RESUMEN

Hyperketonemia (HYK) in early lactation can have a different impact on health and productivity depending on the timing of HYK onset. While specific metabolites measured during the dry period may serve as biomarkers of HYK, the correlations between metabolites represent a challenge for the use of metabolic profiles dataset, and little has been explored on HYK. This exploratory cohort study aimed a) to characterize the correlations among metabolites measured during the late dry period in dairy cows, and b) to identify biomarkers in the late dry period associated with the onset of HYK at the first (wk1) and second (wk2) week of lactation. Individual blood samples from 440 Holstein dairy cows were collected at 21 ± 3 days before expected parturition. From each sample, 36 different metabolites were measured in serum and plasma. Hyperketonemia was diagnosed in wk1 and wk2 of lactation based on the blood concentration of beta-hydroxybutyrate (BHB > 1.2 mmol/L). Principal component analysis (PCA) was performed to reduce metabolites to a smaller number of uncorrelated components. Multivariable logistic regression models were applied to assess the associations between principal components (PC) and HYK at wk1 only (HYK+ wk1), wk2 only (HYK+ wk2), or both weeks (HYK+ wk1-2). The incidence of HYK was 16.2% in the first week, 13.0% in the second week, and 21.2% within the first two weeks of lactation. The results of PCA highlighted 10 PCs from which two were associated with HYK+ wk1 as compared with cows without HYK during the first two weeks of lactation (non-HYK); the PC a2 led by bilirubin and non-esterified fatty acids (OR = 1.29; 95%CI: 1.02-1.68), and the PC a5 led by alkaline phosphatase (ALP) and gamma-glutamyl transferase (GGT) (OR = 2.77; 95%CI: 1.61-4.97). There was no evidence of an association between any PC and HYK+ wk2 (vs. non-HYK cows). Cows with elevated PC a5 (led by ALP and GGT) in the dry period were 3.18 times more likely to be HYK+ wk1 than HYK+ wk2 (OR: 3.18, 95%CI: 1.34-8.73; P = 0.013). Overall, the main hypothesis generated by our exploratory study suggests that cows with biomarkers of liver dysfunction (ALP, GGT, bilirubin) assessed by PCA at 3 weeks before calving are more likely to develop HYK during the first week of lactation compared to the second week. In addition, results suggest that cows with HYK in both of the first two weeks of lactation had an overall metabolic disbalance during the onset of the late dry period, which based on PCs, encompass biomarkers related to glucogenic and ketogenic metabolic pathways as well as liver dysfunction and fatty liver. Further research is needed to determine the underlying mechanisms associated with the different adaptations between cows that develop HYK during the first and second week of lactation.

