Growing conditions and seasonal fluctuations are critical factors affecting fruit and vegetable nutritional quality. The effects of two partially overlapping cropping seasons, early (ECS; January-May) and full (FCS; March-July), on the main carpometric traits and bioactive components of different watermelon fruits were investigated in the open field. Four watermelon genotypes, comprising of three commercial cultivars 'Crimson Sweet', 'Dumara', 'Giza', and the novel hybrid 'P503 F1', were compared. The carpometric traits varied significantly between genotypes. Soluble solids and yield were higher under FCS than ECS. The variation affecting colour indexes between the two growing seasons exhibited a genotype-dependent trend. The antioxidant components and radical scavenging activity of watermelon fruits were also significantly affected by differences in received solar energy and temperature fluctuations during the trial period. The average citrulline, total phenolics and flavonoid contents were 93%, 71% and 40% higher in FCS than in ECS. A genotype-dependent variation trend was also observed for lycopene and total vitamin C between cropping seasons. The hydrophilic and lipophilic radical scavenging activities of the pulp of ripe watermelon fruits of the different genotypes investigated varied between 243.16 and 425.31 µmol Trolox Equivalent (TE) of 100 g-1 of fresh weight (fw) and from 232.71 to 341.67 µmol TE of 100 g-1 fw in FCS and ECS, respectively. Our results, although preliminary, show that the functional quality of watermelon fruits is drastically altered depending on the environmental conditions that characterize the ECS and LCS.

The aim of the present work was to evaluate the influence of genetic impact on the content of some characteristic secondary metabolites in tomato fruits. The study was conducted to screen 14 different tomato genotypes for antioxidant capacity and quality attributes (polyphenols, tocopherols, carotenoids, vitamin C) to select the genetic lines with high biological value, multiple disease resistance, and high yield capacity for further usage in crop improvement programs. Significant differences were found among the different hybrids and cultivation seasons for the phytochemical content of the fruits. The varieties developed by breeding to increase their resistance were found to differ significantly. During a 3-year cultivation experiment, the level of lycopene in freshly harvested fruits ranged between 10.38 and 63.23 mg.kg-1 fwt for outdoor-cultivated Naik114 and Naik352, respectively. As for ß-carotene, its content varied between 0.49 mg.kg-1 for Unorosso and 11.66 mg.kg-1 for Cherrola F1. The highest level of vitamin C (702.59 mg.kg-1) was recorded for Cherrola F1, while the lowest level (1.36.86 mg.kg-1) was determined in fruits of Unorosso. Neither polyphenol nor vitamin C showed positive correlation with antioxidant activity. In the three cultivation seasons, the highest concentration of polyphenols (579.19-804.12 mg.kg-1 fwt) was determined in fruits of outdoor-cultivated Cherolla F1 variety. The α- and γ-tocopherol content varied between 2.56 and 18.99 mg.kg-1, and 0.28 and 9.35 mg.kg-1, respectively, in fresh tomato fruit. Among the newly bred resistant varieties, the ZKI114 variety was proved to have outstanding features concerning the amounts of the bioactive components.

Tomato-based products are significant components of vegetable consumption. The processing tomato industry is unquestionably in need of a rapid definition method for measuring soluble solids content (SSC) and lycopene content. The objective was to find the best chemometric method for the estimation of SSC and lycopene content from visible and near-infrared (Vis-NIR) absorbance and reflectance data so that they could be determined without the use of chemicals in the process. A total of 326 Vis-NIR absorbance and reflectance spectra and reference measurements were available to calibrate and validate prediction models. The obtained spectra can be manipulated using different preprocessing methods and multivariate data analysis techniques to develop prediction models for these two main quality attributes of tomato fruits. Eight different method combinations were compared in homogenized and intact fruit samples. For SSC prediction, the results showed that the best root mean squared error of cross-validation (RMSECV) originated from raw absorbance (0.58) data and with multiplicative scatter correction (MSC) (0.59) of intact fruit in Vis-NIR, and first derivatives of reflectance (R 2 = 0.41) for homogenate in the short-wave infrared (SWIR) region. The best predictive ability for lycopene content of homogenate in the SWIR range (R 2 = 0.47; RMSECV = 17.95 mg kg-1) was slightly lower than that of Vis-NIR (R 2 = 0.68; 15.07 mg kg-1). This study reports the suitability of two Vis-NIR spectrometers, absorbance/reflectance spectra, preprocessing methods, and partial least square (PLS) regression to predict SSC and lycopene content of intact tomato fruit and its homogenate.

