BACKGROUND: The salutogenic model has been established as a health promoting resource that is related to a strong sense of coherence (SOC), positive subjective health and quality of life (QoL). The aim of the study was to compare internal and external resources, life style factors, perceived health and QoL in Japan and Austria and to determine associations among these factors. METHODOLOGY AND PRINCIPAL FINDINGS: A survey was conducted in a Japanese (N = 460) and an Austrian (N = 421) student sample using the following self-report health questionnaires: Sense of Coherence Scale (SOC-13), Social and Gender Role Scale, Multidimensional Scale of Perceived Social Support (MSPSS), Dutch Eating Behaviour Questionnaire (DEBQ), SF-12 Health Survey, and the Cross-cultural Health Survey. Analyses of data showed that age (ß -0.12), and stress (ß -0.21) were negatively related and SOC (ß 0.47), family support are (ß -0.35) positively related to mental QoL. Significant predictors for emotional strain, were female gender (ß -0.24), older age (ß-0.14), lower SOC (ß 0.28), less traditional gender and social role patterns (ß 0.10), more restrained eating (ß -0.20), more alcohol intake (ß -0.16), and more stress (ß -0.25) explaining 42% of the variance in Austrian students. In Japan stress (ß -0.38) was negatively related and SOC (ß 0.37) positively related to mental QoL. Older age (ß -0.20), lower SOC (ß 0.29) and more stress (ß -0.33) were identified as significant predictors explaining 35% of the variance in Japanese students. CONCLUSIONS AND SIGNIFICANCE: SOC and stress are strongly associated with QoL and perceived health in Austria as well as in Japan. SOC seems to be a crucial predictor for stress, and emotional health independent of the cultural context. A major challenge of cross-cultural research is to understand perceived health and QoL and the extent in which it is individually, socially, or culturally determined.

Light-induced lesions are a powerful tool to study the amazing ability of photoreceptors to regenerate in the adult zebrafish retina. However, the specificity of the lesion towards photoreceptors or regional differences within the retina are still incompletely understood. We therefore characterized the process of degeneration and regeneration in an established paradigm, using intense white light from a fluorescence lamp on swimming fish (diffuse light lesion). We also designed a new light lesion paradigm where light is focused through a microscope onto the retina of an immobilized fish (focused light lesion). Focused light lesion has the advantage of creating a locally restricted area of damage, with the additional benefit of an untreated control eye in the same animal. In both paradigms, cell death is observed as an immediate early response, and proliferation is initiated around 2 days post lesion (dpl), peaking at 3 dpl. We furthermore find that two photoreceptor subtypes (UV and blue sensitive cones) are more susceptible towards intense white light than red/green double cones and rods. We also observed specific differences within light lesioned areas with respect to the process of photoreceptor degeneration: UV cone debris is removed later than any other type of photoreceptor in light lesions. Unspecific damage to retinal neurons occurs at the center of a focused light lesion territory, but not in the diffuse light lesion areas. We simulated the fish eye optical properties using software simulation, and show that the optical properties may explain the light lesion patterns that we observe. Furthermore, as a new tool to study retinal degeneration and regeneration in individual fish in vivo, we use spectral domain optical coherence tomography. Collectively, the light lesion and imaging assays described here represent powerful tools for studying degeneration and regeneration processes in the adult zebrafish retina.

