Polyphenol-rich grape pomace (GP) represents a valuable processing by-product with considerable potential as sustainable livestock feed. This study aimed to investigate the effects of different levels of GP on the growth performance and nitrogen utilization efficiency, antioxidant activity, and rumen and rectum microbiota of Angus bulls. Thirty Angus bulls were allocated three dietary treatments according to a completely randomized design: 0% (G0), 10% (G10), and 20% (G20) corn silage dry matter replaced with dried GP dry matter. The results showed that the average daily gain (ADG) of the G0 group and G10 group was higher than that of the G20 group (p < 0.05); urinary nitrogen levels decreased linearly with the addition of GP (linear, p < 0.05). In terms of antioxidants, the levels of catalase (CAT) in the G10 group were higher than in the G0 and G20 groups (p < 0.05), and the total antioxidative capacity (T-AOC) was significantly higher than that in the G20 group (p < 0.05). In addition, in the analysis of a microbial network diagram, the G10 group had better microbial community complexity and stability. Overall, these findings offer valuable insights into the potential benefits of incorporating GP into the diet of ruminants.

This study aims to explore the different growth performances of the Angus bull on potato vine and leaf mixed silage in the early fattening period and to provide a reference animal production trial. Thirty-six 13-month-old Angus bulls were divided into three groups with 403.22 ± 38.97 kg initial body weight and fed with three different silage diets: (1) control: whole-plant corn silage as control (CS); (2) treatment 1: 50% whole-plant corn +50% potato vine and leaf silage (PVS1); and (3) treatment 2: 75% potato vine and leaf +15% rice straw +10% cornmeal silage (PVS2). After the 14 days pre-feeding, the formal experiment was carried out for 89 days. The result showed that the ash content of the potato vine and leaf mixed silage (PVS) in the treatment groups was higher than that in control group, and the ash content of PVS1 and PVS2 even reached 10.42% and 18.48% (DM%), respectively, which was much higher than that of the CS group at 4.94%. The crude protein content in silage also increased with the additional amount of potato vine and leaf. The apparent crude protein digestibility of the PVS groups was also significantly higher than that of the CS group (p < 0.05). In terms of serum biochemical indexes, blood urea nitrogen (BUN) in the experimental groups was significantly higher than in the control group (p < 0.05). Compared with PVS2, cholesterol (CHO) was significantly lower in the CS and PVS1 groups (p < 0.05). Moreover, the high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) of PVS2 were significantly higher than those of the CS and PVS1 group (p < 0.05), and daily gain (ADG) as a key production index had a significantly negative correlation with the CHO (r = -0.38, p < 0.05) and HDL-C (r = -0.40, p < 0.05) of cattle. In conclusion, PVS had higher crude protein content and ash but less starch than whole-corn silage. The PVS could replace whole-plant corn silage at the same dry matter status and did not affect the weight gain in this trial.

Objective: Osteoporosis (OP) can be considered a chronic complication of type 2 diabetes mellitus (T2DM). Aberrant activation of the nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome is associated with the pathogenesis of various inflammation-related diseases, e.g., T2DM and OP. Vitamin D affects the inflammatory pathway and inhibits an excessive inflammatory response. The current study investigated the inter-relationship between vitamin D and inflammasome activation in T2DM. Method: Hepatocellular carcinoma (HepG2) cells and bone marrow stromal cells (BMSCs) were treated with Conditioned Medium of bone marrow mesenchymal stem cells after VitD treatment (CM-VitD), as well as phosphoinositide 3-kinase (PI3K) specific agonist, 740Y-P, or the PI3K specific inhibitor, LY294002, respectively, or both. 40 Eight-week-old female Sprague Dawley rats were selected and established as a DM model. The rats were injected with CM-VitD, as well as the 740Y-P specific agonist, or the LY294002 inhibitor, respectively, or both. A quantitative reverse transcription polymerase chain reaction and western blotting were conducted to evaluate the expression of messenger ribonucleic acid and protein in the RUX2 gene, alkaline phosphatase (ALP), OsteoPontiN (OPN), peroxisome proliferator-activated receptor gamma (PPARγ), fatty acid-binding protein 4 (FABP4), protein kinase B (AKT), PI3K, NLRP3, apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC), caspase-1, interleukin (IL)-1 beta (ß), IL-18, and tumor necrosis factor alpha (TNF-α) in the BMSCs and liver tissue of rats. Enzyme-linked immunosorbent assay was used to detect the concentration of inflammatory factors in the cell supernatant and serum of rats. Results: An isolated co-culture of HepG2/insulin-resistance cells and BMSCs promoted the adipogenic transformation of the latter and inhibited the transformation of BMSCs into osteogenesis. The PI3K specific agonist, 740Y-P, significantly increased the expression of PI3K, AKT, NLRP3, ASC and Caspase-1 while the PI3K specific inhibitor, LY294002, does the opposite. Additionally, CM-VitD reduced the expression of NLRP3, ASC, caspase-1, IL-1ß, and IL-18 in BMSCs and rat liver via the PI3K/AKT pathway. Conclusion: Vitamin D can inhibit the inflammatory response induced by T2DM and promote the osteogenesis of BMSCs, which may play a key role in the treatment of type 2 diabetes patients with OP.

