RESUMEN

Fipronil is a persistent insecticide known to transfer into hen eggs from exposure from animal drinking water and feed, but some questions remain regarding its transfer behavior and distribution characteristics. Therefore, the dynamic metabolism, residue distribution and transfer factor (TF) of fipronil were investigated in 11 edible tissues of laying hens and eggs over 21 days. After a continuous low-dose drinking water exposure scenario, the sum of fipronil and all its metabolites (defined as fipronilT) quickly transferred to each edible tissue and gradually increased with exposure time. FipronilT residue in eggs first appeared at 3 days and then gradually increased. After a single high-dose feed exposure scenario, fipronilT residue in edible tissues first appeared after 2 h, quickly peaked at 1 day, and then gradually decreased. In eggs, fipronilT residue first appeared at 2 days, peaked 6-7 days and then gradually decreased. The TF values followed the order of the skin (0.30-0.73) > egg yolk (0.30-0.71) > bottom (0.21-0.59) after drinking water exposure, and the order of the skin (1.01-1.59) > bottom (0.75-1.1) > egg yolk (0.58-1.10) for feed exposure. Fipronil sulfone, a more toxic compound, was the predominant metabolite with higher levels distributed in the skin and bottom for both exposure pathways. FipronilT was distributed in egg yolks rather than in albumen owing to its lipophilicity, and the ratio of egg yolk to albumen may potentially reflect the time of exposure. The distinction is that the residues after feed exposure were much higher than that after drinking water exposure in edible tissues and eggs. The study highlights the residual characteristics of two exposure pathways, which would contribute to the tracing of contamination sources and risk assessment.

Currently, the pharmaceutical and personal care products (PPCPs) have posed great challenge to advanced oxidation techniques (AOTs). In this study, we decorated sponge iron (s-Fe0) with Cu and Pd (s-Fe0-Cu-Pd) and further optimized the synthesis parameters with a response surface method (RSM) to rapidly degrade diclofenac sodium (DCF). Under the RSM-optimized conditions of Fe: Cu: Pd = 100: 4.23: 0.10, initial solution pH of 5.13, and input dosage of 38.8 g/L, 99% removal of DCF could be obtained after 60 min of reaction. Moreover, the morphological structure of trimetal was characterized with high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM), X-ray diffraction (XRD), X-ray photoelectron spectra (XPS). Electron spin resonance (ESR) signals have also been applied to capture reactive hydrogen atoms (H*), superoxygen anions, hydroxyl radicals, and single state oxygen (1O2). Furthermore, the variations of DCF and its selective degradation products over a series of s-Fe0-based bi(tri)metals have been compared. Additionally, the degradation mechanism of DCF has also been explored. To our best knowledge, this is the first report revealing the selective dechlorination of DCF with low toxicity over Pd-Cu co-doped s-Fe0 trimetal.

Aeromonas hydrophila infections are common in aquaculture. Our previous studies found that the A. hydrophila B11 strain can survive in fish macrophages for at least 24 h and the two-component system EnvZ/OmpR may be involved in intracellular survival. To reveal the role and mechanism of the two-component system EnvZ/OmpR in intracellular survival of A. hydrophila, the genes of envZ/ompR were silenced by shRNAi. The results showed that the survival rates of the envZ-RNAi and ompR-RNAi strains were only 2.05% and 3.75%, respectively, which were decreased by 91% and 83.6% compared with that of the wild-type strain. The escape ability of envZ-RNAi and ompR-RNAi was also decreased by 51.4% and 19.7%, respectively. The comparative transcriptome analysis revealed that the functional genes directly related to bacterial intracellular survival mainly included the genes related to anti-stress capacity, and the genes related to Zn2+ and Mg2+ transport. Further research confirmed that two-component system EnvZ/OmpR can regulate the expression of the important molecular chaperones, such as groEL, htpG, dnaK, clpB and grpE. The expression of these molecular chaperones in wild-type strain was up-regulated with the increase in H2 O2 concentrations, while the expression of these molecular chaperones in silent strains did not change significantly. Cells that phagocytosed wild-type strain had higher ROS content than cells that phagocytosed silent strains. Two-component system EnvZ/OmpR could also regulate zinc transporter (znuA, znuB, znuC) and zinc efflux protein (zntA) to maintain zinc homeostasis in cells, thus affecting the ability of bacteria to survive in phagocytes. Moreover, two-component system EnvZ/OmpR could affect the growth and intracellular survival of A. hydrophila by regulating the expression of MgtA, MgtC and MgtE and participating in bacterial Mg2+ homeostasis in fish macrophages.

