RESUMEN

A enrofloxacina é um dos antimicrobianos mais utilizados na avicultura industrial, e a deposição de resíduos em produtos avícolas, como os ovos, são de grande importância para a saúde pública. Na legislação brasileira não existe padronização do período de carência para o seu uso na produção avícola e não há Limite Máximo de Resíduo (LMR) fixado para enrofloxacina em ovos. Neste estudo, foi utilizado o kit de ELISA comercial (Bioo Scientific(r)) e a LC-MS/MS na pesquisa de enrofloxacina em ovos de 30 galinhas tratadas previamente via água de bebida, com 10mg/kg de enrofloxacina, durante cinco dias. Seis ovos foram coletados diariamente e analisados durante o tratamento e após a sua suspensão, durante 15 dias. A deposição de resíduos obteve níveis máximos no quinto dia de tratamento das aves, declinando gradativamente até não ser detectada a partir do nono dia de suspensão do tratamento. Considerando como base o LMR de 100µg/kg fixado pelo Brasil para tecidos comestíveis de aves e pela União Europeia para músculo, gordura e pele, após seis dias de suspensão do tratamento, os níveis de resíduos foram inferiores a esse limite, tendo como médias 37,43µg/kg na LC-MS/MS e 14,731µg/kg no ELISA. Dentro das condições deste estudo, um período de carência de seis dias seria mais adequado para utilização dos ovos para consumo humano. Foram detectados valores de resíduos nos ovos menores no ELISA em relação à LC-MS/MS para a mesma amostra, mas os dois métodos apresentaram concordância estatística entre si. A LC-MS/MS é o teste recomendado pela legislação brasileira para a análise de resíduos em alimentos; entretanto, pelos resultados obtidos, o kit de ELISA utilizado também pode ser aplicado na detecção de resíduos de enrofloxacina em ovos, com as vantagens de rapidez e simplicidade...


Enrofloxacin is one of the most used antibiotics in the poultry industry and the deposition of residues in poultry products, such as eggs, are of great concern to public health. In Brazilian law there is no standard withdrawal period for enrofloxacin in eggs and there is no Maximum Residue Limit (MRL) established for this antimicrobial in eggs. In this study, (Bioo Scientific(r)) commercial ELISA kit and LC-MS/MS were used to investigate enrofloxacin in eggs of 30 hens pretreated via drinking water at 10mg/kg of enrofloxacin for five days. Six eggs were collected daily and analyzed during treatment and after the end of treatment, for 15 days. Residues obtained maximum levels on the fifth day of treatment, declined gradually and were no longer detected from the ninth day to the end of treatment. Based on the MRL of 100mg/kg established for edible tissues of poultry by Brazillian law and for muscle, fat and skin, by the European Union, after six days of treatment withdrawal, the residue levels were below that limit, with the average of 37.43mg/kg in LC-MS/MS and 14.731mg/kg in ELISA. Within the conditions of this study, a withdrawal period of six days would be more appropriate to use the eggs for human consumption. The values obtained by ELISA for residues in eggs were lower than those obtained in LC-MS/MS for the same sample, however both methods showed statistical agreement. LC-MS/MS is the recommended method by Brazilian legislation for analysis of residues in food, however, according to the results the ELISA kit used can also be applied to the detection of enrofloxacin residues in eggs, with the advantages of speed and simplicity...

Polonium-210 ((210)Po) concentration was quantified in the muscle tissue and organs of two predatory marine fishes (Genypterus brasiliensis and Cynoscion microlepidotus) from Cabo Frio, Rio de Janeiro, Brazil. The species C. microlepidotus, a benthic carnivore, registered higher (210)Po in its tissue. The organs associated with digestion displayed the maximum radionuclide compared with other organs. The average activity was 2 mBq kg(-1) for G. brasiliensis and it was 6 mBq kg(-1) for C. microlepidotus. The activity concentrations varied significantly between the species and among organs.