From the metagenome of a carbamazepine amended selective enrichment culture the genome of a new to science bacterial species affiliating with the genus Nocardioides was reconstructed. From the same enrichment an aerobic actinobacterium, strain CBZ_1T, sharing 99.4% whole-genome sequence similarity with the reconstructed Nocardioides sp. bin genome was isolated. On the basis of 16S rRNA gene sequence similarity the novel isolate affiliated to the genus Nocardioides, with the closest relatives Nocardioides kongjuensis DSM19082T (98.4%), Nocardioides daeguensis JCM17460T (98.4%) and Nocardioides nitrophenolicus DSM15529T (98.2%). Using a polyphasic approach it was confirmed that the isolate CBZ_1T represents a new phyletic lineage within the genus Nocardioides. According to metagenomic, metatranscriptomic studies and metabolic analyses strain CZB_1T was abundant in both carbamazepine and ibuprofen enrichments, and harbors biodegradative genes involved in the biodegradation of pharmaceutical compounds. Biodegradation studies supported that the new species was capable of ibuprofen biodegradation. After 7 weeks of incubation, in mineral salts solution supplemented with glucose (3 g l-1) as co-substrate, 70% of ibuprofen was eliminated by strain CBZ_1T at an initial conc. of 1.5 mg l-1. The phylogenetic, phenotypic and chemotaxonomic data supported the classification of strain CBZ_1T to the genus Nocardioides, for which the name Nocardioides carbamazepini sp. nov. (CBZ_1T = NCAIM B.0.2663 = LMG 32395) is proposed. To the best of our knowledge, this is the first study that reports simultaneous genome reconstruction of a new to science bacterial species using metagenome binning and at the same time the isolation of the same novel bacterial species.

The results showed that soil electrical conductivity, (EC2: 7 dS/m) increased soluble solids, lycopene content, total phenolic content, hydrophilic and lipophilic radical scavenging activities (HRSA and LRSA) by 14.2, 149, 20, 46.4, and 19.0%, respectively, compared with control. Under 0.5% spent engine oil (SEO), flavonoid content decreased by 21.7% compared with the control. HRSA and LRSA of fruits subjected to EC2/SEO1 treatment were, respectively, 45.9 and 35.5% lower than control. The a*/b* ratio was positively and significantly (P < 0.01) correlated with ß-carotene (R = 0.78), lycopene (R = 0.68), total vitamin C (R = 0.71), α-tocopherol (R = 0.83), γ-tocopherol (R = 0.66), HRSA (R = 0.93), LRSA (R = 0.80), and soluble solids (R = 0.84) suggesting that it may be a promising indicator of fruit quality in areas affected by such constraints. The research revealed that combined stresses induce responses markedly different from those of individual treatments, which strain the need to focus on how the interaction between stresses may affect the functional quality of tomato fruits.

An efficient and rapid liquid chromatographic method was developed for the separation of carotenoids and their geometrical isomers in tomato products using a core C30 column of 2.6 µm particles with gradient elution of tert-butyl-methyl-ether in 2% water in methanol. Excellent separation of the major carotenoids such as lycopene, ß-carotene and lutein as well as their geometrical isomers and oxygen-containing derivatives with resolution factors ranging between 0.78 and 4.0 and selectivity of 1.01-1.63 was achieved. Validation of the developed method met the acceptance criteria concerning linearity, recovery, precision and limit of detection and quantification. Calibrations were linear with correlation coefficient (R2) values between 0.9966 and 0.9999. The limit of detection and quantification values were found to be 0.008 and 0.017 and 0.029 and 0.056 µg/mL, respectively. Recovery of 94.3-99.9%, intraday precision of 1.81-4.45% and interday precision of 3.13-6.86% were obtained. The hyphenation of liquid chromatography with diode-array and mass spectrometry was helpful in the identification of the separated carotenoids particularly the unusual di-hydroxy cyclolycopene adduct and di-methoxy lycopene determined for the first time in tomato products. Commercially available kinds of tomato juice and ketchup were evaluated based on their carotenoid content.

Eclipta prostrata (L.) is an important and well-known medicinal plant due to its valuable bioactive compounds. Microorganisms, including arbuscular mycorrhizal fungi (AMF), and salinity could directly impact plant metabolome, thus influencing their secondary metabolites and the efficacy of herbal medicine. In this study, the role of different single AMF species (Funneliformis mosseae, Septoglomus deserticola, Acaulospora lacunosa) and a mixture of six AMF species in plant growth and physio-biochemical characteristics of E. prostrata under non-saline conditions was investigated. Next, the most suitable AM treatment was chosen to examine the impact of AMF on physio-biochemical features and polyphenol profiles of E. prostrata under saline conditions (100 and 200 mM NaCl). The findings indicated that AMF mixture application resulted in more effective promotion on the aboveground part of non-saline plants than single AMF species. AM mixture application improved growth and salt tolerance of E. prostrata through increasing the activity of catalase, peroxidase (at 4 weeks), proline, and total phenolic content (at 8 weeks). Such benefits were not observed under high salinity, except for a higher total phenolic concentration in mycorrhizal plants at 8 weeks. Through high-performance liquid chromatography, 14 individual phenolic compounds were analyzed, with wedelolactone and/or 4,5-dicaffeoylquinic acid abundant in all treatments. Salinity and mycorrhizal inoculation sharply altered the polyphenol profiles of E. prostrata. Moderate salinity boosted phenolic compound production in non-AM plants at 4 weeks, while at 8 weeks, the decline in the content of phenolic compounds occurred in uncolonized plants subjected to both saline conditions. Mycorrhization augmented polyphenol concentration and yield under non-saline and saline conditions, depending on the growth stages and salt stress severity. Plant age influenced polyphenol profiles with usually a higher content of phenolic compounds in older plants and changed the production of individual polyphenols of both non-AM and AM plants under non-stress and salt stress conditions. A better understanding of factors (involving mycorrhiza and salinity) affecting the phenolic compounds of E. prostrata facilitates the optimization of individual polyphenol production in this medicinal plant.