The surface properties of the adsorbents and the acidic environment have an influence effect on Pb adsorption. In order to further improve the adsorption performance of biochar, we herein reported an effective method to synthesize high-adsorbed biochar by co-doping with nitrogen and phosphorus. After atom doping, the N/P co-doped biochar (NP-BC) showed the enhanced adsorption capacity for lead ion (Pb2+). The adsorption kinetics, isotherm, pH value, and influencing factors were studied. The results show that the synthesized NP-BC has high Pb2+ adsorption capacity in aqueous solution, and can be maintained with various environmental interference factors including pH, natural organic matter, and other metal ions. High adsorption performance shows that the material may be well used to remove Pb2+ in various water bodies. Various characterization experiments prove that surface properties contribute to Pb2+ adsorption, and the high performance of NP-BC is mainly due to the surface complexation between functional groups and Pb2+. This work demonstrates that the surface functional groups of biochar are critical to the development of high-performance heavy metal adsorbents.

Abstract Fluoroquinolones are an important class of antimicrobial agents to manage infectious diseases. However, knowledge about how host bile acids are modified by fluoroquinolones is limited. We investigated and compared the impact of fluoroquinolones on circulating bile acid profiles and gut microbiota from in vivo studies. We administered ciprofloxacin (100 mg/kg/day) or moxifloxacin (40 mg/kg/day) orally to male Wistar rats for seven days. Fifteen bile acids (BAs) from the serum and large intestine were quantified by HPLC-MS/MS. The diversity of gut microbiota after ciprofloxacin and moxifloxacin treatment was analyzed using high-throughput, next-generation sequencing technology. The two fluoroquinolone-treated groups had different BA profiles. Ciprofloxacin significantly reduced the hydrophobicity index of the BA pool, reduced secondary BAs, and increased taurine-conjugated primary BAs in both the serum and large intestine as compared with moxifloxacin. Besides, ciprofloxacin treatment altered intestinal microbiota with a remarkable increase in Firmicutes to Bacteroidetes ratio, while moxifloxacin exerted no effect. What we found suggests that different fluoroquinolones have a distinct effect on the host BAs metabolism and intestinal bacteria, and therefore provide guidance on the selection of fluoroquinolones to treat infectious diseases.

The potential effects of extracellular polymeric substances (EPS) on the behavior and toxicity of silver nanoparticle (Ag-NPs) and silver sulfide nanoparticle (Ag2S-NPs) remains ambiguous. The interaction of EPS from Bacillus subtilis with Ag2S-NPs, metallic Ag-NPs, or ionic Ag, and the associated plant safety had been examined in this study. The biological impacts of Ag-NPs and Ag2S-NPs were Ag form-dependent and highly influenced by microbial EPS. Compared with metallic Ag-NPs, Ag2S-NPs exert inert biological impacts, as revealed by 3.44 times lower Ag bioaccumulation in wheat (Triticum aestivum L.) seedlings and nearly reduce plant biomass when wheat was subjected to 1.0 mg-Ag L-1 of Ag-NPs and Ag2S-NPs with the transfer factors of 151.56-930.87 vs. 12.52-131.81, respectively. These observations were coincident with the low dissolved Ag ([Ag]diss) in the Ag2S-NPs treatment than the Ag-NPs treatment (114.0 vs. 0.0791, µg L-1). Compared with the enhanced toxicity of Ag2S-NPs to wheat, Bacillus subtilis EPS significantly alleviate the phytotoxicity of Ag-NPs, as revealed by the relative root elongation (7.15-45.40% decrease vs. 2.39-11.75% increase), and malondialdehyde (1.47-83.22% increase vs. 8.57-25.25% decrease) and H2O2 (11.27-71.78% increase vs. 5.16-36.67% decrease) contents. These constrasting plant responses of B. subtilis EPS are mainly caused by their complexation property with toxic Ag+ and nutrient elements for wheat stressed by Ag-NPs and Ag2S-NPs, respectively. Our findings highlight the importance of rhizospheric EPS in affecting the biogeochemistry and ecotoxicity of metal nanoparticles including Ag-NPs and Ag2S-NPs in agricultural systems.

