Previous studies with broiler breeders indicate a P retention threshold when fed daily dietary levels of non-phytate P (NPP) exceeding 320 mg. Fibroblast growth factor 23 (FGF23) is a hormone secreted by osteocytes which modulates P retention and could be the biological agent which controls the P threshold in breeders. To evaluate the relationship between FGF23 and the P retention threshold, a 4-wk study with 32-wk-old breeders was conducted with 6 dietary treatments with daily NPP intake of 216 to 576 mg/d/h with increments of 80 mg/kg diet. The goals were 1) to elucidate how plasma FGF23 corresponds with the P retention threshold in broiler breeders and 2) to determine the amount of P for optimal egg production and bone health. Results showed that between daily 288 mg and 360 mg dietary NPP intake, P retention decreased from 33 to 26% but FGF23 levels increased from 130 pg/mL to 220 pg/mL with increasing NPP. The elevation of plasma FGF23 between the range of 288 mg to 360 mg dietary NPP/d intake suggests that FGF23 is related to the P retention threshold and may be the major hormone for regulating physiological P levels when intake of daily dietary P levels are increased above 288 mg NPP.

Our objective was to determine the within and between laboratory performance of an enzymatic spectrophotometric method for milk urea nitrogen (MUN) determination. This method first uses urease to hydrolyze urea into ammonia and carbon dioxide. Next, ammonia (as ammonium ions) reacts with 2-oxoglutarate, in the presence of reduced nicotinamide-adenine dinucleotide phosphate (NADPH) and the enzyme glutamate dehydrogenase (GlDH), to form l-glutamic acid, water, and NADP+. The change in light absorption at 340 nm due to the conversion of NADPH to NADP+ is stoichiometrically a function of the MUN content of a milk sample. The relative within (RSDr) and between (RSDR) laboratory method performance values for the MUN enzymatic spectrophotometric method were 0.57% and 0.85%, respectively, when testing individual farm milks. The spectrophotometric MUN method demonstrated better within and between laboratory performance than the International Dairy Federation differential pH MUN method with a much lower RSDr (0.57 vs. 1.40%) and RSDR (0.85 vs. 4.64%). The spectrophotometric MUN method also had similar method performance statistics as other AOAC International official validated chemical methods for primary milk component determinations, with the average of all RSDr and RSDR values being <1%. An official collaborative study of the enzymatic spectrophotometric MUN method is needed to achieve International Dairy Federation, AOAC International, and International Organization for Standardization official method status.

The food production system needs to be sustainable including poultry sector to feed the increasing global population. An accepted economical and environmental approach of broiler production is to produce larger broilers faster while using less feed. Broiler production is aimed at producing consumable meat and meat products. The global broiler meat market has evolved over the years with increasing selection pressure shifted toward attaining yield characteristics for increased cut-up parts such as breast and thighs. There is a shift toward a big bird market in the U.S. with approximately 70% of the broiler meat produced from large birds (>2.72 kg). Genetic selection of broilers for quantitative traits such as growth rate and lean muscle mass without increasing the fat mass has altered broiler physiological homeostasis to adapt toward the larger rates of muscle protein turnover. Physiological stresses created due to selection pressures in broilers have produced several muscle myopathies including an emerging one called woody breast myopathy. The sustainable broiler production practice may require humane consideration of raising broilers in less stressful grow-out regimes that will have minimal impact on broiler metabolic health. Another sustainability approach of broiler production toward feed efficiency lies on understanding dietary formulation approach of amino acids and energy that promote optimal nutrient utilization and minimal nutrient output to environment while also fulfilling the growth demands and body composition changes associated with increased protein gain in current meat broilers brought by the genetic progress.

