RESUMEN

Infectious bronchitis virus (IBV), the causative agent of infectious bronchitis, is responsible for major economic losses in the poultry industry worldwide. While IBVs can usually be passaged in primary chicken embryonic fibroblasts (CEFs), most of the wild ones cannot adapt to passaged cell lines. In this study, the wild strain CK/CH/MY/2020 was used to infect primary CEF and immortalize DF-1 CEF cells. Results indicated that IBV was able to cause lesions and pass onto CEF, but not DF-1. Indeed, the virus could enter DF-1 cells and synthesize the associated structural gene but could not assemble into complete viral particles for release. Furthermore, transcriptome sequencing analysis showed significant differences in gene expression between CEF and DF-1 cells after viral infection, although the corresponding antiviral responses could be activated in both cell types. The biggest difference was in terms of the amino acid biosynthesis pathway and the cytokine receptor interaction pathway, which were significantly and specifically activated in CEF. This could actually explain why intact viruses can be assembled but not in DF-1. In addition, SLBP and P2RX7 affect the replication of IBV's structural genes to some extent. Overall, IBV can enter CEF and DF-1 cells, but the complex intracellular cytokine interactions affect the assembly and release of viral particles. The insight will be useful for the study of IBV through in vitro transmission and pathogenesis. IMPORTANCE: Infectious bronchitis virus (IBV) is responsible for high morbidity and mortality as well as substantial economic losses worldwide. Transcriptome sequencing of IBV-infected chicken embryonic fibroblast and DF-1 cells revealed that the virus elicits antiviral immunity in cells after viral infection, but IBV cannot activate DF-1 cells to produce sufficient amounts of viral structures to assemble into complete virions, which may be caused by the interactions between cytokines. The study of IBV cellular adaptations is important for vaccine development and investigation of the pathogenesis of IBV.

BACKGROUND: In HR+ /HER2- breast cancer patients with ≤ 3 positive axillary lymph nodes (ALNs), genomic tests can streamline chemotherapy decisions. Current studies, centered on tumor metrics, miss broader patient insights. Automated Breast Volume Scanning (ABVS) provides advanced 3D imaging, and its potential synergy with radiomics for ALN evaluation is untapped. OBJECTIVE: This study sought to combine ABVS radiomics and clinical characteristics in a nomogram to predict ≤ 3 positive ALNs in HR+ /HER2- breast cancer patients with 1-2 positive sentinel lymph nodes (SLNs), guiding clinicians in genetic test candidate selection. METHODS: We enrolled 511 early-stage breast cancer patients: 362 from A Hospital for training and 149 from B Hospital for validation. Using LASSO logistic regression, primary features were identified. A clinical-radiomics nomogram was developed to predict the likelihood of ≤ 3 positive ALNs in HR+ /HER2- patients with 1-2 positive SLNs. We assessed the discriminative capability of the nomogram using the ROC curve. The model's calibration was confirmed through a calibration curve, while its fit was evaluated using the Hosmer-Lemeshow (HL) test. To determine the clinical net benefits, we employed the Decision Curve Analysis (DCA). RESULTS: In the training group, 81.2% patients had ≤ 3 metastatic ALNs, and 83.2% in the validation group. We developed a clinical-radiomics nomogram by analyzing clinical characteristics and rad-scores. Factors like positive SLNs (OR=0.077), absence of negative SLNs (OR=11.138), lymphovascular invasion (OR=0.248), and rad-score (OR=0.003) significantly correlated with ≤ 3 positive ALNs. The clinical-radiomics nomogram, with an AUC of 0.910 in training and 0.882 in validation, outperformed the rad-score-free clinical nomogram (AUCs of 0.796 and 0.782). Calibration curves and the HL test (P values 0.688 and 0.691) confirmed its robustness. DCA showed the clinical-radiomics nomogram provided superior net benefits in predicting ALN burden across specific threshold probabilities. CONCLUSION: We developed a clinical-radiomics nomogram that integrated radiomics from ABVS images and clinical data to predict the presence of ≤ 3 positive ALNs in HR+ /HER2- patients with 1-2 positive SLNs, aiding oncologists in identifying candidates for genomic tests, bypassing ALND. In the era of precision medicine, combining genomic tests with SLN biopsy refines both surgical and systemic patient treatments.

