RESUMEN

The objective of this randomized controlled trial was to evaluate the efficacy of supplementing bovine colostrum replacer during weaning to reduce intestinal permeability and improve gain. For this experiment, 65 calves were enrolled and housed individually until 70 d of age. Calves were fed milk replacer (150 g/L) 3 times daily with 9 L/d, 10.5 L/d, 11.25 L/d, and 12 L/d offered from d 1-7, d 8-14, d 15-21, d 22-56, respectively. Calves were weaned over 8 d from d 57-64, receiving a total of 7.8 L in 2 meals/d from d 57-60 and 3.8 L/d in one feeding from d 61-64. At d 57, calves were blocked by birth weight and randomly assigned to one of 2 treatments, equal in metabolizable energy, which were fed once daily during weaning from d 57-64: control (CON; n = 31 calves): 3.8 L milk replacer (150 g/L) fed by nipple bottle, or colostrum supplementation (COL; n = 34): a mixture of 1 L bovine colostrum replacer (125 g/L) and 3 L milk replacer (150 g/L) with 3.8 L of the mixture fed by nipple bottle. Serum IgG was measured within 48 h of birth and body weight was taken at d 0, 57, 60, 64, 70, 77, and 84. Starter intake and bovine respiratory disease (BRD) score were measured daily from d 50-70 and fecal consistency was examined daily from d 56-70. Serum ß-hydroxybutyrate (BHB) and lung consolidation were evaluated at d 57, 64, and 70 and intestinal permeability was assessed by recovery of chromium-EDTA, lactulose, and d-mannitol from plasma after oral administration at d 56 and 65. There was no difference in body weight between treatment groups at the start of weaning, but COL were 2.79 kg (95% CI: 0.90-4.68) and 2.76 kg (95% CI: 0.86-4.65) heavier than CON at d 77 and 84, respectively. Additionally, COL tended to gain 100.00 g/d more than CON calves (95% CI: -10.41-207.13) from d 57-84. There were no differences in any of the other variables measured. Supplementation of bovine colostrum replacer during weaning may improve weight gain, but the mechanism of action is not clear.

Establishing accurate illness and treatment rates in dairy calves is crucial, yet calf health records are often incomplete. Thus, the objective of this study was to investigate barriers for dairy farmers for recording calf illnesses and treatments on dairy farms in Ontario, Canada. An online survey was completed by a convenience sample of 88 Ontario dairy farms in 2022, with 34 questions regarding farm demographics, current practices surrounding record keeping and analysis, and factors that would improve recording compliance. Multivariable models were built to assess associations between explanatory variables and the following outcomes: likelihood of making management or treatment protocol changes based on records analysis, factors that would increase the use of electronic recording methods, and whether all calf illnesses and treatments are recorded. Pearson's chi-squared tests were also used to investigate associations between explanatory variables and whether the respondent agreed or disagreed with a proposed reason for why a calf illness or treatment would not be recorded on their farm. Producers had 3.45 times greater odds of recording all antimicrobial treatments if they used a computer software system compared with those that did not. With respect to anti-inflammatory treatments, producers had 3.11 times greater odds of recording these treatments if records were located in the calf barn than elsewhere. Nonfamily employees had 6.08 times greater odds of recording all supportive therapy treatments than farm owners. When calf health records were kept in the calf barn, respondents were less likely to report that illnesses were not recorded due to time constraints (5% vs. 36% if records were elsewhere) or because calf health records were not analyzed (10% vs. 34% if records were elsewhere). On farms that recorded calf treatments in a paper booklet, respondents were more likely to report that treatments were not recorded because calf health records were not analyzed (44% for paper records vs. 21% for other systems). The most commonly indicated factors that would increase recording of illness were recording with a mobile app (27% of respondents) and for the recording system to be easy to use (31% of respondents). Overall, these data indicate that recording may be improved by keeping calf health records in close proximity to the calves and using a recording method that allows for data analysis. An easy-to-use mobile app may also improve recording if it could be used in the calf barn, provide data analytics, and allow for time-efficient data entry.

