RESUMEN

Toxoplasma gondii causes morbidity, mortality, and disseminates widely via cat sexual stages. Here, we find T. gondii ornithine aminotransferase (OAT) is conserved across phyla. We solve TgO/GABA-AT structures with bound inactivators at 1.55 Å and identify an inactivator selective for TgO/GABA-AT over human OAT and GABA-AT. However, abrogating TgO/GABA-AT genetically does not diminish replication, virulence, cyst-formation, or eliminate cat's oocyst shedding. Increased sporozoite/merozoite TgO/GABA-AT expression led to our study of a mutagenized clone with oocyst formation blocked, arresting after forming male and female gametes, with "Rosetta stone"-like mutations in genes expressed in merozoites. Mutations are similar to those in organisms from plants to mammals, causing defects in conception and zygote formation, affecting merozoite capacitation, pH/ionicity/sodium-GABA concentrations, drawing attention to cyclic AMP/PKA, and genes enhancing energy or substrate formation in TgO/GABA-AT-related-pathways. These candidates potentially influence merozoite's capacity to make gametes that fuse to become zygotes, thereby contaminating environments and causing disease.

Eimeria tenella is a major cause of caecal coccidiosis in commercial poultry chickens worldwide. Here, we report chromosomal scale assembly of Eimeria tenella strain APU2, a strain isolated from commercial broiler chickens in the U.S. We obtained 100× sequencing Oxford Nanopore Technology (ONT) and more than 800× Coverage of Illumina Next-Seq. We created the assembly using the hybrid approach implemented in MaSuRCA, achieving a contiguous 51.34 Mb chromosomal-scale scaffolding enabling identification of structural variations. The AUGUSTUS pipeline predicted 8060 genes, and BUSCO deemed the genomes 99% complete; 6278 (78%) genes were annotated with Pfam domains, and 1395 genes were assigned GO-terms. Comparing E. tenella strains (APU2, US isolate and Houghton, UK isolate) derived Houghton strain of E. tenella revealed 62,905 high stringency differences, of which 45,322 are single nucleotide polymorphisms (SNPs) (0.088%). The rate of transitions/transversions among the SNPs are 1.63 ts/tv. The strains possess conserved gene order but have profound sequence heterogeneity in a several chromosomal segments (chr 2, 11 and 15). Genic and intergenic variation in defined gene families was evaluated between the two strains to possibly identify sequences under selection. The average genic nucleotide diversity of 2.8 with average 2 kb gene length (0.145%) at genic level. We examined population structure using available E. tenella sequences in NCBI, revealing that the two E. tenella isolates from the U.S. (E. tenella APU2 and Wisconsin, "ERR296879") share a common maternal inheritance with the E. tenella Houghton. Our chromosomal level assembly promotes insight into Eimeria biology and evolution, hastening drug discovery and vaccine development.

Strains of Eimeria maxima, an enteric parasite of poultry, vary in virulence. Here, we performed microscopy and RNA sequencing on oocysts of strains APU-1 (which exhibits more virulence) and APU-2. Although each underwent parallel development, APU-1 initially approached maturation more slowly. Each strain sporulated by hour 36; their gene expression diverged somewhat thereafter. Candidate biomarkers of viability included 58 genes contributing at least 1000 Transcripts Per Million throughout sporulation, such as cation-transporting ATPases and zinc finger domain-containing proteins. Many genes resemble constitutively expressed genes also important to Eimeria acervulina. Throughout sporulation, the expression of only a few genes differed between strains; these included cyclophilin A, EF-1α, and surface antigens (SAGs). Mature and immature oocysts uniquely differentially express certain genes, such as an X-Pro dipeptidyl-peptidase domain-containing protein in immature oocysts and a profilin in mature oocysts. The immature oocysts of each strain expressed more phosphoserine aminotransferase and the mature oocysts expressed more SAGs and microneme proteins. These data illuminate processes influencing sporulation in Eimeria and related genera, such as Cyclospora, and identify biological processes which may differentiate them. Drivers of development and senescence may provide tools to assess the viability of oocysts, which would greatly benefit the poultry industry and food safety applications.

