RESUMEN

The reproductive physiology of fish can be changed by the presence of pollutants in the water, which act as endocrine disrupting compounds (EDC). We evaluated the impacts of water contaminants in polluted reservoirs acting as possible EDC on the reproductive physiology of Astyanax fasciatus and Hoplias malabaricus males. We used biomarkers with different levels of biological organization. Hoplias malabaricus adult males were collected in the summer and winter at five different sites in the Tietê River Basin: the Ponte Nova reservoir (PN), considered a reference site due to the low anthropogenic influence; the Billings reservoir (BIL) at two different branches; and the Guarapiranga reservoir (GUA) at two different branches. Astyanax fasciatus adult males were collected at PN and BIL. BIL and GUA are subjected to great anthropogenic action. We analyzed gonadal histomorphology, testosterone (T), 11-ketotestosterone (11-KT), estradiol (E2) plasma levels, and gene expression of hepatic vitellogenin (vtgA) and pituitary follicle stimulating hormone (fshß). In the PN reservoir (reference), the biomarkers analyzed in both species did not differ between the periods analyzed. This is an evidence that the animals keep the same reproductive activity during both seasons. The changes in the plasma concentration of gonadal steroids in both species in polluted reservoirs suggest the presence of EDC compounds in the water and/or adjusts of the physiological setpoint to allow the reproduction in such adverse conditions. The use of vtgA as biomarker suggests the presence of estrogenic compounds, mainly in BIL, but with a more evident response of H. malabaricus. However, even considering physiological changes, both species present testes during the maturation phase that allow the reproduction in an environment with a high degree of pollution.

Diclofenac (DCF) and caffeine (CAF) are persistent pharmaceuticals that occur in mixtures in the aquatic ecosystems causing effects in the reproductive physiology of aquatic organisms. This study evaluated the physiological reproductive responses of Astyanax altiparanae males exposed to nominal concentrations of DCF (3.08 mg L-1) and CAF (9.59 mg L-1) separately and combined, for 96 h. The steroids profile, estrogenic biomarker vitellogenin (vtgA), testes and liver morphology, and also mortality of males were assessed. DCF and CAF degradation was 5% of the initial concentration for 24 h. The LC50 of the DCF and CAF were 30.8 mg L-1 and 95.9 mg L-1, respectively. Males exposed to DCF and CAF exhibited a reduction of 17ß-Estradiol (E2) concentration compared to control (CTL). Similarly, testosterone (T) was also reduced in the DCF treatment, but this response was not observed in 11-Ketotestosterone (11-KT). Males exposed to DCF + CAF combined did not exhibit differences in T, E2 and 11-KT steroids. The vtgA gene expression and the sperm concentration did not change among the treatments. Moreover, acute exposure revealed a hypertrophy of hepatocytes cells in the DCF and DCF + CAF treatments. In conclusion, DCF and CAF, isolated, exhibit an endocrine disruptive activity in A. altiparanae male, an opposite response observed with the mixture of both compounds that abolishes the endocrine disruptive effects. DCF seems to be more toxic for this species, altering also hepatocytes morphology.

Although interspecific trophic interactions plays a principal role within elasmobranch communal nurseries, little is known over variation in foraging strategies adopted by young-of-year of sympatric species. To test the hypothesis of dietary resource partitioning between batoids within a communal nursery, we investigated two cownose ray species, Rhinoptera bonasus and R. brasiliensis, which occur in heterospecific groups, a strategy predicted to increase survival and foraging success. Using two biochemical tracers, fatty acids (FA) and stable isotopes (δ15 N and δ13 C), the combined effects of maternal investment and the formation of heterospecific groups implying competition for, or partitioning of available food resources were investigated. Through univariate and multivariate analyses of biochemical tracers in several tissues (fin clip, muscle, liver, red blood cells; RBC) and plasma, our results revealed significant interspecific differences in tracers between the two species. Total FAs (∑saturated FA, ∑monounsaturated FA and ∑polyunsaturated FA) and trophic biomarkers (i.e., docosahexaenoic acid, arachidonic acid, oleic acid and δ15 N) were the principle tracers responsible for the differences detected. These data revealed that R. brasiliensis was less enriched in physiologically important essential FAs than R. bonasus. Our findings suggest that these congeneric species differ in maternal investment strategy and moderately partition food resources over relatively fine spatial scales within a single nursery habitat to limit competition. These results provide further knowledge on the foraging strategies adopted by batoids in communal nursery areas, information that is required for improving spatial conservation and management planning.

