This study aims to develop a nano-sized fluoridated layered double hydroxide (LDH)-based release system via hydrothermal treatment for the controlled delivery of fluoride (F-) ions in the oral environment. The synthesis of conventional LDH-type (C-LDH) precursor nanomaterials was conducted using a co-precipitation method at constant pH, and the nanoparticulate-LDH (N-LDH) was synthesized by a hydrothermal procedure. Fluoride LDH (F-LDH) products were obtained through indirect synthesis using the precursor ion-exchange technique by varying the agitation time (2 and 24 h) and temperature (25 and 40 °C) to produce 12 material samples. The materials were characterized by energy dispersive x-ray, hexamethyldisilazane, digital radiography x-ray, Fourier-transform infrared, thermogravimetric analysis, and scanning electron microscopy. Additionally, the F-release kinetic profile was evaluated for 21 d in neutral and acid media with mathematical model analysis. Products with varying F-quantities were obtained, revealing specific release profiles. In general, there was a higher F-release in the acid medium, with emphasis on F-LDH-8. Fluoride-LDH and controlled fluoride delivery was successfully obtained, proving the potential of these nanomaterials as alternative anti-caries agents.

Introducción: Cryptococcus neoformans es una levadura encapsulada, que se encuentra en fuentes ambientales, incluyendo excrementos de palomas. Tiene una gran relevancia clínica porque es el agente etiológico de la criptococosis, considerada una causa importante de mortalidad en personas inmunocomprometidas en todo el mundo. Objetivo: determinar la prevalencia y susceptibilidad de C. neoformans aislados del suelo y excremento de palomas en la ciudad de Maceió ­ Alagoas, Brasil. Metodología: Se recolectaron 150 muestras (50 de excretas secas, 50 de excretas frescas y 50 del suelo) y cultivaron en Agar Dextrosa Sabouraud. Las colonias aisladas sugestivas de C. neoformans se sometieron a identificación y caracterización mediante análisis morfológicos, actividad de la enzima fenol-oxidasa, sensibilidad a la cicloheximida, desarrollo a 37°C, hidrólisis de urea, asimilación de carbono y nitrógeno y fenotipificación en medio canavanina-glicina-azul de bromotimol. La prueba de susceptibilidad antifúngica se realizó utilizando la técnica de difusión en agar. Resultados: se obtuvieron 36 (24%) muestras positivas para C. neoformans, de las cuales 33 fueron de excretas secas y 3 del suelo cercano al excremento. El perfil de susceptibilidad fue de 100.0% para anfotericina B y 87.4% para ketoconazol, no obstante, hubo un alto porcentaje de resistencia a fluconazol (91.5%) e itraconazol (80.0%). Conclusión: estos hallazgos confirman que las excretas de palomas secas son reservorios de C. neoformans en el medio ambiente, caracterizando un problema de salud única. Además, la anfotericina B exhibió una alta actividad in vitro, representando buena alternativa en el tratamiento de la criptococosis.


Introdução: Cryptococcus neoformans é um fungo leveduriforme encapsulado, encontrado em fontes ambientais, incluindo excretas de pombos. Apresenta grande relevância clínica por ser agente etiológico da criptococose, considerada importante causa de mortalidade em indivíduos imunocomprometidos em todo o mundo. Objetivo: determinar a prevalência e suscetibilidade de C. neoformans isolados do solo e de excretas de pombos na cidade de Maceió, Alagoas. Metodologia: foram coletadas 150 amostras (50 de excretas secas, 50 de excretas frescas e 50 do solo) e cultivadas em Ágar Sabouraud Dextrose. As colônias isoladas sugestivas de C. neoformans foram submetidas à identificação e caracterização por meio de análises morfológicas, atividade da enzima fenoloxidase, sensibilidade à cicloheximida, termotolerância à 37ºC, hidrólise da ureia, assimilação de carbono e nitrogênio e quimiotipagem em ágar L-canavanina-glicina-azul de bromotimol. O teste de suscetibilidade antifúngica foi realizado por meio da técnica de difusão em ágar. Resultados: foram obtidas 36 (24%) amostras positivas para C. neoformans, das quais 33 (91,6%) foram provenientes de excretas secas e 3 (8,3%) do solo próximo a excrementos. O perfil de suscetibilidade foi de 100,0% para a anfotericina B e 87,4% para o cetoconazol, não obstante, verificou-se alto percentual de resistência ao fluconazol (91,8%) e itraconazol (80%). Conclusão: estes achados confirmam que excretas de pombos secas são reservatórios de C. neoformans no ambiente, caracterizando um problema de saúde única. Além disso, a anfotericina B apresentou elevada atividade, in vitro, representando boa alternativa no tratamento da criptococose.

