BACKGROUND: Data-driven development of medical biomarkers of bone requires a large amount of image data but physical measurements are generally too restricted in size and quality to perform a robust training. PURPOSE: This study aims to provide a reliable in silico method for the generation of realistic bone microstructure with defined microarchitectural properties. Synthetic bone samples may improve training of neural networks and serve for the development of new diagnostic parameters of bone architecture and mineralization. METHODS: One hundred-fifty cadaveric lumbar vertebrae from 48 different male human spines were scanned with a high resolution peripheral quantitative CT. After prepocessing the scans, we extracted 10,795 purely spongeous bone patches, each with a side length of 32 voxels (5 mm) and isotropic voxel size of 164 µm. We trained a volumetric generative adversarial network (GAN) in a progressive manner to create synthetic microstructural bone samples. We then added a style transfer technique to allow the generation of synthetic samples with defined microstructure and gestalt by simultaneously optimizing two entangled loss functions. Reliability testing was performed by comparing real and synthetic bone samples on 10 well-understood microstructural parameters. RESULTS: The method was able to create synthetic bone samples with visual and quantitative properties that effectively matched with the real samples. The GAN contained a well-formed latent space allowing to smoothly morph bone samples by their microstructural parameters, visual appearance or both. Optimum performance has been obtained for bone samples with voxel size 32 × 32 × 32, but also samples of size 64 × 64 × 64 could be synthesized. CONCLUSIONS: Our two-step-approach combines a parameter-agnostic GAN with a parameter-specific style transfer technique. It allows to generate an unlimited anonymous database of microstructural bone samples with sufficient realism to be used for the development of new data-driven methods of bone-biomarkers. Particularly, the style transfer technique can generate datasets of bone samples with specific conditions to simulate certain bone pathologies.

Background and objectives: The use of delirium screening instruments (DSIs) is recommended in critical care practice for a timely detection of delirium. We hypothesize that the patient-related factors "level of sedation" and "mechanical ventilation" impact test validity of DSIs. Materials and Methods: This is a prospective, bi-center observational study (clinicaltrials.gov: NCT01720914). Critically ill patients were screened for delirium daily for up to seven days after enrollment using the Nursing Delirium Screening Scale (Nu-DESC), Intensive Care Delirium Screening Checklist (ICDSC), and Confusion Assessment Method for the Intensive Care Unit (CAM-ICU). Reference standard for delirium diagnosis was the neuropsychiatric examination using the criteria of the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Text Revision (DSM-IV-TR). Immediately before delirium assessment, ventilation status and sedation levels were documented. Results: 160 patients were enrolled and 151 patients went into final analysis. Delirium incidence was 23.2%. Nu-DESC showed a sensitivity and specificity of 88.5%, a positive predictive value (PPV) of 71.9%, and a negative predictive value (NPV) of 95.8%. ICDSC had a sensitivity of 62.5%, a specificity of 92.4%, a PPV of 71.4%, and a NPV of 89.0%. CAM-ICU showed a sensitivity of 75.0%, a specificity of 94.7%, a PPV of 85.7%, and a NPV of 90.0%. For Nu-DESC and ICDSC, test validity was significantly better for non-sedated patients (Richmond Agitation Sedation Scale (RASS) 0/-1), whereas test validity for CAM-ICU in a severity scale version showed no significant differences for different sedation levels. No DSI showed a significant difference in test validity between noninvasively and invasively ventilated patients. Conclusions: Test validities of DSIs were comparable to previous studies. The observational scores ICDSC and Nu-DESC showed a significantly better performance in awake and drowsy patients (RASS 0/-1) when compared with other sedation levels. Physicians should refrain from sedation whenever possible to avoid suboptimal performance of DSIs.