Monitoring the body condition score (BCS) of dairy cows is a management strategy that can assist dairy producers in decision-making. The BCS and its variations reflect the level of body fat reserves and fat mobilization throughout the different stages of lactation. Cows that mobilize excessive amounts of fat reserves in response to the increased energy requirements of the transition period are more likely to have higher beta-hydroxybutyrate (BHB) concentration in blood, leading to a higher incidence of hyperketonemia postpartum. In this study, our main objective was to evaluate how both BCS (at 21 d prior to the expected calving date, -21 BCS) and change in BCS during the late dry period (-21 d to calving, ∆BCS) are associated with temporal patterns of blood BHB concentrations during the first two weeks of lactation. Our secondary objective was to characterize the relationship between the change in BCS in the late dry period, and milk yield and milk composition in the first milk test postpartum. In this retrospective cohort study, we assessed BCS at 21 (±3) days before the expected calving date and within three days after calving. Blood BHB concentration was measured at days 3 (±1), 7 (±1), and 14 (±1) postpartum. Hyperketonemia (HYK) was defined as blood BHB ≥ 1.2 mmol/L. To evaluate how -21 BCS and ∆BCS during the late dry period were associated with BHB in early lactation, linear mixed-effects regression models with an unstructured covariate matrix were performed. The association between ∆BCS and incidence of postpartum HYK were determined using a multivariable log-binomial model. A linear regression model was used to evaluate the association between ∆BCS and milk yield and milk composition in the first monthly test-day. Covariates used for model adjustment include parity, season, and baseline BCS. We observed that cows with BCS ≥ 4.0 at 21 d before their expected calving date had the highest BHB concentration postpartum, but no evidence that BCS ≥ 4.0 at 21 d was associated with fluctuations of BHB over time. Cows that experienced a large BCS loss (larger than 0.5 units) during the late dry period had a 61% (95% CI: 1.04, 2.50) higher risk of developing HYK in early lactation and had higher BHB concentrations during early lactation compared with cows with no ∆BCS prepartum. These associations were observed independently of the BCS at -21 d prepartum (baseline). In addition, cows that lost more than 0.5 BCS unit in the late dry period produced 3.3 kg less milk (95% CI: -7.06, 0.45) at the first milk test compared to cows that had no ∆BCS during the late dry period. No evidence of an association between -21 BCS and ∆BCS in the late dry period and milk composition was observed in our study. These results suggest that dynamic measures of BCS during the late dry period, such as ∆BCS, are better at evaluating blood BHB patterns in early lactation than BCS measured at a single time point. Cows with larger BCS loss during the late dry period and with greater parity are more likely to have higher concentrations of blood BHB postpartum, with the highest concentrations reported at 7 d post-calving.

RESUMEN

The need for consortial programs to provide advanced education in food animal veterinary production medicine has been recognized and lauded for nearly three decades. This article describes one effort to create a dairy production medicine curriculum funded by a United States Department of Agriculture (USDA) Higher Education Challenge Grant. This National Center of Excellence in Dairy Production Medicine Education for Veterinarians is housed at the Dairy Education Center of the University of Minnesota and the project was a collaboration of the University of Minnesota, the University of Illinois, the University of Georgia, and Kansas State University. The article reviews the need for innovative ways to educate students who will optimally serve the dairy industry, provides a broad overview of the process of developing and delivering the eight-week dairy production medicine curriculum, and describes the challenges faced and lessons learned as a result of offering such a program.

RESUMEN

A narrative integrative review on the evidence for hoof trimming found 16 articles on efficacy, frequency, and associations with behavior and physiologic parameters. Review of these studies revealed (1) hoof trimming is associated with behavior and physiologic changes; (2) increasing the frequency of hoof trimming seems to decrease the incidence of hoof lesions; (3) there is limited research to support any particular technique; and (4) descriptions of the hoof trimming techniques used is inadequate in most articles. To increase scientific support for hoof trimming practices, current knowledge gaps in technique, timing, and frequency of hoof trimming need to be addressed.

Therapeutic hoof blocks have been recommended for treatment of sole ulcers in dairy cattle; however, they are underutilized in the industry. Twenty Holstein cows were randomly assigned to receive a wooden hoof block applied to the left hind leg (n = 5), to the right hind leg (n = 5), or no hoof blocks (n = 10). Accelerometers were affixed to both hind legs of 10 blocked cows and 5 control cows. Cows were observed for locomotion scores, and milk production was recorded. Mixed models were constructed to determine effects of block application on behavior, milk production, and locomotion. Activity was altered by day, with cows being most active on Day - 1. The mean number of daily lying bouts and lying bout duration did not differ by treatment. Locomotion scores were higher for blocked cows on Days 1, 2, and 3 compared with baseline; however, milk production did not differ between treatments. In conclusion, although block application affected the appearance of locomotion, it did not appear to adversely affect the behavior or milk production of sound dairy cows.