Thimerosal (TH), an organomercurial compound, is used as a preservative in vaccines and cosmetics. Its interaction with human hemoglobin (Hb) was investigated under physiological conditions using biophysical and biological assays, aiming to evaluate hazardous effects. TH interacts spontaneously with Hb (stoichiometry 2:1, ligand-protein), preferably by electrostatic forces, with a binding constant of 1.41 × 106 M-1. Spectroscopic data allows to proposing that TH induces structural changes in Hg, through ethylmercury transfer to human Hb-Cys93 residues, forming thiosalicylic acid, which, in turn, interacts with the positive side of the amino acid in the Hb-HgEt adduct chain. As a consequence, inhibition of Hb-O2 binding capacity up to 72% (human Hb), and 50% (human erythrocytes), was verified. Dose-dependent induction of TH forming advanced glycation end products (AGE) and protein aggregates (amyloids) was additionally observed. Finally, these results highlight the toxic potential of the use of TH in biological systems, with a consequent risk to human health.

Purpose: Quercetin is a flavonoid known for its therapeutic properties and for forming complexes. Although the antimony-quercetin (SbQ) complex has been produced before, no previous exploration of its characteristics has been published in literature. Thus, this study aimed to characterize this complex, assess its stability and investigate its complexation site through its antibacterial activity. Methods: The SbQ complex was synthetized using Sb(III) potassium tartrate trihydrate and quercetin anhydrous (1:1) (v/v) as a solution and dried using three methods: rotaevaporation, lyophilization and spray drying. The material, in solution, was analyzed by UV-vis and fluorimetry; and, in the powder, by X-ray diffraction (XRD), both scanning electronic and fluorescence microscopy and infrared spectroscopy (FT-IR). Antimicrobial activity was evaluated via broth microdilution. Results: UV-vis exhibited a shoulder peak at 291 nm indicating metal chelation at C-ring of quercetin and confirmed 1:1 stoichiometry. Spectrofluorimetry showed an increase of intensity with the complex formation with an emission band (525 nm). After drying, XRD and SEM indicated loss of crystallinity and a difference in shape and size of the complex compared to its precursors. FT-IR suggested by a shift of frequency of the carbonyl group (1661 cm-1) that the quercetin bond to antimony by the C-3, followed by positions C-5 and C-4 carbonyl, which has been confirmed by MIC through the structure-activity relationship of the antibacterial activity of quercetin. Conclusion: These results provided a characterization of SbQ complex with the confirmation of its binding site, working as a guide for future studies involving this complex.

Layered double hydroxides have received more attention from researchers due to their range of applications, ease of synthesis and low cost of production. With broader knowledge about solar radiation effects on the body, the use of sunscreens has become even more important. The ability of some nanostructures, such as layered double hydroxides, to act as matrices has made it possible to obtain improvements in photoprotective formulations, with solutions to problems caused by radiation and sunscreens. This review article brings together the most recent advances of these clays, the layered double hydroxides, applied to photoprotection.

RESUMO Os hidróxidos duplos lamelares vêm ganhando maior atenção de pesquisadores, em decorrência da gama de aplicações, da facilidade de síntese e do baixo custo de produção. Com o crescente conhecimento sobre diversos efeitos da radiação solar sobre o organismo, a utilização de fotoprotetores tem se tornado indispensável. A capacidade de nanoestruturas, como os hidróxidos duplos lamelares, atuarem como matrizes possibilitou melhorias nas formulações fotoprotetoras, tendo em vista os problemas provocados pela radiação, bem como por alguns filtros solares. Este artigo de revisão reúne os avanços mais recentes destas argilas, os hidróxidos duplos lamelares, aplicados à fotoproteção.

