RESUMEN

The present study describes the community diversity and gives a seasonal distribution of an intertidal macroalgal assemblage at Prainha Beach, Arraial do Cabo City in Rio de Janeiro state, Brazil. Sampling was performed during four seasons of a 1-year period. Organisms were sampled and photographed using photoquadrats to quantify relative coverage. Ninety-six taxa of macroalgae were registered, including 23 chlorophytes, 19 ochrophytes, and 54 rhodophytes, while three barnacles, seven mollusks, one isopod, and one polychaete were recorded among the zoobenthos. In the upper intertidal zone, the coverage was frequently dominated by invertebrates. Macroalgae always dominated the middle and lower zones, covering almost 100 % in both zones. The highest values of species diversity (H'), richness (S), and turnover rates were found during the transition from spring to summer, which coincided with the upwelling period. A comparison with the 1980's flora revealed that the major changes in the macroalgal assemblage were among species belonging to the same genera, unless new additions were provided. The combination of photoquadrats and minimally destructive sampling allowed a detailed description of the composition and structural characteristics of the intertidal zone, a methodology that should be applied to study protected marine areas.

Prorocentrum lima is a widely distributed marine benthic dinoflagellate that produces diarrhetic toxins, okadaic acid (OA) and its analogs, that may promote damage on bivalve tissues and cellular responses. Cultivation of the brown mussel Perna perna represents an important economic activity in the tropical and subtropical regions, where mussels may co-occur with P. lima. This study aimed to assess the behavioral, cellular immune responses, and pathological condition of P. perna following a short-term experimental exposure to P. lima. The toxic dinoflagellate treatment was compared to a non-toxic exposure to the chlorophyte Tetraselmis sp. at similar concentrations. The prevalence of pathological conditions and parasites were assessed, and a pathological index was applied by scoring the prevalences into four levels. Reaction time and the number of stimuli necessary for shell-valve closure response significantly increased after 72 h of P. lima exposure. Circulating hemocyte concentration was significantly lower in P. lima exposed mussels than in control mussels at 48- and 96 h of incubation, while hemocyte relative size in exposed mussels was significantly higher than that in control mussels. Comparatively, phagocytic activity and ROS production by hemocytes was significantly higher in mussels exposed to P. lima at 48- and 96 h of incubation, respectively. In addition, exposed mussels significantly presented exacerbated hemocytic infiltration in digestive organs, higher prevalence of moderate to severe atrophy in digestive tubules, and higher pathological index which suggests an impairment of mussel immunologic responses. A lower prevalence of Rickettsia-like organisms (RLOs), trematodes and copepods in P. lima exposed mussels suggests a direct toxic effect of OA on parasites. The exposure of mussels to P. lima is likely to occur frequently and may lead to constraints on mussel behavior, physiology, and pathological condition.

Kleptoplasty is the retention of plastids obtained from ingested algal prey, which can remain temporarily functional and be used for photosynthesis by the predator. With a new approach based on cell cycle analysis, we have addressed the question of whether the toxic, bloom-forming dinoflagellate Dinophysis norvegica practice kleptoplasty or if they replicate their own plastid DNA. Dividing (G2) and non-dividing (G1) D. norvegica cells from a natural population were physically separated with a flow cytometer based on their DNA content. Average numbers of nuclear and plastid rDNA copies were quantified with real-time PCR both in the G1 and G2 group. Cells from the G1 group contained 5800 +/- 340 copies of nuclear rDNA and 1300 +/- 200 copies of plastid rDNA; cells from the G2 group contained 9700 +/- 58 copies of nuclear rDNA and 1400 +/- 220 copies of plastid rDNA (mean +/- SD, n = 3). The ratio G2/G1 in average rDNA copies per cell was 1.67 for nuclear DNA and 1.07 for plastid DNA. These ratios show that plastid acquisition in D. norvegica is either uncoupled with the cell cycle, or plastids accumulate rapidly in the beginning of the cell cycle owing to feeding, as would be expected in a protist with kleptoplastic behaviour but not in a protist with own plastid replication. In addition, flow cytometry measurements on cells from the same population used for real-time PCR showed that when kept without plastidic prey, live Dinophysis cells lost on average 36% of their plastid phycoerythrin fluorescence in 24 h. Together these findings strongly suggest that D. norvegica does not possess the ability for plastid replication.