Dispersion encoded full range (DEFR) optical coherence tomography (OCT) has become highly attractive as it is a simple way to increase the measurement range of OCT systems. Full range OCT is especially favorable as it does not only increase the measurement range but also shifts the highest sensitivity into the center of the measurement range. While the early versions of DEFR were highly computational expensive, new versions reduce the number of necessary Fourier transforms. Recently it has been shown that a GPU based algorithm can perform DEFR with more than 20,000 A-lines per second. We present a new version of the DEFR algorithm that requires only one Fourier transform per A-scan and uses convolution in z-space instead of multiplication in k-space, therefore reducing the computational effort considerably. While dispersion encoding has so far only been used to suppress mirror artifacts, we show that, with dispersion encoding and only one more Fourier transform, autocorrelation terms can be removed likewise. Since very high values of dispersion reduce the effective measurement range in dispersion encoded OCT, we present an estimate for a sufficient amount of dispersion for a successful image recovery, which is depending on the thickness of the scattering layers. Furthermore, we demonstrate the usability of ZnSe as a new dispersive material with a very high dispersion and describe a simple method to extract the dispersive phase from the measurement of a single reflex of a glass surface. Using a standard consumer PC, an artifact-free recovery of 1000 - 2000 A-scans per second with 2048 depth values including autocorrelation removal was achieved. The dynamic range (sensitivity) is not reduced and the suppression ratio of mirror artifacts and autocorrelation signals is more than 50dB using ZnSe.

OBJECTIVE: To immunohistochemically evaluate matrix metalloproteinase (MMP)-9 expression in benign and malignant mammary gland tumors (MMTs) in dogs and relate expression to prognostic factors and patient outcome. ANIMALS: 118 female dogs with naturally occurring mammary gland tumors and 8 dogs without mammary gland tumors. PROCEDURES: 24 benign mammary gland tumors and 94 MMTs (1/affected dog) were obtained during surgical treatment; control mammary gland tissue samples were collected from unaffected dogs after euthanasia for reasons unrelated to the study. Tumors were evaluated for proliferation, invasive growth, histologic grade, and metastatic capacity; expression of MMP-9 was determined immunohistochemically, and its relationship with clinical and histologic findings was investigated. For dogs with MMTs, follow-up continued for 2 years; data were used to compute overall survival time and disease-free interval and construct survival curves. RESULTS: MMTs had significantly higher MMP-9 expression in stromal cells and in neo-plastic cells than did the benign neoplasms. Stromal MMP-9 expression was also higher in highly proliferative tumors and in tumors with invasive growth, high histologic grade, and metastatic capacity. Furthermore, tumors from patients with shorter overall survival times and disease-free intervals had higher expression of MMP-9 in stromal cells. CONCLUSIONS AND CLINICAL RELEVANCE: In dogs with MMTs, level of MMP-9 expression by stromal cells was related to factors of poor prognosis and shorter overall survival times and disease-free intervals. These results suggested that MMP-9 produced by tumor-adjacent stromal cells contributed to MMT progression in female dogs and that assessment of MMP-9 expression may be a valuable prognostic factor.

Recent studies of canine malignant mammary tumours suggest that reduction of E-cadherin and/or beta-catenin correlates with invasive behaviour and lymph node metastasis. The aims of this study were to examine the interrelationships between the expression of E-cadherin and beta-catenin, and the relationship between the expression of E-cadherin and/or beta-catenin and the mode of growth and metastatic capacity of canine malignant mammary tumours. 90 spontaneous malignant tumours and local and regional lymph nodes were studied. A significant relationship was evidenced between membranous expression of E-cadherin and beta-catenin (p=0.0027), but not between E-cadherin and cytoplasmic beta-catenin. Only E-cadherin as a separate factor was significantly related to tumour invasion (p=0.0072) and lymph node metastasis (p=0.0001). Neither membranous nor cytoplasmic beta-catenin expression was significantly related to either of these phenomena.

The relationship between MIB-1 labelling indices (LI), as detected by immunohistochemical methods, and other clinico-pathological characteristics was studied in a series of 77 malignant mammary tumours surgically removed from 47 female dogs. The immunostaining was assessed on the basis of the estimated percentage of positive cells in the areas of highest labelling. Multivariate logistic regression demonstrated no influence of breed, age, previous pregnancies, previous progestin administration, histological type or location of the tumour on MIB-1 LI. MIB-1 LI was significantly related to the size of the tumour, necrosis, invasive growth and histological grade, but not with ulceration, lymph node metastasis, skin fixation or E-cadherin expression. The significant relationship between MIB-1 LI and other known factors of poor prognosis suggests that a high LI may have prognostic value in canine malignant mammary tumours.