ABSTRACT It is widely known that high fat diet (HFD) can contribute to the advent of health problems. Recent studies have indicated that obesity imposes a hemodynamic overload to the kidneys. In order to further investigate such injuries, two groups of six Swiss mice each were fed with a controlled AIN93G diet or a high fat (AIN93G modified) diet for eight weeks. Blood samples were collected to determine the hormonal, lipid profile, glucose, urea, and creatinine levels. Histopathological and immunohistochemical analysis were carried out to analysis the kidney damage. Fractions of renal membranes were prepared to assess the Na,K-ATPase activity, lipid peroxidation, total cholesterol, and phospholipid content. The results indicated that the blood lipid profile, urea and creatinine was not altered by the HFD. On the other hand, it was observed in HFD diet mice elevated glucose blood levels along with an augment on insulin and a decrease on corticosterone release. HFD provoked a reduction in the diameter of the convoluted tubules and cell volume in Bowman's capsule and an increased number of positive cells with Na,K-ATPase, but reduced the Na,K-ATPase activity and the cholesterol content in the kidney cell membrane but favored the lipid peroxidation.

The ever-increasing demand for natural products and biotechnology derived from bees and ultra-modernization of various analytical devices has facilitated the rational and planned development of biotechnology products with a focus on human health to treat chronic and neglected diseases. The aim of the present study was to prepare and characterize polymeric nanoparticles loaded with Brazilian red propolis extract and evaluate the cytotoxic activity of "multiple-constituent extract in co-delivery system" for antileishmanial therapies. The polymeric nanoparticles loaded with red propolis extract were prepared with a combination of poly-ε-caprolactone and pluronic using nanoprecipitation method and characterized by different analytical techniques, antioxidant and leishmanicidal assay. The red propolis nanoparticles in aqueous medium presented particle size (200-280 nm) in nanometric scale and zeta analysis (-20 to -26 mV) revealed stability of the nanoparticles without aggregation phenomenon during 1 month. After freeze-drying method using cryoprotectant (sodium starch glycolate), it was possible to observe particles with smooth and spherical shape and apparent size of 200 to 400 nm. Attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and thermal analysis revealed the encapsulation of the flavonoids from the red propolis extract into the polymeric matrix. Ultra performance liquid chromatography coupled with diode array detector (UPLC-DAD) identified the flavonoids liquiritigenin, pinobanksin, isoliquiritigenin, formononetin and biochanin A in ethanolic extract of propolis (EEP) and nanoparticles of red propolis extract (NRPE). The efficiency of encapsulation was determinate, and median values (75.0 %) were calculated using UPLC-DAD. 2,2-Diphenyl-1-picryhydrazyl method showed antioxidant activity to EEP and red propolis nanoparticles. Compared to negative control, EEP and NRPE exhibited leishmanicidal activity with an IC50 value of ≅38.0 µg/mL and 31.3 µg/mL, 47.2 µg/mL, 154.2µg/mL and 193.2 µg/mL for NRPE A1, NRPE A2, NRPE A3 and NRPE A4, respectively. Nanoparticles loaded with red propolis extract in co-delivery system and EEP presented cytotoxic activity on Leishmania (V.) braziliensis. Red propolis extract loaded in nanoparticles has shown to be potential candidates as intermediate products for preparation of various pharmaceutical dosage forms containing red propolis extract in the therapy against negligible diseases such as leishmaniasis. Lipophorin (Lp) is the main haemolymphatic lipoprotein in insects and transports lipids between different organs. In adult females, lipophorin delivers lipids to growing oocytes. In this study, the interaction of this lipoprotein with the ovaries of Rhodnius prolixus was characterised using an oocyte membrane preparation and purified radiolabelled Lp (125I-Lp). Lp-specific binding to the oocyte membrane reached equilibrium after 40-60 min and when 125I-Lp was incubated with increasing amounts of membrane protein, corresponding increases in Lp binding were observed. The specific binding of Lp to the membrane preparation was a saturable process, with a Kdof 7.1 ± 0.9 x 10-8M and a maximal binding capacity of 430 ± 40 ng 125I-Lp/µg of membrane protein. The binding was calcium independent and pH sensitive, reaching its maximum at pH 5.2-5.7. Suramin inhibited the binding interaction between Lp and the oocyte membranes, which was completely abolished at 0.5 mM suramin. The oocyte membrane preparation from R. prolixus also showed binding to Lp from Manduca sexta. When Lp was fluorescently labelled and injected into vitellogenic females, the level of Lp-oocyte binding was much higher in females that were fed whole blood than in those fed blood plasma.

