RESUMEN

Pasture-based beef cattle are raised in a range of production environments. Some paddocks may contain trees and other objects that allow for grooming, hence being naturally enriching, whilst others may be barren without these opportunities. Additionally, it is not uncommon for cattle to move between these enriched and barren environments as part of routine management. While the benefits of enrichment are well studied, how this 'enrichment loss' impacts cattle welfare as access to stimuli is removed is unknown. This trial assessed the impacts of the loss of an enriching object (grooming brush) on grazing beef cattle welfare and production characteristics. When grooming brush access was blocked, cattle became dirtier, showed reduced average daily gain, and had elevated faecal cortisol metabolites, although this varied according to the degree of initial individual brush use. Additionally, allogrooming and grooming on other objects were reduced when access to the brush was returned, potentially indicating a rebound effect. These results demonstrate that the loss of adequate grooming objects can impair the overall welfare of grazing cattle; however, further work is needed to determine exactly which natural or artificial objects provide adequate grooming opportunities.

RESUMEN

OBJECTIVE: We propose an efficient approach based on a convolutional denoising autoencoder (CDA) network to reduce motion and noise artifacts (MNA) from corrupted atrial fibrillation (AF) and non-AF photoplethysmography (PPG) data segments so that an accurate PPG-signal-derived heart rate can be obtained. Our method's main innovation is the optimization of the CDA performance for both rhythms using more AF than non-AF data for training the AF-specific CDA model and vice versa for the non-AF CDA network. METHODS: To evaluate this unconventional training scheme, our proposed network was trained and tested on 25-sec PPG data segments from 48 subjects from two different databases-the Pulsewatch dataset and Stanford University's publicly available PPG dataset. In total, our dataset contains 10,773 data segments: 7,001 segments for training and 3,772 independent segments from out-of-sample subjects for testing. RESULTS: Using real-life corrupted PPG segments, our approach significantly reduced the average heart rate root mean square error (RMSE) of the reconstructed PPG segments by 45.74% and 23% compared to the corrupted non-AF and AF data, respectively. Further, our approach exhibited lower RMSE, and higher sensitivity and PPV for detected peaks compared to the reconstructed data produced by the alternative methods. CONCLUSION: These results show the promise of our approach as a reliable denoising method, which should be used prior to AF detection algorithms for an accurate cardiac health monitoring involving wearable devices. SIGNIFICANCE: PPG signals collected from wearables are vulnerable to MNA, which limits their use as a reliable measurement, particularly in uncontrolled real-life environments.