Multiple infections by HPV genotypes are frequently detected in HPV+ cervical lesions but the interaction between each viral genotype during carcinogenesis is poorly understood. Here we carried out a comprehensive study to characterize the multiple HPV expression and integration by RNA-seq analyses of 19 invasive cervical carcinomas coinfected by several HPV genotypes. Analysis of tumor DNA by a hybridization assay indicated multiple infections ranging from 2 to 6 different HPV genotypes. RNA-seq analysis showed that a single HPV genotype was preferentially expressed. Finally, the search for HPV/human chimeric transcripts indicated integration from preferentially expressed genotypes. In conclusion, the present study indicated that, in invasive cervical carcinomas infected by multiple HPV genotypes, one HPV was preferentially expressed, supporting the hypothesis that a single HPV genotype was associated with cancer development.

Material e métodos: Para citotoxicidade foram utilizadas concentrações a partir de 10% do extrato aquoso seguindo uma sequência de diluições. A leitura das placas se procedeu com corante de vermelho neutro em espectofotometro. O ensaio anti-inflamatório ocorreu por indução da inflamação no conduto auditivo de ratos induzidos por óleo de cróton 5% em acetona. Os animais foram alocados em grupos: extrato aquoso de W. douradinha 100%, 50% e 25%, e grupos controle positivo (betametasona), e controle negativo (solução fisiológica de NaCl 0,9%). O potencial anti-inflamatório dos tratamentos foi avaliado através de medição do bordo caudal da pinna auricular, assim como pelo peso de amostra obtida por punch. Também foram pesquisados compostos com ação anti-inflamatória comprovada através de análise cromatográfica. Objective: To evaluate the in vitro cytotoxic effect caused by cell exposure to Waltheria douradinha aqueous extract, and to test the topical anti-inflammatory potential in an experimental model of inflammation. Material and methods: For cytotoxicity, concentrations from 10% of the aqueous extract were used following a dilution sequence. Plates were read with red-neutral dye in spectrophotometer. The antiinflammatory assay was by induction of inflammation in the ear canal of rats induced by 5% cróton-oil in acetone. The animals were allocated in groups: aqueous extract of W. douradinha 100%, 50% and 25%, and positive control groups (betamethasone) and negative control (0.9% NaCl physiological solution). The anti-inflammatory potential of the treatments was evaluated by measuring the caudal edge of the pinna, as well as the sample weight obtained by punch. Compounds with proven antiinflammatory action through chromatographic analysis were also investigated. Results: when compared to control, cells exposed to concentrations from 0.1% to 1% of the aqueous extract did not show cytotoxicity, being characterized as toxic concentrations of 10, 5 and 1%. For the anti-inflammatory assay, rodents treated with 50% aqueous extract of W. douradinha were equivalent to the control group with allopathic anti-inflammatory. Also flavonoid compounds of antiinflammatory action were identified: Luteolin, Kercetin, Kaempferol and Rutina. Conclusion: The aqueous extract of W. douradinha has cytotoxic activity at concentrations greater than 1%, besides demonstrating possibilities for effect as an edematogenic compound when administered to the outer ear of croton-oil induced rats.

In this contribution, we examine the photophysical properties of 15 totally trans-trans 1,4-distyrylbenzene derivatives (DSBs) functionalized with different electron-donating (ED) and electron-withdrawing (EW) groups by experimental and computational methodologies. We use UV-vis and fluorescence spectroscopies to determine the experimental optical properties such as the maximum absorption (λabsexp) and emission (λemexp) wavelengths, the highest occupied molecular orbital-lowest unoccupied molecular orbital (HOMO-LUMO) energy gaps (ΔEabsexp), the molar extinction coefficients (ε), the fluorescence quantum yields (Φf), and the fluorescence lifetimes (τ). We also calculate the experimental spontaneous emission decay rate (krexp) and correlate all of these magnitudes to the corresponding calculated properties, maximum absorption (λabscal) and emission (λemcal) wavelengths, vertical transition energies (ΔEabscal), oscillator strength (Fosc), and spontaneous emission decay rate (krcal), obtained by the time-dependent density functional theory method. We analyze the effect of the electronic nature of the substituents on the properties of the DSBs, finding that the ED and EW groups lead to bathochromic shifts. This is consistent with the decrease of ΔE values as the strength of ED and EW substituents increases. We find excellent correlations between calculated and experimental values for λabs, λem, and ΔEabs (r ∼ 0.99-0.95). Additionally, the correlations between the relative ε with Fosc values and the kr values are in good agreement (r ∼ 0.88-0.72) with the experimental properties. Overall, we find that for substituted 1,4-DSBs, computational chemistry is an excellent tool to predict structure-property relationships, which can be useful to forecast the properties of their polymeric analogues, which are usually difficult to determine experimentally.