Collibacillosis is considered one of the major diseases of the modern poultry industry, due to the significant losses it causes. Escherichia coli contributes not only to the disease itself, by causing weight loss of the birds, but also to the increase in carcasses condemnation during slaughter and processing. Detection of virulence factors in E. coli strains of the APEC pathotype contributes to the characterization and pathogenicity of this agent. PCR techniques have been very helpful in the search for genes that encode those virulence factors. This study aimed to detect the gene Fel A of E. coli by PCR and relate its positivity to low weight in broiler flocks with airsacculitis as diagnosed by the health inspection service. The study involved 40 flocks of broilers slaughtered in a single poultry slaughterhouse, under Federal Sanitary Inspection, located in the state of Rio Grande do Sul, Brazil. Three broilers were randomly selected to obtain one "pool" of three tracheas for each PCR. DNA was extracted using phenol-chloroform and amplified using a pair of primers specific to gene Fel A of E. coli. Of the 40 flocks analyzed by PCR, 35% (14/40) were positive for the gene Fel A. PCR was an effective technique for the detection of gene Fel A in broiler flocks. A colibacilose é considerada uma das principais doenças da indústria avícola moderna, devido aos grandes prejuízos econômicos causados. A Escherichia coli contribui não só para a doença em si, levando à perda de peso das aves, bem como para o aumento da taxa condenação de carcaças durante o abate e processamento. A detecção de fatores de virulência de cepas de E. coli do patotipo APEC colabora para a caracterização de sua patogenicidade e as técnicas de PCR têm sido muito úteis na pesquisa de genes que os codificam. Este estudo objetivou diagnosticar E. coli pela detecção o gene Fel A por PCR e relacionar a positividade para este agente com o baixo peso em frangos de corte provenientes de lotes condenados por aerossaculite. Foram estudados 40 lotes de frangos de corte abatidos em um matadouro avícola sob Inspeção Sanitária Federal, localizado no Estado do Rio Grande do Sul. Foram colhidos aleatoriamente 3 frangos e obtidos "pools" de três traqueias em cada um deles para PCR. O DNA foi extraído pelo método de fenol-clorofórmio e amplificado com pares de "primers" específicos para gene Fel A de E. coli. Dos 40 lotes analisados pela PCR, 35% (14/40) foram positivos para o gene Fel A. A PCR foi eficaz para a detecção do gene Fel A em lotes de frangos de corte e houve relação entre a presença do gene Fel A, a queda de peso e aumento na taxa de aerossaculite...

The production of chicken feet is primarily intended for foreign markets, and there is still no specific legislation in Brazil that determines the quality standard of these products. The bacteriological quality of chicken feet was evaluated as a product for human consumption at different steps of the technological processes. Eighty broiler feet from 20 lots at 4 steps of processing were collected for quantitative analysis, total count of aerobic mesophilic bacteria, and determining the most probable number of coliforms and fecal coliforms. Thirty-eight pools of 15 broiler feet each from 19 lots were used for qualitative analysis and the isolation of Salmonella enterica spp. and Escherichia coli O157:H7. Escherichia coli O157:H7 was not found in any of the samples. Salmonella spp. were isolated in 68% (13/19) of the lots. The Salmonella Schwarzengrund serotype was found in 12 of the 13 lots of positive samples and the Salmonella Anatum and Salmonella Corvallis serotypes were identified in the remaining lot. Processing is effective in reducing contamination by mesophilic bacteria, coliforms, and Salmonella spp. in these products. This work constitutes the first study in Brazil on microbiological quality of chicken feet.

The phytoplankton assemblages from eight reservoirs of the Paranapanema River were studied during two consecutive years. Chlorophyceae and Bacillaryophyceae dominated in richness. The observed high number of taxa, 234, reflects the extensive sampling programme and evidences the necessity of considering the whole hydrograph basin to assess the biodiversity status of inland water ecosystems. The dams had a negative effect on phytoplankton richness, with higher number of taxa associate to riverine (non-regulated) stretches. The tributary rivers also exhibited high species richness, showing the importance of considering the lateral dimension, in addition to the longitudinal one, for aquatic biota inventories in large river basins/reservoirs. Richness and diversity were also positively influenced by the connectivity with lateral wetlands (macrophyte-dominated lakes) due to the periphyton influence. The phytoplankton abundance/biomass was not influenced by higher water retention time. Higher values occurred in the middle basin stretches (river-passage reservoirs) due to the increase in the trophic conditions. There was a positive correlation with phosphorus. Poorer light conditions in the cascade do not limit the phytoplankton biomass, with assemblages dominated by species tolerant to turbulent conditions and high mineral turbidity. Bacillariophyceae and Cryptophyceae dominated numerically. The first group (unicellular forms) was prominent in the large and oligotrophic upstream reservoirs. The second was highly abundant in the river-passage (low retention time), and more eutrophic, reservoirs. Cyanophyceae growth is probably controlled by advection processes (wash-out effect). The zooplankton does not control the phytoplankton biomass and the diversity of both groups is positively associated in the cascade. The structure of the phytoplankton assemblages showed to be a good indicator of the operationally distinct reservoirs of the Paranapanema cascade and also reflected the changes in the trophic conditions along the basin.