BACKGROUND: The prevalence of atrial fibrillation (AF) increases with age and can lead to stroke. Therefore, older adults may benefit the most from AF screening. However, older adult populations tend to lag more than younger groups in the adoption of, and comfort with, the use of mobile health (mHealth) apps. Furthermore, although mobile apps that can detect AF are available to the public, most are designed for intermittent AF detection and for younger users. No app designed for long-term AF monitoring has released detailed system design specifications that can handle large data collections, especially in this age group. OBJECTIVE: This study aimed to design an innovative smartwatch-based AF monitoring mHealth solution in collaboration with older adult participants and clinicians. METHODS: The Pulsewatch system is designed to link smartwatches and smartphone apps, a website for data verification, and user data organization on a cloud server. The smartwatch in the Pulsewatch system is designed to continuously monitor the pulse rate with embedded AF detection algorithms, and the smartphone in the Pulsewatch system is designed to serve as the data-transferring hub to the cloud storage server. RESULTS: We implemented the Pulsewatch system based on the functionality that patients and caregivers recommended. The user interfaces of the smartwatch and smartphone apps were specifically designed for older adults at risk for AF. We improved our Pulsewatch system based on feedback from focus groups consisting of patients with stroke and clinicians. The Pulsewatch system was used by the intervention group for up to 6 weeks in the 2 phases of our randomized clinical trial. At the conclusion of phase 1, 90 trial participants who had used the Pulsewatch app and smartwatch for 14 days completed a System Usability Scale to assess the usability of the Pulsewatch system; of 88 participants, 56 (64%) endorsed that the smartwatch app is "easy to use." For phases 1 and 2 of the study, we collected 9224.4 hours of smartwatch recordings from the participants. The longest recording streak in phase 2 was 21 days of consecutive recordings out of the 30 days of data collection. CONCLUSIONS: This is one of the first studies to provide a detailed design for a smartphone-smartwatch dyad for ambulatory AF monitoring. In this paper, we report on the system's usability and opportunities to increase the acceptability of mHealth solutions among older patients with cognitive impairment. Environmental enrichment can improve livestock welfare through increasing environmental complexity to promote a greater range of natural behaviours. However, there is limited understanding of the need for and impacts of enrichments for extensively managed beef cattle that can sometimes be kept in grassed paddocks devoid of additional natural and artificial features, i.e., 'barren pastures'. This trial assessed which enrichments beef cattle preferred and utilised in a barren paddock environment. Eight groups of seven Angus steers housed on pastured paddocks devoid of natural or artificial features were observed during daylight hours for two days a week over a period of three weeks, after being presented with four enrichments simultaneously: a cattle brush, a piece of hanging rope, a tree stump, and a woodchip pile. Although enrichment use generally decreased over time, the brush, stump, and woodchip maintained a higher level of use than the rope, based on the frequency of interactions and number of displacements around the enrichments (both p < 0.001). This suggests that the brush, stump, and woodchip pile were more valuable resources to the cattle, allowing for grooming and lying behaviours, although oral manipulations also occurred on the stump, woodchip, and rope. The inclusion of these enrichments can increase the complexity of barren pasture environments and allow for the increased expression of natural behaviours, potentially contributing to improved welfare.

OBJECTIVE: With the increasing use of wearable healthcare devices for remote patient monitoring, reliable signal quality assessment (SQA) is required to ensure the high accuracy of interpretation and diagnosis on the recorded data from patients. Photoplethysmographic (PPG) signals non-invasively measured by wearable devices are extensively used to provide information about the cardiovascular system and its associated diseases. In this study, we propose an approach to optimize the quality assessment of the PPG signals. METHODS: We used an ensemble-based feature selection scheme to enhance the prediction performance of the classification model to assess the quality of the PPG signals. Our approach for feature and subset size selection yielded the best-suited feature subset, which was optimized to differentiate between the clean and artifact corrupted PPG segments. CONCLUSION: A high discriminatory power was achieved between two classes on the test data by the proposed feature selection approach, which led to strong performance on all dependent and independent test datasets. We achieved accuracy, sensitivity, and specificity rates of higher than 0.93, 0.89, and 0.97, respectively, for dependent test datasets, independent of heartbeat type, i.e., atrial fibrillation (AF) or non-AF data including normal sinus rhythm (NSR), premature atrial contraction (PAC), and premature ventricular contraction (PVC). For independent test datasets, accuracy, sensitivity, and specificity rates were greater than 0.93, 0.89, and 0.97, respectively, on PPG data recorded from AF and non-AF subjects. These results were found to be more accurate than those of all of the contemporary methods cited in this work. SIGNIFICANCE: As the results illustrate, the advantage of our proposed scheme is its robustness against dynamic variations in the PPG signal during long-term 14-day recordings accompanied with different types of physical activities and a diverse range of fluctuations and waveforms caused by different individual hemodynamic characteristics, and various types of recording devices. This robustness instills confidence in the application of the algorithm to various kinds of wearable devices as a reliable PPG signal quality assessment approach.