Grafting by vegetables is a practice with many benefits, but also with some unknown influences on the chemical composition of the fruits. Our goal was to assess the effects of grafting and storage on the extracted juice of four orange-fleshed Cantaloupe type (Celestial, Donatello, Centro, Jannet) melons and two green-fleshed Galia types (Aikido, London), using sensory profile analysis and analytical instruments: An electronic tongue (E-tongue) and near-infrared spectroscopy (NIRS). Both instruments are known for rapid qualitative and quantitative food analysis. Linear discriminant analysis (LDA) was used to classify melons according to their varieties and storage conditions. Partial least square regression (PLSR) was used to predict sensory and standard analytical parameters. Celestial variety had the highest intensity for sensory attributes in Cantaloupe variety. Both green and orange-fleshed melons were discriminated and predicted in LDA with high accuracies (100%) using the E-tongue and NIRS. Galia and Cantaloupe inter-varietal classification with the E-tongue was 89.9% and 82.33%, respectively. NIRS inter-varietal classification was 100% with Celestial variety being the most discriminated as with the sensory results. Both instruments, classified different storage conditions of melons (grafted and self-rooted) with high accuracies. PLSR showed high accuracy for some standard analytical parameters, where significant differences were found comparing different varieties in ANOVA.

The present paper reports about a comparative survey on the chemical composition, antioxidant activity and in vitro antimicrobial activity of selected propolis samples collected in Hungary. The total levels of polyphenolic compounds including flavonoids in ethanolic extracts of propolis were assessed. The major constituents of ethanolic extracts of propolis were analysed by gas chromatography/mass spectrometry analysis. Total phenolic content was determined spectrophotometrically using a Folin-Ciocalteu reagent. Free radical scavenging activities were evaluated by means of 2,2-diphenyl-1-picrylhydrazyl assay. In vitro inhibitory activity was investigated against eight different bacterial strains by agar well diffusion assay. An extensive comparison was carried out regarding general parameters and specific polyphenolic components. The experimental data led to the observation that there is considerable variability in terms of the quality and the biological value of the distinctive propolis samples. These findings confirm the hypothesis of the study; versatile experimental results are required for proper, well-reasoned, balanced and standardised industrial applications. The major flavonoid components were found to be chrysin and pinocembrin; however, versatile minor components were also detected. The total polyphenol content of ethanolic extracts of propolis ranged between 104.6 mg/g and 286.9 mg/g (gallic acid equivalent). The radical scavenging activity of ethanolic extracts of propolis varied between 101.7 mg/g and 286.9 mg/g (ascorbic acid equivalent). As the quality of propolis depends on the season, vegetation and the area of collection, marked differences were found among the different products examined in terms of both composition and general characteristics. The studied samples exhibited significant differences in term of antimicrobial activities.

A Gram-stain-negative, obligately aerobic, non-motile, non-sporulating, rod-shaped bacterium, designated TZCO2T, was isolated from the soil of an irrigated coffee plantation in Arusha, Tanzania, East Africa. Phylogenetic analysis, based on 16S rRNA gene sequences, indicated that the isolate is affiliated with the genus Taibaiella in the family Chitinophagaceae. Its closest relative is Taibaiella koreensis THG-DT86T (96.7%). The pH and temperature ranges for growth were pH 6.0-8.5 (optimum 7.0-7.5) and 10-35 °C (optimum 30 °C, respectively. The predominant fatty acids were iso-C15:0 (32.4%), iso-C15:1 G (22.6%), iso-C17:0 (15.1%) and iso-C17:0 3-OH (10.0%) The only isoprenoid quinone detected in strain TZCO2T was menaquinone-7 (MK-7); the major polar lipids were phosphoaminolipid, phosphatidylethanolamine, unidentified aminolipids and lipids. The DNA G+C content was 51.9 mol%. Physiological and chemotaxonomic data further confirmed that strain TZCO2T is distinct from other members of the genus Taibaiella. Thus, strain TZCO2T is considered to represent a novel species of the genus, for which the name Taibaiella coffeisoli sp. nov. is proposed. The type strain is TZCO2T (=NCAIM B 02601T=CCM 8601T).

