RESUMEN

We previously reported that female rats placed on a diet containing refined carbohydrates (HCD) resulted in obesity and reproductive abnormalities, such as high serum LH concentration and abnormal ovarian function. However, the impacts at the hypothalamic-pituitary (HP) function, specifically regarding pathways linked to reproductive axis modulation are unknown. In this study, we assessed whether subacute feeding with HCD results in abnormal reproductive control in the HP axis. Female rats were fed with HCD for 15 days and reproductive HP axis morphophysiology was assessed. HCD reduced hypothalamic mRNA expression (Kiss1, Lepr, and Amhr2) and increased pituitary LHß+ cells. These changes likely contribute to the increase in serum LH concentration observed in HCD. Blunted estrogen negative feedback was observed in HCD, with increased kisspeptin protein expression in the arcuate nucleus of the hypothalamus (ARH), lower LHß+ cells and LH concentration in ovariectomized (OVX)+HCD rats. Thus, these data suggest that HCD feeding led to female abnormal reproductive control of HP axis.

Abstract 18. Introduction: Tooth development results from a highly coordinated epithelial-mesenchyme interaction in which mesenchyme cells originate the dental papilla and dental follicle, while ectodermal cells originate the enamel organ. Simultaneously, bone tissue is formed around the developing tooth, trapping it in a bony crypt. Tooth eruption requires the resorption of the coronal part of the bony crypt, followed by degradation of the lamina propria, most likely by metalloproteinases (MMPs) activity. Objectives: The aim of this research was to determine MMP-2 expression in the dental germ cells (ameloblasts, odontoblasts, dental papilla and dental follicle) and surrounding tissues (alveolar bone and lamina propria) of rat molars throughout the eruptive process. Material and Methods: A total of 24 rats (4,6,9,11,14 and 16 days old) were used in this study. MMP-2 was detected through immunohistochemistry. A qualitative analysis was performed to investigate the expression of MMP2 in the dental germ cells, lamina propria, and coronal and basal regions of the bony crypt. Results: MMP-2 expression was observed in the dental papilla cells, dental follicle, ameloblasts, odontoblasts and bone cells from the coronal and basal regions of the bony crypt. MMP-2 was also detected in the lamina propria during the mucosal penetration stage of tooth eruption. Conclusion: We conclude that MMP-2 may be important for the extracellular matrix rearrangement necessary for tooth development and secretion of its mineralized tissues. We also conclude that MMP-2 may play a role in the extensive tissue remodeling during the intra-and-extra-osseous phases of the tooth eruption process.

Introducción: el desarrollo del diente resulta de una interacción epitelial-mesénquima altamente coordinada en la cual las células mesénquima originan la papila dental y el folículo dental, mientras que las células ectodérmicas originan el órgano del esmalte. Simultáneamente, el tejido óseo se forma alrededor del diente en desarrollo y lo atrapa en una cripta ósea. La erupción dentaria requiere la resorción de la parte coronal de la cripta ósea, seguida de la degradación de la lámina propia, muy probablemente por la actividad metaloproteinasas (MMPs). Objetivos: el objetivo de esta investigación fue determinar la expresión de MMP-2 en las células germinales dentales (ameloblastos, odontoblastos, papila dentaria y folículo dentario) y tejidos circundantes (hueso alveolar y lámina propia) de molares de rata a lo largo del proceso eruptivo. Material y métodos: en este estudio se utilizó un total de 24 ratas (4,6,9,11,14 y 16 días de edad). MMP-2 se detectó a través de inmunohistoquímica. Un análisis cualitativo fue realizado para investigar la expresión de MMP-2 en las células de germen dentales, el lámina propria, y las regiones coronales y basales de la cripta ósea. Resultados: la expresión de MMP2 fue observada en las células de la papila dental, el folículo dental, el ameloblastos, el odontoblastos y las células del las regiones basales y coronales de la cripta ósea. La expresión de MMP-2 también se detectó en la lámina propia durante la etapa de penetración de la mucosa de la erupción dental. Conclusión: Concluimos que MMP-2 puede ser importante para el cambio extracelular de la matriz necesario para el desarrollo del diente y la secreción de sus tejidos mineralizados. También concluimos que MMP-2 puede desempeñar un papel en la remodelación extensa del tejido durante las fases intra y extraósea del proceso de erupción dental.