OBJECTIVE: We have developed a peak detection algorithm for accurate determination of heart rate, using photoplethysmographic (PPG) signals from a smartwatch, even in the presence of various cardiac rhythms, including normal sinus rhythm (NSR), premature atrial contraction (PAC), premature ventricle contraction (PVC), and atrial fibrillation (AF). Given the clinical need for accurate heart rate estimation in patients with AF, we developed a novel approach that reduces heart rate estimation errors when compared to peak detection algorithms designed for NSR. METHODS: Our peak detection method is composed of a sequential series of algorithms that are combined to discriminate the various arrhythmias described above. Moreover, a novel Poincaré plot scheme is used to discriminate between basal heart rate AF and rapid ventricular response (RVR) AF, and to differentiate PAC/PVC from NSR and AF. Training of the algorithm was performed only with Samsung Simband smartwatch data, whereas independent testing data which had more samples than did the training data were obtained from Samsung's Gear S3 and Galaxy Watch 3. RESULTS: The new PPG peak detection algorithm provides significantly lower average heart rate and interbeat interval beat-to-beat estimation errors-30% and 66% lower-and mean heart rate and mean interbeat interval estimation errors-60% and 77% lower-when compared to the best of the seven other traditional peak detection algorithms that are known to be accurate for NSR. Our new PPG peak detection algorithm was the overall best performers for other arrhythmias. CONCLUSION: The proposed method for PPG peak detection automatically detects and discriminates between various arrhythmias among different waveforms of PPG data, delivers significantly lower heart rate estimation errors for participants with AF, and reduces the number of false negative peaks. SIGNIFICANCE: By enabling accurate determination of heart rate despite the presence of AF with rapid ventricular response or PAC/PVCs, we enable clinicians to make more accurate recommendations for heart rate control from PPG data.

To test the hypothesis that complexed trace mineral supplementation would increase antioxidant capacity and decrease muscle oxidative stress and damage in young horses entering an exercise training program, Quarter Horses (mean [Formula: see text] SD; 9.7 ± 0.7 mo) balanced by age, sex, and BW were assigned to receive complexed (CTM; n = 8) or inorganic (INORG; n = 8) trace minerals at -12 week relative to this study. Blood and muscle samples were collected before (week 0) and after 12 week of light exercise training surrounding a 1.5-h trailer stressor. Muscle glutathione peroxidase (GPx) activity was higher for CTM than INORG horses (P ≤ 0.0003) throughout the study. Following both trailer stressors, serum creatine kinase increased (P < 0.0001) and remained elevated through 24 h post-trailering (P < 0.0001). At week 0, muscle malondialdehyde, expression of superoxide dismutase 2, and whole blood GPx activity increased (P [Formula: see text] 0.003) following trailering but trailering did not affect these measures at week 12. Young horses supplemented with CTM had higher muscle GPx activity than horses receiving INORG, but CTM did not affect damage markers following a stressor. Dietary CTM may be useful for improving antioxidant capacity during exercise training in young equine athletes.

Conjugated linoleic acid (CLA) improves oxidative stress and mitochondrial biogenesis in various species but has not been thoroughly investigated in horses. We collected blood and muscle samples from lightly exercising horses before and 6 and 12 wk after receiving either soybean oil (CON; n = 5) or CLA (CLA; n = 5) supplementation. Samples were analyzed for markers of mitochondrial characteristics, antioxidant status, oxidative stress, and muscle damage. Data were analyzed using a linear model with repeated measures. In the triceps brachii (TB), citrate synthase (CS) activity was higher in CON than CLA horses (P = 0.003) but was unaffected by diet in the gluteus medius (GM). Integrative (relative to mg protein) cytochrome c oxidase (CCO) activity was higher in TB than the GM (P < 0.0001), while intrinsic (relative to CS) CCO was lower in the TB than the GM (P = 0.02) and tended to be lower in CON than CLA horses (P = 0.06). Neither CS nor integrative CCO activities were affected by time. In the GM, superoxide dismutase activity tended to increase in CON through week 12 (P = 0.10). Over both muscle groups, glutathione peroxidase activity tended to be higher in CON compared with CLA at week 12 (P = 0.06). Malondialdehyde was higher in the TB than the GM (P = 0.0004) but was unaffected by diet, while serum creatine kinase activity tended to be lower in CLA than CON horses (P = 0.07). These results suggest that CLA supplementation may lead to mitochondrial adaptations and prevent myofiber perturbation in skeletal muscle of young, lightly exercised horses.