The aim of this experiment was to evaluate the effect of undenatured type II collagen supplementation on inflammation and cartilage degeneration after exercise in healthy dogs. Forty healthy Labrador Retrievers (20 male/20 female; range 5-12 yr; average 8 yr) were sorted into two groups: undenatured type II collagen group receiving 40 mg UC-II (10 mg collagen type II/min. 3% undenatured type II collagen; Lonza Consumer Health, Inc.) and placebo group receiving 40 mg maltodextrin daily by capsule. After 2-wk loading, all dogs began an 11-wk endurance exercise regimen consisting of two weekly runs, starting at 5 km and increasing incrementally to 8 km, with one final 16 km run. Blood samples were collected at baseline, pre and post first 5 km run, and pre- and post-16 km run. Activity per kilometer was greater in male undenatured type II collagen vs. male placebo over all runs (P = 0.004), and average moving speed was greater in all undenatured type II collagen dogs compared with placebo over all runs (P < 0.001). Hematology analysis indicated that during the first insult, undenatured type II collagen dogs had a greater lymphocyte count (P < 0.001) and lymphocyte percentage (P = 0.001) vs. placebo dogs. Undenatured type II collagen dogs had a lesser neutrophil percentage (P = 0.042) and neutrophil to lymphocyte ratios (P = 0.001) compared to placebo dogs. For the final insult, undenatured type II collagen dogs had greater lymphocyte percentage (P = 0.013) and lesser mean corpuscular hemoglobin concentration (P = 0.043) compared with placebo dogs. Both groups had significant changes between timepoints for several hematological parameters. Biomarker IL-6 was lesser in undenatured type II collagen dogs compared with placebo at post 5 km (P = 0.037). Cartilage oligomeric matrix protein (COMP) was lesser in undenatured type II collagen dogs at post 16 km (P = 0.023), and only the placebo dogs had a significant increase in COMP from pre to post 16 km (P = 0.021). In summary, Labrador Retrievers supplemented with undenatured type II collagen had decreased inflammation and cartilage degeneration compared with nonsupplemented dogs during exercise.

Our first objective was to redesign a modified 14-sample milk calibration sample set to obtain a well-distributed range of milk urea nitrogen (MUN) concentrations while maintaining orthogonality with variation in fat, protein, and lactose concentration. Our second objective was to determine the within- and between-laboratory variation in the enzymatic spectrophotometric method on the modified milk calibration samples and degree of uncertainty in MUN reference values, and then use the modified milk calibration samples to evaluate and improve the performance of mid-infrared partial least squares (PLS) models for prediction of MUN concentration in milk. Changes in the modified milk calibration sample formulation and manufacturing procedure were made to achieve the desired range of MUN concentrations. A spectrophotometric enzymatic reference method was used to determine MUN reference values, and the modified milk calibration samples were used to calibrate 3 mid-infrared milk analyzers. The within- and between-laboratory variation in the reference values for MUN were 0.43 and 0.77%, respectively, and the average expanded analytical uncertainty for the mean MUN value of the 14-sample calibration set was (mean ± SD) 16.15 mg/100 g ± 0.09 of milk. After slope and intercept adjustment to achieve a mean difference of zero with the calibration samples, it could be seen that the standard deviation of the differences of predicted versus reference MUN values among 3 different instruments and their PLS models were quite different. The orthogonal sample set was used (1) to determine when a PLS model did not correctly model out the background variation in fat, true protein, or anhydrous lactose; (2) to calculate an intercorrection factor to eliminate that effect, and (3) to improve the model performance (i.e., 50% reduction in standard deviation of the difference between instrument predictions and reference chemistry values for MUN).