Nowadays, skin disease is becoming one of the most malignant diseases that threaten people's health. Computer aided diagnosis based on deep learning has become a widely used technology to assist medical professionals in diagnosis, and segmentation of lesion areas is one of the most important steps in it. However, traditional medical image segmentation methods rely on numerous pixel-level labels for fully supervised training, and such labeling process is time-consuming and requires professional competence. In order to reduce the costs of pixel-level labeling, we proposed a method only using image-level label to segment skin lesion areas. Due to the lack of lesion's spatial and intensity information in image-level labels, and the wide distribution range of irregular shape and different texture on skin lesions, the algorithm must pay great attention to the automatic lesion localization and perception of lesion boundary. In this paper, we proposed a Self-Guided Multiple Information Aggregation Network (SG-MIAN). Our backbone network MIAN utilizes the Multiple Spatial Perceptron (MSP) solely using classification information as guidance to discriminate the key classification features of lesion areas, and thereby performing more accurate localization and activation of lesion areas. Additionally, adjunct to MSP, we also proposed an Auxiliary Activation Structure (AAS) and two auxiliary loss functions to further self-guided boundary correction, achieving the goal of accurate boundary activation. To verify the effectiveness of the proposed method, we conducted extensive experiments using the HAM10000 dataset and the PH2dataset, which demonstrated superior performance compared to most existing weakly supervised segmentation methods.

Objective: To study the clinical application value of contrast-enhanced ultrasound (CEUS) in diagnosing renal space-occupying lesions. Methods: Sixty-seven patients with renal space-occupying lesions detected by routine ultrasound examination received the contrast-enhanced ultrasound examination. When observing the perfusion mode of the mass, we analyzed the perfusion characteristics of contrast-enhanced ultrasound and compared them with the surgical pathological results. Results: Sixty-seven lesions, which were identified in 67 patients with renal space-occupying lesions, included 55 renal malignant tumors and 12 benign ones. The sensitivity of qualitative diagnosis by CEUS imaging was 96.4%, the specificity was 66.7%, and the accuracy was 91.0%. Conclusion: The real-time blood supply of renal space-occupying lesions helps judge their nature according to the enhancement mode. It has high clinical application value in diagnosing benign and malignant lesions.

A novel protocol of gold-catalyzed N-aryl propargylamines to construct tetrahydroquinolines and 5,6-dihydro-4H-pyrrolo[3,2,1-ij]quinolines in moderate to good yields has been developed through the tandem reactions of intramolecular hydroarylation and transfer hydrogenation. The strategy has the advantages of easy access to raw materials, simple reaction conditions, good substrate compatibility, high efficiency, and excellent regioselectivity.

Medical image segmentation plays a crucial role in computer-aided diagnosis. However, due to the large variability of medical images, accurate segmentation is a highly challenging task. In this paper, we present a novel medical image segmentation network named the Multiple Feature Association Network (MFA-Net), which is based on deep learning techniques. The MFA-Net utilizes an encoder-decoder architecture with skip connections as its backbone network, and a parallelly dilated convolutions arrangement (PDCA) module is integrated between the encoder and the decoder to capture more representative deep features. Furthermore, a multi-scale feature restructuring module (MFRM) is introduced to restructure and fuse the deep features of the encoder. To enhance global attention perception, the proposed global attention stacking (GAS) modules are cascaded on the decoder. The proposed MFA-Net leverages novel global attention mechanisms to improve the segmentation performance at different feature scales. We evaluated our MFA-Net on four segmentation tasks, including lesions in intestinal polyp, liver tumor, prostate cancer, and skin lesion. Our experimental results and ablation study demonstrate that the proposed MFA-Net outperforms state-of-the-art methods in terms of global positioning and local edge recognition.