In a national survey of fresh, unfrozen, American pasture-raised lamb and pork, the prevalence of viable Toxoplasma gondii was determined in 1500 samples selected by random multistage sampling (750 pork, 750 lamb) obtained from 250 retail meat stores from 10 major geographic areas in the USA. Each sample consisted of a minimum of 500g of meat purchased from the retail meat case. To detect viable T. gondii, 50g meat samples of each of 1500 samples were bioassayed in mice. Viable T. gondii was isolated from 2 of 750 lamb samples (unweighted: 0.19%, 0.00-0.46%; weighted: 0.04%, 0.00-0.11%) and 1 of 750 pork samples (unweighted: 0.12%, 0.00-0.37%; weighted: 0.18%, 0.00-0.53%) samples. Overall, the prevalence of viable T. gondii in these retail meats was very low. Nevertheless, consumers, especially pregnant women, should be aware that they can acquire T. gondii infection from ingestion of undercooked meat. Cooking meat to an internal temperature of 66°C kills T. gondii.

Abstract Despite the global importance of the zoonotic parasite Toxoplasma gondii, little is known regarding its infection in the Democratic Republic of São Tomé and Príncipe (DRSTP). This is the first report of antibodies to T. gondii in cattle, sheep, goats and pigs from the DRSTP. Antibodies were assessed by the modified agglutination test (MAT), with a cut-off titer of 100 for cattle and 20 for sheep, goats and pigs. The present study revealed an overall seroprevalence of 55.8%; 27.1% in 48 cattle, 68.4% in 98 sheep, 70.1% in 97 goats and 43.7% in 103 pigs. The south geographical area for cattle, the central area for sheep, and adult age and living in the central region for goats were found to be risk factors for seropositivity to T. gondii. These results support the scenario of a considerable presence of sporulated oocysts as well as of infected intermediate hosts in the local environment. Consumption of raw or undercooked meat should be considered as an important potential source of infection for animals and humans in the DRSTP.


Resumo Apesar da importância global do parasita zoonótico Toxoplasma gondii, pouco se conhece sobre sua infecção na República Democrática de São Tomé e Príncipe (RDSTP). Esse é o primeiro relato de anticorpos para T. gondii em bovinos, ovinos, caprinos e suínos da RDSTP. Os anticorpos foram pesquisados pelo teste de aglutinação direta modificada (TADM), com um título de corte de 100 para bovinos e de 20 para ovinos, caprinos e suínos. O presente estudo revelou uma soroprevalência global de 55,8%: 27,1% em 48 bovinos, 68,4% em 98 ovinos, 70,1% em 97 caprinos e 43,7% em 103 suínos. A área geográfica sul para os bovinos, a área central para os ovinos, bem como a idade adulta e a região central para os caprinos foram considerados fatores de risco para soropositividade a T. gondii. Esses resultados suportam o cenário de uma considerável presença de oocistos esporulados, bem como de hospedeiros intermediários infectados no ambiente local. O consumo de carne crua ou mal passada deve ser considerado como uma importante fonte potencial de infecção para animais e seres humanos na RDSTP.

Abstract Toxoplasma gondii and Leishmania infantum are zoonotic protozoal parasites. Serum samples were obtained from 186 donkeys (Equus africanus asinus) from Portugal and assessed for antibodies to T. gondii by the modified agglutination test (MAT). For titration of antibodies to Leishmania spp. the direct agglutination test was used (DAT). Eleven donkeys were seropositive for T. gondii with titres of 20 (n = 7), 80 (n = 2), 640 (n = 1) and ≥ 2560 (n = 1). One donkey was seropositive for Leishmania spp. (titre of 800). Donkeys in Portugal are exposed to and can be infected with T. gondii and Leishmania spp.


Resumo Toxoplasma gondii e Leishmania infantum são protozoários parasitas com potencial zoonótico. Foram obtidas amostras de soro de 186 jumentos (Equus africanus asinus) e avaliadas para anticorpos anti-T. gondii pelo teste de aglutinação direta modificada (TADM), em Portugal. Para a titulação de anticorpos anti-Leishmania spp. foi usado o teste de aglutinação direta (TAD). Onze jumentos foram soropositivos para T. gondii com títulos de 20 (n = 7), 80 (n = 2), 640 (n = 1) e ≥ 2560 (n = 1). Um jumento foi soropositivo para Leishmania spp. (título de 800). Os jumentos em Portugal estão expostos e podem ser infectados com T. gondii e Leishmania spp.