Three isotopic tracers ([2,3,3-2H3]-L-serine, [2H11]-L-betaine, and [1-13C]-L-methionine) were administered by amnion injection into 18-day-old chick embryos to investigate the kinetics of methionine metabolism. The embryos utilized were from eggs collected from 34-week-old Cobb 500 broiler breeders that were fed either a control diet containing folic acid (1.25 mg/kg diet) and pyridoxine HCl (5 mg/kg diet) or diets devoid of supplemental pyridoxine or folic acid. Intermediate metabolites of methionine metabolism and polyamines were analyzed in 18-day-old chick embryos. There were no differences in hepatic [2H2] methionine or [2H3] cysteine enrichments or in physiological concentrations of sulfur amino acids for chick embryos from breeders fed the control diet and embryos from breeders fed diets containing no pyridoxine or folic acid. Supplementation of B6 or folic acid did not affect the production of methionine and cysteine in chick embryos. However, breeders fed the control diet with both folic acid and pyridoxine supplementation produced embryos with a two-fold reduction of hepatic homocysteine and increased spermine compared with embryos from breeders fed diets containing no supplemental pyridoxine or folic acid (P < 0.05). Hepatic S-adenosylmethionine for embryos from breeders fed no supplemental B6 was half the concentration compared with embryos from breeders fed the control diet. Embryos from breeders fed the control diet were utilized to determine the proportion of homocysteine going through remethylation and transsulfuration and also to determine the pathway of remethylation. Sixty-five percent of the methyl groups used for homocysteine remethylation from control embryos was via the MFMT pathway. Alternatively, 61% of homocysteine from control embryos was remethylated via the MFMT and the BHMT reactions and 39% of homocysteine was catabolized to cysteine via the transsulfuration pathway. These data show that in embryos, intermediate metabolites of methionine and polyamines increase in concentration when pyridoxine levels are provided in deficient concentrations to the breeder hen. In addition, this research demonstrates that folic acid deficient embryos conserve methionine, rather than catabolize it to cysteine.

An initial study profiled non-cellulosic polysaccharide (NCP) levels in feed ingredient samples-corn, soybean meal (SBM), whole soybean, and distillers dried grains with solubles (DDGS). A separate NCP digestibility assay was performed in broilers at day 21 (grower phase) and day 42 (finisher phase) fed corn-soy based diets with and without composite enzymes (phytases, multi-carbohydrases, and proteases). Negative control (NC), NC + composite enzymes (NC+E), and positive control (PC) diets were tested. Negative control and NC + E diets were isocaloric, 3,020 kcal/kg ME at grower phase and 3,026 kcal/kg ME at finisher phase. Positive control diet was formulated to meet the Cobb standard nutrient specifications. Diets, pooled digesta, and excreta samples from all treatment diets were collected from 21- and 42-day-old birds and NCP content analyzed as soluble NCP (S-NCP) and insoluble NCP (I-NCP) fractions. Digestibility coefficient (DC) values were determined for all dietary treatments for both the feeding periods. Results from the ingredient analysis showed NCP levels of ∼7 to 10% in corn samples, ∼8 to 11% in SBM samples, ∼11 to 14% (including pectin level) in whole soybean, and ∼12 to 17% in DDGS samples, suggesting variation (P < 0.05) in NCP levels existed within ingredient samples. Digestibility assays showed that enzyme treated (NC + E) diet improved DC values at day 21 from 6 to 10 units and 6 to 9 units for ileal and total tract collection, respectively, for I-NCP fraction (P < 0.05) compared to DC values for NC or PC diets. The ileal DC values at day 42 were not different between treatment diets (∼0.6) but total tract DC values improved 9 to 11 units for broilers fed NC + E diet compared to NC or PC diets. Higher DC values for S-NCP were observed for all treatments for both feeding periods (∼0.7-ileal and ∼0.8-total tract) compared to DC values for I-NCP. Overall, the use of supplemental enzymes in corn-soy-based diets showed improvement in total NCP digestion.

Digestibility coefficients of nutrients, metabolizable energy (ME), net energy (NE) and the ratio of NE to ME (NE/ME) of 20 diets were measured in broiler chickens (1 to 21 d). Dietary nutrients were formulated to keep similar ME/nutrient ratios, except for dietary protein, fat, and fiber using corn, soybean meal, animal protein blend, barley, poultry oil and an enzyme mixture of xylanase, glucanase, and phytase. Digestibility coefficients of nutrients and ME were measured in battery cages under free-access of feed, while NE was measured in floor pens feeding 75% of recommended ME intake each day. NE for maintenance was calculated on basis of mean metabolic weight using a coefficient from a previous study and NE for gain was calculated by body protein and fat gains using dual-energy x-ray absorptiometry. Digestibility coefficients of protein and neutral detergent fiber (NDF) were curvilinearly related to dietary protein and NDF, respectively, while digestibility coefficients of fat and starch were linearly correlated to dietary fat and starch, respectively. The inclusion of enzymes increased the digestion coefficient of NDF to predict the digestibility of protein, NDF, fat, and starch. MEn/gross energy ratio averaged 72.5% and was correlated to protein, fat, NDF, and starch. ME values were accurately predicted from chemical characteristics, where best equations were obtained from digestible nutrients. Energetic efficiencies of ME were 72% (NE/MEn) and 68% (NE/ME) and varied by about 20 and 18%, respectively. Ratios of energetic efficiency were 68% for digestible carbohydrates; 86% for digestible fat; and 76% (NE/MEn) and 59% (NE/ME) for digestible protein. According to the lowest residual standard deviation the best nutrient components to predict energy were digestible nutrients for predicting ME values (41 kcal/kg); digestible protein intake, fecal organic matter, and body fat and protein for predicting heat increment values (111 kcal/kg); and combination of ME and crude nutrient for predicting NE values (140 kcal/kg).