Dissolved black carbon (DBC) is ubiquitous in aquatic systems, being an important subgroup of the dissolved organic matter (DOM) pool. Nevertheless, its aquatic photoactivity remains largely unknown. In this study, a range of spectroscopic indices of DBC and humic substance (HS) samples were determined using UV-Vis spectroscopy, fluorescence spectroscopy, and proton nuclear magnetic resonance. DBC can be readily differentiated from HS using spectroscopic indices. It has lower average molecular weight, but higher aromaticity and lignin content. The apparent singlet oxygen quantum yield (Φsinglet oxygen) of DBC under simulated sunlight varies from 3.46% to 6.13%, significantly higher than HS, 1.26%-3.57%, suggesting that DBC is the more photoactive component in the DOM pool. Despite drastically different formation processes and structural properties, the Φsinglet oxygen of DBC and HS can be well predicted by the same simple linear regression models using optical indices including spectral slope coefficient (S275-295) and absorbance ratio (E2/E3) which are proxies for the abundance of singlet oxygen sensitizers and for the significance of intramolecular charge transfer interactions. The regression models can be potentially used to assess the photoactivity of DOM at large scales with in situ water spectrophotometry or satellite remote sensing.

The transcription factor, interferon regulatory factor 4 (IRF4), serves an essential role in the regulation of immune responses, and has been reported to act as a diagnostic and prognostic marker for various hematological malignancies. The present study aimed to investigate whether IRF4 could exert effects on human non­small cell lung cancer (NSCLC) and to explore the underlying mechanism. The mRNA and protein expression of IRF4 was detected in NSCLC tissues using reverse­transcription quantitative polymerase chain reaction and western blotting, respectively. In the in vitro experiment, IRF4 expression was knocked down or overexpressed using lentivirus in human lung adenocarcinoma A549 and lung squamous cell carcinoma LC­AI cell lines. Cell proliferation and colony number were analyzed using MTT and colony formation assays, respectively. The expression levels of IRF4 mRNA and protein were significantly higher in NSCLC tissues (n=54) compared with that in adjacent non­tumor tissues. Similarly, the expression levels of Notch1 and Notch2 mRNA were significantly higher in NSCLC tissues. Furthermore, the expression level of IRF4 mRNA was positively correlated with the levels of Notch1 and Notch2 mRNA in NSCLC tissues. Consequently, using NSCLC cell lines, it was demonstrated that the knockdown of IRF4 expression significantly reduced the cell proliferation rate and colony formation, whereas IRF4­overexpression significantly increased them. Notably, the IRF4 knockdown significantly decreased the expression levels of Notch1 and Notch2 mRNA, and phosphorylated protein kinase B (AKT), whereas IRF4 overexpression resulted in the opposite. The results of the present study indicate that IRF4 is overexpressed and serves as a tumor promoter in human NSCLC, at least partially, through activating the Notch­Akt signaling pathway.

