RESUMEN

The African Swine Fever Virus (ASFV) is responsible for causing African Swine Fever (ASF), a severe contagious disease characterized by hemorrhagic symptoms. The p30 protein of ASFV is the most abundantly expressed viral protein. It is reported to be antigenic and has recognized phosphorylation, glycosylation, and membrane attachment sites, which also shows that the C-terminal region of p30 is more active than the N-terminal region. The present study reports the unique RNase activity of recombinant p30. The RNase activity of p30 was stable at an optimum temperature of 37 °C, and the maximum activity was recorded at pH 7-9 in the presence of monovalent salts. The mutant of p30 (p30m), where cysteine was mutated to alanine at position 109, showed a loss of RNase activity. Our understanding of ASFV biology is significantly less; until now, we have little knowledge about the functions of its proteins. The results of the present study will assist in exploring the biology of ASFV and the role of its protein in counteracting the host immune response.

An increased pathogenic waste post-COVID-19 pandemic forced policymakers to treat biomedical waste (BMW) similar to municipal solid waste (MSW) to dispose into dumpsites and MSW landfills across the globe. The granular bentonite of geosynthetic clay liners (GCLs) does not completely seal the macro-voids upon saturation due to the loss of osmotic potential in the salt environment from the leachate. Such behavior of GCLs can lead to advection-dominant virus migration through the liner system. A knowledge of the fate and transport of coronavirus and other viral pathogens in compacted clays is essential for safe disposal of the viral pathogens in MSW landfills. Although the attenuation and transport parameters for coronavirus have been recently evaluated theoretically, experimental backup is currently lacking. The present work uses Newcastle disease virus (NDV) as a surrogate to coronavirus due to structural similarities for studying the fate and transport in the compacted natural clays. This study also implicitly addresses the waste management facilities for waste generated from NDV outbreaks through poultry litter and carcasses. The interaction of bentonite and kaolin clays with the NDV was studied by varying the virus concentration, interaction time, and clay dose using batch sorption tests. The studied clays showed excellent attenuation efficiency for the NDV. Design parameters, viz., the diffusion coefficient and retardation factor, were evaluated, affirming the suitability of these clays for exclusive pathogenic waste disposal protocols that are discussed in this article.

BACKGROUND: India recorded the first outbreak of African swine fever (ASF) in North-eastern region (NER) in the year 2020. AIM: The current study was undertaken to investigate the transmission of African swine fever virus (ASFV) in the wild boars of Northeast India, particularly of Assam. MATERIAL AND METHODS: ASF suspected mortal tissue remains and blood samples of wild boars collected from different locations of Assam were screened for molecular detection of swine viruses which includes Classical swine fever virus, Porcine Circovirus 2, Porcine reproductive and respiratory syndrome virus and ASFV. RESULTS: One sample each from Manas and Nameri National Parks was detected positive for ASFV. Besides this, one of the samples was positive for CSFV and one of the ASFV positive samples was also positive for PCV2. Several striking gross and microscopic alterations were noticed in different organs of ASFV infected animals. Sequencing and phylogenetic analysis of B646L gene confirmed the presence of ASFV genotype-II in wild boars. Circulation of similar genotype in domestic pigs of NER in the contemporary period as well as locations near to the aforementioned national parks indicates the transmission of ASFV from domestic to wild boars. CLINICAL RELEVANCE: The detection of ASFV in the wild boars of Assam is alarming as it is an impending threat to pig population and other endangered species (particularly Pygmy hog), making it increasingly daunting to control the disease. CONCLUSION: Chances are high for ASFV to become endemic in Assam region if stringent measures are not taken at proper time.

Newcastle disease virus (NDV) affects commercial poultry as well as other avian species in the wild and in captivity. Although the diversity of NDV in domestic chickens has been well understood, little light has been shed on NDV outbreaks in other avian species. We provide an annotated sequence of NDV/Owl/Guwahati/01/20, a virulent strain of NDV isolated from barn owls in captivity from Guwahati in Northeast India. The complete genome is 15,192 base pairs long with a fusion protein (F) cleavage site 112KRQKR↓F117. The isolate showed 97.67% identity with its closest match, another highly virulent strain from Indonesia isolated from vaccinated commercial chickens; however, they differ in the F cleavage site. The NDV isolate from the owl shares 83.02% and 81.88% identity with the vaccine strains R2B and LaSota, respectively. Phylogenetic analysis with F gene as well as whole-genome nucleotide sequence reveals that the NDV isolate from owl belongs to genotype VII, subgenotype VII.2, and differs significantly from all other isolates of NDV from India.

Recently, multimodal detection of analytes through a single nanoprobe has become an eminent approach for researchers. Herein a fluorescent nanoprobe, functionalized-GQD (F-GQD), has been designed through edge functionalization of graphene quantum dots (GQDs) by 2,6-diaminopyridine molecules. The fluorescence of F-GQD is quite sensitive to medium pH, making it a suitable pH sensor within the pH range 2-6. Interestingly, F-GQD shows dual sensing of Pb2+ and ClO- by entirely different pathways; Pb2+ exhibits fluorescence turn-on performance while ClO- triggers turn-off fluorescence quenching. The fluorescence enhancement may originate from the Pb2+-induced aggregation of the nanodots. The limit of detection (LOD) was also impressive, 1.2 µM and 12.6 nM for Pb2+ and ClO-, respectively. The detailed mechanistic investigations reveal that both dynamic and static quenching effects operate together in the F-GQD-ClO- system. The dynamic quenching was attributed to the energy migration from F-GQD to ClO- through hydrogen bonding interaction (static quenching) between the amine group at the F-GQD surface and ClO-. The F-GQD nanodot reveals excellent sensitivity toward the detection of ClO- in real samples. Moreover, the F-GQDs also serve as multicolor fluorescent probes for cell imaging; the probe can easily penetrate the cell membrane and successfully detect intracellular ClO-.

A N-doped carbon dot (NCD) has been synthesized via a simplistic one-step hydrothermal technique using l-aspartic acid and 3,6-diaminoacridine hydrochloride. The NCDs exhibit a high quantum yield (22.7%) and excellent optical stability in aqueous media. Additionally, NCDs display good solid-state yellowish-green emission and are suitable for security ink applications. The remarkable fluorescence (FL) properties of NCDs are further applied to develop a multifunctional sensor for bilirubin (BR) and vitamin B12 (VB12) via fluorescence quenching. We have systematically studied the FL quenching mechanisms of the two analytes. The primary quenching mechanism of BR is via the Förster resonant energy transfer (FRET) pathway facilitated by the H-bonding network between the hydrophilic moieties existing at the surface of BR and NCDs. In contrast, the inner filter effect (IFE) is mainly responsible for the recognition of VB12. The practicability of the nanoprobe NCDs is further tested in real-sample analysis for BR (human serum and urine samples) and VB12 (VB12 tablets, human serum, and energy drink) with a satisfactory outcome. The in vitro competency is also verified in the human cervical cancer cell line (HeLa cell) with negligible cytotoxicity and significant biocompatibility. This result facilitates the application of NCDs for bioimaging and recognition of VB12 in a living organism.