The supplementation of dairy cows with tannins can reduce the ruminal degradation of dietary protein and urine N excretion, but high concentration in the diet can impair ruminal function, diet digestibility, feed intake, and milk yield. This study evaluated the effect of low concentrations (0, 0.14, 0.29, or 0.43% of diet in DM basis) of a tannin extract from the bark of Acacia mearnsii (TA) on milking performance, dry matter intake (DMI), digestibility, chewing behavior, ruminal fermentation, and N partition of dairy cows. Twenty Holstein cows (34.7 ± 4.8 kg/d, 590 ± 89 kg, and 78 ± 33 d in lactation) were individually fed a sequence of 4 treatments in 5, 4 × 4 Latin squares (with 21-d treatment periods, each with a 14-d adaptation period). The TA replaced citrus pulp in the total mixed ration and other feed ingredients were kept constant. Diets had 17.1% crude protein, mostly from soybean meal and alfalfa haylage. The TA had no detected effect on DMI (22.1 kg/d), milk yield (33.5 kg/d), and milk components. The proportions in milk fat of mixed origin fatty acids (16C and 17C) and the daily secretion of unsaturated fatty acids were linearly reduced and the proportion of de novo fatty acids was increased by TA. Cows fed TA had linear increase in the molar proportion of butyrate and linear reduction in propionate in ruminal fluid, whereas acetate did not differ. There was a tendency for the ratio of acetate to propionate to be linearly increased by TA. Cows fed TA had a linear reduction in the relative ruminal microbial yield, estimated by the concentrations of allantoin and creatinine in urine and body weight. The total-tract apparent digestibility of neutral detergent fiber, starch, and crude protein also did not differ. The TA induced a linear increase in meal size and duration of the first daily meal and reduced meal frequency. Rumination behavior did not differ with treatment. Cows fed 0.43% TA selected against feed particles >19 mm in the morning. There were tendencies for linear decreases in milk urea N (16.1-17.3 mg/dL), urine N (153-168 g/d and 25.5-28.7% of N intake), and plasma urea N at 6, 18, and 21 h postmorning feeding, and plasma urea N 12 h postfeeding was reduced by TA. The proportion of N intake in milk (27.1%) and feces (21.4%) did not differ with treatment. Reductions in urine N excretion and milk and plasma urea N suggest that TA reduced ruminal AA deamination, whereas lactation performance did not differ. Overall, TA up to 0.43% of DM did not affect DMI and lactation performance, while there was a tendency to reduce urine N excretion.

Chemically defined diets are commonly used in amino acid (AA) requirement studies to allow for tight control of AA delivery. However, those diets are not representative of commercial diets in the market and are unpalatable. Methionine (Met) is usually the first limiting AA in cat diets, but little is known about its requirement for adult cats. Thus, the objectives of this study were: 1) to develop a semisynthetic diet limiting in Met and evaluate its effect on acceptance and feeding behavior in cats; and 2) to evaluate the effect of different sources and inclusions of Met on preference in cats fed the semisynthetic diet. A semisynthetic diet deficient in Met and total sulfur AA (TSAA) was developed. Healthy adult male cats (n = 9) were fed (0800 and 1600 h) the semisynthetic diet top dressed with DL-Met solution (T-DLM), to meet 120% of the TSAA requirement, for 8 d. Feed intake was measured and a 30-min video recording was taken at the 0800 h feeding to evaluate feeding behavior of the cats. Following the acceptability trial, two bowl tests were performed where first choice was recorded and intake ratio was calculated as consumed food (A/A + B). Three combinations were tested: semisynthetic diet deficient in Met (T-BASAL) vs. T-DLM; T-BASAL vs. diet sufficient in Met provided 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA); and T-DLM vs. T-MHA. Average feed intake remained high throughout the acceptability period (94.5% intake of total offered), but some cats decreased intake, resulting in a decrease in BW (≤2.5% of initial BW) over time (P < 0.05). Behaviors were similar among days (P > 0.05) with the exception of grooming the chest and body (P < 0.05). No preferences were observed towards a specific treatment (Met source and level) during the two-bowl tests (P > 0.05) and agreed with the cats expressing similar feeding behaviors during the preference tests (P > 0.05). In conclusion, a semisynthetic diet deficient in Met was successfully developed and can be used in studies to evaluate the effects of low protein and AA supplemented diets. Cats seem to show no preference for Met source and/or inclusion level in a semisynthetic diet application, which is of benefit for future studies aiming to determine the Met requirement in this species.


Previous studies that determined the requirement of amino acids (AA) in cats utilized experimental diets that do not represent commercial cat diets available in the market. Using this type of diets can present a challenge when applying AA requirements to commercial diet production. Thus, the goals of this study were to: 1) develop a semisynthetic diet deficient in methionine (Met) for adult cats with the inclusion of intact ingredients and to evaluate the effect of diet on behavior and acceptance; and 2) investigate the preference of a semisynthetic diet with different levels (deficient and sufficient) and sources [DL-Met and 2-hydroxy-4-(methylthio)-butanoic acid] of Met. The semisynthetic diet was well accepted by most cats. No major differences were observed in feeding behavior and preference towards Met source and level of inclusion. However, improvement in texture is recommended to increase acceptance and prevent removal of cats in feeding studies up to three weeks.

Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry provides accurate species-level identification of many, microorganisms retrieved from bovine milk samples. However, not all those microorganisms are pathogenic. Our study aimed to: (1) determine the species-specific prevalence of microorganisms identified in bovine milk of apparently healthy lactating quarters vs. quarters with clinical mastitis (CM); and (2) map current information and knowledge gaps on udder health relevance of microorganisms retrieved from bovine milk samples. A mixed study design (meta-analysis and mapping review) was chosen. We gathered several large Canadian, US and Brazilian data sets of MALDI-TOF results for organisms cultured from quarter milk samples. For meta-analysis, two datasets (apparently healthy quarters vs. CM samples) were organized. A series of meta-analyses was conducted to determine microorganisms' prevalence. Then, each species reported was searched through PubMed to investigate whether inflammation (increased somatic cell count (SCC) or signs of CM) was associated with microorganism's recovery from milk. A total of 294 different species of microorganisms recovered from milk samples were identified. Among 50,429 quarter-milk samples from apparently healthy quarters, the 5 most frequent species were Staphylococcus chromogenes (6.7%, 95% CI 4.5-9.2%), Aerococcus viridans (1.6%, 95% CI 0.4-3.5%), Staphylococcus aureus (1.5%, 95% CI 0.5-2.8%), Staphylococcus haemolyticus (0.9%, 95% CI 0.4-1.5%), and Staphylococcus epidermidis (0.7%, 95% CI 0.2-1.6%). Among the 43,924 quarter-milk CM samples, the 5 most frequent species were Escherichia coli (11%, 95% CI 8.1-14.3%), Streptococcus uberis (8.5%, 95% CI 5.3-12.2%), Streptococcus dysgalactiae (7.8%, 95% CI 4.9-11.5%), Staphylococcus aureus (7.8%, 95% CI 4.4-11.9%), and Klebsiella pneumoniae (5.6%, 95% CI 3.4-8.2%). When conducting the PubMed literature search, there were 206 species identified by MALDI-TOF for which we were not able to find any information regarding their association with CM or SCC. Some of them, however, were frequently isolated in our multi-country dataset from the milk of quarters with CM (e.g., Citrobacter koseri, Enterococcus saccharolyticus, Streptococcus gallolyticus). Our study provides guidance to veterinarians for interpretation of milk bacteriology results obtained using MALDI-TOF and identifies knowledge gaps for future research.

