RESUMEN

Metal-organic framework composites have the advantages of large surface area, high porosity, strong catalytic efficiency and good stability, which provide a great possibility of finding excellent electrode materials for electrochemical sensors. However, MOF composites still face various challenges and difficulties, which limit their development and application. This paper reviews the application of MOF composites in electrochemical sensors, including MOF/carbon composites, MOF/metal nanoparticle composites, MOF/metal oxide composites and MOF/enzyme composites. In addition, the application challenges of MOF composites in electrochemical sensors are summarized. Finally, the application prospect for MOF composites is considered to promote the synthesis of more MOF composites with excellent properties.

RESUMEN

Acute influenza infection has been reported to be associated with neurological symptoms such as influenza-associated encephalopathy (IAE). Although the pathophysiology of this condition remain unclear, neuroinflammation and associated alterations in the central nervous system (CNS) are usually induced. Microglia (MGs), CNS-resident macrophages, are generally the first cells to be activated in response to brain infection or damage. We performed reverse transcriptase droplet digital PCR (RT-ddPCR) and luminex assays to investigate virus proliferation and immune reactions in BV2 MGs infected with influenza A(H1N1)pdm09 virus. Furthermore, isobaric tags for relative and absolute quantitation (iTRAQ)-based quantitative proteomics methods were used to investigate the dynamic change in the protein expression profile in BV2 MGs to gain insight into the CNS response to influenza A (H1N1) pdm09 infection. Our results showed that the influenza A(H1N1)pdm09 virus was replicative and productive in BV2 MG cells, which produced cytokines such as interleukin (IL)-1ß, IL-6, tumour necrosis factor (TNF)-α and monocyte chemoattractant protein (MCP)-1. The expression of osteopontin (OPN) in the influenza A (H1N1) pdm09-infected BV2 MGs was upregulated at 16 and 32 h post-infection (hpi) compared to that in the control group, resulting in aggravated brain damage and inflammation. Our study indicates that OPN signalling might provide new insights into the treatment of CNS injury and neurodegenerative diseases in IAE.

RESUMEN

Premature ventricular contraction (PVC) is associated to the risk of ventricular dysfunction and cardiovascular events. Its diagnosis depends on a long-time monitoring, and computational tools for PVC recognition can provide significant assistance to specialists. For this purpose, we present an automatic algorithm for the recognition PVC beat based on long-term 12-lead ECG.A total of 249 patients with PVC were included in this study. Initially, a novel QRS onset detection function was used to automatically extract QRS complexes from massive original ECG data. Then, non-personalized but shared QRS-width features of 12-lead QRS complexes were extracted and fed to a binary classifier based on SVM. In order to verify the model, 17, 512 normal beats and 17, 690 PVC beats extracted from 35 patients were used for training, and another 215 normal beats and 291 PVC beats selected randomly from the remaining 214 patients were used for testing. As a result, the achieved accuracy, sensitivity, specificity in training data and testing data are 98.9%, 98.3%, 99.5% and 97.2%, 97.7%, 96.7%, respectively. The high accuracy of PVC recognition makes it promising to be an efficient technique being used in clinical settings to automatically analyze huge ECG data so as to replace the tedious manual interpretation.

RESUMEN

A new electrochemical sensor was fabricated for the sensitive and selective detection of acetaminophen (AP), based on a bare glassy carbon electrode (GCE) modified with poly caffeic acid (PCA) and Zn/Ni-ZIF-8-800 using an electro-polymerization method. The carbon material of Zn/Ni-ZIF-8-800 derived from bimetal-organic framework was synthesized by one-step pyrolysis method. Then, the as-prepared materials and the corresponding fabricated sensors were fully characterized by XRD, FTIR, BET, SEM, TEM and XPS analyses. In addition, the electron transfer abilities of the modified sensors were investigated by CV and EIS. Subsequently, the parameters of polymerization time, pH value and scan rate were systematically studied and optimized. Under the optimum conditions, the desirable linear relationships were obtained between the peak current and the AP concentration at the PCA@ Zn/Ni-ZIF-8-800/GCE sensor with a detection limit of 0.0291 µM. The outstanding electrochemical performance of the proposed sensor can be attributed to the good electrocatalytic activity of PCA, the large surface area and high conductivity of Zn/Ni-ZIF-8-800 as well as the synergistic effect of PCA and Zn/Ni-ZIF-8-800. Moreover, the fabricated sensor was also successfully applied for the determination of AP in pharmaceutical dosage forms and human urine sample, and satisfactory recoveries were obtained.

RESUMEN

A newly emerged H7N9 influenza virus poses high risk to human beings. However, the pathogenic mechanism of the virus remains unclear. The temporal response of primary human alveolar adenocarcinoma epithelial cells (A549) infected with H7N9 influenza virus and H1N1 influenza A virus (H1N1, pdm09) were evaluated using the proteomics approaches (2D-DIGE combined with MALDI-TOF-MS/MS) at 24, 48 and 72 hours post of the infection (hpi). There were 11, 12 and 33 proteins with significant different expressions (P<0.05) at 24, 48 and 72hpi, especially F-actin-capping protein subunit alpha-1 (CAPZA1), Ornithine aminotransferase (OAT), Poly(rC)-binding protein 1 (PCBP1), Eukaryotic translation initiation factor 5A-1 (EIF5A) and Platelet-activating factor acetylhydrolaseⅠb subunit beta (PAFAH1B2) were validated by western-blot analysis. The functional analysis revealed that the differential proteins in A549 cells involved in regulating cytopathic effect. Among them, the down-regulation of CAPZA1, OAT, PCBP1, EIF5A are related to the death of cells infected by H7N9 influenza virus. This is the first time show that the down-regulation of PAFAH1B2 is related to the later clinical symptoms of patients infected by H7N9 influenza virus. These findings may improve our understanding of pathogenic mechanism of H7N9 influenza virus in proteomics.

RESUMEN

To explore the mechanisms of influenza H7N9 virus pathogenesis, influenza H7N9 virus and H1N1 influenza A virus(H1N1pdm09)-infected A549 cellular models were established, and differential protein expression in A549 cells infected with the two strains were investigated.A549 cells were infected with H7N9 and H1N1pdm09influenza virus at a multiplicity of infection(MOI)of 0.001.The temporal response of A549 cells infected with the two strains was evaluated using the proteomics approaches(2DDIGE combined with MALDI-TOF-MS/MS)at 24,48 and 72hours post infection(hpi). There were 11,12 and 33proteins with significantly different expression at 24,48 and 72hpi,respectively.Compared with H1N1pdm09 infection, functional analysis revealed that the down-regulation of proteins in H7N9 infection including F-actin-capping protein subunit alpha-1(CapZ-α1), ornithine aminotransferase(OAT),poly(rC)-binding protein 1(PCBP1)and eukaryotic translation initiation factor 5A-1(eIF5A)produced cytopathic effects. The down-regulation of platelet-activating factor acetylhydrolaseIb subunit beta(PAFAH1B2)in H7N9-infection may be related to the clinical symptoms of patients infected by the influenza H7N9 virus.