BACKGROUND: Sucrose transporters (SUTs) mediate sucrose phloem loading in source tissue and sucrose unloading into sink tissue in potatoes and higher plants, thus playing a crucial role in plant growth and development. In potatoes, the physiological function of the sucrose transporters StSUT1 and StSUT4 has been clarified, whereas the physiological role of StSUT2 is not yet fully understood. METHODS AND RESULTS: This study analyzed the relative expression of StSUT2 compared to that of StSUT1 and StSUT4 in different tissues from potatoes and its impact on different physiological characteristics by using StSUT2-RNA interference lines. Here, we report a negative effect of StSUT2-RNA interference on plant height, fresh weight, internodes number, leaf area, flowering time, and tuber yield. However, our data indicate that StSUT2 is not involved in carbohydrate accumulation in potato leaves and tubers. In addition, the data of the RNA-seq between the StSUT2-RNA interference line and WT showed that 152 genes were differentially expressed, of which 128 genes were upregulated and 24 genes were downregulated, and the GO and KEGG analyses revealed that differentially expressed genes were mainly related to cell wall composition metabolism. CONCLUSIONS: Thus, StSUT2 functions in potato plant growth, flowering time, and tuber yield without affecting carbohydrate accumulation in the leaves and tubers but may be involved in cell wall composition metabolism.

OBJECTIVES: A considerable number of benign lesions, especially category 4a lesions on Breast Imaging Reporting and Data System Magnetic Resonance Imaging (BI-RADS-MRI), were biopsied according to BI-RADS-MRI, which was a diagnostic imaging challenge. This study aimed to evaluate the diagnostic performance of ultrasound elastography (UE) assisted Breast Imaging Reporting and Data System (BI-RADS) for BI-RADS-MRI category 4a lesions. METHODS: Between January 2017 and December 2019, 228 breast lesions categorized as BI-RADS-MRI 4a were included. Conventional ultrasound (US) and UE were performed to evaluate each lesion. Pathology results were used as the gold standard. The diagnostic performances of different UE methods and our re-assessment proposal were evaluated. RESULTS: When BI-RADS-MRI category 4a, BI-RADS-US category 3-4a, the stiffness of soft or intermediate in elasticity assessment according to the fifth edition of the BI-RADS atlas, strain ratio < 1.335, age ≤ 52 years, and the maximum diameter of lesion ≤20 mm were simultaneously met, an ultrasound-guided empty needle biopsy was not recommended, but short-term ultrasound follow-up for 3-6 months was recommended, and biopsy was performed after changes in evaluation. In this way, 95 of 228 BI-RADS-MRI category 4a lesions avoided biopsies, and the number of patients with biopsies decreased by 41.7%. CONCLUSIONS: UE offers benefits in the characterization of BI-RADS-MRI category 4a lesions. Ultrasound and elastography can help optimize therapy recommendations for BI-RADS-MRI category 4a lesions by our re-assessment proposal.

A low reproductive rate coupled with human activities has endangered the giant panda, a species endemic to southwest China. Although giant pandas feed almost exclusively on bamboo, they retain carnivorous traits and suffer from carnivorous diseases. Additionally, their immune system is susceptible to aging, resulting in a reduced ability to respond to diseases. This study aimed to determine the genes and pathways expressed differentially with age in blood tissues. The differentially expressed genes in different age groups of giant pandas were identified by RNA-seq. The elderly giant pandas had many differentially expressed genes compared with the young group (3 years old), including 548 upregulated genes and 401 downregulated genes. Further, functional enrichment revealed that innate immune upregulation and adaptive immune downregulation were observed in the elderly giant pandas compared with the young giant pandas. Meanwhile, the immune genes in the elderly giant pandas changed considerably, including genes involved in innate immunity and adaptive immunity such as PLSCR1, CLEC7A, CCL5, CCR9, and EPAS1. Time series analysis found that giant pandas store glycogen by prioritizing fat metabolism at age 11, verifying changes in the immune system. The results reported in this study will provide a foundation for further research on disease prevention and the energy metabolism of giant pandas.

Infectious bronchitis virus (IBV) causes avian infectious bronchitis (IB) and there are multiple serotypes worldwide originating from deletions, insertions, point mutations, and RNA recombination. In this study, a recombinant IBV, named CK/CH/MY/2020, was isolated from southwest China. Sequencing and phylogenetic analysis revealed that CK/CH/MY/2020 consists of 27,614 nucleotides and belongs to the GI-28 genotype. Moreover, the strain is a recombination product originating from three live attenuated vaccine strains (H120, 4/91, and LDT3-A). The recombination is complicated involving at least nine recombination sites; the first 3/5 portion is mainly composed of H120 and 4/91, and the second 2/5 contains LDT3-A. Pathogenicity analysis showed that CK/CH/MY/2020 could cause respiratory and kidney diseases in chickens resulting in moderate mortality. Therefore, the recombinant strain is more virulent than the attenuated vaccine strains. This study shows that even in the absence of wild strains, the recombination and revirulence of multiple attenuated vaccines could occur simultaneously, which also highlights the continuous evolution in IBV.

