RESUMEN

The objective of this research was to analyze the miR-145 function in thyroid papillary carcinoma cells and explore its possible mechanism. For this purpose, the TPC-1 cell line was selected, miR-145 overexpression and rab5c shRNA lentiviral vector were constructed, and transfected into PTC cells. Luciferase reporter gene was performed to determine the relationship between miR-145 and rab5c, Western blot and qPCR were performed to detach the expression of the related genes, CCK-8 cell proliferation assay and Transwell cell invasion assay were used to determine the proliferation and invasion ability of PTC-1 cells. Results showed that MiR-145 overexpression inhibited the wt-rab5c (wild-type rab5c)luciferase activity, decreased the expression of rab5c mRNA and protein levels in the TPC-1 cell line, inhibited the proliferation and invasion of PTC cell line TPC-1(P < 0.05). In TPC-1 cells, both miR-145 overexpression and RNA interference with rab5c could increase the expression of the p-ERK protein (P < 0.05). In conclusion, MiR-145 inhibits the proliferation and invasion of PTC cells by downregulating rab5c and activating MAPK/ERK pathway in vitro.

Atherosclerosis (AS) often occurs in cardiovascular disease, which is a chronic vascular disease and is harmful to human health. Oxidative stress is involved in its etiology. This study aimed to determine the effectiveness of Isoflavones from semen sojae preparatum (ISSP) in inhibiting oxidative stress and its important molecular mechanisms through in vivo and in vitro experiments. ApoE-/- mice were used to establish atherosclerosis models through a high-fat diet, and endothelial cells were used to establish oxidative stress injury models through ox-LDL induction. The degree of oxidative stress damage was assessed by detecting changes in ET-1, LDH, SOD, and MDA indicators. It was observed that after ISSP treatment, the oxidative stress damage of mice and endothelial cells was improved. The Nrf2/AER signaling pathway is an important antioxidant pathway that has attracted our attention. Western blotting and qRT-PCR were used to detect the expression of Nrf2, HO-1, and NQO1 in mice aortae and endothelial cells. The results showed that the Nrf2 signaling pathway was activated after ISSP intervention. In addition, in this study, after preantagonizing the estrogen receptors GPR30 and ERß, it was observed that the effects of ISSP in treating endothelial cell oxidative damage and activating the Nrf2 signaling pathway were weakened. After silencing Nrf2 by Nrf2-siRNA transfection, the effect of ISSP in treating endothelial cell oxidative damage was inhibited. This study shows that ISSP may reduce oxidative stress damage and atherosclerosis through the Nrf2 signaling pathway, and this effect may involve the GPR30 and ERß estrogen receptors.

BACKGROUND: Papillary thyroid carcinoma is the most frequent histological subtype of thyroid cancer with a high incidence. We aimed to explore the function of circular RNA_0039411 (circ_0039411) and its associated mechanism in papillary thyroid carcinoma progression. METHODS: Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and Western blot assay were conducted to determine the expression of RNA and protein, respectively. The colony formation ability, migration, invasion, and apoptosis were analyzed by colony formation assay, transwell migration assay, transwell invasion assay, and flow cytometry. Cell glycolytic metabolism was analyzed using fluorescence-based glucose assay kit and fluorescence-based lactate assay kit. Dual-luciferase reporter assay and RNA-Pull-Down Assay were performed to validate the binding between microRNA-423-5p (miR-423-5p) and circ_0039411 or SRY-box transcription factor 4 (SOX4). The xenograft tumor model was used to assess the role of circ_0039411 in the tumor growth in vivo. RESULTS: Circ_0039411 was highly expressed in papillary thyroid carcinoma tissues and cell lines compared with adjacent normal tissues and NTHY-ORI3.1 cells. Circ_0039411 interference suppressed the colony formation ability, migration, invasion, and glycolysis but promoted the apoptosis of papillary thyroid carcinoma cells. MiR-423-5p was a target of circ_0039411 in papillary thyroid carcinoma cells. Circ_0039411 knockdown-mediated effects in papillary thyroid carcinoma cells were largely overturned by the silence of miR-423-5p. MiR-423-5p bound to the 3' untranslated region (3'UTR) of SOX4. SOX4 overexpression largely reversed circ_0039411 silencing-mediated effects in papillary thyroid carcinoma cells. Circ_0039411 positively regulated SOX4 expression by sponging miR-423-5p in papillary thyroid carcinoma cells. Circ_0039411 silencing notably suppressed the growth of xenograft tumors in vivo. CONCLUSION: Circ_0039411 promoted the malignant behaviors of papillary thyroid carcinoma cells partly depending on the regulation of the miR-423-5p/SOX4 axis.