Numerous animals have declining populations due to habitat loss, illegal wildlife trade, and climate change. The cotton-top tamarin (Saguinus oedipus) is a Critically Endangered primate species, endemic to northwest Colombia, threatened by deforestation and illegal trade. In order to assess the current state of this species, we analyzed changes in the population of cotton-top tamarins and its habitat from 2005 to 2012. We used a tailor-made "lure strip transect" method to survey 43 accessible forest parcels that represent 30% of the species' range. Estimated population size in the surveyed region was approximately 2,050 in 2005 and 1,900 in 2012, with a coefficient of variation of approximately 10%. The estimated population change between surveys was -7% (a decline of approximately 1.3% per year) suggesting a relatively stable population. If densities of inaccessible forest parcels are similar to those of surveyed samples, the estimated population of cotton-top tamarins in the wild in 2012 was 6,946 individuals. We also recorded little change in the amount of suitable habitat for cotton-top tamarins between sample periods: in 2005, 18% of surveyed forest was preferred habitat for cotton-top tamarins, while in 2012, 17% percent was preferred. We attribute the relatively stable population of this Critically Endangered species to increased conservation efforts of Proyecto Tití, conservation NGOs, and the Colombian government. Due to continued threats to cotton-top tamarins and their habitat such as agriculture and urban expansion, ongoing conservation efforts are needed to ensure the long-term survival of cotton-top tamarins in Colombia.

O uso de fitoterápicos é uma alternativa de baixo custo e de fácil acesso para o tratamento de feridas cutâneas. Objetivou-se avaliar a ação do extrato oleoso de urucum na cicatrização de feridas cutâneas abertas. Inicialmente, identificaram-se os principais ácidos graxos do óleo de urucum. Foi realizado ensaio citotóxico para determinar as concentrações a serem utilizadas no ensaio in vivo. No experimento, feridas cutâneas em ratos Wistar foram diariamente tratadas com: extrato de urucum 0,1% (U 0,1%), extrato de urucum 0,01% (U 0,01%), vaselina (V) e solução fisiológica (SF), por até 21 dias. Aos quatro, sete, 14 e 21 dias, foi avaliada clinicamente a presença de exsudato, crosta e epitelização. Determinaram-se as áreas da lesão, e amostras de pele, fígado e rins foram coletadas para avalição histológica. Aos 21dias, amostras de pele foram coletadas para análise tensiométrica. Clinicamente, todos os grupos de tratamento apresentaram evolução cicatricial fisiológica. Os grupos U 0,1% e U 0,01% apresentaram maior presença de epitelização aos sete dias e maior retração cicatricial aos quatro dias. Na histologia, U 0,1% e U 0,01% apresentaram aos quatro e sete dias maior quantidade de fibrina e inflamação que V e SF, e, nos demais momentos, não houve diferenças entre os grupos. Quanto à fase cicatricial, aos quatro dias todos os grupos encontravam-se na fase inflamatória, aos sete dias U 0,1% e U 0,01% permaneciam na fase inflamatória, diferindo de SF e V, que se caracterizavam na fase proliferativa. Aos 14 dias, os grupos apresentavam-se em transição de fase proliferativa para maturação e, aos 21dias, estavam todos na fase de maturação. Os grupos tratados com urucum expressaram menor resistência à tensão que V e SF. Concluiu-se com este estudo que o extrato oleoso de urucum acelera o processo cicatricial nos primeiros dias, mas proporciona uma cicatriz de baixa qualidade.

