RESUMEN

This work is an evaluation of metal contamination degree in Phorcus turbinatus and a monitoring of metals impact on metallothioneins functioning. The gastropod was sampled from six stations along the northeastern and eastern coasts of Tunisia during four seasons (2014-2015). Our results suggested that sedimentological metallic contents (Copper, Zinc and Cadmium) vary significantly depending on stations and not significantly according to seasons except for copper. Nevertheless, differences were no significant between stations and significant between seasons as for ambient factors, metallic intrinsic contents and metallothioneins rates. The monodonta tissue seems to be enriched in the four analyzed metals (Cu, Zn, Cd and Hg) and this species seems to have an ability to accumulate metals. Metal effect on the protein induction may be linked to physicochemical factors (temperature, O2 and Cu contents in sediment). In fact, metallothioneins levels were positively correlated to the dissolved oxygen rates and negatively linked to temperature.

Impacts of chemical contaminants associated with dumping of dredged urban river sediments at a coastal disposal area in Saronikos Gulf (Eastern Mediterranean) were investigated through a combined approach of sediment toxicity testing and active biomonitoring with caged mussels. Chemical analyses of aliphatic hydrocarbons (AHs), polycyclic aromatic hydrocarbons (PAHs), Cu, and Zn in combination with the solid phase Microtox® test were performed on sediments. Concentrations of PAHs, AHs, Cu, and Zn as well as multiple biomarkers of contaminant exposure and/or effects were measured in caged mussels. Sediments in the disposal and neighboring area showed elevated PAHs and AHs concentrations and were characterized as toxic by the solid-phase Microtox® test during and after dumping operations. Biomarker results in the caged mussels indicated sublethal effects mainly during dumping operations, concomitantly with high concentrations of PAHs and AHs in the caged mussel tissues. Cu and Zn concentrations in sediments and caged mussels were generally not elevated except for sediments at the site in the disposal area that received the major amount of dredges. High PAHs and AHs levels as well as sublethal effects in the caged mussels were not persistent after termination of operations. The combined bioassay-biomarker approach proved useful for detecting toxicological impacts of dredged river sediment disposal in sediments and the water column. Nevertheless, further research is needed to evaluate whether sediment toxicity will have long-term effects on benthic communities of the disposal area.

Metal concentrations were determined in surface sediments and transplanted mussels in a shallow semi-enclosed bay, Pagassitikos Gulf, Aegean Sea. Cu, Zn, Pb, Hg, and As levels in sediments were enriched close to point sources of pollution, i.e., Volos Port and cement plant. Fe, Mn, Cr, and Ni spatial distributions in sediments were rather uniform with increasing concentrations towards deeper sites, suggesting natural enrichment, whereas levels found in the port, although elevated, were not the highest in the study area. Spatial variations in concentrations of most metals in mussels transplanted nearshore were generally in accordance to those in sediments, with the highest Fe, Mn, Cr, Ni, and Cu levels in the port and the highest Hg levels close to the cement plant. Concentrations of metals in sediments and transplanted mussels were comparable to those reported in Mediterranean areas. Results indicate that pollution point sources contribute to anthropogenic metal enrichment in the study area.

We studied if the levels of copper released from antifouling treated nets used in finfish mariculture could affect the immune defense mechanism and/or induce oxidative stress in Dicentrarchus labrax, after short term exposure in laboratory experiments. Dissolved copper concentration released from the treated nets, copper bioavailability and a set of biomarkers responses were measured. Biomarkers included hemoglobin concentration, activities of lysozyme, total complement, respiratory burst, glutathione S-transferase and acetycholinesterase and concentration of thiobarbituric acid reactive substances. Results indicated elevated copper concentration in seawater (184 µg L(-1)) but low concentration in muscle (1.5 µg g(-1)) and liver (117 µg g(-1)). Copper bioavailability was independent of copper complexes with dissolved organic carbon. However, formation of copper complexes with other matrices could neither be excluded nor justified. The released copper from the treated nets did not induce oxidative stress but affected the immediate immune defense mechanism of the exposed fish making them more easily vulnerable to diseases. Consequently, copper-based antifouling treated nets could be a risk factor for D. labrax health.

An integrated approach using biomarkers and contaminant levels in mussels Mytilus galloprovincialis L. was employed to assess chemical contamination in Greek coastal waters within the framework of the MYTIMED program. Biomarkers (metallothioneins, glutathione S-transferase, catalase, acetylcholinesterase and RNA:DNA ratio) have been previously described in mussels caged at 14 sites in coastal areas influenced by different types of anthropogenic activities. This study applied a biomarker index, the Integrated Biomarker Response (IBR) to summarize biomarker responses and relate stress levels to concentrations of organochlorine compounds (PCBs, DDTs), polycyclic aromatic hydrocarbons (PAHs), and metals (Cu, Ni, Fe, Zn) measured in the mussel tissues. The IBR index indicated environmental stress at sites near cities and industries and was overall related to organic contaminants, but also elevated metal concentrations at certain sites. Slightly increased IBR values at few sites away from known pollution sources were not accompanied with increased contaminant levels suggesting stress possibly caused by natural factors. Results confirmed the usefulness of integration of biological effects measurements and chemical analysis for the assessment of chemical contamination in coastal waters.

Mussels are worldwide recognized as pollution bioindicators and used in Mussel Watch programs, because they accumulate pollutants in their tissues at elevated levels in relation to pollutant biological availability in the marine environment. The present study deals with the use of Mytilus galloprovincialis as a local bioindicator of heavy metal and (137)Cs contamination in an estuarine ecosystem (Thermaikos gulf, Greece in Eastern Mediterranean). M. galloprovincialis samples were collected monthly from two aquaculture farms during the period April to October 2000. Analyses for the heavy metals Cu, Cr, Ni, Zn, Fe, Mn and (137)Cs showed that the concentrations measured were low and similar to those from other non-polluted Mediterranean areas. In terms of the two sampling stations, there were no statistically significant differences between them. On the contrary, the seasonal evolution of either heavy metals or (137)Cs levels presented high variation. The levels were found to increase during the cold period of the year, especially for Cu, Zn, Mn and Cr which are essential for life. Stable metals were positively inter-related and moreover, metals more involved in biochemical activities seem to present more correlations than others with less significant role in the metabolism of the organisms.

Statistical power analysis has been used to determine the sensitivity of the mussel-watch programme in Hellenic seas, with particular focus placed on the data obtained for metal content in mussels from the Saronicos Gulf during the period 1985-1997. Sample collection was carried out quarterly. Soft tissues of 60-100 similarly sized mussels (Mytilus galloprovincialis) were pooled into six composite samples and analysed by Atomic Absorption Spectroscopy (AAS). The study concludes that the resolution of monitoring programmes employed thus far is sufficiently high to detect temporal trends related to changes in contaminant levels caused either by drastic environmental control (such as those imposed on point-source pollutants) or by sudden increases in contaminant load. However, the detection of more subtle trends, which may be expected in the near future as a result of global pollution-control policies, is likely to require the adoption of much more sensitive programmes. In addition, the review of up-to-date knowledge on metal biology in mussels suggests that factors related to growth and reproduction should be taken into account to remarkably increase the resolution of mussel-watch programmes. Indicative maximum values for components of error to be complied with during routine monitoring are suggested.