Endocrine disrupting compounds (EDCs) have the potential to alter fish reproduction at various levels of organization. The aim of this study was to assess the impact of a natural environment with heavily anthropogenic influence on the physiological processes involved in reproduction in the freshwater fish lambari (Astyanax fasciatus) using different biomarkers. Adult males and females were collected in different seasons from two distinct sites in the same watershed: Ponte Nova Reservoir (PN) considered a pristine or small anthropogenic influence reference point; and Billings Reservoir (Bil), subjected to a large anthropogenic impact. Biological indices, such as hepatosomatic index and gonadosomatic index (GSI), gonadal histomorphology, fecundity, and biomarkers such as plasma levels of estradiol (E2) as well as hepatic gene expression of its alfa nuclear receptor (ERα), were analyzed. Hepatic vitellogenin (VTG) gene expression was evaluated in both sexes, as an indicator of xenoestrogen exposure. Females collected at PN presented a typical annual variation reflected in GSI, whereas for those sampled at Bil the index did not change through the seasons. The higher concentration of E2 in males collected at Bil during spring/2013, together with the detection of VTG gene expression, suggest the presence of EDCs in the water. These EDCs may have also influenced fecundity of females from Bil, which was higher during winter and spring/2013. Gene expression of ERα and ovarian morphology did not differ between fish from both sites. Water conditions from Bil reservoir impacted by anthropic activity clearly interfered mainly with biomarkers of biological effect such as plasma E2 levels and absolute and relative fecundity, but also altered biomarkers of exposure as VTG gene expression. These facts support the notion that waterborne EDCs are capable of causing estrogenic activity in A. fasciatus.

This study aimed at analyzing the energetic substrate (ES) in the main storage tissues of Steindachneridion parahybae, throughout the reproductive cycle in captivity. Differently from wild, in captivity, feeding is not interrupted during the reproductive period, the females do not spawn spontaneously, and they are sedentary. Adult females were sampled monthly and based on their histology and gonadosomatic index (GSI), ovaries were classified into: previtellogenic (PRV), vitellogenic (VTG), and regression (REG) stages. Ovaries at the VTG stage showed higher protein and lipids levels than at the PRV stage with a positive correlation between these substrates and the GSI. Muscle was the main source of proteins transferred to the ovaries, according to the negative correlation between these organs. Lipids remained unchanged in the liver, which is an important supplier in vitellogenesis, a pattern that probably occurs due to the continuous feeding. Muscular glycogen levels were higher at the VTG and REG than at the PRV stages. Plasma triglycerides were also higher during REG, while glucose levels were more elevated during the VTG stage. These results suggest that with constant food supply, the pattern of deposition of ES in S. parahybae is different from that described for other wild potamodromous species.(AU)


O objetivo deste estudo foi analisar a composição do substrato energético (SE) nos principais tecidos de armazenamento de Steindachneridion parahybae, durante todo o ciclo reprodutivo em cativeiro. Diferentemente do ambiente natural, em cativeiro, a alimentação desses animais não é interrompida durante o período reprodutivo, as fêmeas não desovam espontaneamente, e são sedentárias. Fêmeas adultas foram amostradas mensalmente e baseada na histologia e no índice gonadossomático (IGS), os ovários foram classificados: estádios pré-vitelogênico (PRV), vitelogênico (VTG) e regressão (REG). Os ovários no estádio VTG apresentaram uma maior concentração de lipídeos e proteínas em relação ao estágio PRV. Esses substratos correlacionaram-se positivamente com o IGS. O músculo foi a principal fonte de proteína transferida aos ovários, como foi confirmado pela análise de correlação negativa entre esses órgãos. Os lipídeos mantiveram-se inalterados no fígado, considerado um importante órgão fornecedor de lipídeos para a vitelogênese, padrão que possivelmente ocorreu devido à contínua alimentação. A concentração do glicogênio muscular foi mais elevada durante os estágios VTG e REG em relação ao PRV. A concentração de triglicerídeos plasmática apresentou maiores valores no estádio REG enquanto a concentração de glicose no plasma foi maior durante os estádios VTG. Esses resultados sugerem que com alimentação constante, as fêmeas de S. parahybae apresentam um distinto padrão de mobilização dos substratos energéticos em relação ao que já foi descrito para outras espécies potamódromas de ambiente natural.(AU)