Abstract Neospora spp. are intracellular protozoa with worldwide distribution and closely related to Toxoplasma gondii, which can infect a variety of mammals including horses. From September 2013 to June 2014, 185 horses from northern, central and southern parts of mainland Portugal were randomly sampled and tested for detection of immunoglobulin (Ig) G antibodies to Neospora spp. using an indirect multi-species enzyme-linked immunosorbent assay (ELISA) commercial test (ID Screen® Neospora caninum Indirect Multi-species; ID.vet Innovative Diagnostics, Grabels, France). Two horses (1.1%; CI: 0.1-3.8%), one male and one female, were found to be seropositive for Neospora spp. Both seropositive animals were horses housed indoors but with access to outdoors, used for leisure activities and were apparently healthy, with good body condition and with no alterations at physical examination. This was the first serologic survey of antibodies to Neospora spp. carried out in horses from Portugal.


Resumo Neospora spp. são protozoários intracelulares com distribuição mundial e estreitamente relacionados com Toxoplasma gondii, que podem infectar uma variedade de mamíferos, incluindo cavalos. De setembro de 2013 a junho de 2014, 185 cavalos de áreas do Norte, Centro e Sul de Portugal continental foram aleatoriamente amostrados e testados para a detecção de anticorpos imunoglobulinas (Ig) G anti-Neospora spp., utilizando-se um ensaio imunoenzimático (ELISA) indireto multi-espécies comercial (ID Screen® Neospora caninum Indirect Multi-species; ID.vet Innovative Diagnostics, Grabels, France). Dois cavalos (1,1%; IC: 0,1-3,8%), um macho e uma fêmea, foram detectados como seropositivos para Neospora spp. Ambos os animais seropositivos eram cavalos mantidos em cocheiras mas com acesso aos piquetes, eram utilizados para atividades de lazer e estavam aparentemente saudáveis, com boa condição corporal e sem alterações ao exame físico. Essa é o primeiro rastreio de anticorpos para Neospora spp. realizado em cavalos de Portugal.

Abstract Neospora caninum is protozoan parasite with domestic and wild dogs, coyotes and grey wolves as the definitive hosts and many warm-blooded animals as intermediate hosts. It was cultivated and named in 1988. Neosporosis is a major disease of cattle and has no public health significance. Since 1990's N. caninum has emerged as a major cause of abortion in cattle worldwide, including in Brazil. N. caninum also causes clinical infections in several other animal species. Considerable progress has been made in understanding the biology of N. caninum and there are more than 200 papers on this subject from Brazil. However, most of the reports on neosporosis from Brazil are serological surveys. Overall, little is known of clinical neosporosis in Brazil, particularly cattle. The few reports pertain to sporadic cases of abortion with no information on epidemics or storms of abortion. The objective of the present review is to summarize all reports from Brazil and suggest topic for further research, including prevalence of N. caninum oocysts in soil or in canine feces, and determining if there are additional definitive hosts, other than the domestic dog. There is need for a national survey in cattle using defined parameters. Future researches should focus on molecular characterization of N. caninum strains, possibility of vaccine production and relationship between wildlife and livestock epidemiology.


Resumo Neospora caninum é um protozoário parasita que possui os canídeos domésticos e selvagens, coiotes e lobos cinzentos como hospedeiros definitivos e vários animais de sangue quente como hospedeiros intermediários. Foi cultivado e nomeado em 1988. A neosporose é uma das principais doenças em bovinos e não tem significância em saúde pública. Desde 1990, N. caninum tem emergido como uma das principais causas de aborto em bovinos em todo o mundo, inclusive no Brasil. N. caninum também causa infecções clínicas em várias outras espécies animais. Consideráveis avanços foram feitos na compreensão da biologia desse parasita e há mais de 200 trabalhos sobre o assunto no Brasil. No entanto, a maioria dos relatos de neosporose do Brasil são relacionados a sorologia. Em geral, pouco se sabe sobre a neosporose clínica no Brasil, particularmente em bovinos. Os poucos relatos referem-se a casos esporádicos de aborto sem informações sobre epidemias ou surtos de aborto. O objetivo da presente revisão é resumir todos os relatos sobre N. caninum no Brasil e sugerir tópicos para pesquisas futuras, incluindo a prevalência de oocistos de N. caninum no solo ou em fezes caninas e determinar se há hospedeiros definitivos adicionais, exceto o cão doméstico no país. Uma pesquisa nacional em bovinos usando parâmetros definidos seria de grande importância. Pesquisas futuras deveriam ser focadas na caracterização de cepas de N. caninum, possibilidade de produção de vacinas e a relação epidemiológica entre a vida selvagem e o gado.

