The trial of experimental vaccine consisting of protective protein antigens of P. aeruginosa cell wall was carried out on 114 volunteers. The vaccine proved to be faintly reactogenic and induced the formation of specific humoral immunity in 98% of the volunteers who retained a high level of anti-P. aeruginosa antibodies in their blood for up to 5 months (the term of observation after the course of immunization was over.

A preclinical study of seven batches of lactoglobulin, a new biological preparation against opportunistic bacteria and salmonellae, has been carried out. High antibacterial activity of the preparation with respect to the virulent forms of Salmonella typhimurium, Salmonella enteritidis, Pseudomonas aeruginosa, Klebsiella pneumoniae, Proteus vulgaris, Proteus mirabilis has been established. The preparation has been shown to be safe and nontoxic. The 4-year term of its storage at a temperature of 6 degrees +/- 4 degrees C has been substantiated.

Synthetic polysaccharide (S-PS) containing aglycone-spacer with a free amino group was really alpha 1,6-mannan with Cn approximately 10. S-PS was transformed into isothiocyanate derivative by treating it with thiophosgene and engaged into reaction with amino group of bovine serum albumin (BSA) lysine residues. Rabbits were immunized with S-PS-BSA conjugate and antibodies to S-PS titres were estimated by means of ELISA. S-PS-BSA conjugate was proved to provoke specific anti-polysaccharide antibodies formation in rabbits.

Two variants of cell-free protein vaccine have been prepared from the mixture of 4 P. aeruginosa strains, serovars 02, 06, 07 and 011, and from a single P. aeruginosa strain, serovar 02. The preparation contains proteins with molecular weight ranging from 20,000 to 100,000 and the admixture of lipopolysaccharide in negligible amounts (not exceeding 0.08% of dry weight). The vaccine produces no signs of toxicosis in laboratory animals. The vaccine effectively protects mice challenged with P. aeruginosa of different O-serotypes and stimulates the formation of specific protective antibodies in rabbits.

Partially purified water extract was obtained from the initial water extract by ultracentrifugation. Nine protein fractions differing in molecular weight, homogeneity and the content of lipopolysaccharide (LPS) were obtained by stepwise precipitation with ammonium sulfate and subsequent fractionation in columns packed with Sephadex G-100 and DEAE cellulose. Two protein fractions with a molecular weight of 30000 and 40000 daltons were practically free of LPS. These fractions were homogeneous as shown by analytical centrifugation and formed a single precipitation line with P. aeruginosa antiserum; both fractions were found to be antigenically identical. In the enzyme immunoassay these two fractions proved to be least active in comparison with the other protein fractions, but when used for the immunization of rabbits, they induced the formation of specific protective (for mice) antibodies. Both antisera were equally active in the experiments of the passive protection of mice. The isolated LPS-free proteins are supposed to be the proteins of the outer membrane of P. aeruginosa cell wall and have the properties of protective antigens.

To determine the content of lipopolysaccharide (LPS) in P. aeruginosa aqueous extracts the ELISA was used. Membrane filtration and ultracentrifugation followed by precipitation with ammonium sulfate at 80% saturation and gel filtration on Sephadex G-100 have proved to be the most effective methods for purification of the aqueous extract from LPS.

The preliminary study of the safety, reactogenicity and immunological effectiveness of polyvalent vaccine prepared from Pseudomonas aeruginosa protective antigens was carried out. In this study the vaccine was used for the immunization of volunteer donors. No postvaccinal complications were revealed. After the immunization with pyoimmunogen the titer of antibodies to P. aeruginosa increased 2-fold in comparison with their initial level.

A total of 191 batches of commercial human gamma globulin were studied with the aim of detecting natural antibodies by means of cross and countercurrent electrophoresis, immunodiffusion in agar gel and the passive hemagglutination test. Commercial human gamma globulin preparations were found to contain, to a greater or lesser extent, natural antibodies to the cell-wall O-antigen of Ps. aeruginosa and the antigens of its extracellular slime (capsule); the titers of these antibodies were 1 : 64 to 1 : 256 in 75-85% of cases, 1 : 512 and greater in 10-11% of cases. Commercial gamma globulin preparations also contained antibodies protecting mice from experimental Pseudomonas infection. No correlation was detected between the degree of serological activity and the protective activity of gamma globulin preparations.

Artificial antigens were obtained on the basis of the polysaccharide component of P. aeruginosa complexed with an indifferent protein. Immunological study indicated that the specific polysaccharide of P. aeruginosa lipopolysaccharide contained two structures, high molecular and low molecular, having qualitative and quantitative differences in their hydrocarbon composition. Artificial complex antigens possessed serological and immunogenic properties, the low molecular polysaccharide fraction complexed with protein having less pronounced serological and immunogenic activity than polysaccharide and the high molecular fraction complexed with protein. Antificial complex antigens exerted no protective effect in generalized P. aeruginosa infection in rats.

Antigenic complexes extracted from S. typhi Ty24446 in tryptic proteolysis displayed heterogeneity by molecular weight. The O-antigenic activity of the preparation was associated with the components with a mol. weight of 6.7-10(4)--2-10(6), Vi-antigenic activity--with the components with a mol. weight of 6.7-10(4)--2-10(6) and the components with a mol. weight of 1.75-10(4)--6.7-10(6). Components with a mol. weight below 1.75-10(4) possessed no Vi-antigenic activity. Correlations between the high- and low-molecular components in the "tryptic" antigen depended on the regimen of proteolysis.