Eutrophication results in a deficiency of n-3 LC-PUFA (long-chain polyunsaturated fatty acids) in aquatic food chains, affecting fish nutrition and physiology. The trophic transfer of FA (fatty acids) to fish species of different feeding habits was investigated in two reservoirs in southeast Brazil-the mesotrophic Ponte Nova Reservoir (PN) and the hypereutrophic Billings Reservoir (Bil). Total FA profile of stomach contents and adipose tissue, triacylglycerols (TAG), and phospholipids (PL) from liver and muscle of the omnivorous Astyanax fasciatus and the carnivorous Hoplias malabaricus were analyzed by gas chromatography. A prevalence of n-6PUFA, as 18:2n-6 (linoleic acid) and 20:4n-6 (arachidonic acid, ARA) was observed in the stomach contents and in the tissues of A. fasciatus from the PN reservoir. In contrast, n-3 LC-PUFA, as 20:5n-3 (eicosapentaenoic acid, EPA) was accumulated in fish tissues from Bil, resulting in higher n3/n6 and EPA/ARA ratios, compared to fish from PN. This differential FA accumulation was also observed for H. malabaricus, but differences were slightly minor, and no changes were observed in the EPA/ARA ratios between fish from both reservoirs. Regardless reservoir, FA profiles of TAG resembled that of their diet, whereas FA profiles of PL were more conservative and mainly comprised by LC-PUFA. We conclude that reservoir trophic status affected the FA composition of food resources available to these fish species, resulting in differential allocation of n-3 and n-6 FA. As expected, FA profile of the investigated fish species also reflected their feeding habit and physiological demands.

The amphibian order Gymnophiona is poorly known, and studies about their reproduction are mainly comparative and descriptive, focusing on the structure of testes, ovaries and oviducts. However, to understand the reproductive processes, including those of the oviparous dermatophagic species, it is important to know the dynamics of storage and mobilization of energetic substrates to gonads and skin during the reproductive cycle of males and females, as well as the endocrine control associated. For the present study, total lipids and proteins were measured during the annual cycle in the plasma, liver, muscle, testes, ovaries and skin of Siphonops annulatus in captivity. Plasma levels of gonadal steroids (estradiol, testosterone and progesterone) were quantified by radioimmunoassay. Histological analyses of ovaries and testes were performed to classify the maturation stages of the animals during the reproductive cycle. Gonadal maturation in males and females of S. annulatus was accompanied by metabolic changes in reserve tissues, which supported gonadal development and prepared the females' epidermis for skin feeding by the offspring. Even in confinement conditions, females and males synchronized the reproductive period. However, due to the absence of environmental cues in captivity inadequate levels of the hormones responsible for gamete release were triggered, leading to a lack of reproductive success.

Down syndrome (DS) is the most common form of mental retardation of genetic etiology. Several polymorphisms in genes involved with the folic acid cycle have been associated to the risk of bearing a DS child; however, the results are controversial. Betaine-homocysteine methyltransferase (BHMT) is a key enzyme of folate pathway, and catalyzes the remethylation of homocysteine into methionine. Recent studies suggest that the polymorphism BHMT 742G>A may be associated with a decreased risk of having a DS child. We herein investigate the association of this polymorphism with the occurrence of DS in a Brazilian population. We have genotyped 94 mothers of DS infants (DSM) and 134 control mothers (CM) for this polymorphism through PCR-RFLP, and found significant differences for both BHMT 742G>A genotype (P=0.04) and allele (P=0.03) frequencies between DSM and CM. The observed genotypic frequencies were GG=0.45; GA=0.45 and AA=0.10 in CM, and GG=0.54; GA=0.38 and AA=0.02 in DSM. Allelic frequencies were G=0.68 and A=0.32 in CM and G=0.78 and A=0.22 in DSM. The presence of the mutant BHMT 742 A allele decreases 40% the risk of bearing a DS child (OR=0.61; 95% CI: 0.40-0.93; P=0.03), and the risk is diminished up to >80% in association with the homozygous genotype (OR=0.17; 95% CI: 0.04-0.80; P=0.01). Our results indicate that women harboring the single nucleotide polymorphism BHMT 742G>A have a decreased risk of a DS pregnancy, and further studies are necessary to confirm this protective effect.

