RESUMEN

Avulsion injury results in motoneuron death due to the increased excitotoxicity developing in the affected spinal segments. This study focused on possible short and long term molecular and receptor expression alterations which are thought to be linked to the excitotoxic events in the ventral horn with or without the anti-excitotoxic riluzole treatment. In our experimental model the left lumbar 4 and 5 (L4, 5) ventral roots of the spinal cord were avulsed. Treated animals received riluzole for 2 weeks. Riluzole is a compound that acts to block voltage-activated Na+ and Ca2+ channels. In control animals the L4, 5 ventral roots were avulsed without riluzole treatment. Expression of astrocytic EAAT-2 and that of KCC2 in motoneurons on the affected side of the L4 spinal segment were detected after the injury by confocal and dSTORM imaging, intracellular Ca2+ levels in motoneurons were quantified by electron microscopy. The KCC2 labeling in the lateral and ventrolateral parts of the L4 ventral horn was weaker compared with the medial part of L4 ventral horn in both groups. Riluzole treatment dramatically enhanced motoneuron survival but was not able to prevent the down-regulation of KCC2 expression in injured motoneurons. In contrast, riluzole successfully obviated the increase of intracellular calcium level and the decrease of EAAT-2 expression in astrocytes compared with untreated injured animals. We conclude that KCC2 may not be an essential component for survival of injured motoneurons and riluzole is able to modulate the intracellular level of calcium and expression of EAAT-2.

RESUMEN

We present the design and implementation of a new, modular gas target suitable for high-order harmonic generation using high average power lasers. To ensure thermal stability in this high heat load environment, we implement an appropriate liquid cooling system. The system can be used in multiple-cell configurations, allowing us to control the cell length and aperture size. The cell design was optimized with heat and flow simulations for thermal characteristics, vacuum compatibility, and generation medium properties. Finally, the cell system was experimentally validated by conducting high-order harmonic generation measurements using the 100 kHz high average power HR-1 laser system at the Extreme Light Infrastructure Attosecond Light Pulse Source (ELI ALPS) facility. Such a robust, versatile, and stackable gas cell arrangement can easily be adapted to different experimental geometries in both table-top laboratory systems and user-oriented facilities, such as ELI ALPS.

RESUMEN

We present a novel polarization alignment technique based on windowed Fourier-transform (WFT) spectral interferometry to determine the wavelength-dependent orientation of the principal polarization axes of photonic crystal fibers (PCFs). To test the technique, a commercially available, 82.5-cm-long HC-800-02 type hollow-core PCF was measured. The angles belonging to the fast and the slow principal axes of the fiber were determined from the peak intensity values of the ridges in the WFT map at different wavelengths. We demonstrate that the orientation of the principal polarization axes of the tested PCF is wavelength-dependent. The precision of the angle measurement was better than 0.3°.

RESUMEN

We present a detailed study on the generation of widely tunable visible light through four wave mixing in specifically designed micro-structured fibers. The fiber's properties are optimized for an efficient conversion to the visible and near infrared with a combined tunability from 620 to 910 nm of a picosecond Yb-doped tunable source for biomedical applications.

Applications based on photonic crystal fibers depend strongly on their dispersion properties that might differ from the desired specifications due to deficiencies in the manufacturing process. Since dispersion characteristics might also be affected by the placement of the fiber, in this paper the effect of various placements on the chromatic dispersion properties of a commercially available HC-800-02 photonic crystal fiber was investigated between 760 and 870 nm with Fourier-transform spectral interferometry. To test the scaling of dispersion with fiber length, samples of different lengths ranging from 10 to 97 cm were used in the measurements. It was found that the dispersion properties of the orthogonal directions were different. The dispersion parameter showed small dependence on the placement and fiber length. The polarization-mode dispersion (PMD) of the fiber was measured using an indirect and a direct technique. To retrieve the PMD directly in the case of the shorter fibers where the fringes were too sparse for the Fourier method, the so-called minima-maxima method was employed. The precision was comparable with both techniques; however, the direct approach proved to be more accurate when longer samples were measured, and the indirect method seemed to be more reliable in the case of shorter fibers.