A study was conducted to determine the effect of different feeding programs and energy intakes on the body composition of broiler breeders (BB) from 22 wk of age to 65 wk of age. Cobb 500 BB that had been reared using three growth curves: (1) the control group followed Cobb 500 guidelines (SBW) for target body weights (BW), (2) the second group target BW was 20% heavier (HBW) than the SBW group, and (3) the third group target BW was 20% lighter (LBW) than the SBW group. At 21 wk of age, pullets from each growth curve were assigned to be fed one of six treatments. Diets were formulated and allocated to provide 330, 360, 390, 420, 450, or 480 kcal ME/hen/d and 24 g protein/hen/d, at peak intake. Body composition (lean mass, fat mass, and mineral content) was measured by dual-energy x-ray absorptiometry throughout the production period. Significantly, greater (P < 0.05) amounts of fat mass and lower amounts of lean mass were evident at the beginning of the production period for HBW hens compared with SBW and LBW hens. Higher levels of energy intake also affected body composition. Namely, fat deposition was greatest throughout the production period in hens consuming 480 kcal/d at peak intake (P < 0.05). Critically, fluctuations to lean and fat mass throughout the production period are suggestive of underlying regulatory processes. Specifically, hens appears to maintain consistent lean mass throughout the production period with a coefficient of variation of <12% across all birds. The results of this study provide important BB body composition information from management and nutrition studies that showed the dynamics of body composition change during the production period.

OBJECTIVES: Postmenopausal (PM) women taking therapies using estrogens plus progestogens (EPTs) can experience side effects (breast pain, vaginal spotting/bleeding). Sensitivity of the European Quality of Life Five Dimension Five Level (EQ-5D-5L) in measuring quality of life of PM women experiencing side effects of EPTs is unknown. A crosswalk between the Menopause-Specific Quality of Life (MENQOL) questionnaire and the EQ-5D-5L was assessed. METHODS: The measures were administered to 352 PM women (side effects = 202; control = 75; untreated = 75) in a non-interventional study. MENQOL total scores, treated as continuous and categorical predictors, were mapped onto EQ-5D-5L utilities using regression. Ordinary least-squares regression using averaged scores over time, goodness of fit, and estimated coefficients was also assessed. RESULTS: Mean age was 53.7 years. The first model (MENQOL as a continuous variable) showed a moderate correlation (-0.589) and statistically significant relationship with the EQ-5D-5L (p < 0.001), with an equation of EQ-5D-5L = 0.992 - 0.042 × MENQOL. The EQ-5D-5L mean scores were comparable (side effects = 0.854; control = 0.927; untreated = 0.836) to MENQOL mean scores estimated in the first model (side effects = 0.865 [standard deviation 0.07]; control = 0.909; untreated = 0.833). Linearity assumptions were supported with MENQOL scores as a categorical predictor. Goodness of fit was moderate (R2 = 0.347; root mean squared error = 0.093). CONCLUSION: The crosswalk supports conversion of MENQOL scores to EQ-5D-5L-derived health utilities for group-level analyses in PM women.