Metternichia princeps, a tree that belongs to the Solanaceae family, occurs in the Atlantic Forest of Brazil, from the state of Rio de Janeiro to Bahia. In the county of Itaguaí, state of Rio de Janeiro, M. princeps was suspected to cause a lethal kidney disease in goats. The poisoning was experimentally reproduced in goats confirming the suspicion. Experiments were then performed in rabbits. Leaves of M. princeps were collected in the county of Itaguaí, where the poisoning in goats occurred. Eleven rabbits received the dried plant material, ten received the fresh plant and one rabbit served as control. The dehydrated leaves were ground and mixed with water. Initially the suspension was administered by stomach tube to nine rabbits and caused death of seven of them at doses from 0.125g/kg on. Two rabbits that got sick, but survived at doses of 0.0625g/kg and 0.125g/kg, received doses of 0.5 and 0.25g/kg respectively, two months later and died, indicating that no tolerance to the plant poisoning effects developed. Additionally the dried leaves administered six months after collection to two rabbits at doses of 0.5 and 1g/kg respectively, caused death only of the rabbit that received the dose of 1g/kg, indicating that the stored leaves lost toxicity. Fresh sprouts caused death in three of six rabbits at doses of 1.55g/kg to 3g/kg. Fresh mature leaves caused death in one of four rabbits at the dose of 2g/kg, what suggests that there is no difference in toxicity between mature leaves and sprouts. The dried leaves caused death of rabbits at smaller doses than fresh leaves, what can be explained by the way of administration of the plant; dried leaves were given within a few minutes, whilst fresh leaves were ingested during one or two days, a fact that may influence absorption of the toxic principle. In all experiments with the dried and fresh leaves (sprouts and mature leaves) of M. princeps, liver and heart lesions predominated. The course of the poisoning...


Metternichia princeps, árvore que pertence à família Solanaceae, ocorre na Mata Atlântica desde o estado do Rio de Janeiro até a Bahia. É conhecida popularmente com os nomes de "café-do-mato", "trombeteira" e "jasmin-do-morro". Em um estabelecimento no município de Itaguaí, RJ, suspeitou-se que M. princeps era responsável por doença renal letal em caprinos. A intoxicação foi reproduzida experimentalmente nessa espécie, confirmando a suspeita. Foram então realizados experimentos em coelhos. Folhas de M. princeps foram coletadas no município de Itaguaí/RJ, local onde foi diagnosticada a intoxicação em caprinos. Onze coelhos receberam a planta dessecada, dez receberam a planta fresca e um coelho serviu como controle. As folhas dessecadas de M. princeps foram trituradas e misturadas com água. Inicialmente, a suspensão foi administrada por via intragástrica através de sonda a nove coelhos e causou a morte de sete coelhos nas doses a partir de 0,125g/kg. Dois coelhos que adoeceram mas sobreviveram às doses de 0,0625 e de 0,125g/kg, dois meses mais tarde receberam doses de 0,5 e 0,25g/kg, respectivamente, e morreram, o que indica que não houve desenvolvimento de tolerância à toxidez da planta. Adicionalmente, a planta dessecada, administrada a dois coelhos nas doses de 0,5 e 1g/kg seis meses após a coleta, causou a morte somente do coelho que recebeu a dose de 1g/kg, indicando que a planta armazenada perde em toxidez. Os brotos frescos causaram a morte de três dos seis coelhos, nas doses de 1,55g/kg a 3g/kg. As folhas maduras frescas causaram a morte de um dos quatro coelhos, na dose de 2g/kg, o que sugere que não há diferença na toxidez entre a brotação e as folhas maduras. As folhas dessecadas causaram a morte dos coelhos em doses menores que as folhas frescas, o que talvez possa ser explicado pelo modo de administração da planta; as folhas dessecadas eram administradas em poucos minutos, enquanto que as folhas frescas eram ingeridas durante um a dois dias...