This study evaluated the effect on dairy cows of the partial replacement of whole plant corn silage (WPCS) with corn ear fibrous coproduct (CEFC) in diets with concentrate coproducts from citrus and corn on dry matter intake (DMI), lactation performance, digestibility, and chewing behavior. Holstein dairy cows (n = 20) in 5, 4 × 4 Latin squares (21-d periods) were fed a combination of strategies for feeding fibrous coproducts in a 2 × 2 factorial arrangement of the following treatments: (1) forage feeds: the partial replacement of WPCS (CS) with CEFC (CO), and (2) concentrate feeds: the partial replacement of wet corn gluten feed (GF) with a blend of pelleted citrus and corn distillers dried grains (CD) to have isonitrogenous diets. The concentrations of physically effective neutral detergent fiber (NDF; peNDF>8) were (% of dry matter): 21.8% for CS, 19.2% for CO, 20.7% for GF, and 20.2% for CD. Cows fed diet CS-CD had the highest yield of energy-corrected milk (30.0 kg/d) relative to the other diets (28.4 kg/d). Milk fat concentration was reduced on CO relative to CS. Cows fed the CO diets had higher DMI (21.2 vs. 20.2 kg/d) and digestible organic matter intake and tended to have a lower ratio of energy-corrected milk to DMI than cows fed CS. Diets CO reduced the daily intake of peNDF>8 and the intake as percent of body weight of peNDF>8, forage NDF, and total NDF relative to CS. Cows fed CO had greater meal frequency and lower daily meal time, meal duration, meal size, and duration of the largest meal than cows fed CS. The CO diet reduced rumination and total chewing in minutes per day and minutes per kilogram of DMI. When expressed per unit of peNDF>8 intake, rumination and total chewing were not affected by forage source. The total-tract starch digestibility coefficient was lower for cows fed CO than CS, but the intake of digestible starch was higher on CO than CS. Cows fed GF had reduced milk yield (29.6 vs. 30.8 kg/d), tended to have reduced DMI (20.4 vs. 21.0 kg/d), and had reduced digestible organic matter intake than cows fed CD. Feed efficiency was not affected by source of concentrate. The type of concentrate did not affect the intake of forage NDF and peNDF>8, but cows fed GF had higher intake of total NDF as percent of body weight than cows fed CD. The GF increased meal frequency and reduced meal size and largest meal duration and size. Cows fed GF had higher rumination and total chewing than cows fed CD (min/d, min/kg of DMI, and min/kg peNDF>8). Starch digestibility was higher and the intake of digestible starch tended to be higher on cows fed GF than CD. Plasma urea-N was higher, milk urea-N tended to be higher, and N utilization efficiency tended to be lower on cows fed GF than CD. Ruminal microbial yield was not affected by any treatment. All strategies evaluated were nutritionally viable and CEFC was a feasible partial replacement for WPCS.

This review synthesizes research findings on the pain and welfare of dairy calves undergoing disbudding procedures. We describe disbudding practices in North America as well as the use and perceptions of pain control for these procedures. Governing bodies across Canada and the United States, including each country's veterinary medical association and nationwide initiatives such as proAction and Farmers Assuring Responsible Management (FARM), recommend or require the use of a local anesthetic, a nonsteroidal anti-inflammatory drug (NSAID), and a sedative for disbudding procedures. Although the use of pain relief for disbudding has increased over the past decade or so, some in the dairy industry still do not believe that pain control for disbudding is necessary. As a painful procedure, disbudding has numerous welfare impacts on the calf both during and following the procedure that can be categorized under all 3 principles of animal welfare: natural living, biological functioning, and affective state. The use of pain control for disbudding; namely, a local anesthetic and NSAID, can improve welfare outcomes such as procedure-induced pain behavior, cortisol concentrations, mechanical nociceptive threshold, emotional states, and so on, compared with no pain control for the procedure. Although extensive research exists on pain control practices for disbudding, this review identified further gaps in knowledge and areas for future research. Mechanical nociceptive threshold can be evaluated around the disbudding wounds and is a reliable test in older calves; however, this outcome in very young calves after caustic paste disbudding has been reported to be inconclusive compared with that in older calves. As well, research evaluating xylazine sedation for disbudding has reported both potentially positive and negative results that are difficult to interpret or base suggestions on for the use of this drug. Finally, wounds caused by disbudding take a long time to heal (up to 13 wk) and have increased sensitivity for the entire healing process. Therefore, future research should aim to (1) determine accurate behavioral tests for calves under 1 wk of age undergoing disbudding to better understand their experience, (2) further attempt to understand the effects of xylazine sedation for disbudding and potential impacts of providing this medication, and (3) determine more ways to reduce the healing time and pain experienced by the calf after disbudding procedures.