OBJECTIVES: Postmenopausal women (PMW) can experience side-effects (breast pain/tenderness and vaginal spotting/bleeding) associated with estrogen plus progestin therapies (EPTs). To assess these outcomes, the Breast Pain and Tenderness Daily Diary (BPT-DD) and the Vaginal Bleeding and Spotting Daily Diary (VBS-DD) were developed for electronic completion (eDiaries). This study evaluated the psychometric properties of the eDiaries. METHODS: The eDiaries were completed daily for 28 days by 202 PMW experiencing breast pain/tenderness and/or vaginal spotting/bleeding while on EPTs. Confirmatory factor analysis (CFA) investigated the BPT-DD structure. Response distributions, test-retest reliability (intraclass correlation coefficient [ICC]), internal consistency (BPT-DD only), and construct validity (via known groups and convergent validity analyses) were assessed. RESULTS: Completion rates were high: over 90% of women missed <3 daily entries. CFA supported the BPT-DD unidimensional structure (Bentler's Comparative Fit Index >0.98). BPT-DD inter-item correlations (r = 0.77-0.89) and internal consistency (Cronbach's alpha = 0.95-0.97) were high and good test-retest reliability was demonstrated (ICC ≥ 0.70). The eDiaries correlated moderately (>0.40), in a logical pattern with other instruments, supporting convergent validity. Known-groups analyses indicated both measures demonstrated significant differences between patients of differing severity (p < 0.001). CONCLUSION: The study provides evidence of strong psychometric properties for the BPT-DD and VBS-DD to assess breast pain/tenderness and spotting/bleeding in PMW.

An experiment was conducted using non-colostomized and colostomized broiler breeder hens to determine the effects of feeding limestone of 2 different mean particle sizes (185 microns and 3490 microns) on P excretion, total P and Ca retention, and urinary P and Ca excretion during a 6-week feeding study. Additionally, changes in plasma inorganic P (iP) and ionic Ca (Ca++) and urinary excretion of P and Ca were determined in one egg laying cycle of 24 hours. One-hundred-fifty non-colostomized and 6 colostomized broiler breeder hens, 30 wk of age, were divided into 2 groups and fed broiler breeder diets supplemented with either small particle or large particle limestone. Two % acid insoluble ash (Celite) was added to the feed as a marker. Diets, excreta, and urine samples were analyzed for total P and Ca by ionic coupling plasma (ICP) analysis. The non-colostomized breeders fed large particle limestone compared to small limestone particles produced a significant increase in percent tibia ash (P < 0.0001) and egg specific gravity (P = 0.0382), but P excretion approached a tendency of being reduced (P = 0.1585). The urinary total P and Ca (∼18 and 9%, respectively) of total P and Ca excretion for breeders fed both sizes of limestone was not significantly different in the colostomized breeders. In plasma, both iP and Ca++ reached a peak during 18 to 20 h and 20 to 24 h post oviposition for smaller and larger particle sized limestone fed groups, respectively. The maximal excretion of urinary P was found during 11 to 20 h post oviposition, whereas urinary Ca peaked during 0 to 11 h post oviposition for both smaller and larger particle sized limestone supplemented groups. In summary, the findings indicate that the particle size (smaller and larger) of calcium source did not significantly influence the quantitative total urinary excretion of Ca and P but did influence the timing of Ca and P excretion.

Woody breast (WB) is a myopathy of the Pectoralis major muscle in broilers that is characterized by increased hardness of the raw fillet. This defect increases economic losses due to associated decreased meat quality and the addition of training skilled laborers to identify problematic fillets. Therefore, the objectives of this study were to determine if compression force (CF) could be used as a non-invasive method of detecting WB and to assess changes in meat attributes during cold storage. Deboned fillets (n = 121) were collected from 8-wk-old broilers, and categorized as normal (NORM, n = 42), mild (MILD, n = 39), and moderate/severe (SEV, n = 40) for WB. Right fillet weight, drip loss, and CF of the cranial region were measured on the day of processing (day 0) and 1, 2, 3, 4, and 8 d of cold storage (4°C). Left fillets were sampled for sarcomere length (SL) and gravimetric fragmentation index (GFI) analysis following 1, 3, and 8 d of cold storage. Initial breast fillet weight, CF, cumulative drip loss, and SL increased as severity of WB category increased (P < 0.05). Compression force was highly correlated to WB severity (rs = 0.79). Compression force, SL, and GFI declined with storage time (P < 0.05), whereas cumulative drip loss increased (P<0.05). Cumulative drip loss, SL, and GFI had interactions between storage length and WB category (P < 0.05), where SEV generally changed more than NORM over time. The results of this study indicate that increased severity of WB fillets is associated with higher CF compared to normal fillets. Furthermore, there is a softening effect of the meat over time and data suggest that moisture (drip loss) and myofibril proteolysis (fragmentation) contribute to these changes. Using instrumental CF to assess fillet hardness can be used as a reliable tool to identify WB. However, adaptability in the commercial processing require additional studies prior to the incorporation into the processing plant for online grading purposes.