Blood samples were collected from 3449 cows on 57 representative Ontario dairy herds during the summer of 1998 and analysed for antibody to Neospora caninum using an ELISA. Forty-eight herds (2742 cattle) contained at least one N. caninum-seropositive animal. Two composite milk samples were collected from all cattle: the first on the day of blood collection and the second 68 to 365 days later. All milk samples were submitted for bacteriological culture. Ontario Dairy Herd Improvement Corporation (DHI) data were available for 3162 cattle in the 57 herds at the time of bleeding. Furthermore, complete DHI data were available for 1658 cattle that were culled between 12 and 24 months following blood collection. Using a standardised ELISA sample-to-positive (S/P) cut-off of > or = 0.45, the corrected seroprevalence was 8.2% overall and 10.1% within seropositive herds. At blood collection the odds of N. caninum-seropositive cows having a high linear score (> or = 4.0; equivalent to a somatic cell count > or = 200,000 cells/ml) was 27% less than for seronegative animals. Similarly, at the time of culling, the odds of having a high linear score was 22% less in N. caninum-seropositive cattle. Overall, linear score was lower in N. caninum-seropositive cattle at culling. After controlling for herd, parity, days in milk, and the interval between collection of milk samples, the odds of N. caninum-seropositive cattle testing positive for an environmental pathogen (i.e. environmental Streptococcus species and coliforms) on the second milk sample was 56% less than for seronegative animals. The odds were 83% less at a higher ELISA S/P cut-off of > or = 0.70. Finally, the odds of N. caninum-seropositive cattle developing a new infection with a major pathogen (environmental or contagious) were 60% less than seronegative cows using the higher ELISA S/P cut-off.

OBJECTIVE: To determine whether Neospora caninum serostatus was associated with milk production among Holstein cattle in Ontario. DESIGN: Case-control study and cross-sectional observational study. ANIMALS: 3,702 Holstein cows in 83 herds (case-control study) and 3,162 Holstein cows in 57 herds. PROCEDURE: Herds in the case-control study were grouped on the basis of N. caninum abortion status. Herds in the observational study were considered representative of Ontario dairy herds. The N. caninum serostatus of individual cows was determined with a kinetic ELISA. Milk production was modeled to compare seropositive with seronegative animals while controlling for parity, days since parturition, and herd clustering. RESULTS: In the case-control study, 305-day milk production of seropositive cows was significantly less than milk production of seronegative cows in herds with abortions attributable to N. caninum infection and in herds with abortions attributable to pathogens other than N. caninum, but not in herds without abortion problems. In the observational study, 305-day milk production for seropositive cows was not significantly different from milk production of seronegative cows. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the association between N. caninum serostatus and milk production in Ontario Holstein dairy cattle may depend on abortion status of the herd. In herds with abortion problems, regardless of cause, N. caninum-seropositive cattle produced less milk, whereas in herds without abortion problems, N. caninum-seropositive cattle produced the same amount of milk as seronegative cattle.

OBJECTIVE: To determine whether time until culling or risk of culling was associated with Neospora caninum serostatus among Holstein cattle in dairy herds in Ontario. DESIGN: Retrospective cohort study. ANIMALS: 3,412 cows in 56 herds. PROCEDURE: Blood samples were collected, and serum was tested for antibodies against N. caninum. Information on cows that were culled was collected during the 1- to 2-year period that producers were unaware of serostatus of individual cows in their herds. RESULTS: Herd prevalence of N. caninum-seropositive cows ranged from 0 to 68.3% (median, 7.0%). During the time of the study, 184 of 359 (51.3%) N. caninum-seropositive cows were culled, compared with 1,388 of 3,053 (45.5%) seronegative cows. Mean time from blood sample collection to culling for seronegative cows (289 days; 95% confidence interval, 280 to 299 days) was not significantly different from mean time for seropositive cows (296 days; 95% confidence interval, 269 to 323 days). Survival analysis indicated that N. caninum serostatus was not associated with time to culling or risk of culling. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that N. caninum serostatus of Holstein cows in Ontario was not significantly associated with either time to culling or risk of culling. Thus, N. caninum serostatus alone should not be used to determine whether cows should be culled.
