RESUMEN

Escherichia coli releases outer membrane vesicles (OMVs) into the extracellular environment. OMVs, which contain the outer membrane protein, lipopolysaccharides (LPS), and genetic material, play an important role in immune response modulation. An isobaric tag for relative and absolute quantitation (iTRAQ) analysis was used to investigate OMV constituent proteins and their functions in burn trauma. OMV sizes ranged from 50 to 200 nm. Proteomics and Gene Ontology analysis revealed that ΔrfaC and ΔrfaG were likely involved in the upregulation of the structural constituent of ribosomes for the outer membrane and of proteins involved in protein binding and OMV synthesis. ΔrfaL was likely implicated in the downregulation of the structural constituent of the ribosome, translation, and cytosolic large ribosomal subunit. Kyoto Encyclopedia of Genes and Genomes analysis indicated that ΔrfaC and ΔrfaG downregulated ACP, ACEF, and ADHE genes; ΔrfaL upregulated ACP, ACEF, and ADHE genes. Heat map analysis demonstrated upregulation of galF, clpX, accA, fabB, and grpE and downregulation of pspA, ydiY, rpsT, and rpmB. These results suggest that RfaC, RfaG, and RfaL proteins were involved in outer membrane and LPS synthesis. Therefore, direct contact between wounds and LPS may lead to apoptosis, reduction in local cell proliferation, and delayed wound healing.

Sericin, a waste product of the silk textile industry, has favorable physicochemical and biological properties. In this study, we extracted a low molecular weight (MW) sericin (LMW-sericin; below 10 kDa) by a performing high-temperature and high-pressure method and confirmed the MW using matrix-assisted laser desorption ionization-time of flight and liquid chromatography-mass spectrometry. Furthermore, we determined its biological effects on macrophages and human adipose stem cells (hASCs) as cell models to investigate the biocompatibility, immunomodulation behavior, and potential signaling pathway-related wound healing via analyses of gene expression of focal adhesion and human cytokines and chemokines using quantitative real-time polymerase chain reaction and cytokine assay. LMW-sericin showed good biocompatibility both in macrophages and hASCs. Macrophages cultured with 0.1 mg/ml LMW-sericin displayed an improved inflammatory response shown by the upregulation of CXCL9, IL12A, BMP7, and IL10, which developed Th1 and Th2 balance. LMW-sericin also improved the differentiation of macrophages toward the M2 phenotype by significantly enhancing the expression of Arg-1, which is conducive to the repair of the inflammatory environment. Moreover, the gene expression of hASCs showed that LMW-sericin promoted the secretion of beneficial adhesion molecules that potentially activate the gene transcription of differentiation and migration in hASCs, as well as significantly enhanced the levels of PKCß1, RhoA, and RasGFR1 as fruitful molecules in wound healing. These findings provide insights into LMW-sericin application as a potential biomaterial for wound management.

There have been numerous recent advances in wound care management. Nevertheless, the assessment of hemostatic dressing is essential to enable surgeons and other physicians and healthcare professionals to make the correct decisions regarding the disposition of severe hemorrhage. Here, we investigated the relative efficacies of chitosan-based and conventional gauze dressings in a rat model of femoral artery hemorrhage and in patients with surgical wounds. Dressing effectiveness was evaluated based on hemostatic profiles, biocompatibility, antimicrobial activity, and blood factor responses in coagulation. Relative to standard gauze dressing, the chitosan fiber (CF) dressing treatment significantly shortened the time to hemostasis in injured rats. Moreover, the CF dressing significantly prolonged partial thromboplastin time, enhanced blood absorption, and reduced antithrombin production without altering the prothrombin ratio. Unlike regular gauze bandages, the CF dressing demonstrated remarkable antibacterial activity. The results of this study indicate the effectiveness of chitosan as a hemostatic dressing and elucidate its underlying mechanism. It is possible that chitosan surgical dressings could serve as first-line intervention in hospital emergency care for uncontrolled hemorrhage.

The existence of red, inflammatory, and chronic itchy condition in the skin is commonly speculated as the presence of Atopic Dermatitis (AD) in patients. The use of silk clothing as a non-pharmacological approach in the management of AD has been noticed as an effective alternative therapy; however, the evidence based on its usage is poorly served. Hence, we aim to evaluate the effectiveness of using pure silk clothing in the therapy of AD patients. The clinical trial was performed by recruiting 30 patients with AD for up to 8 weeks of observation. They were instructed to wear pure silk clothing for the whole day without any additional medication and were investigated using the AD-related questionnaires. The findings revealed a significant decrease of AD occurrence along with a great improvement of patient's quality of life at each time point. Our investigation demonstrated that this treatment promotes good skin appearance, comfort, and remarkable improvement in the quality of life. This promising preliminary outcome warrants a further study; hence, it can be a potential non-pharmacological treatment choice for controlling the severity of AD.

BACKGROUND AND PURPOSE: Limited information is available regarding the role Panton-Valentine leukocidin (PVL) plays in methicillin-susceptible Staphylococcus aureus (MSSA). In this study, we compared the frequency of the PVL gene between MSSA strains isolated from patients with MSSA infections and MSSA strains isolated from patients with evidence of MSSA nasal colonization. We also explored the role that the PVL toxin plays in the ability of MSSA to cause disease as well as the phylogenetic relationship between these S aureus strains. METHODS: The presence of MSSA strains was screened among children aged <18 years during routine health maintenance visits and among children aged <7 years at daycare centers or kindergartens during the 2003 to 2008 period. At the same time, clinical MSSA isolates were recovered from patients with various types of bacterial infections. Polymerase chain reaction was applied to detect the presence of the PVL and SEB genes in these strains. The strains were also subjected to pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST) studies. Clinical features were compared between patients with PVL-positive and those with PVL-negative isolates. RESULTS: A total of 495 colonizing MSSA and 71 clinical MSSA isolates were used. The prevalence of PVL-positive S aureus was significantly higher among clinical isolates than among colonizing isolates (14/71, 19.7%; 5/495, 1.0%; p < 0.05). In addition, we found that patients with PVL-positive MSSA infections had a significantly longer duration of fever and tended to have higher C-reactive protein levels than patients with PVL-negative MSSA infections. MLST typing of the 19 PVL-positive MSSA isolates revealed ST59, a strain that is similar to the MLST type of community-associated methicillin-resistant Staphylococcus aureus found in Taiwan. The PFGE typing of PVL-positive/ST59 MSSA isolates revealed multiple pulsotypes. CONCLUSION: The prevalence of the PVL gene was significantly higher among clinical strains of MSSA (19.7%) than among colonizing strains (1.0%). In addition, patients infected with PVL-positive MSSA strains had fever for a significantly longer duration and tended to have higher C-reactive protein levels than patients with PVL-negative MSSA infections. Our findings imply that PVL may play an important role in the pathogenesis of S aureus infection.