The objective of this study was to determine the effect of a biologically normal plane of nutrition compared with a limited plane on the primary outcome wound healing, and one dose of nonsteroidal anti-inflammatory drug (NSAID) compared with 2 on the secondary outcomes: lying behavior, haptoglobin concentrations, and mechanical nociceptive threshold (MNT) in calves disbudded via cautery iron. Eighty female Holstein calves were enrolled at birth, individually housed, and fed via a Calf Rail system (Förster Technik). A 2 × 2 factorial design was used to assess the effect of plane of nutrition and an additional NSAID. Calves were randomly assigned to a biologically normal plane of nutrition (BN; offered up to 15 L/d) or a limited plane (LP; offered up to 6 L/d) and to receive one or 2 doses of meloxicam. All calves received a lidocaine cornual nerve block and a subcutaneous injection of meloxicam 15 min before cautery disbudding at 18 to 25 d of age, and half the calves received an additional injection of meloxicam (0.5 mg/kg) 3 d after disbudding. Tissue type present, wound diameter, and wound depth were evaluated 2 times per week for 7 to 8 wk as measures of wound healing, lying behavior was recorded beginning 1 to 2 wk before disbudding until 7 to 8 wk after as a behavioral indicator of pain, haptoglobin concentrations were measured once per day for 7 d after disbudding, and MNT was evaluated 2 times/wk for 3 wk. Survival analyses were analyzed using Cox regression models (wound healing) and continuous data were analyzed using mixed-effect linear regression models. Only 12% of horn buds were completely healed by 7 to 8 wk after disbudding and 54% had re-epithelized at this time. At any time, wounds from BN calves were more likely to have had re-epithelization occur compared with wounds from LP calves (hazard ratio: 1.93, 95% CI: 1.18-3.14). Wounds from calves that received only one dose of NSAID were more likely to have re-epithelization occur, compared with wounds from calves given 2 doses (hazard ratio: 1.87, 95% CI: 1.15-3.05). Wounds from BN calves had smaller diameters and depths over time beginning on wk 3 compared with LP calves. Wounds from calves that received an additional NSAID had larger diameters and depths over time beginning on wk 4 and 3 respectively, compared with calves that only received one dose of NSAID. Calves that received an extra NSAID tended to be less sensitive 7, 10, and 17 d after disbudding compared with calves that only received one dose and spent less time lying in the week after disbudding. Calves on the BN milk program were more active compared with LP calves with lower lying times, fewer lying bouts per day, and longer average lying bouts. Our results indicate that a BN milk feeding program for calves can result in faster healing times and more activity, and that providing an extra NSAID 3 d after disbudding appears to slow the healing process but may result in less pain experienced by the calf 1 to 2 wk after the procedure. This study is also among the first to demonstrate that after the complete removal of the horn bud, wounds can take more than 8 weeks to re-epithelize and fully heal.

Male dairy calf welfare is a key issue in the Canadian cattle industry. The welfare of male dairy calves can be explored through the aspects of health and biological functioning, affective states, and natural living. Presently, the main welfare issues associated with the production of male dairy calves include morbidity and mortality, colostrum and feeding management, transportation, isolation, castration and disbudding, and euthanasia. Opportunities to improve male dairy calf welfare include improving accepted industry practices, enhancing education and compliance with industry codes of practice, and increasing veterinarian involvement in on-farm animal welfare. The benefits of improving male dairy calf welfare include maintenance of the cattle industry's social license and improved producer mental health and occupational satisfaction. The main barriers to improving male dairy calf welfare are economics and cultural attitudes within the industry towards male dairy calves.

While the raw pet food market continues to grow, the risk of bacterial contamination in these types of diets is a major concern, with Salmonella enterica and Listeria monocytogenes being the most frequently associated pathogens in raw pet food product recalls. dl-Methionine is included in some commercial feline kibble and canned diets to improve protein quality; however, an alternative to this is a liquid methionine supplement, 2-hydroxy-4-(methylthio)-butanoic acid (HMTBa), which is also an organic acid. 2-Hydroxy-4-(methylthio)-butanoic acid has previously demonstrated similar efficacy to formic acid against pathogens in a liquid environment and may be a good candidate to inhibit S. enterica and L. monocytogenes in raw ground meat. First, the minimum inhibitory concentration and minimum bactericidal concentration of HMTBa against these pathogens under laboratory growth conditions were determined by measuring growth of pathogens over 36 h when exposed to 10 concentrations of HMTBa (0.10% to 1.00%) mixed with tryptic soy broth. 2-Hydroxy-4-(methylthio)-butanoic acid included at ≥0.50% was bactericidal to S. enterica and L. monocytogenes (P < 0.05). Next, five levels of HMTBa (0.50% to 1.25%) were included in raw ground meat mixtures inoculated with cocktails of S. enterica or L. monocytogenes, and contamination levels were determined at four timepoints: immediately, and after refrigerated storage (4 °C) at 24, 48, and 72 h after removal from freezer (24 h at -20 °C). 2-Hydroxy-4-(methylthio)-butanoic acid included as 1.25% of the meat mixture reduced S. enterica and L. monocytogenes compared with the control (P < 0.05); however, it did not result in total kill of either of these pathogens. Following this, feeding behaviors of seven domestic cats were assessed when offered a raw chicken diet treated with or without 1.25% HMTBa for 5 d each, after which a 2-d 2-choice preference test was conducted. Cats demonstrated a preference for raw diets without HMTBa, but still readily consumed diets with 1.25% HMTBa, suggesting that such a diet was still palatable to them.