This is the first report of Toxoplasma gondii infection in black vultures (Coragyps atratus), which are obligate scavengers found throughout the Americas. Serum samples from 121 wild black vultures caught in urban areas of the city of São Paulo, Brazil, were tested for the presence of T. gondii antibodies using the modified agglutination test (MAT; cutoff point 1:5). T. gondii antibodies were found in 16 birds (13.2%), with titers of 1:5 (6 birds), 1:10 (8 birds), and 1:20 (2 birds).(AU)


Este é o primeiro relato de infecção por Toxoplasma gondii em urubus (Coragyps atratus) que são aves carniceiras obrigatórias, encontradas no continente americano. Amostras de soro de 121 urubus, capturados em área urbana da cidade de São Paulo, Brasil, foram testadas quanto à presença de anticorpos anti-T. gondii pelo teste de aglutinação modificada (MAT, ponto de corte de 1:5). Anticorpos foram encontrados em 16 (13,2%) aves com títulos de 1:5 (6 aves), 1:10 (8 aves) e 1:20 (2 aves).(AU)

A vigilância e monitoramento de doenças em animais silvestres são imprescindíveis no contexto ambiental e de saúde pública, pois estes animais agem como sentinelas, refletindo alterações ambientais precocemente, o que proporciona maior eficácia no monitoramento ambiental e permite o acesso rápido a informações sobre as condições da área. Neste contexto, as aves são importantes no ciclo biológico do Toxoplasma gondii e na epidemiologia da toxoplasmose, principalmente porque seus tecidos representam importantes fontes de proteína na alimentação de felídeos e humanos. Objetivou-se detectar anticorpos anti-T. gondii, por meio do teste de aglutinação modificada em aves silvestres de três Unidades de Conservação (UC) Federais dos Estados da Paraíba e Bahia. No período de dezembro de 2011 a outubro de 2013 foram capturadas com redes de neblina 222 aves silvestres pertencentes a 67 espécies, 27 famílias e 12 ordens. Após a captura, foi colhido sangue de cada animal e separado o soro, que foi submetido ao Teste de Aglutinação Modificada (MAT≥1:25) utilizando taquizoítos inativados na formalina e 2-mercaptoetanol. Dentre as 222 amostras analisadas, três (1,3%) foram sororreagentes: 1 de 16 (6,2%) pipira-preta Tachyphonus rufus (título 50), 1 de 5 (20%) juriti-gemedeira Leptotila rufaxilla (título 50) e 1 de 1 (100%) caneleiro-enxofre Casiornis fuscus (título 25). Este é o primeiro relato da ocorrência de anticorpos anti-T. gondii nas referidas espécies de aves silvestres de vida livre nas duas UC Federais estudadas.


Surveillance and monitoring of wildlife pathogens are essential in the environmental context and human public health, as these animals act as sentinels, reflecting environmental changes early on, whath gives more efficient environmental monitoring and allows quick access to information on the conditions of area. Birds are important in the epidemiology and life cycle of Toxoplasma gondii, because their tissues are important source of protein in the diet of felids and humans. The objective was to determine antibodies to Toxoplasma gondii in wild birds from three Federal Conservation Units of the states of Paraíba and Bahia by Modified Agglutination Test (MAT). From December 2011 to October 2013, 222 wild birds of 67 species from 27 families and 12 Orders were captured with mist nets. Blood samples were then collected and the serum was separated by centrifugation. The sera were tested (MAT≥1:25) using formalin-fixed whole tachyzoites and 2-mercaptoethanol. Antibodies to T. gondii were found in 3 of 222 (1.3%) birds: in 1 of 16 (6.2%) white-lined tanager (Tachyphonus rufus, titer 50), in 1 of 5 (20%) gray-fronted dove (Leptotilla rufaxila, titer 50), and in 1 of 1 (100%) ashy-throated casiornis (Casiornis fuscus, titer 25). This is the first report of occurrence of antibodies to T. gondii in these tree bird species from two Federal Conservation Units.