Fatty acids have been used in marine biogeochemistry as food chain biomarkers, but in freshwater these studies are rare. In order to evaluate the fatty acid potential as biomarkers in freshwater, their profile was analyzed during vitellogenesis in two fish species, in both waterfall and reservoir environments of the Paraíba do Sul River Basin. Detrivorous Hypostomus affinis and omnivorous Geophagus brasiliensis seem to elongate and desaturate polyunsaturated fatty acids (PUFA) and transfer them to the ovaries' phospholipids. Waterfall Geophagus brasiliensis have more highly unsaturated fatty acids in the liver, but in the reservoir, accumulation mainly occurs in muscle and ovary triglycerides, suggesting trophic opportunism and a plasticity during vitellogenesis. In Hypostomus affinis, PUFA alteration occurs only in the reservoir, suggesting a high phytoplankton occurrence. Eutrophication and water speed is reflected in Hypostomus affinis ovaries by higher PUFAn3 and bacterial fatty acids. As in marine environments, analysis of mono- and polyunsaturated fatty acids during vitellogenesis can be used as a tool in food chain studies in freshwater.

Foram revisados casos de intoxicação por samambaia (Pteridium aquilinum) em bovinos de 10 pequenas propriedades rurais de sete municípios da Região Central do Rio Grande do Sul. O estudo abrangeu 6.256 necrospias de bovinos realizadas num período de 43 anos e incluiu 15 necropsias de bovinos que morreram em conseqüência da intoxicação aguda por samambaia. As taxas de morbidade e mortalidade foram 17,9 por cento e a letalidade era virtualmente de 100 por cento. Em 40 por cento das propriedades a doença ocorreu em pequenos surtos afetando vários bovinos; em 60 por cento das propriedades apenas um bovino era afetado. Os principais sinais clínicos incluíam febre (40-42ºC), apatia, salivação e hemorragias, principalmente pelas gengivas, pela cavidade nasal e pelo trato gastrintestinal. Múltiplas petéquias eram observadas nas mucosas e na pele. Ocasionalmente observou-se hematúria e sangue no leite. A doença era invariavelmente fatal após um curso clínico de dois dias. Alterações hematológicas consistiam de leucopenia por neutropenia, anemia normocítica normocrômica e trombocitopenia arregenerativa. Os achados de necropsia incluíam hemorragias de intensidade variável em vários órgãos e infartos no fígado. Tanto as hemorragias quanto os infartos foram confirmados histologicamente; agregados de bacilos e vasos trombosados foram observados em associação com os infartos. Aplasia da medula óssea era um achado consistente nos quatro casos em que esse órgão foi examinado.(AU)


Cases of acute poisoning by bracken fern (Pteridium aquilinum) in cattle from 10 small farms of seven municipalities of the Central Region of Rio Grande do Sul State, Brazil, were reviewed. The study encompassed 6,256 necropsies of cattle considering a 43-year period and included 15 necropsies of cattle dying from acute bracken poisoning. Morbidity and mortality were 17.9 percent and lethality was virtually 100 percent. In 40 percent of the farms the disease occurred in small outbreaks affecting several cattle per farm and in 60 percent only one bovine was affected in each farm. Main clinical signs include fever (40-42ºC), apathy, drooling, and hemorrhages; the latter were mainly from the gums, nostrils, and gastrointestinal tract. Multiple petechiae were observed in several mucosae and in the skin. Occasionally hematuria and blood in the milk were reported. The disease was invariably fatal after a course of approximately two days. Hematological changes included severe neutropenia, non-regenerative normocytic normochromic anemia and thrombocytopenia. Necropsy findings include hemorrhages o varying degrees in several organs and infarcts in the liver. Both hemorrhages and hepatic infarcts were confirmed histologically; clusters of bacterial rods and thrombosed vessels were associated with the infarcts. Marked bone marrow aplasia was a consistent finding in the four cases in which the marrow was evaluated.(AU)

We investigated in vitro skin penetration and permeation of fluconazole from emulsions containing different penetration enhancers. Fluconazole permeation was high (15-65% of the applied dose) across hairless mouse skin and low (8-9%) across pig ear skin. Permeation across mice skin from a formulation containing propyleneglycol and isopropyl myristate was significantly higher than that observed with the paraffin oil and propyleneglycol or Transcutol emulsions. With pig skin, the paraffin oil or isopropyl myristate and propyleneglycol emulsions showed similar skin permeation and penetration. However, these emulsions provided epidermal concentrations higher than the minimal inhibitory concentrations for most dermatophytes.