The purpose of this study was to assess the oil extract of Bixa orellana L. as a healing agent in the rat model of open wound healing. Initially, the oil was obtained and characterized through gas chromatography. Furthermore, the cytotoxic potential of the oil was verified in cell cultures to determine the doses used in animal experiments. Wounds were surgically produced in Wistar rats, these were treated with the oil extract at 0.1% (U 0.1%), 0.01% (U 0.01%), petrol jelly (V) and saline (SF) for up to 21 days. At four, seven and 14 days of treatment the wounds were assessed clinically regarding the presence of exudate, crust and epithelialization. The wound area was also determined and skin, kidney and liver tissues were harvested for histopathology. At 21 days of treatment the skins were also harvested for tension resistance assessment. Clinically, all groups evolved similarly, however, those treated with U 0.1% and U 0.01% had a greater amount of epithelialized wounds by day seven, and grater shrinkage by day four. Histopathologicaly, the skin samples of oil treated wounds had more lesions in the inflammatory phase at seven days, when compared to the controls, which were majorly in the proliferation phase. By 14 days no difference was observed among groups, which were all in the transition from the proliferation to the maturation phase. By day 21, all wounds were in the maturation phase. Oil treated wounds also had more fibrin in the first two assessment dates, when compared to the controls. Tension resistance of the oil treated wounds was, however, inferior to that of the controls. This study shows that B. orellana L. oil will hasten the onset of the healing process and its initial phases, but will ultimately produce a scar of poorer quality.

By 14 days no difference was observed among groups, which were all in the transition from the proliferation to the maturation phase. By day 21, all wounds were in the maturation phase. Oil treated wounds also had more fibrin in the first two assessment dates, when compared to the controls. Tension resistance of the oil treated wounds was, however, inferior to that of the controls. This study shows that B. orellana L. oil will hasten the onset of the healing process and its initial phases, but will ultimately produce a scar of poorer quality.(AU)

While Triticum sp. has been shown to act in wound healing, stimulating collagen synthesis by fibroblasts, the use of this plant extract has yet to be assessed in vivo, in commercially viable presentations. This study used rabbits and assessed, on days seven, 14, and 21, the presence or absence of granulation tissue and epithelialization, histopathological structures, and scar quality through the breaking and tension strength. Treatments, performed for 21 days, were aqueous extract of T. aestivum at a concentration of 2mg/mL (group I) and 10mg/mL (group II) and a nonionic cream (control group). We demonstrate that the formation of granulation tissue was not significantly different between treatments. In the analysis of epithelial tissue, wounds in group II differed from other treatments by day 7. On days 14 and 21 there was no significant clinical difference between groups. In the histopathological evaluation, scar quality and rupture strength did not differ between the groups in the studied period. In the tension strength evaluation, group I differed from the others, presenting a higher tension strength overall. O extrato de trigo (Triticum sp.) vem sendo usado na cicatrização de feridas por estimular a síntese de fibroblastos, entretanto a sua aplicabilidade in vivo em apresentações comercialmente viáveis ainda tem de ser demonstrada. Neste estudo, avaliaram-se feridas cutâneas de coelhos tratadas com extrato aquoso de T. aestivum quanto à presença de tecido de granulação e epitelização, estruturas histológicas, qualidade cicatricial, além de ensaio tensiométrico. As feridas foram tratadas diariamente, por 21 dias, com diferentes concentrações do extrato (grupo I = 2mg/mL; grupo II = 10mg/mL) ou apenas o veículo (grupo controle = creme não iônico), e avaliadas nos dias sete, 14 e 21. A formação de tecido de granulação não diferiu entre os tratamentos. A epitelização aconteceu em menor tempo em feridas do grupo II, mas aos 14 dias já não havia diferença neste parâmetro. Na avaliação histopatológica, a qualidade cicatricial e a força de ruptura não diferiram no período estudado, entretanto a resistência tensiométrica das feridas do grupo I foi maior que a dos demais tratamentos. Tomato apex necrosis virus (ToANV) is a new virus that causes important damage in tomato crops from the Culiacan Valley, Sinaloa, Mexico. To understand the relationship between ToANV and its vector Bermisia tabaci (Hemiptera: Aleyrodidae) (Gennadius) biotype B, laboratory and greenhouse trials were completed to: 1) determine the acquisition and inoculation access periods of ToANV by B. tabaci from tomato to tomato, 2) understand the transmission efficiency at different B. tabaci population densities, 3) estimate the time from inoculation of the virus at different B. tabaci densities to manifestation of symptoms in the plants, and 4) determine the retention time of the virus by the insect vector. The presence of the virus in plants was determined by reverse transcription-polymerase chain reaction amplification ofa 795-bp fragment (GenBank JN704068), which is phylogenetically related to ToANV (GenBank EF063242). The results showed that B. tabaci is an effective vector for ToANV with relatively long acquisition (12 h) and inoculation (9 h) access periods; a single adult is capable of transmitting and retaining the virus for up to 7d, suggesting a persistent mode of transmission. These results will help in the development of management strategies for controlling the vector and the disease.