OBJECTIVE: To detect differences in the prevalence of dyslipidemia in adult ambulatory laboratory tests from three different health care providers, in Salvador-Brazil, 1995. METHODS: The study was carried out using a probabilistic sample of 24 out of 104 laboratories (approximately equal to 25%) which performed the same enzymatic method for lipid tests with the same quality control. Laboratory tests results were separated into three groups, according to health care provider: Sistema Unico de Saúde (SUS, governamental), Health Maintenance Organizations (HMO), and Private or Patients Health Insurance (PHI). Criteria for dyslipidemias in mg/dl were: total cholesterol > or = 240; LDL-c > or = 160; HDL-c < 35 and triglycerides > or = 200. Prevalence rates, 95% confidence intervals (CI) and chi (2) test were used in the analysis. RESULTS: From the 5464 adult subjects, 14.8% were from SUS, 31.6%. from HMO and 53.6% PHI. The highest prevalence for hypercholesterolemia--28.0% (CI 24.0; 32.0) and for LDL-c > or = 160mg/dL--30.4% (CI 27.6; 33.2), were found in the HMO group. PHI had the highest prevalence for HDL-c < 35mg/dL--12.3% (CI 10.0; 13.8), and for hypertrygliceridemia--17.8% (CI 16.3; 19.3). Most of the differences among health providers were statistically significant. CONCLUSION: The best lypid profile observed in subjects from SUS suggests social differences in the prevalence of dyslipidemias. As compared to other dyslipidemias, HDL-c < 35mg/dL prevalence was lower than expected in all three groups. The data may provide insights to medical doctors and other health care professionals regarding the questions of dyslipidemias. It can also provide objective information to the patients and encourage them to change their life styles.

OBJETIVO - Detectar diferenças nas prevalências das dislipidemias em adultos da demanda laboratorial de três diferentes prestadores de assistência médica, em Salvador, 1995. MÉTODOS - Estudo seccional, clínico epidemiológico, baseado em resultados das dosagens de lípides séricos de uma amostra probabilística de 25 'por cento' dos 104 laboratórios que usavam o mesmo método de dosagem e o mesmo padräo para controle de qualidade. O critério de dislipidemias foi o do II Consenso Brasileiro de Dislipidemias para adultos. Os resultados foram agrupados conforme procedência: Sistema Unico de Saúde (SUS), convênios (C) e seguros privados + particulares (SP). Análise: prevalência com intervalos de confiança (IC) a 95 'por cento', e teste do X². RESULTADOS - Dos 5.464 adultos com mensäo da procedência, 14,8 'por cento' do SUS, 31,6 'por cento' dos C e 53,6 'por cento' S + P. As mais elevadas prevalências de hipercolesterolemia - 28,0 'por cento' (IC 24,0; 32,0) e de LDL-c elevado - 30,4 'por cento' (IC 27,6; 33,2) foram detectados no grupo C. No grupo S + P concentram-se as maiores prevalências de HDL-c anormal - 12,3 'por cento' (IC 10,0; 13,8) e hipertrigliceridemia - 17,8 'por cento' (IC 16,3; 19,3). Quase todas as diferenças entre as prevalências foram estatisticamente significantes, favoráveis às taxas mais baixas do SUS. CONCLUSÄO - O melhor perfil lipídico do grupo SUS sugere diferenças sociais nas prevalências das dislipidemias. Para os três prestadores de assistência, a prevalência do HDL-c menor que 35mg/dL foi baixa quando comparada às das demais dislipidemias. Os resultados podem ser inferidos para a prática médica dos respectivos prestadores de assistência e indicam a necessidade de informaçäo e de orientaçäo dos usuários para a reduçäo das dislipidemias e melhoria do estilo de vida.

OBJETTIVO - Determinar a prevalência das dislipidemias em adultos da demanda laboratorial não-hospitalar da cidade de Salvador (BA). MÉTODOS - Casuística procedente de amostra probalística de 25 'por cento' dos laboratórios não-hospitalares da cidade que usavam o método enzimático para dosagem dos lípides séricos e controle de qualidade da Sociedade Brasileira de Patologia Clínica (93 'por cento'); LDL estimado pela fórmula de Friedewald. Sorteados os meses ímpares de 1995 para o estudo. Critérios para dislipidemias, em mg/dl: colesterol 'maior ou igual' 240; LDL 'maior ou igual' 160; HDL 'menor' 35 e triglicérides 'maior ou igual' 200. Análise: prevalências. seus intervalos de confiaça (IC) a 95 'por cento' e qui-quadrado (X²). RESULTADOS - Excluídos uma recusa e um laboratório que arquivava laudos só por 3 meses, foram analisados dados de 24 dos 26 laboratórios amostrados. Dos 7392 adultos, 65,5 'por cento' eram mulheres. Prevalências estimadas e valores ... "Fórmulas"... Todas as prevalências diferiram significantemente inter-gênero (p,0,05 a p<0,001). Também foram mais freqüentes nas mulheres os níveis indesejáveis do colesterol e LDL. CONCLUSÄO - As dislipidemias são importante fator de risco para aterosclerose na demanda laboratorial de Salvador em ambos os gêneros. Os resultados subsidiam os médicos para incentivar mudanças no estilo de vida que conduzam seua pacientes a níveis lipídicos desejáveis