This narrative literature review summarizes findings regarding the associations of clinical and subclinical hypocalcemia with postpartum health, reproduction, and milk production. To better understand the effects of hypocalcemia, we reviewed clinical and subclinical presentations of the condition and the dynamics of blood Ca concentration in the early postpartum period. We summarize and discuss the associations between hypocalcemia and performance of dairy cows. Up to 50% of dairy cows suffer from at least one disease event in the transition period. The important roles of calcium in muscle contraction and immune function make it a key component of metabolism, inflammation, and defense against infection. Although the effect of clinical hypocalcemia (milk fever) on health and performance is clear, the definition of subclinical hypocalcemia (SCH) and its consequences for health and performance are still ambiguous. Differences in study designs, sampling protocols, Ca concentration thresholds, and sample sizes that may be underpowered for health and reproduction outcomes lead to inconsistent conclusions on the effects of SCH. On current evidence, classification of SCH should be based on at least 2 measurements of blood calcium, using cutpoints supported with relevant data, which may vary depending on the outcome of interest. Arbitrary or poorly supported interpretative thresholds for blood Ca concentrations should be abandoned. Transient SCH appears to be associated with greater milk yield, whereas SCH that is present several days after calving is associated with lesser production and greater disease risk. However, when blood calcium is measured days after calving, primary effects of calcium metabolism may be confounded by feed intake, inflammation, or disease, which might either contribute to or be a consequence of hypocalcemia. Additional research is needed to refine sampling schemes to classify SCH, and to better inform the goals and means of prevention of SCH.

The use of local anesthesia and a nonsteroidal anti-inflammatory drug (NSAID) can reduce indicators of pain and inflammation and encourage self-rewarding behavior in calves following disbudding. Although the use of sedation may be recommended as a best practice for disbudding, there is little research in this area. The objective of this study was to evaluate the effects of xylazine sedation in conjunction with a local anesthetic and an NSAID in calves undergoing cautery disbudding. One hundred twenty-two group-housed female and male Holstein calves fed milk with automated feeders, aged 13 to 44 d, were enrolled over 9 replicates and randomly allocated to 1 of 2 treatments: (1) sedated: lidocaine cornual nerve block, 0.5 mg/kg meloxicam (administered subcutaneously) and 0.2 mg/kg xylazine (administered intramuscularly), or (2) nonsedated: lidocaine cornual nerve block and meloxicam. Outcomes collected consisted of feeding behavior (collected using automated milk feeders), latency to drink milk following disbudding, play behavior (induced by adding bedding), lying behavior, mechanical nociceptive threshold (MNT, measured using a pressure force algometer), struggling behavior during disbudding, length of time to administer the nerve block, length of time to disbud, and serum haptoglobin concentrations. Data were analyzed using mixed models with a fixed effect for baseline values and a random effect for trial replicate. Linear regression was used to assess continuous outcomes, logistic regression for binary outcomes, and Poisson and negative binomial models for count data with negative binomial models used if the over dispersion term was significant. There were no detected differences between the treatment groups in mean daily milk consumption in the 72-h following disbudding. Sedated calves had reduced average milk drinking speed from 0 to 24 h and 24 to 48 h following disbudding compared with nonsedated calves, but no difference was detected from 48 to 72 h. Sedated calves had reduced MNT at 0, 60, and 240 min after disbudding, but no differences were detected between groups at 24 h after disbudding. Nonsedated calves had 4.5 times the odds (95% CI: 1.5-13.2) of struggling more than twice during the disbudding procedure compared with sedated calves, and it took less time to administer a nerve block to sedated calves compared with nonsedated. At +3 h, nonsedated calves were 79 times (95% CI: 22.4 to 279.2) more likely to play compared with sedated calves, and 24 h after disbudding, sedated calves were 2 times more likely to play compared with nonsedated calves (95% CI: 0.93-4.3). The results indicate that calves sedated with xylazine for cautery disbudding responded less to painful stimuli (disbudding and MNT) both during and following the procedure and had a higher rate of play behavior 24 h following sedation compared with the nonsedated calves, but xylazine may also have a prolonged carryover effect that affects suckling behavior for 48 h following sedation.