Bacillus species are among the most investigated beneficial bacteria and widely used in agricultural systems as biological control agents. Its biocontrol efficacy is controlled by diverse regulators. Cyclic diguanylate (c-di-GMP) is a nearly universal second messenger in bacteria and modulates various important physiological processes, including motility, biofilm formation, antifungal antibiotic production and host colonization. However, the impact of c-di-GMP on biocontrol efficacy of beneficial bacteria is unknown. Bacillus velezensis PG12 is an effective biocontrol strain against apple ring rot disease caused by Botryosphaeria dothidea. In this study, the contribution of c-di-GMP to biocontrol efficacy of B. velezensis PG12 was investigated. Deletion of single gene encoding diguanylate cyclase or phosphodiesterase did not affect its biocontrol efficacy against apple ring rot. However, artificial modulation of c-di-GMP level in the cells leads to a significant change of biocontrol efficacy, suggesting that c-di-GMP positively regulates biocontrol efficacy of B. velezensis PG12 against apple ring rot disease. More evidences indicate that c-di-GMP does not affect the antagonistic activity of B. velezensis PG12 against B. dothidea in vitro and in vivo, but positively regulates biofilm formation of B. velezensis PG12 and its colonization on apple fruits. Importantly, deletion of ydaK could rescue the inhibition of biofilm formation, bacterial colonization and biocontrol efficacy caused by low c-di-GMP level, indicating that YdaK is the potential c-di-GMP receptor to regulate biofilm formation, colonization and effective biological control. However, YdaK did not affect the antagonistic activity of B. velezensis PG12 against B. dothidea. Based on these findings, we propose that c-di-GMP regulates biofilm formation, subsequently the bacterial colonization on apple fruits and thus biocontrol efficacy of B. velezensis through its receptor YdaK. This is the first report showing that c-di-GMP plays a role in biocontrol efficacy of beneficial bacteria.

OBJECTIVE: To study the clinical diagnostic value of contrast-enhanced ultrasound (CEUS) in bladder occupied lesions. METHODS: 38 cases of conventional-ultrasound-found bladder occupied lesions did color Doppler flow imaging (CDFI) and CEUS checks. By comparing the difference between two types of blood flow imaging technologies in displaying the flow of bladder occupied lesions and observing the perfusion modes of contrast agents to enter lesions, the perfusion characteristics of CEUS were analyzed. Finally, they were contrasted with the surgical pathology results. RESULTS: Of all the 38 cases, there were 51 bladder occupied lesions, including 43 bladder malignant tumors, 2 bladder inverted papillomas, and 6 glandular cystitis lesions. The blood flow display rate of bladder occupied lesions was 100% using CEUS. Apparently, it was higher than that of CDFI (62.7%), and the result of these showed a statistically significant difference (P < 0.05). Using CEUS, 46 malignant lesions and 5 glandular cystitis lesions were indicated, and the diagnostic accuracy rate was 86.3%. CONCLUSION: CEUS can improve the blood flow display rate of bladder occupied lesions, and it can also observe the real-time blood flow of these lesions. It can help judge their nature and has a higher clinical value in differentiating the benign from the malignant.