This study aimed to evaluate the effects of different proportions of alfalfa silage on the fermentation quality, in vitro digestibility, and aerobic stability of total mixed ration (TMR) silage. Three TMRs were prepared with different silage contents on a fresh matter basis: (1) 60% alfalfa silage (AS60), (2) 40% alfalfa silage (AS40), and (3) 20% alfalfa silage (AS20). The lactic acid in AS60 did not increase after 30 days of ensiling (p > 0.05). Butyric acid was detected in the AS20 group after 14 days of ensiling. The AS60 group showed significantly higher in vitro dry matter digestibility than the AS20 group (p < 0.05). The aerobic stability of TMR silage gradually increased with a decreasing percentage of alfalfa silage (p < 0.05). Unlike AS60, which directly gained an acidic environment from the alfalfa silage, AS40 developed a stable acidic environment during ensiling and further improved aerobic stability. However, when the percentage of alfalfa silage was reduced to 20%, a risk of clostridial spoilage occurred in the TMR silage. Therefore, the addition of 40% alfalfa silage to TMR is optimal and could achieve both good fermentation quality and considerable resistance to aerobic deterioration in TMR silage.

A set of photo-switchable monopeptides derived from cis-ß-dibenzodiazocine-l-alanine (cis-DBDAA) have been designed and synthesized, which are capable of photo-click reacting with diaryltetrazoles or diarylsydnones in a hydrophobic phospholipid bilayer environment. The DBDAA monopeptides include both a hydrophobic tail on C-terminal, providing high affinity toward lipid membrane, and a modularized functional moiety on N-terminal, enabling rapid optimization of the self-assembly strength to form multifunctional supramolecules. With the cis-DBDAA monopeptides photo-switched into trans-configuration, we were able to disrupt the supramolecular assembly through an efficient photo-click reaction across the lipid bilayer of liposomes. We reveal that the performance of the photo-click reactions between the monopeptides and photo-generated nitrile imine intermediates is significantly enhanced by enrichment of both reactants in the hydrophobic membrane lamel of liposomes. Enrichment of the DBDAA monopeptide in lipid phase serves as a convenient method to introduce bioorthogonal chemical handles on live cell membranes, which enables fluorescence labelling of single cell's membrane with high spatiotemporal resolution to facilitate the studies on cell membrane dynamics.

RESUMEN

A photo-induced 1,3-dipolar cycloaddition between nitrile imine and highly ring-strained N[double bond, length as m-dash]N double bond as a dipolarophile was discovered. The photo-isomerization of carbon-bridged octocyclic azobenzene (CBOA) into its trans-configuration accelerates the ligation reaction at a very rapid rate (28 400 M-1 s-1). The CBOA-based photo-click reaction was proved to be bioorthogonal. In addition, the NoxaB peptide was successfully cross-linked by a CBOA stapler which plays a dual role: photo-control of the conformation of the peptide and photo-conjugation of probes in live cells.

Ultra-fast and selective covalent-bond forming reactions with spatiotemporal controllability are foundational for developing a bioorthogonal approach with high manipulability. However, it is challenging to exploit a reporter functional group to achieve these requirements simultaneously. Here, 11H-Dibenzo[c,f][1,2]diazepine and a set of heterocyclic analogues are investigated for both their photo-switching natures and their ability to serve as dipolarophiles in photo-click reactions with diarylsydnone. Sulfur-containing dibenzothiadiazepine (DBTD) is discovered to be an excellent chemical reporter in cycloaddition with visible-light excitation for in-situ ring-strain loading via its (Z) → (E) photo-isomerization. The bioorthogonal utility of the DBTD tag in spatiotemporally controlled ligation for protein modifications on live cells is also demonstrated.

We constructed a library of diarylsydnone (DASyd) candidates in search of a photoclickable reaction toward alkynes, enabling an ultra-accelerated reactivity, while suppressing the background cycloaddition in the dark. The in vitro and in vivo protein labelling experiments revealed that the photo-accelerated DASyd-alkyne cycloaddition exhibits robust selectivity.

A library of 12N,9-Diaryl 2-methyl-8-azaadenine (DAMA) compounds was designed and constructed through an aryl-pairing combination strategy for identifying a nucleobase-containing molecular switch that functions by the pH-regulated Dimroth rearrangement. By utilizing 2D thin-layer chromatography/mass spectrometry (2D-TLC-MS), the DAMA compounds were easily screened to identify which compounds could be used as molecular switches. The pH-switching ability of the DAMA was achieved by incorporating the acridine group as the key structural unit, as well as dual-modal colorimetric/fluorometric on/off properties as the probe functions. The real-time tracing of the switching process clearly indicated that the paired aromatics on both terminals of the DAMA molecule play a key role in tuning the switching kinetics.

Use of tight ultrafiltration (UF) membranes has created a new pathway in fractionation of dye/salt mixtures from textile wastewater for sustainable resource recovery. Unexpectedly, a consistently high rejection for the dyes with smaller sizes related to the pore sizes of tight UF membranes is yielded. The potential mechanism involved in this puzzle remains unclear. In this study, seven tailored UF membranes with molecular weight cut-offs (MWCOs) from 6050 to 17530 Da were applied to separate dye/salt mixtures. These UF membranes allowed a complete transfer for NaCl and Na2SO4, due to large pore sizes. Additionally, these UF membranes had acceptably high rejections for direct and reactive dyes, due to the aggregation of dyes as clusters for enhanced sizes and low diffusivity. Specifically, the membrane with an MWCO of 7310 Da showed a complete rejection for reactive blue 2 and direct dyes. An integrated UF-diafiltration process was subsequently designed for fractionation of reactive blue 2/Na2SO4 mixture, achieving 99.84% desalination efficiency and 97.47% dye recovery. Furthermore, reactive blue 2 can be concentrated from 2.01 to 31.80 g·L-1. These results indicate that UF membranes even with porous structures are promising for effective fractionation of dyes and salts in sustainable textile wastewater treatment.

RESUMEN

A small library of diarylsydnones (DASyds) was constructed based on aryl-pairing combinations and subjected to click reaction toward alkenes under photoirradiation with high efficiency. We were able to demonstrate the utility of DASyds for highly fluorescent turn-on ligation targeting the trans-cyclooct-4-en-1-ol moieties on protein.