An isotope dose technique was utilized (i) to determine endogenous amino acid (AA) and protein losses and (ii) to propose adjusted values for AA requirements. The endogenous flow rate was calculated from the pool of enrichment in plasma AA, assuming similitude to enrichment of endogenous AA. In experiment 1, chicks were orally administered D4-lysine at 2% of estimated lysine intake from 16 to 24 days to find the isotopic steady state of the atom percent excess (APE) of lysine for plasma and jejunal and ileal digesta. The APE of D4-lysine in plasma, jejunal digesta and ileal digesta reached the isotopic steady state at 5.5, 3.4 and 2.0 days, respectively, by using the broken-line model. It was assumed that the isotopic steady state at 5 days identified for D4-lysine is also representative for the 15N-labeled AA. In experiment 2, chicks were fed diets from 1 to 21 days with increasing levels of fat (6%, 8%, 12%, 13% extract ether), protein (26%, 28.5%, 31% CP) or fiber (14%, 16%, 18% NDF) by adding poultry fat, soybean meal, blended animal protein or barley. Chicks were orally administered 15N-threonine, 15N-cysteine, 15N-methionine, 15N-lysine and 15N-leucine at 2% of estimated daily intake for 5 days from 17 to 21 days of age. Dietary nutrients influenced endogenous losses (EL), where dietary fat stimulated EL of lysine (P=0.06), leucine and protein (P=0.07); dietary protein enhanced EL of leucine and protein; and finally the dietary fiber increased EL of leucine. Dietary nutrients also affected apparent ileal digestibility (AID). Dietary fat increased AID of cysteine but decreased AID of lysine. Dietary protein reduced AID of protein, threonine, lysine and leucine, and similarly dietary fiber decreased AID of protein, threonine, methionine, lysine and leucine. In contrast, dietary fat or protein did not affect real ileal digestibility (RID) of protein and AA except threonine and leucine. The dietary fiber reduced the RID of protein, threonine and leucine. This indicate that variations of some endogenous AA and protein losses due to dietary nutrients almost eliminates the effects of RID, and thus the EL coming from the body should be utilized to adjust the AA requirement instead of changing the true digestible nutrients of ingredients. The present data suggest that 5 days' feeding labeled AA was enough to reach the isotopic steady state and AA requirements should be adjusted when additional dietary protein, fat or fiber is fed.