Background: Because the halo around the tumor in shear wave elastography (SWE) is defined as the "stiff rim" sign, the diagnosis of breast lesions with the stiff rim sign is popular. However, only a few studies have described the stiff rim sign quantitatively. Objective: This study aimed to investigate the usefulness of the stiff rim sign in the diagnosis and tumor, node, metastasis stage of breast cancer. Methods: Two hundred and ten breast lesions were analyzed retrospectively. The maximum, mean, minimum Young's modulus (YM), and the YM standard deviation in the lesion, the peritumoral stiffness (shell), and the region containing lesion and shell were obtained. The suspicious SWE feature with the best diagnostic performance was chosen to downgrade or upgrade the Breast Imaging Reporting and Data System (BI-RADS) classification. The coincidence rates of SWE and B-mode ultrasound in T staging and their positive predictive value (PPV) for T staging were compared. Results: The presence of "stiff rim" sign was selected to upgrade or downgrade the BI-RADS classification because of its best performance. In pathological benign lesions, 18.9% (25 of 132) of lesions should undergo biopsy if BI-RADS combined with the stiff rim sign were referred while it was 57.6% (76 of 132) if BI-RADS alone was referred. The coincidence rate of T2 staging evaluated by SWE was significantly higher than B-mode ultrasound (about 30% increase, P < 0.001). The PPVs of SWE for T1 and T2 staging were higher than B-mode ultrasound (P < 0.05). Conclusions: BI-RADS combined with "stiff rim" sign is expected to improve the diagnostic performance of breast lesions to avoid unnecessary biopsy. The maximum diameter of the lesion measured in SWE is more accurate than B-mode ultrasound in the estimation of T staging, which is beneficial to the treatment and prognosis of breast cancer.

OBJECTIVE: To discuss the clinical application value of contrast-enhanced ultrasound (CEUS) in testicular occupied lesions. METHODS: Nine conventional-ultrasound-found testicular occupied lesions which underwent CEUS meantime were analyzed retrospectively. The CEUS perfusion pattern was compared with the surgical pathological result or follow-up findings. RESULTS: Among all the 9 testicular occupied lesions, there were 5 testicular malignant tumors, 1 testicular benign tumor, 1 testicular tuberculosis, and 2 testicular hematomas. CEUS diagnosed 6 testicular malignant tumors, 1 testicular benign tumor, and 2 testicular hematomas, and its diagnostic accuracy was about 88.9%. CONCLUSION: CEUS has high clinical application value in the differential diagnoses of benign and malignant testicular occupied lesions.

The widespread nature and economic importance of Infectious bronchitis virus (IBV) and interactions between IBV and the host immune response remain poorly understood. Understanding the mechanism of virus recognition via innate immunity can help resist IBV invasion. Retinoic acid-induced gene I-like receptor (RLRs) recognize virus RNA in virus infection, and LGP2 is a member of RLRs. According to the current studies, LGP2 exhibited certain inhibition in the virus, and there is a lack of investigation for chicken's LGP2. It is important to figure out the role of chLGP2 in host immune recognition of IBV. Our results showed that chLGP2 inhibited the proliferation of IBV Beaudette in cells. Also, chLGP2 can identify and combine with IBV RNA. The domains of chLGP2 were separately expressed and inspired by related literature, and the chLGP2 K30A mutant was constructed. Our results suggested its structural integrity and the adenosine triphosphatase (ATPase) activity are critical for IBV inhibiting activity. chTRBP was selected after CO-IP and Mass spectrometry test. We found chTRBP and chLGP2 are the interacting partners and promote mutual expression. Our study showed that chTRBP could also suppress IBV infections via chLGP2, which provided a basis for future innate immunity research for IBV.

The use of ultrasound images to acquire breast cancer diagnosis information without invasion can reduce the physical and psychological pain of breast cancer patients and is of great significance for the diagnosis and treatment of breast cancer. There are some differences in the texture of breast cancer between benign and malignant cases. Therefore, this paper proposes an adaptive learning method based on ultrasonic image texture features to identify breast cancer. Specifically, firstly, we used dictionary learning and sparse representation to learn the ultrasonic image texture dictionary of benign and malignant cases, respectively, and then used the combination of the two dictionaries to represent the test image to obtain the texture distribution characteristics of the test image under the two dictionary representations, which called the sparse representation coefficient. Finally, these above features were filtered by sparse representation and sent to sparse representation classifier to establish benign and malignant classification model. 128 cases were randomly divided into training and testing sets according to 2: 1 for training and testing. The proposed method has achieved state-of-the-art results, with an accuracy of 0.9070 and the area under the receiver operating characteristic curve of 0.9459. The results demonstrate that the proposed method has the potential to be used in the clinical diagnosis of benign and malignant breast cancer.

