosystems are well documented on metazoans, we have a poor understanding of their impact on microbial communities at the base of the food web. We use metabarcoding to characterize protist community composition in sites of varying fisheries management schemes and benthic profiles across the island of Lombok, Indonesia. Our study shows that rubble coverage and net primary productivity are the strongest explainers of variation in protist communities across Lombok. More specifically, rubble fields are characterized by increases in small heterotrophic protists, including ciliates and cercozoans. In addition to shifts in heterotrophic protist communities, we also observed increases in diatom relative abundance in rubble fields, which corresponded to sites with higher net primary productivity. These results are the first to characterize protist communities in tropical marine rubble fields and provide insight on environmental factors potentially driving these shifts on a local scale.

Climate change will likely intensify hurricane activity in coastal regions. A thorough understanding of hurricane impacts to marine fauna is necessary to prepare for and mitigate potential impacts to social systems dependent upon adjacent fauna. Yet, research attention, conservation funding, and policy all can be biased toward taxa of societal interest, potentially favoring a limited understanding of hurricane impacts. Here, we analyzed the frequency of mentions of taxa in newspaper articles in relation to hurricane activity at three coastal US locations coupled with analysis of long-term fisheries-independent data. While economically important taxa dominate media discourse, we observed long-term hurricane-related abundance declines in ecologically important taxa having little direct human utility. We conclude that there is a potential for research and policy biases related to hurricane impacts. Preparation and mitigation efforts will benefit from researchers and managers making directed efforts to identify and incorporate hurricane sensitive taxa into their work.

Marine food webs are structured through a combination of top-down and bottom-up processes. In coral reef ecosystems, fish size is related to life-history characteristics and size-based indicators can represent the distribution and flow of energy through the food web. Thus, size spectra can be a useful tool for investigating the impacts of both fishing and habitat condition on the health and productivity of coral reef fisheries. In addition, coral reef fisheries are often data-limited and size spectra analysis can be a relatively cost-effective and simple method for assessing fish populations. Abundance size spectra are widely used and quantify the relationship between organism size and relative abundance. Previous studies that have investigated the impacts of fishing and habitat condition together on the size distribution of coral reef fishes, however, have aggregated all fishes regardless of taxonomic identity. This leads to a poor understanding of how fishes with different feeding strategies, body size-abundance relationships, or catchability might be influenced by top-down and bottom-up drivers. To address this gap, we quantified size spectra slopes of carnivorous and herbivorous coral reef fishes across three regions of Indonesia representing a gradient in fishing pressure and habitat conditions. We show that fishing pressure was the dominant driver of size spectra slopes such that they became steeper as fishing pressure increased, which was due to the removal of large-bodied fishes. When considering fish functional groups separately, however, carnivore size spectra slopes were more heavily impacted by fishing than herbivores. Also, structural complexity, which can mediate predator-prey interactions and provisioning of resources, was a relatively important driver of herbivore size spectra slopes such that slopes were shallower in more complex habitats. Our results show that size spectra slopes can be used as indicators of fishing pressure on coral reef fishes, but aggregating fish regardless of trophic identity or functional role overlooks differential impacts of fishing pressure and habitat condition on carnivore and herbivore size distributions.

The nonapeptide arginine vasotocin (AVT) regulates osmotic balance in teleost fishes, but its mechanisms of action are not fully understood. Recently, it was discovered that nonapeptide receptors in teleost fishes are differentiated into two V1a-type, several V2-type, and two isotocin (IT) receptors, but it remains unclear which receptors mediate AVT's effects on gill osmoregulation. Here, we examined the role of nonapeptide receptors in the gill of the euryhaline Amargosa pupfish (Cyprinodon nevadensis amargosae) during osmotic acclimation. Transcripts for the teleost V1a-type receptor v1a2 were upregulated over fourfold in gill 24 h after transferring pupfish from 7.5 ppt to seawater (35 ppt) or hypersaline (55 ppt) conditions and downregulated after transfer to freshwater (0.3 ppt). Gill transcripts for the nonapeptide degradation enzyme leucyl-cystinyl aminopeptidase (LNPEP) also increased in fish acclimating to 35 ppt. To test whether the effects of AVT on the gill might be mediated by a V1a-type receptor, we administered AVT or a V1-type receptor antagonist (Manning compound) intraperitoneally to pupfish before transfer to 0.4 ppt or 35 ppt. Pupfish transferred to 35 ppt exhibited elevated gill mRNA abundance for cystic fibrosis transmembrane conductance regulator (cftr), but that upregulation diminished under V1-receptor inhibition. AVT inhibited the increase in gill Na+/Cl- cotransporter 2 (ncc2) transcript abundance that occurs following transfer to hypoosmotic environments, whereas V1-type receptor antagonism increased ncc2 mRNAs even without a change in salinity. These findings indicate that AVT acts via a V1-type receptor to regulate gill Cl- transport by inhibiting Cl- uptake and facilitating Cl- secretion during seawater acclimation.

Pupfishes (genus Cyprinodon) evolved some of the broadest salinity tolerances of teleost fishes, with some taxa surviving in conditions from freshwater to nearly 160 ppt. In this study, we examined transcriptional dynamics of ion transporters and aquaporins in the gill of the desert Amargosa pupfish (Cyprinodon nevadensis amargosae) during rapid salinity change. Pupfish acclimated to 7.5 ppt were exposed to freshwater (0.3 ppt), seawater (35 ppt), or hypersaline (55 ppt) conditions over 4 h and sampled at these salinities over 14 d. Plasma osmolality and Cl- concentration became elevated 8 h after the start of exposure to 35 or 55 ppt but returned to baseline levels after 14 d. Osmolality recovery was paralleled by increased gill Na+/K+-ATPase activity and higher relative levels of messenger RNAs (mRNAs) encoding cystic fibrosis transmembrane conductance regulator (cftr) and Na+/K+/2Cl- cotransporter-1 (nkcc1). Transcripts encoding one Na+-HCO3- cotransporter-1 isoform (nbce1.1) also increased in the gills at higher salinities, while a second isoform (nbce1.2) increased expression in freshwater. Pupfish in freshwater also had lower osmolality and elevated gill mRNAs for Na+/H+ exchanger isoform-2a (nhe2a) and V-type H+-ATPase within 8 h, followed by increases in Na+/H+ exchanger-3 (nhe3), carbonic anhydrase 2 (ca2), and aquaporin-3 (aqp3) within 1 d. Gill mRNAs for Na+/Cl- cotransporter-2 (ncc2) also were elevated 14 d after exposure to 0.3 ppt. These results offer insights into how coordinated transcriptional responses for ion transporters in the gill facilitate reestablishment of osmotic homeostasis after changes in environmental salinity and provide evidence that the teleost gill expresses two Na+-HCO3- cotransporter-1 isoforms with different roles in freshwater and seawater acclimation.