OBJETIVO: Avaliar a ação cicatrizante de própolis verde e vermelha, correlacionando ao teor de flavonoides MÉTODOS: Foi realizada a quantificação de flavonoides totais dos extratos etanólicos de própolis verde e vermelha para posterior incorporação em pomada base a 20%. Utilizou-se 20 ratos Wistar distribuídos em quatro grupos: solução salina a 0,9% (S), pomada base (B), pomada própolis verde (G), pomada própolis vermelha (R), todos submetidos à confecção de lesões excisivas na região mediana do dorso. Os ratos foram tratados diariamente durante 15 dias. Neste período foram observados peso; temperatura corporal; diâmetro das feridas. Para análise histológica, amostras das feridas foram coletadas. Ao fim do experimento foram realizadas coleta sanguínea e remoção do rim e fígado para análises bioquímica e histológica. RESULTADOS: Os teores de flavonoides totais das própolis verde (4,50 %) e vermelha (5,92 %) foram elevados (>2 %), mas, embora a segunda apresente teor maior que a primeira, a evolução da própolis verde, macro e histologicamente, foi melhor na reparação das feridas. Não foram observadas nefro ou hepatotoxicidade, resultado corroborado pelos ensaios bioquímicos (TGP e albumina). A própolis influiu na redução de colesterol total, triglicerídeos e glicemia. CONCLUSÃO: Não foi possível correlacionar o teor de flavonoides totais com a ação cicatrizante da própolis. O dado revela a necessidade da elucidação dos flavonoides encontrados em cada classe de própolis para desvendar qual (ou quais) flavonoide(s) seria(m) representativo(s) no processo cicatricial.

This reveals the need for elucidation of the flavonoids found in each class of propolis to unravel which one(s) would be important for the healing process.

Despite the advent of molecular genetics, cell therapy and automation of procedures, the possibility of transmission ofinfection by bacterial contamination of blood components improves in importance, observing as an adverse event of bloodtransfusion the transfusion reaction known as bacterial contamination. During the analysis performed by the qualitycontrol of Hemoterapy Service it was found that there is a variable incidence of bacterial contamination of bloodcomponents in blood bags. These data showed that the proportion of patients undergoing blood transfusions could beexposed to microorganisms present in blood bags assessed as positive by the culture procedure and thuscould be acquiredsome kind of infection due to this contamination. Apesar del advenimiento de la genética molecular,la terapia celular y la automatización de los procedimientos, laposibilidad de transmisión de la infección por contaminación bacteriana de los componentes de la sangre crece enimportancia, al ser visto como un evento adverso dela reacción a la transfusión conocido como reacción transfusional porcontaminación bacteriana. Se constató durante las pruebas realizadas para el control de calidad del Departamento deHematología que hay una incidencia variable de la contaminación bacteriana de los productos de la sangre en las bolsas desangre. Estos datos mostraron que la mayoría de lospacientes que se someten a transfusiones de sangrepodrían estarexpuestos a los microorganismos presentes en las bolsas de sangre evaluadas como positivas por el procedimiento decultura y, así, adquirir algún tipo de infección debido a esta contaminación. Entre todas las medidaspara prevenir ycontrolar la contaminación bacteriana de los productos sanguíneos, deben ser destacados los procedimientos de asepsiaadecuados para el sitio de punción del brazo del donante, así como los de limpieza de las manos, que habitualmentedeben ser incorporados en todos los bancos de sangre; además del uso de guantes desechables durante laejecución detodas las actividades.