We present a set of data on human and chicken Toxoplasma gondiiseroprevalence that was investigated and analysed in light of groundwater vulnerability information in an area endemic for waterborne toxoplasmosis in Brazil. Hydrogeological assessment was undertaken to select sites for water collection from wells for T. gondiioocyst testing and for collecting blood from free-range chickens and humans for anti-T. gondiiserologic testing. Serologic testing of human specimens was done using conventional commercial tests and a sporozoite-specific embryogenesis-related protein (TgERP), which is able to differentiate whether infection resulted from tissue cysts or oocysts. Water specimens were negative for the presence of viable T. gondiioocysts. However, seroprevalence in free-range chickens was significantly associated with vulnerability of groundwater to surface contamination (p < 0.0001; odds ratio: 4.73, 95% confidence interval: 2.18-10.2). Surprisingly, a high prevalence of antibodies against TgERP was detected in human specimens, suggesting the possibility of a continuous contamination of drinking water with T. gondiioocysts in this endemic setting. These findings and the new proposed approach to investigate and analyse endemic toxoplasmosis in light of groundwater vulnerability information associated with prevalence in humans estimated by oocyst antigens recognition have implications for the potential role of hydrogeological assessment in researching waterborne toxoplasmosis at a global scale.

Os zoológicos modernos são instituições destinadas à manutenção da fauna selvagem com o objetivo de promover a conservação, pesquisa científica, lazer, recreação e educação ambiental. A ampla variedade de espécies selvagens, vivendo em condições diferentes do seu habitat natural, representa um ambiente propício à disseminação de doenças, muitas delas zoonóticas. Devido à escassez de dados e à relevância dos mamíferos selvagens neste contexto epidemiológico, tanto na toxoplasmose, quanto na leptospirose, foi efetuado o inquérito sorológico para toxoplasmose e leptospirose em mamíferos selvagens neotropicais do Zoológico de Aracaju, Sergipe, Brasil. Para tanto foram colhidas amostras sanguíneas de 32 animais, adultos, de ambos os sexos incluindo: 14 macacos-prego (Cebus libidinosus), quatro macacos-prego-do-peito-amarelo (Cebus xanthosternus), três onças-suçuaranas (Puma concolor), uma onça-pintada (Pantheraonca), uma raposa (Cerdocyon thous), seis guaxinins (Procyon cancrivorus), dois quatis (Nasua nasua) e um papa-mel (Eira barbara). Para a pesquisa de anticorpos anti-Toxoplasma gondii foi utilizado o Teste de Aglutinação Modificada (MAT ³"1:25) e para pesquisa de anticorpos anti-Leptospira spp. foi utilizado o teste de Soroaglutinação Microscópica (ponto de corte ³1:100) com uma coleção de antígenos vivos que incluiu 24 variantes sorológicas de leptospiras patogênicas e duas leptospiras saprófitas. Dentre os 32 mamíferos, 17 (53,1 por cento) apresentaram anticorpos anti-T. gondii e quatro (12,5 por cento) foram positivos para anticorpos anti-Leptospira spp. De acordo com o sexo, 60 por cento (9/15) dos machos e 47,1 por cento (8/17) das fêmeas foram soropositivos para T. gondii e 26,7 por cento (4/15) dos machos apresentaram anticorpos anti-Leptospira spp. Dos mamíferos que apresentaram anticorpos anti-T. gondii, 47 por cento (8/17) nasceram no zoológico, 41,2 por cento (7/17) foram oriundos de outras instituições e dois (11,8 por cento) ...


