RESUMEN

Alzheimer's disease (AD) is one of the most common neurodegenerative diseases among the elderly and it accounts for nearly 80% of all dementias. The pathogenesis of AD is complicated and enigmatic thus far. The mitochondrial cascade hypothesis assumes that mitochondrial damage may mediate, drive, or contribute to a variety of AD pathologies and may be the main factor in late-onset AD. Currently, there are no widely recognized drugs able to attenuate mitochondrial damage in AD. Notably, increasing evidence supports the efficacy of acupuncture for improving the mitochondrial structure and protecting mitochondrial functions in AD. This review reports the mechanisms by which acupuncture regulates mitochondrial dynamics, energy metabolism, calcium homeostasis and apoptosis. In conclusion, these findings suggest that AD mitochondrial dysfunction represents a reasonable therapeutic target and acupuncture could play a significant role in preventing and treating AD.

Evacuation dynamics of pedestrians in a square room with one exit is studied. The movement of the pedestrians is guided by the static floor field model. Whenever multiple pedestrians are trying to move to the same target position, a game theoretical framework is introduced to address the conflict. Depending on the payoff matrix, the game that the pedestrians are involved in may be either hawk-dove or prisoner's dilemma, from which the reaped payoffs determine the capacities, or probabilities, of the pedestrians occupying the preferred vacant sites. The pedestrians are allowed to adjust their strategies when competing with others, and a parameter κ is utilized to characterize the extent of their self-interest. It is found that self-interest may induce either positive or negative impacts on the evacuation dynamics depending on whether it can facilitate the formation of collective cooperation in the population or not. Particularly, a resonance-like performance of evacuation is realized in the regime of prisoner's dilemma. The effects of placing an obstacle in front of the exit and the diversity of responses of the pedestrians to the space competition on the evacuation dynamics are also discussed.

Chaperone-mediated autophagy (CMA) is one of the three types of autophagy. In recent years, CMA has been shown to be associated with the pathogenesis of several types of cancer. However, whether CMA is involved in the pathogenesis of colorectal cancer (CRC) remains unclear. In this study, we investigated CMA activity in tissue specimens from CRC patients and mouse models of colitis-associated CRC (induced by administration of AOM plus DSS). In addition, we down-regulated CMA in CT26 colon carcinoma cells stably transfected with a vector expressing a siRNA targeting LAMP-2A, the limiting component in the CMA pathway, to explore the role of CMA in these cells. Apoptosis was detected using TUNEL assay, and the apoptosis-related proteins were detected using western blotting. Cell proliferation was assessed using MTT assay, Ki-67 labelling and western blotting for PCNA. We found that LAMP-2A expression was significantly increased in CRC patients and mouse models and varied according to the stage of the disease. Inhibition of CMA in CT26 cells facilitated apoptosis, as evidenced by increased TUNEL immunolabeling, increased expression of Bax and Bnip3, and decreased expression of Bcl-2. Cell proliferation assays showed that inhibition of CMA impeded the proliferation of CT26 cells. These data support the hypothesis that CMA is up-regulated in CRC, and inhibition of CMA may be a new therapeutic strategy for CRC patients.