Investigation of effects of Meis homeobox 2 (MEIS2) on proliferation and apoptosis of thyroid cancer (TC) cells and its specific molecular mechanism is the main purpose of this study. In this study, we found that the expression of MEIS2 was down-regulated in TC tissues and cell lines (B-CPAP, TPC-1 and K1), compared to adjacent histologically normal tissues and normal thyroid cell (Nthy-ori 3-1). Then, overexpression of MEIS2 promoted B-CPAP cell apoptosis and decreased cell proliferation, viability and cell cycle progression. Further studies confirmed that overexpression of MEIS2 could significantly decrease p65 expression in the nucleus of B-CPAP cells. However, the opposite results were presented after interference of MEIS2 expression. Taken together, MEIS2 expression was significantly down-regulated in TC. In addition, MEIS2 could inhibit NF-κB pathway activation, so as to perform both suppression of the viability and proliferation of TC cells and promotion of apoptosis.

RESUMEN

A novel optical reverse mapping (ORM) method and an ORM criterion are proposed to evaluate the relevance between the directional backlight (DB) 3D light-field display system aberration and the crosstalk. Based on the ORM criterion, the space-division-multiplexed catadioptric integrated backlight (SCIB) and symmetrical triplet-compound lenticular array (triplet LA) are designed. The SCIB is composed of hybrid Fresnel integrated backlight unit (hybrid Fresnel unit) and space-division-multiplexed microprism unit (microprism unit). The hybrid Fresnel unit is used to provide the directional light, and the divergence angle is 2.4-degrees. The average uniformity of 83.02% is achieved. The microprism unit is used to modulate the directional light distribution into three predetermined directions to establish a 90-degree viewing area. Combined with SCIB, the triplet LA is used to suppress the aberrations and reduce the crosstalk. In the experiment, a DB 3D light-field display system based on SCIB and triplet LA is set up. The displayed light-field 3D image can be observed in a 90-degree viewing angle. Compared to the conventional DB 3D display system, the light-field 3D image is aberration-suppressed, and the SSIM values are improved from 0.8462 to 0.9618. Meanwhile, the crosstalk measurement results show that the average crosstalk is 3.49%. The minimum crosstalk is 2.31% and the maximum crosstalk is 4.52%. The crosstalk values in 90-degree are lower than 5%.

A novel catadioptric retroreflector floating device (CRA) used in the 3D floating light-field system is proposed. The floating light-field image constructed by the CRA is aberration-suppressed. The luminance and the contrast of the image are substantially improved in a 90-degree viewing angle. The CRA is constituted of the designed catadioptric retroreflector (CR). The CR consists of three lenses, the first and the second lens is to refract the light, and the rear surface of the third lens is coated with reflective coating in order to reflect the incident light. The CRA is processable and the fabrication process using UV embossing is also described. A spectrophotometer is utilized to measure the retroreflective efficiency of the CRA. The average retroreflective efficiency of the CRA is 80.1%. A beam quality analyzer is utilized to measure the beam spot quality of the CRA, and the image quality can satisfy the requirements of human eye observation. In the experiment, compared to the floating light-field image constructed by the micro-beads type retroreflector floating device (MRA), the image quality of the floating light-field image constructed by the CRA is significantly enhanced. In the quantitative computer simulation, the PSNR values of the images are increased from 23.0185 to 32.1958.