Squamous cell carcinoma is a prevalent head and neck tumor with high mortality. We studied the role played by laminin alpha1 chain peptide AG73 on migration, invasion, and protease activity of cells (OSCC) from human oral squamous cell carcinoma. Immunohistochemistry and immunofluorescence analyzed expression of laminin alpha1 chain and MMP9 in oral squamous cells carcinoma in vivo and in vitro. Migratory activity of AG73-treated OSCC cells was investigated by monolayer wound assays and in chemotaxis chambers. AG73-induced invasion was assessed in Boyden chambers. Invasion depends on MMPs. Conditioned media from cells grown on AG73 was subjected to zymography. We searched for AG73 receptors related to these activities in OSCC cells. Immunofluorescence analyzed AG73-induced colocalization of syndecan-1 and beta1 integrin. Cells had these receptors silenced by siRNA, followed by treatment with AG73 and analysis of migration, invasion, and protease activity. Oral squamous cell carcinoma expresses laminin alpha1 chain and MMP9. OSCC cells treated with AG73 showed increased migration, invasion, and protease activity. AG73 induced colocalization of syndecan-1 and beta1 integrin. Knockdown of these receptors decreased AG73-dependent migration, invasion, and protease activity. Syndecan-1 and beta1 integrin signaling downstream of AG73 regulate migration, invasion, and MMP production by OSCC cells.

Migration, invasion and protease activity are essential for tumor progression and metastasis. Metastatic cells rely on invadopodia to degrade and invade extracellular matrix (ECM). Invadopodia are membrane protrusions with enzymes required for ECM degradation. These protrusions contain cortactin and membrane type 1 matrix metalloproteinase (MT1-MMP) superimposed to areas of digested matrix. Here we characterized invadopodia in a cell line (CAC2) derived from human adenoid cystic carcinoma. We carried out fluorescent-substrate degradation assay to assess in situ protease activity of CAC2 cells. Digestion spots in fluorescent substrate appear as black areas in green background. Cells were cultured on Matrigel-gelatin-FITC and fixed after 1 h and 3 h. CAC2 cells were double labeled to actin and cortactin. Cells were also double stained to actin and MT1-MMP. Samples were studied by laser scanning confocal microscopy. In all time points CAC2 cells showed actin, cortactin, and MT1-MMP colocalized with digestion spots in fluorescent substrate. We searched for other proteases involved in invadopodia activity. We have previously demonstrated that MMP9 influences adenoid cystic carcinoma behavior. This prompted us to investigate role played by MMP9 on invadopodia formation. CAC2 cells had MMP9 silenced by siRNA. After 1 h in fluorescent substrate, cells with silenced MMP9 showed clear decrease in matrix digestion compared with controls. No differences were found in cells with silenced MMP9 grown for 3 h on fluorescent substrate. Our results showed that CAC2 cells exhibit functional invadopodia containing cortactin and MT1-MMP. Furthermore, MMP9 would be required in the initial steps of invadopodia formation.

We studied the induction of protease activity by the laminin alpha1-derived peptide AG73 in cells from adenoid cystic carcinoma (CAC2) and myoepithelioma (M1), respectively a malignant and a benign salivary gland tumors. Laminin alpha1 chain and MMP9 were immunolocalized in adenoid cystic carcinoma and myoepithelioma in vivo and in vitro. Cells grown inside AG73-enriched laminin-111 exhibited large spaces in the extracellular matrix, suggestive of remodeling. The broad spectrum MMP inhibitor GM6001 decreased spaces induced by AG73 in CAC2 and M1 cells. This result strongly suggests that AG73-mediated matrix remodeling involves matrix metalloproteinases. CAC2 and M1 cells cultured on AG73 showed a dose-dependent increase of MMP9 secretion, as detected by zymography. Furthermore, siRNA silencing of MMP9 decreased remodeling in 3D cultures. We searched for AG73 receptors regulating MMP9 activity in our cell lines. CAC2 and M1 cells grown on AG73 exhibited colocalization of syndecan-1 and beta1 integrin. siRNA knockdown of syndecan-1 expression in these cells resulted in decreased adhesion to AG73 and reduced protease and remodeling activity. We investigated syndecan-1 co-receptors in both cell lines. Silencing beta1 integrin inhibited adhesion to AG73, matrix remodeling and protease activity. Double-knockdown experiments were carried out to further explore syndecan-1 and beta1 integrin cooperation. CAC2 cells transfected with both syndecan-1 and beta1 integrin siRNA oligos showed significant decrease in adhesion to AG73. Simultaneous silencing of receptors also induced a decrease in protease activity. Our results suggest that syndecan-1 and beta1 integrin signaling downstream of AG73 regulate adhesion and MMP production by CAC2 and M1 cells.
