Broiler breeder chickens are commercially feed restricted to slow their growth and improve their health and production, however, there is research demonstrating that this leads to chronic hunger resulting in poor welfare. A challenge in these studies is to account for possible daily rhythms or the effects of time since last meal on measures relating hunger. To address this, we used 3 feed treatments: AL (ad libitum fed), Ram (restricted, fed in the morning), and Rpm (restricted, fed in the afternoon) to control for diurnal effects. We then conducted foraging motivation tests and collected home pen behavior and physiological samples at 4 times relative to feeding throughout a 24-h period. The feed treatment had the largest influence on the data, with AL birds weighing more, having lower concentrations of plasma NEFA, and mRNA expression of AGRP and NPY alongside higher expression of POMC in the basal hypothalamus than Ram or Rpm birds (P < 0.001). R birds were more successful at and had a shorter latency to complete the motivation test, and did more walking and less feeding than AL birds in the home pen (P < 0.01). There was little effect of time since last meal on many measures (P > 0.05) but AGRP expression was highest in the basal hypothalamus shortly after a meal (P < 0.05), blood plasma NEFA was higher in R birds just before feeding (P < 0.001) and glucose was higher in Ram birds just after feeding (P < 0.001), and the latency to complete the motivation test was shortest before the next meal (P < 0.05). Time of day effects were mainly found in the difference in activity levels in the home pen when during lights on and lights off periods. In conclusion, many behavioral and physiological hunger measures were not significantly influenced by time of day or time since the last meal. For the measures that do change, future studies should be designed so that sampling is balanced in such a way as to minimize bias due to these effects.

Animal welfare assessments are essential for the identification of welfare hazards and benchmarking of welfare improvements, though welfare assessments for zoo species are lacking. Bears are commonly housed in zoos but currently no composite welfare assessment tool exists for captive bears. This study describes the development of such a tool for use across hibernating bear species. A draft tool was developed using indicators derived from the literature and a modified Delphi analysis with an international group of bear keepers. A total of 18 bear keepers from 12 zoos were recruited to trial the tool on 24 brown bears and American black bears. The participating keepers assessed their bears three times across a period of nine days. Intraclass correlation coefficients analysis was used to analyse inter-, intra-rater and item reliability. The inter- and intra-rater reliability showed good to excellent levels of agreement (>0.7, p < 0.05). Item reliability was also assessed and showed good to excellent levels of agreement (>0.75, p < 0.05). The resulting bear welfare assessment is an important step in identifying and understanding challenges to bear welfare in captivity.

Broilers have been bred for fast growth which has led to welfare problems such as high mortality, lameness and skin lesions. Slower growing breeds are thought to have better welfare but are not as efficient in production. This study investigated welfare, behaviour, production and meat quality of faster growing broilers from three main commercial broiler companies (breeds FA, FB and FC) with a commercially available slower growing breed (Hubbard JA757, S). Four hundred birds of each breed (total 1600 birds) were reared in pens of 50 birds, 8 per breed (total 32 pens). Home pen behaviour was recorded once a week in hourly scan samples to get behavioural time budgets. Welfare Assessments (WA) were done when the average bird weight per breed was 2.2 and 2.5kg. Birds and feed supplied were regularly weighed by pen and Feed Conversion Ratio (FCR) and Average Daily Gain (ADG) were calculated at 2.2kg. Birds were then slaughtered and meat quality measures were taken. S and FC had lower mortality and culls due to lameness (P<0.05 for both). Breeds FA, FB and FC grew faster, ate less feed and had better FCR and ADG (P<0.05 for all). S had scores indicating higher welfare for the majority of WA measures and spent more time active and less time sitting, feeding and drinking than the other breeds (P<0.05 for all). Faster growing breeds had more breast meat and S had more leg meat; although S had better meat quality scores (P<0.05). Overall, S birds have improved welfare in terms of activity and welfare measure scores compared to the other breeds but take longer to reach slaughter weight and are not as efficient in production measures. However if lower mortality and improved meat quality are taken into account, as well as the premium price paid for these birds, slower growing broilers may be a viable commercial option.