The potato (Solanum tuberosum) tuber is a swollen stem. Sprouts growing from the tuber nodes represent loss of apical dominance and branching. Long cold storage induces loss of tuber apical dominance and results in secondary branching. Here, we show that a similar branching pattern can be induced by short heat treatment of the tubers. Detached sprouts were induced to branch by the heat treatment only when attached to a parenchyma cylinder. Grafting experiments showed that the scion branches only when grafted onto heat- or cold-treated tuber parenchyma, suggesting that the branching signal is transmitted systemically from the bud-base parenchyma to the grafted stem. Exogenous supply of sucrose (Suc), glucose, or fructose solution to detached sprouts induced branching in a dose-responsive manner, and an increase in Suc level was observed in tuber parenchyma upon branching induction, suggesting a role for elevated parenchyma sugars in the regulation of branching. However, sugar analysis of the apex and node after grafting showed no distinct differences in sugar levels between branching and nonbranching stems. Vacuolar invertase is a key enzyme in determining the level of Suc and its cleavage products, glucose and fructose, in potato parenchyma. Silencing of the vacuolar invertase-encoding gene led to increased tuber branching in combination with branching-inducing treatments. These results suggest that Suc in the parenchyma induces branching through signaling and not by excess mobilization from the parenchyma to the stem.

Coronary artery disease has become the most dangerous diseases to human life. And coronary artery segmentation is the basis of computer aided diagnosis and analysis. Existing segmentation methods are difficult to handle the complex vascular texture due to the projective nature in conventional coronary angiography. Due to large amount of data and complex vascular shapes, any manual annotation has become increasingly unrealistic. A fully automatic segmentation method is necessary in clinic practice. In this work, we study a method based on reliable boundaries via multi-domains remapping and robust discrepancy correction via distance balance and quantile regression for automatic coronary artery segmentation of angiography images. The proposed method can not only segment overlapping vascular structures robustly, but also achieve good performance in low contrast regions. The effectiveness of our approach is demonstrated on a variety of coronary blood vessels compared with the existing methods. The overall segmentation performances si, fnvf, fvpf and tpvf were 95.135%, 3.733%, 6.113%, 96.268%, respectively.

GN/BT nanocomposites were fabricated via colloidal processing methods, and ceramics were sintered through two-step sintering methods. The microstructure and morphology were characterized by X-ray diffraction, high-resolution transmission electron microscopy, and field emission scanning electron microscopy. XRD analysis shows that all samples are perovskite phases, and the lattice parameters a and c almost decrease linearly with the increase of graphene nanosheets. The dielectric properties were tested by using precision impedance. The maximum dielectric constant at the Curie temperature for the nanocomposites with graphene addition of 3 wt % is about 16,000, almost 2 times more than that of pure BaTiO3 ceramics. The relaxation, band structure, density of states, and charge density distribution of GN/BT superlattices were calculated using first-principles calculations for the first time, and results showed the strong hybrid interactions between C 2p states and O 2p and Ti 3d orbitals.

Acrylamide is produced in a wide variety of carbohydrate-rich foods during high-temperature cooking. Dietary acrylamide is a suspected human carcinogen, and health concerns related to dietary acrylamide have been raised worldwide. French fries and potato chips contribute a significant proportion to the average daily intake of acrylamide, especially in developed countries. One way to mitigate health concerns related to acrylamide is to develop potato cultivars that have reduced contents of the acrylamide precursors asparagine, glucose and fructose in tubers. We generated a large number of silencing lines of potato cultivar Russet Burbank by targeting the vacuolar invertase gene VInv and the asparagine synthetase genes StAS1 and StAS2 with a single RNA interference construct. The transcription levels of these three genes were correlated with reducing sugar (glucose and fructose) and asparagine content in tubers. Fried potato products from the best VInv/StAS1/StAS2-triple silencing lines contained only one-fifteenth of the acrylamide content of the controls. Interestingly, the extent of acrylamide reduction of the best triple silencing lines was similar to that of the best VInv-single silencing lines developed previously from the same potato cultivar Russet Burbank. These results show that an acrylamide mitigation strategy focused on developing potato cultivars with low reducing sugars is likely to be an effective and sufficient approach for minimizing the acrylamide-forming potential of French fry processing potatoes.