Modern zoological gardens are institutions to wildlife maintenance, aiming its conservation, the performance of scientific research and leisure, recreation and environ-mental education activities. The variety of wild species living in conditions different from the one found in its natural habitats represents a propitious environment for diseases spread, specially the zoonotic ones. Due to data shortage and wild mammals' epidemio-logical relevance both for toxoplasmosis as for leptospirosis, this study aimed to determine the serological survey of toxoplasmosis and leptospirosis in Neotropical wild mammals, from Zoo of Aracaju, Sergipe, Brazil. Blood samples were collected from 32 wild mammals, adults and from both sexes: 14 wild bearded capuchins (Cebus libidinosus), four golden-bellied capuchins (Cebus xanthosternus), three pumas (Puma concolor), one jaguar (Pantheraonca), one crab-eating fox (Cerdocyon thous), six crab-eating raccoons (Procyon cancrivorus), two South American coatis (Nasua nasua), and one tayra (Eira barbara). Sera were tested to Toxoplasma gondii antibodies by the modified agglutination test (MAT e"1:25), and by the microscopic seroaglutination test (cut-off e"1:100) using 24 serovar of pathogenic leptospiras and two serovar of saprophyte leptospiras. Antibodies to T. gondii were found in 17 of 32 (53.1 percent) wild mammals, and antibodies to Leptospira spp. were found in 4 of 32 (12.5 percent) wild mammals. In relation to gender, 9 of 15 (60 percent) males, and 8 of 17 (47.1 percent) females had antibodies to Toxoplasma gondii, and 4 of 15 (26.7 percent) males had antibodies to Leptospira spp. Considering the origin of the seropositive animals to T. gondii, 8 of 17 (47 percent) were born in the zoo, 7 of 17 (41.2 percent) were from other zoos, and 2 (11.8 percent) were from the wild. Com o objetivo de se avaliar a freqüência de caprinos leiteiros soropositivos para Neospora caninum, no estado de São Paulo, e se verificarem possíveis associações com idade, sexo e problemas reprodutivos, nos capris, e, também, presença de cães, nas propriedades, foram obtidos soros de 923 caprinos de ambos os sexos e idade acima de 3 meses. Os animais eram provenientes de 17 propriedades de diferentes municípios. Para o diagnóstico, foi utilizado o teste de aglutinação para Neospora (NATe"25), e, em todos os capris, aplicou-se um inquérito a partir do qual se obtiveram informações epidemiológicas e de esfera reprodutiva. Todos os resultados estatísticos foram discutidos no nível de 5% de significância. Assim, chegou-se à conclusão de que a freqüência percentual de positividade para N. caninum foi de 19,77%, e, em apenas uma propriedade, não houve registro de animal soropositivo, o que revela difusão do agente, no Estado. Não foram verificadas diferenças significativas entre freqüências de positividade quanto ao sexo, idade ou problemas reprodutivos. Porém, ressalta-se que a presença de cães, nos capris, foi associada a uma maior freqüência de caprinos soropositivos a N. caninum. A representação geográfica da distribuição de caprinos soropositivos para o protozoário, em mapa coroplético em hachuras, pode implicar em um ganho considerável para estudos da epidemiologia geográfica, na elaboração de um planejamento de controle da enfermidade.(AU)


In order to assess the frequency of dairy goats seropositive for Neospora caninum in the state of São Paulo, and its relationship with age, sex, presence of dogs and reproductive problems in goat pens, serum samples were obtained from 923 goats of both sexes older than 3 months of age. Samples were collected in 17 properties located in different municipalities. Neospora agglutination test (NATe"25) was used to determine the presence of Neospora caninum in all animals. Epidemiological and reproductive data were obtained through a questionnaire which was administered in all goat pens. Significance level was set at 5% for all statistical tests. N. caninum positivity rate was 19.77%. Only one of the properties had no seropositive animal revealing N. caninum diffusion in the state. Seropositivity frequency did not correlate with sex, age, or reproductive problems. However, the presence of dogs was associated with a higher frequency of N. caninum seropositivity. A choroplethic map with hatch patterns showing the geographical distribution of goats seropositive for Neospora caninum may considerably contribute to geographic epidemiological studies and the development of disease control strategies.(AU)