There is a need for novel mechanical devices for dispatching poultry on farm following the introduction of EU Regulation (EC) no. 1099/2009 On the Protection of Animals at the Time of Killing. We examined three novel mechanical killing devices: Modified Armadillo, Modified Rabbit Zinger, a novel mechanical cervical dislocation device; and traditional manual cervical dislocation. The four killing methods were tested on 230 chickens across four batches. We measured behavioural, electroencephalogram and post-mortem outcomes in anesthetized laying hens and broilers at two life stages (juveniles and adults/slaughter age). Graeco Latin-Square designs systematically randomized killing treatment, bird type, age and kill order. All birds were lightly anaesthetized immediately prior to the killing treatment with inhalation of Sevoflurane. The novel mechanical cervical dislocation method had the highest kill success rate (single application attempt only, with no signs of recovery) of a mechanical method (96%). The Modified Armadillo was the least reliable with 49% kill success. Spectral analysis of electroencephalogram signals at 2 s intervals for successfully killed birds only revealed progressive decreases in median frequency alongside increases in total power. Later, total power decreased as the birds exhibited isoelectric electroencephalogram signal. Latencies to pre-defined spectral ranges associated with unconsciousness showed that birds subjected to manual and novel mechanical cervical dislocation achieved these states sooner than birds subjected to the modified Armadillo. Nevertheless all methods exhibited short latencies (<4 s). The Modified Rabbit Zinger had the shortest duration of reflex persistence for nictitating membrane, pupillary and rhythmic breathing post method application. Of the methods tested, the novel mechanical cervical dislocation device is the most promising mechanical method for killing poultry on-farm based on a range of behavioural, electroencephalogram and anatomical parameters. This device has the potential to fulfil the current need for a mechanical alternative to manual cervical dislocation.

PP-fold peptides such as peptide YY (PYY) and pancreatic polypeptide (PPY) are known to play key roles in vertebrate energy homeostasis. Until recently, no gene sequence was available for avian PYY and therefore a gap in knowledge of regulation of its expression exists in avian species. Here we further evidence the mRNA sequence for chicken PYY and show that the pancreas is the major site of its mRNA expression, with a secondary peak of expression around the distal jejunum, in contrast to mammals where the large intestine is the major site of PYY expression. We also demonstrate that pancreatic PYY expression is responsive to short-term and long-term nutritional state, increasing within hours of feeding, in contrast to intestinal PYY which does not fluctuate to the same extent, and pancreatic PPY which appears to be primarily determined by long-term energy state. Both pancreatic PYY and PPY expression were found to exhibit ontogeny, being evenly distributed throughout the pancreas in young (2wk) chicks but having a decreasing splenic to duodenal gradient by adolescence (12wk).

Broiler breeders (parents of meat chickens) are selected for fast growth and become obese if fed ad libitum. To avoid this and maintain good health and reproductive ability, they are feed restricted to about 1/3 of what they would eat ad libitum. As a result, they experience chronic hunger and exhibit abnormal behaviour patterns that may indicate stress and frustration. One approach to measuring hunger is to observe how much birds will work, such as pecking a key, for access to more or different types of food. However, the sight, smell, and feedback from consumption of the feed reward changes the context and may artificially raise feeding motivation. To avoid this, we tested broiler breeders in an apparatus in which they could work for access to a wooden platform covered in wood shavings by crossing a water runway which increased in length and depth in 8 successive tests. In the wood shavings area, they could perform exploratory and foraging behaviour (the appetitive phase of feeding) but were never rewarded with feed. Sixty birds were divided into three feed quantity treatments: commercial restriction (R), and twice (2R) or three times (3R) this amount. Overall, birds fed R worked harder to reach the wood shavings area (reached it in a larger number of tests) than 2R and 3R birds (P<0.001). More restricted birds took less time to reach the area (P<0.001, R<2R<3R) and spent more time foraging while there (P<0.001, R>2R>3R). This indicates that restricted-fed birds were hungry and willing to work for the opportunity to forage even though food was never provided, suggesting that their motivation to perform the appetitive component of feeding behaviour (foraging/food searching) was sufficient to sustain their response. Thus food restriction in broiler breeders is a welfare concern. However these methods could be used to test alternative feeding regimes to attempt to find ways of alleviating hunger while still maintaining healthy growth and reproduction in these birds.

Feather pecking, commonly found in flocks of laying hens (Gallus gallus), is detrimental to bird welfare. Thought to cause this problem is the normal housing of layers without a floor substrate. Some evidence suggests that early substrate access decreases later feather pecking. However, there has been little research on the immediate effects of a change in substrate availability on bird welfare, although environmental modifications like this are often done when brooding and rearing laying hen chicks. To investigate this, the behavior of two strains of laying hen chicks was recorded for 4 weeks. The study kept the birds on either wire or peat moss for 14 days and then switched half the chicks to the other flooring. Early feather pecking was not significantly different for birds started on peat moss and switched to wire than for birds only on wire (p > .05). Because moving chicks from peat moss to wire did not cause additional welfare problems, the study recommends that chicks be kept on a substrate when young as feather-pecking levels are lower and immediate welfare is improved compared with birds kept only on wire.