The primary goal was to investigate the effects of l-carnitine on fuel efficiency, as an antioxidant, and for muscle recovery in Labrador retrievers. Dogs were split into two groups, with one group being supplemented with 250 mg/d of Carniking™ l-carnitine powder. Two experiments (Expt 1 and Expt 2) were performed over a 2-year period which included running programmes, activity monitoring, body composition scans and evaluation of recovery using biomarkers. Each experiment differed slightly in dog number and design: fifty-six v. forty dogs; one endurance and two sprint runs per week v. two endurance runs; and differing blood collection time points. All dogs were fed a low-carnitine diet in which a fixed amount was offered based on maintaining the minimum starting weight. Results from Expt 1 found that the carnitine dogs produced approximately 4000 more activity points per km compared with the control group during sprint (P = 0·052) and endurance runs (P = 0·0001). Male carnitine dogs produced half the creatine phosphokinase (CPK) following exercise compared with male control dogs (P = 0·05). Carnitine dogs had lower myoglobin at 6·69 ng/ml following intensive exercise compared with controls at 24·02 ng/ml (P = 0·0295). Total antioxidant capacity (TAC) and thiobarbituric acid reactive substance (TBARS) results were not considered significant. In Expt 2, body composition scans indicated that the carnitine group gained more total tissue mass while controls lost tissue mass (P = 0·0006) and also gained lean mass while the control group lost lean mass (P < 0·0001). Carnitine dogs had lower CPK secretion at 23·06 v. control at 28·37 mU/ml 24 h after post-run (P = 0·003). Myoglobin levels were lower in carnitine v. control dogs both 1 h post-run (P = 0·0157; 23·83 v. 37·91 ng/ml) and 24 h post-run (P = 0·0189; 6·25 v.13·5 ng/ml). TAC indicated more antioxidant activity in carnitine dogs at 0·16 mm v. control at 0·13 mm (P = 0·0496). TBARS were also significantly lower in carnitine dogs both pre-run (P = 0·0013; 15·36 v. 23·42 µm) and 1 h post-run (P = 0·056; 16·45 v. 20·65 µm). Supplementing l-carnitine in the form of Carniking™ had positive benefits in Labrador retrievers for activity intensity, body composition, muscle recovery and oxidative capacity.

White striping (WS) and woody breast (WB) are 2 poultry meat quality defects that affect the acceptance of raw breast fillets as well as properties of cooked and further processed products. The present study was intended to evaluate the incidence of these conditions in broilers at different ages and to compare the properties of fillets with different degrees of WS and WB. For this study, 1,920 birds were processed, at 6 and 9 wk of age, in a standard commercial inline processing system. After chilling, carcasses were deboned and butterfly fillets were collected and weighed. Individual fillets were scored for normal, moderate, severe, and very severe degrees of WS and WB, and for petechial hemorrhagic lesions (PHEM, 0 - no lesion to 2 - severe lesion). Representative fillets with NORM-WS/WB, SEV-WS, SEV-WB, and SEV-WS/WB were selected and stored at 4°C. After 24 h, fillet length, width, cranial height, and caudal height, as well as pH, color, and drip loss were recorded. There was an increase in incidence of severe and very severe WS and WB conditions at 9 wk compared to 6 wk of age. The relationship between fillet weight and the myopathies plateaued at 9 wk of age with more fillets showing a higher score. Mean PHEM scores were higher (P < 0.05) in SEV-WS, SEV-WB, and SEV-WS/WB compared to NORM-WS/WB birds, both at 6 and 9 weeks. NORM-WS/WB birds had lower (P < 0.05) live and breast weight, breast yield, and cranial and caudal heights, as well as b* value (yellowness) compared to SEV-WS/WB. NORM-WS/WB had lower (P < 0.05) pH while NORM-WS/WB and SEV-WS samples showed lower (P < 0.05) drip loss when compared to SEV-WB and SEV-WS/WB. The results from this study showed that the severe degrees of WS and WB are associated with heavier and older birds, and thicker breast fillets. Occurrence of severe degrees of WS and/or WB can affect various raw meat quality factors, mainly color and water holding capacity.

The contribution of dietary, mobilized, or newly synthesized fatty acids in yolk formation at different periods of egg production was determined. In an initial experiment, a single dose of 13C-linoleic acid was administered to pullets at the onset of egg production and their presence in follicles determined over the subsequent 10 days. In a second experiment, pullets were fed a daily 15 mg dose of U-13C-glucose beginning 2 wk prior to sexual maturity through the end of the experimental period. A 50 mg meal of U-13C-linoleic acid was orally administered approximately 10 d prior to sexual maturity (defined as first egg) representing body linoleic acid. Upon each hen's first egg, each bird received a 25 mg meal of 2D31-linoleic acid representing dietary linoleic acid. All eggs were collected for the next 10 days. The incorporation of labeled linoleic acid and palmitic acid in egg yolk was then determined using GC-MS. This process was repeated at peak production and at 45 wk of age. At sexual maturity, the deposition of labeled palmitic acid in the yolk was higher compared with its deposition at peak production and 45 wk of age. The deposition of both 13C- and 2D31-linoleic acid increased with hen age. These results suggest that dietary and tissue linoleic acid is utilized to a greater extent in older hens and that lipogenesis (synthesis of palmitic acid) plays a larger role at sexual maturity in the young hen.

