RESUMEN

Three prepackaged solid phase extraction (SPE) cartridges: two modified styrene divinylbenzene, Bond Elut ENV and Bond Elut PPL (Varian), and one N-vinylpyrrolidone (Strata-X, Phenomenex), were assessed for isolation of THM precursors from three surface waters in Manitoba, Canada. The dissolved organic matter (DOM) from the La Salle River (LR), Lake Winnipegosis (LW) and the Waterhen River (WR) were fractionated into hydrophobic (HPO) and hydrophilic (HPI) parts. ENV isolated less DOM (LR = 46.6 ± 1.5%; LW = 36.2 ± 1.4%; WR = 28.6 ± 2.2%) compared to PPL (LR = 50.2 ± 4.4%; LW = 47.9 ± 2.2%; WR = 37.3 ± 2.8%) and Strata (LR = 46.4% ± 1.0; LW = 51.6 ± 0.3%; WR = 31.9 ± 3.9%). The HPO fraction isolated by each SPE was characterized using Fourier Transform Infrared (FTIR) spectrochemical imaging. The FTIR spectra confirmed the HPO fractions were typical of humic-material and largely resembled fulvic acids; however, the PPL and Strata HPO isolates contained slightly more polysaccharides. The THM formation potential (THMFP) confirmed that the HPO fraction formed more THMs than the HPI. The HPO fraction isolated using ENV was found to have the lowest THMFP of all three SPEs in each waterbody; however, the specific THMFP (µgTHM/mgDOM) results indicated that ENV isolated THM precursors more effectively, as the Strata and PPL isolated a greater amount of non-THM forming material. All three SPE showed significant potential for implementation at water treatment plants as a simple tool to monitor THM precursors in source waters, enabling operators to adapt processes to improve drinking water quality.

A recent upgrade to the optics configuration of a thermal source FTIR microscope equipped with a focal plane array detector has enabled rapid acquisition of high magnification spectrochemical images, in transmission, with an effective geometric pixel size of ∼1 × 1 µm(2) at the sample plane. Examples, including standard imaging targets for scale and accuracy, as well as biomedical tissues and microorganisms, have been imaged with the new system and contrasted with data acquired at normal magnification and with a high magnification multi-beam synchrotron instrument. With this optics upgrade, one can now conduct rapid biodiagnostic ex vivo tissue imaging in-house, with images collected over larger areas, in less time (minutes) and with comparable quality and resolution to the best synchrotron source FTIR imaging capabilities.

The critical questions into the cause of neural degeneration, in Alzheimer disease and other neurodegenerative disorders, are closely related to the question of why certain neurons survive. Answers require detailed understanding of biochemical changes in single cells. Fourier transform infrared microspectroscopy is an excellent tool for biomolecular imaging in situ, but resolution is limited. The mid-infrared beamline IRENI (InfraRed ENvironmental Imaging) at the Synchrotron Radiation Center, University of Wisconsin-Madison, enables label-free subcellular imaging and biochemical analysis of neurons with an increase of two orders of magnitude in pixel spacing over current systems. With IRENI's capabilities, it is now possible to study changes in individual neurons in situ, and to characterize their surroundings, using only the biochemical signatures of naturally-occurring components in unstained, unfixed tissue. We present examples of analyses of brain from two transgenic mouse models of Alzheimer disease (TgCRND8 and 3xTg) that exhibit different features of pathogenesis. Data processing on spectral features for nuclei reveals individual hippocampal neurons, and neurons located in the proximity of amyloid plaque in TgCRND8 mouse. Elevated lipids are detected surrounding and, for the first time, within the dense core of amyloid plaques, offering support for inflammatory and aggregation roles. Analysis of saturated and unsaturated fatty acid ester content in retina allows characterization of neuronal layers. IRENI images also reveal spatially-resolved data with unprecedented clarity and distinct spectral variation, from sub-regions including photoreceptors, neuronal cell bodies and synapses in sections of mouse retina. Biochemical composition of retinal layers can be used to study changes related to disease processes and dietary modification.

Amyotrophic Lateral Sclerosis (ALS) is an untreatable, neurodegenerative disease of motor neurons characterized by progressive muscle atrophy, limb paralysis, dysarthria, dysphagia, dyspnae and finally death. Large motor neurons in ventral horns of spinal cord and motor nuclei in brainstem, large pyramidal neurons of motor cortex and/or large myelinated axons of corticospinal tracts are affected. In recent synchrotron Fourier Transform Infrared microspectroscopy (sFTIR) studies of ALS CNS autopsy tissue, we discovered a small deposit of crystalline creatine, which has a crucial role in energy metabolism. We have now examined unfixed, snap frozen, post-autopsy tissue sections of motor cortex, brain stem, spinal cord, hippocampus and substantia nigra from six ALS and three non-degenerated cases with FTIR and micro-X-ray fluorescence (XRF). Heterogeneous pigmented deposits were discovered in spinal cord, brain stem and motor neuron cortex of two ALS cases. The FTIR signature of creatine has been identified in these deposits and in numerous large, non-pigmented deposits in four of the ALS cases. Comparable pigmentation and creatine deposits were not found in controls or in ALS hippocampus and substantia nigra. Ca, K, Fe, Cu and Zn, as determined by XRF, were not correlated with the pigmented deposits; however, there was a higher incidence of hot spots (Ca, Zn, Fe and Cu) in the ALS cases. The identity of the pigmented deposits remains unknown, although the absence of Fe argues against both erythrocytes and neuromelanin. We conclude that elevated creatine deposits may be indicators of dysfunctional oxidative processes in some ALS cases.