OBJECTIVE: The objective of this study was to assess the interleukin 10 (IL-10) concentrations in the sera of dogs suffering from demodicosis. METHODS: Twenty-six dogs were distributed into three groups: demodicosis groups G1, n=11 (recurring disease) and G2, n=6 (first time occurrence), and a control group, G3, n=9 (healthy dogs). All the animals were subjected to skin scrape tests and blood harvesting for serum extraction. In G1 and G2 only those animals with Demodex canis positive skin tests were included, while healthy dogs were included in G3. To assess IL-10 levels the commercial Quantikine Canine IL-10 Immunoassay(®) (R&D Systems) kit was used. RESULTS: The mean IL-10 level obtained for G1 was 269.4 pg/ml (sd=290.8 pg/ml), for G2 it was 28.5 pg/ml (sd=19.7 pg/ml) while the mean for G3 was 11.9 pg/ml (sd=2.3 pg/ml). There was a significant difference between G1 and the other two groups. CONCLUSIONS: According to our results, dogs with reoccurring demodicosis have higher IL-10 levels than healthy dogs and those suffering the disease for the first time.

Avaliou-se o efeito da substituição do feno de capim tifton pela casca de mamona, 0, 33, 66 e 100% na dieta de ovinos, sobre o peso e rendimento dos componentes não constituintes da carcaça e o rendimento de buchada e panelada. Vinte e oito animais não castrados, com sete meses de idade e peso inicial médio de 19,5±2,45kg, foram distribuídos em delineamento de blocos ao acaso, com quatro tratamentos e sete repetições, e abatidos após 70 dias de confinamento. Houve decréscimo linear nos pesos da língua (0,084 a 0,079kg), traqueia+pulmão (0,50 a 0,42kg), coração (0,14 a 0,12kg), timo (0,035 a 0,025kg), rins (0,100 a 0,088kg) e peso total dos órgãos (2,14 a 1,90kg) de acordo com o percentual de substituição. O mesmo comportamento foi verificado para os pesos do esôfago (0,051 a 0,045kg), rúmen-retículo (0,73 a 0,64kg), intestino delgado (0,78 a 0,64kg) e total de vísceras (2,17 a 1,92kg). Para os pesos da cabeça (1,99 a 1,66kg), patas (0,86 a 0,73kg), pele (2,49 a 1,99kg), sangue (1,36 a 1,19kg) e total de subprodutos (7,12 a 5,96kg) verificou-se efeito linear negativo de acordo com a substituição. O mesmo foi observado para os pesos de buchada (4,71 a 4,22kg) e panelada (7,55 a 6,61kg). A casca de mamona não deve substituir o feno de capim tifton na dieta de ovinos por causar decréscimo no peso de órgãos, vísceras e subprodutos e influenciar negativamente os pesos e rendimentos de buchada e panelada.