BACKGROUND: Nivolumab has shown significant survival benefit and a favorable safety profile compared with dacarbazine chemotherapy among treatment-naïve patients with metastatic melanoma in the CheckMate 066 phase III study. Results from the health-related quality of life (HRQoL) analyses from CheckMate 066 are presented. PATIENTS AND METHODS: HRQoL was evaluated at baseline and every 6 weeks while on treatment using the European Organisation for Research and Treatment of Care (EORTC) Core Quality of Life Questionnaire (QLQ-C30) and the EuroQoL Five Dimensions Questionnaire (EQ-5D). Via a multi-step statistical plan, data were analyzed descriptively, cross-sectionally, and longitudinally, adjusting for baseline covariates, in patients having baseline plus ≥1 post-baseline assessment. RESULTS: Baseline-adjusted completion rates for all HRQoL questionnaires across treatment arms were 65% and 70% for dacarbazine and nivolumab, respectively, and remained similar throughout treatment. The mean baseline HRQoL scores were similar for patients treated with nivolumab and dacarbazine. Baseline HRQoL levels with nivolumab were maintained over time. This exploratory analysis showed a between-arm difference in favor of nivolumab on the EQ-5D utility index and clinically meaningful EQ-5D improvements from baseline at several time points for patients receiving nivolumab. Patients treated with nivolumab did not show increased symptom burden as assessed by the EORTC QLQ-C30. No HRQoL change was noted with dacarbazine patients up to week 43, although the high attrition rate after week 13 did not allow any meaningful analyses. Patients receiving nivolumab deteriorated significantly later than those receiving dacarbazine on several EORTC QLQ-C30 scales and the EQ-5D utility index. CONCLUSIONS: In addition to prolonged survival, these exploratory HRQoL results show that nivolumab maintains baseline HRQoL levels to provide long-term quality of survival benefit, compared with dacarbazine in patients with advanced melanoma.

The global poultry industry has been faced with emerging broiler breast meat quality issues including conditions known as white striping (WS, white striations parallel to muscle fibers) and woody breast (WB, hardness of raw fillet). Experiments were conducted to evaluate effects of WS and WB hardness on meat quality traits in broiler breast fillets. In Exp. 1, birds were processed at approximately 9 wk of age and deboned at 4 h postmortem (PM); in Exp. 2, birds were processed at approximately 6 and 9 wk of age and deboned at 2 h PM. Fillets were categorized as: normal for both white striping and woody breast (NORM); moderate for white striping and mild for woody breast (MILD); severe for white striping and mild for woody breast (WS); severe for woody breast and moderate for white striping (WB); or severe for both white striping and woody breast (BOTH). Sarcomere length, gravimetric fragmentation index, marination uptake, cook loss, and Meullenet-Owens razor shear energy (MORSE) values on non-marinated and marinated fillets were assessed. Sarcomeres tended to be longer (P = 0.07) with increasing severity of WS and WB in both experiments and gravimetric fragmentation index did not differ (P > 0.05) among categories. Marinade uptake decreased (P < 0.05) with increasing severity of WS and WB. Cook losses of non-marinated and marinated fillets were greatest (P < 0.05) in the BOTH category. Even though MORSE values did not differ (P > 0.05) in non-marinated fillets, the marinated BOTH fillets had greater MORSE values (P < 0.05) than other categories of fillets in Exp. 1. Non-marinated NORM fillets had greater (P < 0.05) MORSE values than the other categories at 6 wk age; however, MORSE values did not differ (P > 0.05) among categories of marinated breasts. At 9 wk, WS and BOTH were higher (P < 0.05) in MORSE compared to NORM for non-marinated fillets, but similar to NORM marinated fillets. Results suggest that severe degrees of white striping and woody breast, individually or in combination, negatively impact meat quality, especially water holding capacity attributes such as marinade uptake and cook loss.