Autophagy plays a complicated role in tumorigenesis in a variety of cancers. Recently, many studies report that some autophagy-related markers could be detected in several types of human tumors. However, fewer studies have been conducted to evaluate the relationship between autophagy and lung cancer, especially in non-small cell lung cancer (NSCLC). In this study, the expression levels of autophagy-related markers Beclin 1 and p62 were detected by Western blot analysis and cell immunofluorescence staining in three human NSCLC cell lines A549, H1299 and HCC827. Then, tissue microarray and immunohistochemical staining were used to determine Beclin 1 and p62 expression in 104 NSCLC specimens collected from patients. Beclin 1 and p62 were observed to primarily distribute in the cytoplasm of the cells. Beclin 1 was expressed more predominantly in male and heavy-smoker and its expression was significantly correlated with the differentiation and lymph node metastasis. p62 expression was negatively correlated with TNM stage and lymph node metastasis. Univariate Cox regression analysis revealed that low expression of Beclin 1 and high expression of p62 were significantly associated with shorter survival. Meanwhile, multivariate Cox regression analysis indicated that Beclin 1 and p62 were independent risk factors related to overall survival for patients with NSCLC. Collectively, our study suggests that Beclin 1 and p62 could serve as potential indicators for the prognosis of patients with NSCLC.

AIMS: Pleiotrophin (PTN), an angiogenic factor, is associated with various types of cancer, including lung cancer. Our aim was to investigate the possibility of using serum PTN as an early indicator regarding disease diagnosis, classification and prognosis, for patients with non-small cell lung cancer (NSCLC). METHODS: Significant differences among PTN levels in patients with small cell lung cancer (SCLC, n=40), NSCLC (n=136), and control subjects with benign pulmonary lesions (n=21), as well as patients with different pathological subtypes of NSCLC were observed. RESULTS: A serum level of PTN of 300.1 ng/ml, was determined as the cutoff value differentiating lung cancer patients and controls, with a sensitivity and specificity of 78.4% and 66.7%, respectively. Negative correlations between serum PTN level and pathological differentiation level, stage, and survival time were observed in our cohort of patients with NSCLC. In addition, specific elevation of PTN levels in pulmonary tissue in and around NSCLC lesions in comparison to normal pulmonary tissue obtained from the same subjects was also observed (n=2). CONCLUSION: This study suggests that the serum PTN level of patients with NSCLC could be an early indicator for diagnosis and prognosis. This conclusion should be further assessed in randomized clinical trials.

PURPOSE: The aim was to compare the efficacy and toxicity of gemcitabine plus docetaxel (GD) with platinum-based regimens in patients with untreated advanced non-small cell lung cancer (NSCLC). METHODS: We searched PubMed, Embase and the Cochrane Central Register of Controlled Trials databases for relevant trials. Reference lists of original articles and review articles were also examined. Abstracts presented at the ASCO and ESMO meetings were also searched. Studies were evaluated for eligibility and quality, and then, the data were extracted and analyzed. Statistical analyses were conducted by using RevMan 5.1. The primary end point was overall survival (OS). Secondary end points included 1-year survival, time to progression (TTP), overall response rate (ORR) and grade 3­4 toxicity. RESULTS: Nine randomized controlled trials were identified ultimately. The meta-analysis demonstrated that the survival between GD and platinum-based regimens was comparable according to the pooled HR for overall survival (1.04, 95% CI = 0.96­1.12, p = 0.39) and RR for one-year survival (0.94, 95% CI = 0.84­1.06, p = 0.33). Platinum-based regimens had an advantage in TTP (HR = 1.12, 95% CI = 1.02­1.24, p = 0.02) and ORR (RR = 0.86, 95% CI = 0.74­0.99, p = 0.03). However, GD induced less grade 3­4 nausea/vomiting, anemia, neutropenia and febrile neutropenia (RR = 0.36, 95% CI = 0.15­0.86, p = 0.02; RR = 0.35, 95% CI = 0.23­0.53, p = 0.00; RR = 0.68, 95% CI = 0.52­0.88, p = 0.003; RR = 0.53, 95% CI = 0.34­0.82, p = 0.004, respectively). Grade 3­4 diarrhea, sensory neuropathy, fatigue and thrombocytopenia were comparable between the two groups. CONCLUSIONS: GD acquired similar survival with platinum-based regimens in first-line treatment of advanced NSCLC. Platinum-based regimens had an advantage in TTP and ORR with more grade 3­4 nausea/vomiting, anemia, neutropenia and febrile neutropenia compared with GD.
