RESUMEN

Policy evaluations are crucial in promoting effective administrations. However, evaluations of spatial policies are rare, due to its ambiguity and complexity. Especially when a developmental state transitions to democracy, the loss of top-down leadership disables formulation of indicators for evaluations. In such cases, it is essential to search for new evaluation methods and approaches. This paper introduces a Taiwanese and East Asian context, with a unique historical path toward democracy, which is rarely discussed regarding policy evaluation, especially spatial policy evaluation. As the "hard" developmental states democratize and value more human aspects instead of sheer economic development, compared with "hard" methods such as quantitative impact evaluations, "soft" measures like process evaluation, participatory evaluation and self-assessment may enhance cross-departmental dialogues and connections between policies and executors. With humans at the center of space use and policy making, this research explores the possibility of bottom-up evaluation without indicators. The new ideas proposed here bridge the vacancies of power dominance in developmental states during transition, and serve as references for such states in current times, especially in the Asian region.

RESUMEN

The COVID-19 pandemic has impacted the industry immensely and, in some cases, irreversibly. This research pioneers in studying how the pandemic have influenced the survival and spatial distribution of the health-related manufacturing industry (HRMI) in Taiwan. Eight categories of HRMI are examined, with their change in survival performances and spatial concentration between 2018 and 2020. Average Nearest Neighbour and Local Indicators of Spatial Association are conducted, to visualise the distribution of industrial clusters. We found the pandemic did not shock the HRMI in Taiwan, but actually induced its growth and spatial concentration to a certain extent. Additionally, due to it being a knowledge-intensive industry, the HRMI mainly concentrate in metropolitan areas with which universities and science parks may have largely supported. However, the spatial concentration and cluster scope growth do not necessarily accompany the improvement of spatial survival, which may be resulted from the different life cycle stages an industry category is in. This research fills in the gap of medical studies with literatures and data from the field of spatial studies. It provides interdisciplinary insights under the condition of pandemic.

The ClC-2 channel plays a critical role in maintaining ion homeostasis in the brain and the testis. Loss-of-function mutations in the ClC-2-encoding human CLCN2 gene are linked to the white matter disease leukodystrophy. Clcn2-deficient mice display neuronal myelin vacuolation and testicular degeneration. Leukodystrophy-causing ClC-2 mutant channels are associated with anomalous proteostasis manifesting enhanced endoplasmic reticulum (ER)-associated degradation. The molecular nature of the ER quality control system for ClC-2 protein remains elusive. In mouse testicular tissues and Leydig cells, we demonstrated that endogenous ClC-2 co-existed in the same protein complex with the molecular chaperones heat shock protein 90ß (Hsp90ß) and heat shock cognate protein (Hsc70), as well as the associated co-chaperones Hsp70/Hsp90 organizing protein (HOP), activator of Hsp90 ATPase homolog 1 (Aha1), and FK506-binding protein 8 (FKBP8). Further biochemical analyses revealed that the Hsp90ß-Hsc70 chaperone/co-chaperone system promoted mouse and human ClC-2 protein biogenesis. FKBP8 additionally facilitated membrane trafficking of ClC-2 channels. Interestingly, treatment with the Hsp90-targeting small molecule 17-allylamino-17-demethoxygeldanamycin (17-AAG) substantially boosted ClC-2 protein expression. Also, 17-AAG effectively increased both total and cell surface protein levels of leukodystrophy-causing loss-of-function ClC-2 mutant channels. Our findings highlight the therapeutic potential of 17-AAG in correcting anomalous ClC-2 proteostasis associated with leukodystrophy.

RESUMEN

An antimicrobial peptide, LL-37, is found in an innate defense system of humans. Patients who suffer urinary tract infection (UTI) will generate LL-37 and which is released into urine. LL-37 can be used as an indicator for the diagnosis of UTI. We have designed a biosensor with an interdigitated electrode on a printed-circuit board (PCB). The surface of the electrode was modified with 3-mercaptopropionic acid and immobilized with anti-LL37 antibody to improve the specificity of the biosensor. By de-embedding jig impedance, the impedance associated with the change of LL-37 concentration was calculated. The sensitivity of this biosensor for LL-37 in a urine sample can reach 50 µg/mL.