PURPOSE: To determine the prevalence of dyslipidemia in non-hospital laboratory tests of adults from Salvador, Brazil. METHODS: The study was carried out in subjects from a probabilistic sample of 25% of a total of 104 local laboratories that used the same enzymatic method for lipid analysis with the quality control as recommended by the Brazilian Society of Clinical Analyses. These represented 93% of all non-hospital laboratories of Salvador in 1995. The odd months of 1995 were selected for sampling in the present study. Criteria for dyslipidemias were: total cholesterol > or = 240; LDL > or = 160; HDL < 35 and triglycerides > or = 200 mg/dl. Prevalence rates and their 95% confidence intervals (CI) and chi 2 test were used in the analyses. RESULTS: Tests from two of the 26 laboratories were not used in the analyses due to file problems or refusal. 7,392 adults were screened, 65% female. We observed prevalence (95% CI) of hypercholesterolemia in 30.0 (27.8; 32.2)%, high LDL in 30.1 (30.8; 35.4)% and hypertriglyceridemia in 30.4 (29.0; 31.4)% of the females. As to the male subjects, prevalences were; 24.0 (20.5; 27.5)% for hypercholesterolemia, 26.1 (22.4; 29.3)% for high LDL, 27.6 (25.7; 29.5)% for hypertriglyceridemia. All gender differences were significant. Low HDL occurred in 15.9 (14.2; 17.8)% of males and in 8.0 (7.1; 8.9)% of females. CONCLUSION: Dyslipidemia is an important risk factor observed in non-hospital laboratory tests of men and women in Salvador. Our data may provide physicians and other health care professionals with objective information to encourage life-style changes.

210Po and 210Pb concentrations in urine, hair and skin smear samples from individuals using phosphated fertilizers have been compared with a control group of occupationally unexposed individuals. Urine and hair samples of the test group showed slightly higher concentrations of 210Po and 210Pb than those observed for the control group. These concentrations remained, however, lower than those for uranium mine workers. Skin smear values indicated contamination by direct contact with dust from fertilizers and this may contribute to skin cancer induction in this risk population.

210Po and 210Pb concentrations in urine and hair samples from individuals working in different areas of the uranium industrial complex of Poços de Caldas (Brazil) have been determined and compared with a control group of occupationally unexposed individuals. In the control group, smokers showed higher urinary excretion of these radionuclides. Such difference could not be detected in the workers group, which showed an increase in urinary excretion of 210Po for the group working in the chemical laboratory. Accumulation of 210Po and 210Pb in hair showed the same trends as for urinary excretion. The workers group, with the exception of the office employees, showed a 210Po/210Pb ratio > 1 in hair.

Contamination of vegetables and associates soils by 210Pb, originating either by natural fallout from uranium or by the processing mills of Poços de Caldas (CIPC), State of Minas Gerais, Brazil is estimated by comparison with similar control samples collected from areas distant from the mining and milling operations. For lichens and bryophytes, the mean values of 210Pb concentrations were significantly higher than those of the control region. Similarly, vegetables and soils from kitchen-gardens located near the mill contained much higher concentrations of 210Pb than those observed for other regions.

Data are presented for 210Pb and 210Po concentrations in three species of molluscs collected in the intertidal region of the coast of Rio de Janeiro, Brazil. Lead-210 activity in the soft tissue of the edible mussel Perna perna was 20 times higher than that found in its shell. The visceral mass contained the largest activity for both radionuclides, which is attributed to the feeding habits of the molluscs. Animals collected from Ponta Negra beach (open sea) showed higher concentrations of isotopes than those collected from Boa Viagem beach (within Guanabara Bay).

RESUMEN

Bunodosoma caissarum, a carnivorous marine species exclusive to the Brazilian southern coast, recognized as a sensitive bioindicator of artificial radioactive pollution, also shows a great capacity for concentrating natural alpha-emitters. The authors have measured the alpha-emitters 210Po and its precursor 210Pb in this animal. The average concentrations found are greater (by a factor of 4.6) for 210Po measured in samples gathered on Ponta Negra Beach (protected outer coast) than the values obtained in specimens collected on Boa Viagem Beach (closed bay) along the Rio de Janeiro coast, Brazil. The same was also found for 210Pb (a factor of 2.5).