Six hybrids were subjected to chromatographic analyses by HPLC for the determination of phytochemicals such as capsaicinoid, polyphenol, and vitamin C. The dynamics of ripening of 4 of the hybrids were also characterised. Seven capsaicinoids could be separated and determined; the major compounds were nordihydrocapsaicin, capsaicin, and dihydrocapsaicin, while homocapsaicin and homodihydrocapsaicin derivatives were detected as minor constituents. Capsaicin content ranged between 95.5 ± 4.15 and 1610.2 ± 91.46 µg/g FW, and the highest value was found in Bandai (C. frutescens) at the green ripening stage. The major capsaicinoids had a decreasing tendency in Bandai and Chili 3735 hybrids, while no change was observed in Beibeihong and Lolo during ripening. Nine polyphenol compounds were detected including 8 flavonoids and a nonflavonoid compound in the pods of all hybrids. The major components were naringenin-diglucoside, catechin, and vanillic acid-derivative and luteolin-glucoside. Naringenin-diglucoside ranged from 93.5 ± 4.26 to 368.8 ± 30.77 µg/g FW. Except vanillic acid-derivative, dominant polyphenols increased or remained unchanged during ripening. As for vitamin C, its content tended to increase with the advance in ripening in all hybrids included in this study. The highest value of 3689.4 ± 39.50 µg/g FW was recorded in Fire Flame hybrid.

A simple and efficient high-performance liquid chromatographic method was developed and validated for the separation and determination of capsaicin and its major dihydro- and homoderivatives in spice paprika products in 20 min with fluorescent and 35 min with mass-spectrometric detection. The separation was performed on reversed-phase chromatographic adsorbent of cross-linked endcapping with eluent consisting of 1:1 acetonitrile-water or acetonitrile-0.1% formic acid under isocratic conditions. Excellent separation of all the major and minor capsaicinoids with resolution index between 1.08 and 7.34 was achieved. The detection and quantification limits of capsaicinoids in standard material solutions were between 2 and 10 ng/mL. The lowest detectable amount of capsaicin, with fluorescent detection, was found to be <1 µg/g non-pungent spice paprika powder. The naturally occurring capsaicinoids could be distinguished from the non-capsaicinoids compounds appeared on liquid chromatography-fluorescence profile of extract from drastically processed paprika by applying mass spectroscopic detection. Hungarian spice paprika were evaluated as mild to very hot (capsaicinoid content: 334-1,660 µg/g) and chili products as very or extremely hot products (1,543-2,818 µg/g).

This study was conducted to analyze carotenoids from tomatoes by high-performance liquid chromatography using reversed-phase C18 silica having cross-linked end-capping with diode array and mass spectrometric detection. An efficient gradient elution system was developed to achieve good and reliable separation of both major and minor carotenoids as well as their isomers. Resolution of lycopene, ß-carotene and their isomers was 0.91-3.97 and 1.02-2.86 with cross-linked and conventional C18 column, respectively. The % recovery for zeaxanthin, lycopene and ß-carotene was found to be in the range of 89-97%. Limits of detection and quantification of 19.44 and 64.79 ng/mL for zeaxanthin, 15.6 and 52.4 ng/mL for lycopene and 8.28 and 27.61 ng/mL for ß-carotene were determined. More carotenoid compounds could be separated and detected with the new method as compared with conventional C18 column. Hyphenation of HPLC with photodiode array and mass spectrometry detectors assisted in detection of tetra-dehydrocarotenoid and fatty acid diesters of xanthophylls in tomato products. Content of all-trans-lycopene, ß-carotene and total carotenoid in different industrial tomatoes tested was found to range between 41.87 and 84.65, 0.89 and 1.50 and 53.22 and 112.60 µg/g fresh weight, respectively.

To simultaneously detect Sudan dyes and carotenoids in red pepper and tomato products, high-performance liquid chromatography (HPLC) methods with photodiode-array detection are developed and validated. The methods include the use of end-capped and nonend-capped adsorbents with a gradient elution system starting with water containing methanol. Water content of 9% in the starting mobile phase is found to be necessary to ensure sufficient separation of Sudan dyes and to avoid overlapping or interference with the carotenoids of considerable content. The data of the validation reveal the accuracy and precision of the developed methods. A limit of Sudan dyes detection of 1-5 microg/g in red pepper or tomato sauce could be approached. The methods provide excellent separation of the carotenoids from the unsaponified extracts of red pepper and the tomato products.