Our objective was to assess the effects of feeding negative dietary cation-anion difference (DCAD) prepartum diets on milk production, reproductive performance, and culling. Cows from 4 commercial farms in Ontario, Canada were enrolled in a pen-level controlled trial from November 2017 to April 2019. Close-up pens (1 per farm) with cows 3 wk before calving were randomly assigned to a negative DCAD (TRT; -108 mEq/kg of dry matter; target urine pH 6.0-6.5) or a control diet (CON; +105 mEq/kg of dry matter with a placebo supplement). Each pen was fed TRT or CON for 3 mo (1 period), and then switched to the other treatment for the next period (4 periods per farm). Data from 15 experimental units (8 pen treatments in TRT and 7 in CON), with a total of 1,086 observational units (cows), were included. The effect of treatment on milk yield at the first 3 milk recording tests of lactation was assessed with linear regression models accounting for repeated measures. The risk of pregnancy at first artificial insemination and culling by 30, 60, and 305 d in milk (DIM) were analyzed with logistic regression models, and effects on time to first AI, pregnancy, and culling were assessed with Cox proportional hazards models. All models included treatment, parity, and their interactions, accounting for pen-level randomization and clustering of animals within farm with random effects, giving 10 degrees of freedom for treatment effects. Multiparous cows fed TRT produced more milk at the first (42.0 vs. 38.8 ± 1.2 kg/d) and second (44.2 vs. 41.7 ± 1.3 kg/d) milk tests. However, multiparous cows fed TRT tended to have 0.2 percentage units less milk fat content at these tests. Although multiparous cows fed TRT tended to have greater energy-corrected milk at the first test (least squares means ± standard error: TRT = 46.1 ± 0.9 vs. CON = 43.8 ± 1 kg/d), there were no differences observed in energy-corrected milk at the second or third tests. In primiparous cows, there was no effect of treatment on milk production. Multiparous cows fed TRT had greater pregnancy to first insemination (TRT = 42 ± 3 vs. CON = 32 ± 4%) and tended to have shorter time to pregnancy [hazard ratio (HR) = 1.20; 95% CI: 0.96-1.49]. In primiparous cows fed TRT, time to pregnancy was increased (HR = 0.76; 95% CI: 0.59-0.99). Culling by 30 DIM tended to be less in TRT (3.3 ± 1.1%) than CON (5.5 ± 1.8%). No effect of treatment on culling by 305 DIM was detected in primiparous cows, but in multiparous cows, the TRT diets decreased the odds of culling (21.3 ± 1.9 vs. 31.7 ± 2.8%) and daily risk of culling to 305 DIM (HR = 0.64; 95% CI: 0.46 to 0.89). Under commercial herd conditions, prepartum negative DCAD diets improved milk production and reproductive performance, and reduced culling risk in multiparous cows. In primiparous cows, TRT diets had no effect on milk yield or culling, but increased the time to pregnancy. Our results suggest that negative DCAD diets should be targeted to multiparous cows.

Each pen was fed TRT or CON for 3 mo (1 period) then switched to the other treatment for the next period, with 4 periods per farm. Urine pH was measured weekly until calving, and body condition score (BCS) was measured at enrollment and at 5 wk postpartum. Data from 15 experimental units [8 TRT and 7 CON, with 1,086 (TRT: n = 681; CON: n = 405) observational units (cows)] that received the assigned diet for >1 wk were included. The incidence of milk fever (MF), retained placenta (RP), metritis, hyperketonemia (blood ß-hydroxybutyrate >1.2 mmol/L, measured weekly in wk 1 and 2), clinical mastitis within 30 DIM (MAST), displaced abomasum (DA) within 30 d in milk (DIM), purulent vaginal discharge (PVD, assessed once at wk 5), and number of disease events (≥1 or ≥2) were analyzed with logistic regression models with treatment, parity, BCS, and their interactions, accounting for pen-level randomization and clustering of animals within farm with random effects, giving 10 degrees of freedom to test treatment effects. Multiparous cows fed TRT had greater blood calcium between 1 and 4 DIM than multiparous cows fed CON, and the prevalence of subclinical hypocalcemia (total Ca ≤2.14 mmol/L) was lesser when fed TRT compared with CON (d 1: 73 ± 6% vs. 93 ± 4%; d 2: 65 ± 7% vs. 90 ± 5%), with no differences between treatments detected in primiparous cows. We detected interactions of treatment and BCS at enrollment for MF in multiparous cows and of treatment and parity for ≥2 disease events. Overconditioned (BCS ≥3.75) multiparous cows had reduced incidence of MF when fed TRT (TRT: 2 ± 1%, vs. CON: 13 ± 8%). We detected no treatment effects on RP, metritis, hyperketonemia, or PVD incidence. Cows fed TRT had lesser incidence of DA (1.7 ± 0.7% vs. 3.6 ± 1.6%) and tended to have lesser incidence of MAST compared with CON (1.8% ± 0.6% vs. 4.4 ± 1.4%). No treatment effect was detected on ≥1 disease events (TRT: 38 ± 7%, vs. CON: 42 ± 8%); however, multiparous cows on TRT were less likely to have ≥2 disease events than cows on CON (14 ± 4% vs. 23 ± 6%). Under commercial herd conditions, feeding prepartum diets with negative DCAD improved several measures of postpartum health.