In aged humans, low-intensity exercise increases mitochondrial density, function and oxidative capacity, decreases the prevalence of hybrid fibers, and increases lean muscle mass, but these adaptations have not been studied in aged horses. Effects of age and exercise training on muscle fiber type and size, satellite cell abundance, and mitochondrial volume density (citrate synthase activity; CS), function (cytochrome c oxidase activity; CCO), and integrative (per mg tissue) and intrinsic (per unit CS) oxidative capacities were evaluated in skeletal muscle from aged (n = 9; 22 ± 5 yr) and yearling (n = 8; 9.7 ± 0.7 mo) horses. Muscle was collected from the gluteus medius (GM) and triceps brachii at wk 0, 8, and 12 of exercise training. Data were analyzed using linear models with age, training, muscle, and all interactions as fixed effects. At wk 0, aged horses exhibited a lower percentage of type IIx (p = 0.0006) and greater percentage of hybrid IIa/x fibers (p = 0.002) in the GM, less satellite cells per type II fiber (p = 0.03), lesser integrative and intrinsic (p ≤ 0.04) CCO activities, lesser integrative oxidative phosphorylation capacity with complex I (PCI; p = 0.02) and maximal electron transfer system capacity (ECI+II; p = 0.06), and greater intrinsic PCI, ECI+II, and electron transfer system capacity with complex II (ECII; p ≤ 0.05) than young horses. The percentage of type IIx fibers increased (p < 0.0001) and of type IIa/x fibers decreased (p = 0.001) in the GM, and the number of satellite cells per type II fiber increased (p = 0.0006) in aged horses following exercise training. Conversely, the percentage of type IIa/x fibers increased (p ≤ 0.01) and of type IIx fibers decreased (p ≤ 0.002) in young horses. Integrative maximal oxidative capacity (p ≤ 0.02), ECI+II (p ≤ 0.07), and ECII (p = 0.0003) increased for both age groups from wk 0 to 12. Following exercise training, aged horses had a greater percentage of IIx (p ≤ 0.002) and lesser percentage of IIa/x fibers (p ≤ 0.07), and more satellite cells per type II fiber (p = 0.08) than young horses, but sustained lesser integrative and intrinsic CCO activities (p ≤ 0.04) and greater intrinsic PCI, ECI+II, and ECII (p ≤ 0.05). Exercise improved mitochondrial measures in young and aged horses; however, aged horses showed impaired mitochondrial function and differences in adaptation to exercise training.

Background: Atrial fibrillation (AF) is a common heart rhythm disorder that elevates stroke risk. Stroke survivors undergo routine heart rhythm monitoring for AF. Smartwatches are capable of AF detection and potentially can replace traditional cardiac monitoring in stroke patients. Objective: The goal of Pulsewatch is to assess the accuracy, usability, and adherence of a smartwatch-based AF detection system in stroke patients. Methods: The study will consist of two parts. Part I will have 6 focus groups with stroke patients, caretakers, and physicians, and a Hack-a-thon, to inform development of the Pulsewatch system. Part II is a randomized clinical trial with 2 phases designed to assess the accuracy and usability in the first phase (14 days) and adherence in the second phase (30 days). Participants will be randomized in a 3:1 ratio (intervention to control) for the first phase, and both arms will receive gold-standard electrocardiographic (ECG) monitoring. The intervention group additionally will receive a smartphone/smartwatch dyad with the Pulsewatch applications. Upon completion of 14 days, participants will be re-randomized in a 1:1 ratio. The intervention group will receive the Pulsewatch system and a handheld ECG device, while the control group will be passively monitored. Participants will complete questionnaires at enrollment and at 14- and 44-day follow-up visits to assess various psychosocial measures and health behaviors. Results: Part I was completed in August 2019. Enrollment for Part II began September 2019, with expected completion by the end of 2021. Conclusion: Pulsewatch aims to demonstrate that a smartwatch can be accurate for real-time AF detection, and that older stroke patients will find the system usable and will adhere to monitoring.