Due to lack of the accommodation stimulus, an inherent drawback for the conventional glasses-free stereoscopic display is that precise depth cues for the human monocular vision is rent, which results in the well-known convergence-accommodation conflict for the human visual system. Here, a super multi-view light field display with the vertically-collimated programmable directional backlight (VC-PDB) and the light control module (LCM) is demonstrated. The VC-PDB and the LCM are used to form the super multi-view light field display with low crosstalk, which can provide precisely detectable accommodation depth for human monocular vision. Meanwhile, the VC-PDB cooperates with the refreshable liquid-crystal display panel to provide the convergence depth matching the accommodation depth. In addition, the proposed method of light field pick-up and reconstruction is implemented to ensure the perceived three dimensional (3D) images with accurate depth cues and correct geometric occlusion, and the eye tracker is used to enlarge the viewing angle of 3D images with smooth motion parallax. In the experiments, the reconstructed high quality fatigue-free 3D images can be perceived with the clear focus depth of 13 cm in the viewing angle of ± 20°, where 352 viewpoints with the viewpoint density of 1 mm-1 and the crosstalk of less than 6% are presented.

The light field display can provide vivid and natural 3D performance, which can find many applications, such as relics research and exhibition. However, current light field displays are constrained by the viewing angle, which cannot meet the expectations. With three groups directional backlights and a fast-switching LCD panel, a time-multiplexed light field display with a 120-degree wide viewing angle is demonstrated. Up to 192 views are constructed within the viewing range to ensure the right geometric occlusion and smooth parallax motion. Clear 3D images can be perceived at the entire range of viewing angle. Additionally, the designed holographic functional screen is used to recompose the light distribution and the compound aspheric lens array is optimized to balance the aberrations and improve the 3D display quality. Experimental results verify that the proposed light field display has the capability to present realistic 3D images of historical relics in 120-degree wide viewing angle.

OBJECTIVE: To assess the accuracy of a wrist-worn device (Watch-PAT 200) in the diagnosis of obstructive sleep apnea syndrome (OSAHS). METHODS: Forty-three adult subjects with suspected OSAHS simultaneously had a standard in-laboratory polysomnogram (PSG) and wore the Watch-PAT 200 during a full-night recording. PSG sleep and respiratory events were scored according to standard criteria. The PSG recordings were blindly manually analyzed, while Watch-PAT data were scored automatically based on the algorithm developed previously. RESULTS: The mean age of the subjects was (42.2 ± 12.2) years (x(-) ± s), and mean body mass index was (28.0 ± 3.9) kg/m(2). Mean PSG apnea hypopnea index (AHI) was (34.9 ± 29.9) events per hour, and mean PAT-AHI was (36.0 ± 29.2) events per hour. There was a significant correlation between PAT AHI and AHI by PSG (r = 0.931, P < 0.01). A Bland-Altman plot of PAT AHI and PSG AHI was also used to assess the accuracy of Watch-PAT 200. At lower levels of AHI, PAT tended to overestimate disease severity, while at higher levels of AHI, Watch-PAT underestimated severity. To assess sensitivity and specificity of Watch-PAT, constructed receiver operator characteristic curves using a variety of AHI threshold values (5, 15 and 30 events per hour). For AHI ≥ 5 events per hour as threshold value, the Watch-PAT diagnosing rate was 93%, and sensitivity as well as specificity were 94.7% and 80.0%. The misdiagnosis rate and missed diagnosis rate were 20.0% and 5.3%. Optimal combinations of sensitivity and specificity for the AHI threshold values (15 and 30 events per hour) were 82.6% and 100.0%, 95.0% and 95.7% respectively. CONCLUSION: The Watch-PAT 200 may offer an accurate, robust, and reliable ambulatory method for the detection of OSAHS, with minimal patient discomfort.