Body condition change during the dry period (ΔBCS) has been associated with a myriad of transition cow diseases. We used data from 3 studies to assess the relationship between ΔBCS, feeding behavior, and body condition score (BCS) at dry-off. We also studied the mediation effect that dry matter intake (DMI) has on ΔBCS and the association between dry period feeding time and DMI. A total of 100 parous cows were enrolled in 3 studies to investigate differences in dry period diet on behavior, health, and performance pre- and postcalving. Body weight was measured and BCS was assessed by the same trained observer after dry-off and 1 wk from calving date. The ΔBCS was calculated by subtracting the BCS at calving minus the BCS at dry-off. The BCS at dry-off was categorized as overconditioned (≥3.5) or not overconditioned (<3.5); no cows had a BCS <2.75. Feeding behavior data were collected using electronic feed bins. Parity at dry-off (median = 2; min = 1, max = 6) and 305-d milk production (mean = 10,235 kg, SD = 1,625 kg) from the previous lactation were considered. Data sets were split into 2 time periods: d -56 to -22 (early) and -21 to 0 (late) in relation to calving. Selected feeding behaviors (DMI, DMI as a percentage of body weight, and feeding time) were used to evaluate the associations between each feeding behavior and BCS at dry-off in each period using mixed linear regression models. Each model included the following covariates: parity, previous 305-d milk yield, and trial treatment. Experimental day was included as random slope, and cow was included as random intercept. A mediation analysis was used to evaluate the potential causal direct effect of BCS at dry-off on ΔBCS and the potential indirect effect mediated by differences in DMI. The BCS at dry-off was associated with changes in feeding behavior, such that overconditioned cows had lesser daily DMI and feeding time during the early and late dry periods compared with not overconditioned animals. We also noted an effect of previous 305-d milk yield on DMI; cows that produced more milk had greater DMI throughout the dry period. The ΔBCS was only partially mediated by DMI, and BCS at dry-off still had a direct effect on ΔBCS. This result indicated that mechanisms other than DMI were associated with BCS loss during the dry period. Feeding time correlated weakly and moderately with DMI during the early and late dry periods, respectively. To conclude, strategies to improve intake during the dry period should take dry-off BCS into account or, preferably, efforts should be made to minimize the number of overconditioned cows at the end of lactation.

Exposure of lab animals, humans, and pigs to olfactory sensory feed additives may reduce response to stress and anxiety. The objective of this preliminary study was to determine if feeding a citrus-based olfactory sensory functional feed extract (derived from Citrus sinensis) reduces the negative impact of regrouping of lactating dairy cows. Thirty-two (parity = 2.0 ± 1.2; mean ± SD), mid-lactation Holstein dairy cows (169.8 ± 16.8 DIM) were enrolled as focal cows in this study and housed individually in a tie-stall facility where they were randomly assigned to 1 of 2 treatment diets: (1) control total mixed ration (TMR) (control; n = 16; primiparous = 7; multiparous = 9), or (2) control TMR with 4 g/d of citrus extract (CE) (Phodé, Terssac, France) (CE; n = 16; primiparous = 7; multiparous = 9). Cows were fed their experimental diets for 7 d in the tie-stall facility (baseline), then moved to 1 of 2 experimental free-stall pens (containing 29 other cows) for a period of 7 d, where they remained on the same treatment diet as before. Compared with their baseline, primiparous control cows had decreased rumination time on d 1 and 2, had decreased lying time on d 1, and tended to have decreased lying time on d 2 and 3 following regrouping. In contrast, primiparous cows fed the CE diet did not experience a change in rumination and lying time. Primiparous CE cows had greater feeding time on d 1 and tended to have greater feeding time on d 2 after regrouping compared to primiparous control cows. Primiparous control cows had greater idle standing time, as compared to the CE cows, across the 7 d after regrouping. Primiparous CE cows initiated less total competitive behavior after regrouping, but were also displaced more frequently from the feed bunk and from the free stalls on d 1 after regrouping, as compared to the primiparous control cows. For multiparous cows, CE supplementation was not consistently associated with any benefits to behavior or production after regrouping, possibly because these cows were more experienced with social stressors. Results indicate that feeding CE to mid-lactation naïve primiparous dairy cows may reduce the initiation of competitive interactions and lessen the reduction in rumination and lying time after regrouping. These results need to be verified in further studies where potential confounding effects (e.g., pen social dynamics, pen location) are minimized.