BACKGROUND: Many digital health technologies capable of atrial fibrillation (AF) detection are directly available to patients. However, adaptation into clinical practice by heart rhythm healthcare practitioners (HCPs) is unclear. OBJECTIVE: To examine HCP perspectives on use of commercial technologies for AF detection and management. METHODS: We created an electronic survey for HCPs assessing practice demographics and perspectives on digital devices for AF detection and management. The survey was distributed electronically to all members of 3 heart rhythm professional societies. RESULTS: We received 1601 responses out of 73,563 e-mails sent, with 43.6% from cardiac electrophysiologists, 12.8% from fellows, and 11.6% from advanced practice practitioners. Most respondents (62.3%) reported having recommended patient use of a digital device for AF detection. Those who did not had concerns about their accuracy (29.6%), clinical utility of results (22.8%), and integration into electronic health records (19.8%). Results from a 30-second single-lead electrocardiogram were sufficient for 42.7% of HCPs to recommend oral anticoagulation for patients at high risk for stroke. Respondents wanted more data comparing the accuracy of digital devices to conventional devices for AF monitoring (64.9%). A quarter (27.3%) of HCPs had no reservations recommending digital devices for AF detection, and most (53.4%) wanted guidelines from their professional societies providing guidance on their optimal use. CONCLUSION: Many HCPs have already integrated digital devices into their clinical practice. However, HCPs reported facing challenges when using digital technologies for AF detection, and professional society recommendations on their use are needed.

An attention bias test has been developed as a measure of negative affective states in sheep. The test measures an individual's allocation of attention between a threatening (previous location of a dog) and positive (conspecific photo) stimulus over a 3 min period. This study replicated a previously inconclusive study, to determine whether the test could assess positive affective states under more controlled conditions and with a younger population of animals. Pharmacological treatments were used to induce anxious, calm, happy, and control affective states prior to entering the attention bias test arena (n = 20/treatment). We hypothesized that sheep in positive and negative affective states could be differentiated using key measures of attention during testing, including vigilance (head at or above shoulder height) and duration looking towards the valenced stimuli. Anxious sheep were more vigilant than control animals during attention bias testing as predicted (linear mixed effects model, p = 0.002), but the positive groups did not differ from controls (p > 0.05). There was no effect of treatment on looking behaviors (p > 0.05). We suggest this attention bias test paradigm can assess negative but not positive affect in sheep and that modifications to the ethogram or stimuli are needed to more clearly characterize the direction of attention during testing.

Assessment of negative affective states is a key component of animal welfare research. In laying hens, excessive fearfulness results in reduced production and increased sensitivity to stress. Fearfulness can be defined as a response to a known threat, but anxiety is a response to an unknown threat and may have similar negative consequences. The open field test and tonic immobility test are commonly applied to measure fearfulness in laying hens. An attention bias test that measured individual hen's responses to playback of a conspecific alarm call in the presence of food was recently pharmacologically validated using an anxiogenic drug but was confounded by the hen's typical motionless response in a novel environment. The current study used 56-week old free-range layers to further assess the validity of an attention bias test to differentiate ranging treatment groups in comparison with the open field and tonic immobility tests. The selected hens varied in their range use patterns as tracked by radio-frequency identification technology. 'Indoor' hens did not access the range and 'outdoor' hens ranged daily; previous research has confirmed higher fearfulness in hens that remain indoors. The tonic immobility test did not differentiate ranging groups (P = 0.34), but indoor birds were slower to first step (P = 0.03) and stepped less (P = 0.02) in the open field test. The attention bias test occurred in an isolated wooden box using a conspecific alarm call playback (a threat) and mixed grain (a positive stimulus). The behavioural response of latency to resume eating following playback of the alarm call was measured to differentiate the anxiety states of the indoor and outdoor ranging birds. Before the attention bias test could occur, birds had to be habituated to the test box across three separate 5-minute sessions to increase the willingness to feed within the novel test environment. All birds ate faster across time (P < 0.001) but the indoor birds were slower to eat than the outdoor birds (P < 0.001). In this study, the latency to resume eating following an alarm call was determined to be a poor measure for highly anxious birds as they failed to eat at all. Forty-six percent of indoor hens were excluded for not eating across the 5-minute test. Of the birds that did eat, only 7% of indoor hens ate following playback of the alarm call, compared with 36% of outdoor hens. This repetition of an attention bias test for laying hens highlights the challenges in assessing hens with extreme fearful/anxious responses and that information may be missed when non-performing hens are excluded from behavioural tests. We suggest that latency to eat in a novel arena without any alarm call playback is an informative measure of anxious state that can be applied to all hens but consideration must be made of potential differences in food motivation.