The effects of replacing tifton hay with 0, 33, 66 and 100% of castor bean hulls in the diet of sheep on the non-carcass components of weight and dressing and on the "buchada" and "panelada" yield were evaluated in this research. Twenty-eight non castrated sheep averaging seven months of age and with initial weight of 19.5±2.45kg were assigned to a randomized block design with four treatments and seven replicates, and were slaughtered after 70 days of confinement. A linear decrease of tongue (0.084 to 0.079kg), trachea + lungs (0.50 to 0.42kg), heart (0.14 to 0.12kg), thymus (0.035 to 0.025kg), kidneys (0.100 to 0.088kg) weights and total weight of organs (2.14 to 1.90kg) was observed. The same behavior was observed for the esophagus (0.051 to 0.045kg), rumen-reticulum (0.73 to 0.64kg), small intestine (0.78 to 0.64kg) and total viscera (2.17 to 1.92kg) weights. There was a linear effect for the head (1.99 to 1.66kg), feet (0.86 to 0.73kg), skin (2.49 to 1.99kg), blood (1.36 to 1.19kg) and total by-products (7.12 to 5.96kg) weights according to the treatments and also for "buchada" (4.71 to 4.22kg) and "panelada" (7.55 to 6.61kg) weights. Castor bean hulls should not replace tifton hay because it decreases the weight of organs, viscera and by-products and adversely affects the "buchada" and "panelada" weight and dressing.

Repetitive proteins (RP) of Trypanosoma cruzi are highly present in the parasite and are strongly recognized by sera from Chagas' disease patients. Flagelar Repetitive Antigen (FRA), which is expressed in all steps of the parasite life cycle, is the RP that displays the greatest number of aminoacids per repeat and has been indicated as one of the most suitable candidate for diagnostic test because of its high performance in immunoassays. Here we analyzed the influence of the number of repeats on the immunogenic and antigenic properties of the antigen. Recombinant proteins containing one, two, and four tandem repeats of FRA (FRA1, FRA2, and FRA4, respectively) were obtained and the immune response induced by an equal amount of repeats was evaluated in a mouse model. The reactivity of specific antibodies present in sera from patients naturally infected with T. cruzi was also assessed against FRA1, FRA2, and FRA4 proteins, and the relative avidity was analyzed. We determined that the number of repeats did not increase the humoral response against the antigen and this result was reproduced when the repeated motifs were alone or fused to a non-repetitive protein. By contrast, the binding affinity of specific human antibodies increases with the number of repeated motifs in FRA antigen. We then concluded that the high ability of FRA to be recognized by specific antibodies from infected individuals is mainly due to a favorable polyvalent interaction between the antigen and the antibodies. In accordance with experimental results, a 3D model was proposed and B epitope in FRA1, FRA2, and FRA4 were predicted.

Ric c 1 and Ric c 3 are the major castor bean allergens. In order to identify continuous IgE-epitopes in Ric c 1 and Ric c 3, pools of sera from rats immunized with a pool of 2S albumin from these seeds, Ric c 1 and Ric c 3 overlapping synthetic peptides, were used to screen for IgE-binding epitopes. The allergenic properties were monitored by mast cell degranulation assays, histamine quantification and human-IgE binding. Large and small chains isolated from these proteins present allergenic properties. Four continuous epitopes were identified in Ric c 3 and two in Ric c 1. This knowledge may allow the induction of protective antibody responses to antagonize the IgE recognition.

Se realizó una revisión bibliográfica sobre el síndrome de ojo seco, que abarcó su clasificación, diagnóstico y tratamiento. Se tuvo en cuenta que constituye una entidad frecuente en la Oftalmología; se presenta con la finalidad de mostrar el síndrome de manera actualizada y con el objetivo de facilitar su manejo.


A bibliographic review of the dry eye syndrome that included its classification, diagnosis and treatment is made. Taking into account that it is a frequent entity in Ophthalmology, it is presented in order to provide an updating of the syndrome and to facilitate its management.

RESUMEN

We report the case of a 10 year-old girl who had Stevens-Johnson syndrome and cholestasis after ibuprofen therapy. Liver histology was compatible with vanishing bile duct syndrome. She received ursodeoxycholic acid, and liver tests normalized within 7 months. This report confirms that ibuprofen may induce acute vanishing bile duct syndrome.