Calfhood morbidity is an important issue on commercial dairy operations, with substantial production and welfare impacts. The objective of this observational study was to evaluate potential risk factors for morbidity, including disbudding, disbudding pain control, serum total protein (STP) concentrations, and haptoglobin concentrations in young dairy calves. A total of 140 heifer calves from a commercial dairy farm in southwestern Ontario, Canada, were enrolled at 1 to 9 d of age and followed for 1 wk. Calves were scored at enrollment for several health parameters using a scale from 0 to 3, with 0 representing normal and 3 representing severely abnormal. Calves were only included if they were considered healthy enough to participate using cut points of <3 for rectal temperature and fecal score; ≤2 for nasal discharge, ocular discharge, and cough score; and 1 for ear position, navel score, and joint score. To ensure that only calves that were considered healthy on the baseline day were included, 17 calves were excluded from analysis, resulting in a total of 123 calves. These calves were a part of a disbudding trial, with 100 disbudded using a commercial caustic paste and 23 sham disbudded with a placebo paste. The disbudding paste (commercial or sham) was applied to calves on enrollment day (baseline) with health scoring at baseline and 3 and 7 d later. The following health outcomes were analyzed: diarrhea (a fecal consistency score of ≥2), respiratory disease (a total respiratory score of ≥5), and general sickness [suffering from either diarrhea, respiratory disease, or a navel or joint infection (joint or navel score of ≥2)]. The following variables were assessed for association with the outcomes: STP, inadequate transfer of passive immunity (<5.2 g/dL STP), pain control treatment for disbudding (lidocaine nerve block alone, meloxicam alone, lidocaine nerve block and meloxicam, no pain control, sham), disbudding (disbudded or sham), and haptoglobin concentrations (taken on the baseline day, +3 d and +7 d). We did not detect any relationship between the development of a health outcome of interest (diarrhea, respiratory, or sick) and disbudding or pain control on any of the days following disbudding. However, for every 1 g/dL increase in STP concentration, calves had 0.24 and 0.33 times the odds of suffering from diarrhea or any sickness event, respectively, 3 d after disbudding. At 3 d after disbudding, for every 0.1 mg/mL increase in haptoglobin concentration on that day, calves had 1.34 and 1.20 times the odds of having diarrhea or a general sickness, respectively. These findings suggest that disbudding itself or the pain control method does not appear to influence health after the procedure when healthy calves are disbudded. Calves developing an illness 3 d after disbudding were more likely to have had lower STP concentrations and increased haptoglobin concentrations.

Our objective was to validate a point-of-care handheld blood total calcium analyzer (Ca meter, CM; TD-5220 Vet Ca2+, TaiDoc, New Taipei, Taiwan) to estimate circulating Ca concentrations in postpartum dairy cows. Whole blood was collected from 251 multiparous cows between 1 and 4 d in milk from 2 commercial dairy herds in Ontario, Canada. Blood total calcium concentration (tCa) was analyzed in whole blood, fresh plasma, and thawed plasma, and compared with tCa results from thawed serum analyzed in a diagnostic laboratory (using a Cobas Calcium Gen 2 kit, Roche Diagnostics, Indianapolis, IN) as the reference test (RT). Lin's concordance correlation coefficient (ßrho;) and Bland-Altman (B-A) plots were assessed to evaluate the agreement between the RT and CM results in each type of sample. Receiver operating characteristic curve analyses were used to describe the accuracy of each test against the categorized RT results (at a cut-point of ≤2.14 mmol/L). Samples where the meter gave a nonquantitative result ("high" or "low"; thawed plasma: 3/247; fresh plasma: 6/100; and whole blood: 20/98) were not included in the ßrho; and B-A analyses. Lin's correlation coefficients demonstrated poor agreement between tests (thawed plasma: ßrho; = 0.16; fresh plasma: ßrho; = 0.21; and whole blood: ßrho; = 0.23). Fresh plasma (using a cut-point of 2.55 mmol/L as measured on the CM) had the greatest diagnostic sensitivity (72%), specificity (86%), and accuracy (77%) for determining subclinical hypocalcemia, but that would still misclassify 23% of samples. In addition to substantial variability, the B-A plots revealed bias with changing concentrations of calcium. Because of low sensitivity on whole blood (58%) or thawed plasma (56%), measurement with the CM is not recommended on these types of samples. This rapid and low-cost meter was not sufficiently accurate to quantify blood Ca concentration, but when used with fresh plasma it might be useful as a screening tool for subclinical hypocalcemia.

The effects of feeding frequency on postprandial response of circulating appetite-regulating hormones, insulin, glucose and amino acids, and on physical activity, energy expenditure, and respiratory quotient were studied in healthy adult cats. Two experiments were designed as a 2 x 3 replicated incomplete Latin square design. Eight cats, with an average body weight (BW) of 4.34 kg ± 0.04 and body condition score (BCS) of 5.4 ± 1.4 (9 point scale), were fed isocaloric amounts of a commercial adult maintenance canned cat food either once (0800 h) or four times daily (0800 h, 1130 h, 1500 h, 1830 h). Study 1 consisted of three 21-d periods. On day 14, two fasted and 11 postprandial blood samples were collected over 24 hours to measure plasma concentrations of ghrelin, GLP-1, GIP, leptin, PYY, insulin and amino acids, and whole blood glucose. Physical activity was monitored from day 15 to 21 of each period. In Study 2 indirect calorimetry was performed on the last day of each period. Body weight was measured weekly and feed intake recorded daily in both experiments. No effect of feeding regimen on BW was detected. Cats eating four times daily had lesser plasma concentrations of GIP and GLP-1 (P<0.05) and tended to have lesser plasma PYY concentrations (P<0.1). Plasma leptin and whole blood glucose concentrations did not differ between regimens (P>0.1). Cats fed once daily had a greater postprandial plasma amino acid response, and greater plasma ghrelin and insulin concentrations (P<0.05). Physical activity was greater in cats fed four times (P<0.05), though energy expenditure was similar between treatments at fasting and in postprandial phases. Finally, cats eating one meal had a lower fasting respiratory quotient (P<0.05). Overall, these data indicate that feeding once a day may be a beneficial feeding management strategy for indoor cats to promote satiation and lean body mass.