Endothelial cells (ECs) form an active barrier between the circulation and the body. In addition to controlling transport of molecules between these 2 compartments, the endothelium is a major secretory organ, releasing proteins both into the circulation and into the vascular matrix. Although it is clearly important that proteins are correctly sorted into these 2 spaces, we currently know little of the polarization of this secretion or how it is controlled. Here, we present an optimized system for the analysis of polarized secretion and show that it allows the derivation of deep, robust proteomes from small numbers of primary ECs. We present the first endothelial apically and basolaterally secreted proteomes, demonstrating that ECs polarize the secretion of extracellular vesicle cargoes to the apical surface. Conversely, we find that protein secretion at the basolateral surface is focused on components of the extracellular matrix (ECM). Equine research and management is limited to single-housing systems if individual animal intake is to be precisely recorded. Even then, dry forage intake is difficult to quantify accurately due to stomping or mixing hay with fecal matter and bedding. In cattle management, GrowSafe Systems (GrowSafe) is a commonly used tool to closely monitor individual animal feeding data using radio frequency identification (RFID) tag technology. Animals are equipped with a unique RFID tag that is read by the feed bunks each time the animal lowers its head into the bunk to consume feed. The objectives of this pilot study were 1) to test the feasibility of use of the GrowSafe system with horses by measuring intake of dry hay and 2) to characterize feeding behaviors of horses in an individually housed (without competition) or group-housed (with competition) setting. To test the hypothesis that horses would consume more hay when individually (NOCOMP) compared to group-housed (COMP), 10 mature Quarter Horses (14 ± 1.5 yr) were placed in one of four pens containing GrowSafe feed bunks in a 4-wk crossover design consisting of two 2-wk treatment periods. Pen 1 contained five horses with access to two GrowSafe bunks (Period 1: n = 4 mares, n = 1 gelding; Period 2: n = 5 geldings); pens 2, 3, and 4 contained one horse each with access to one bunk. Horses were individually fed 0.25% body weight (BW; dry matter [DM] basis) of a commercial concentrate once per day and were allowed Coastal bermudagrass hay in the GrowSafe bunks ad libitum. Although five horses were used in the group-housed (COMP) pen to more closely mimic a true group environment, only data from horses that experienced both housing systems (n = 3 mares and n = 3 geldings) were used for statistical analyses. Hourly (P = 0.008) and daily (P = 0.003) durations of hay feeding were higher for NOCOMP compared to COMP horses, and total daily intake (g DM/kg BW) of NOCOMP horses tended to be greater (P = 0.09) than COMP horses. Conversely, eating rate (g DM/kg BW/min) was greater (P = 0.04) for COMP compared to NOCOMP mares but was unaffected by housing in geldings. The GrowSafe system may provide an opportunity for efficient and effective monitoring of individual horse feed intake and feeding behavior in group-housing situations in horses.
