RESUMEN

Antimicrobial use (AMU) is critical to preserving animal health and welfare. However, the development of antimicrobial resistance (AMR) represents a public health threat. Although most antimicrobials used on the farm require a veterinarian prescription, farmworkers make daily on-farm treatment decisions. Therefore, farmworker training is vital to promote responsible AMU. This research project aimed to evaluate the effect of farmworker antimicrobial stewardship (AMS) training on the quantity of AMU on dairy farms in Ohio and California. We hypothesized that farms receiving AMS training would have reduced AMU in dairy cattle compared with farms where training wasn't administered. We designed a quasiexperimental study with 18 conventional dairy farms enrolled in Ohio and California. Twelve farms received AMS training, and 6 farms did not. The AMS program included a 12-wk training focused on accurate identification of cows requiring antimicrobial treatment. We quantified AMU by collecting used drug containers, manually counted by researchers during farm visits. Treatment incidence (TI) was calculated using animal daily doses (ADD), and Poisson regression models were used to analyze the TI data. Disease incidence rate (DIR) in lactating cows was 2.2, 1.5, 1.0, 0.4, 0.3, 0.2, and 0.03/1,000 cow-days for mastitis, lameness, metritis, pneumonia, retained placenta, diarrheas, and other diseases (e.g., conjunctivitis, injuries), respectively. The highest TI by antimicrobial class (provided in ADD/1,000 cow-days) was cephalosporin (5.9), penicillin (5.2), tetracyclines (0.4), lincosamides (0.2), and sulfonamides (0.1). Among the trained farms using a within-treatment group analysis, no significant differences were observed in TI pre-intervention (10.9 ADD/1,000 cow-days) compared with TI postintervention (10.3 ADD/1,000 cow-days). Treatment incidence from the training group was 10.8 ADD/1,000 cow-days and although numerically lower, it was not significantly different compared with TI in the control groups at 13.9 ADD/1,000 cow-days (rate ratio = 0.77, CI = 0.25-2.38). Future research on AMS should incorporate farmworker training with social science approaches to overcome barriers and promote on-farm responsible use.

In North America, surplus dairy calves are sold soon after birth and often marketed through a third party (e.g., live auction, livestock dealer) before entering veal or dairy beef production. Previous work has demonstrated that a percentage of calves have failed transfer of passive immunity (FTPI) and clinical signs of disease on arrival at calf-raising facilities, but little is known regarding calf condition during marketing. The objectives of this cross-sectional study were to (1) estimate the prevalence of FTPI and poor health outcomes in surplus calves on arrival at livestock dealers, and (2) investigate the association between calf-level variables (body weight, sex, source) and health outcomes. Two livestock dealers in Ohio were visited 2 to 3 times per week, with approximately 28 calves enrolled in the study per visit for a total of 1,119 calves. One blood sample per calf was obtained to evaluate FTPI by measuring serum total protein concentrations (using a cutoff <5.1 g/dL). Calves were clinically evaluated for signs of arthritis, broken ribs or tail, dehydration, depression, diarrhea, fever, navel inflammation, and respiratory disease by 2 observers; health outcomes were dichotomized using clinically relevant cut points. Descriptive statistics were used to estimate the prevalence of calves with poor health outcomes. Multivariable logistic regression models were built to investigate the effect of body weight, sex, and source on health outcomes. Nineteen percent (206/1,091) of calves had FTPI.

This study aimed to understand the veterinary-client relationship and perspectives in organic dairy production systems. Twenty-three organic dairy producers and 12 veterinarians were interviewed using semi-structured, one-on-one interviews. Consultation with local veterinarians was typically infrequent, as there was greater reliance on other producers and organic industry professionals for advice on dairy herd health management, perhaps due to a shared focus on organic farming principles. Organic producers generally consulted local veterinarians most frequently to develop vaccine protocols, address animal health emergencies, and gain reproductive services. Common reasons organic producers cited for not regularly consulting a local veterinarian included the cost of veterinary services and a perception that local veterinarians have limited experience with organic systems and products. Some producers also expressed the idea that infrequent veterinarian usage is indicative of a healthy herd. Meanwhile, local veterinarians reported that organic dairy producers often delayed consulting them about individual cases until animal health problems were severe. Local veterinarians also identified challenges in servicing organic herds, such as the lack of standardized regulations between different organic certifiers and limited safety and efficacy data for many herd health products used by organic producers. In addition, their formal training generally did not include organic herd health strategies. Organic dairy clients were typically a small fraction of the overall workload for local veterinarians, which precluded their ability to accumulate experience. In contrast, a handful of specialized veterinarians in the organic milk processing industry work more frequently with organic producers and have published most of the available resource materials on organic dairy herd health. Veterinary-client partnerships could be improved through more open communication and discussion of the challenges identified in this study and through participatory research and outreach engagement that includes local veterinarians, organic certifiers, organic industry veterinarians, and producers.

In a more natural setting, dairy cows separate from herdmates and seek a secluded area to give birth. However, on many dairy facilities, cows calve in barren group pens with limited space, which may limit their ability to perform these behaviors. The objective of this study was to determine the effect of stocking density and provision of a blind (a single-sided solid partition) in group bedded pack maternity pens on the behavior of dairy cattle. The study included 4 experimental treatments: (1) high stocking density (7.7-12.9 m2 of lying space/cow) with a blind, (2) low stocking density (15.4-25.8 m2) with a blind, (3) high stocking density without a blind, and (4) low stocking density without a blind. We analyzed the effect of these treatments on locomotor and separation behavior as well as blind use during the 24 h before calving. A total of 127 primiparous heifers and 247 multiparous cows were housed in 16 mixed-parity groups (4 groups per treatment) from approximately 3 wk before to immediately after calving. Locomotor behavior increased from 24 to 2 h before calving regardless of treatment but increased the most in high stocking density pens without a blind. Additionally, heifers performed more locomotion than cows. Animals increased their distance from other cows beginning approximately 4 h before calving. Animals in low stocking density pens had a greater distance from other cows compared with those in high stocking density pens. Additionally, heifers had a greater distance from penmates during the 4 h before calving than cows. More animals calved in the area of the pen that contained a blind compared with the same respective areas of high and low stocking density pens that did not contain a blind. These results suggest that periparturient dairy cattle in group maternity pens are motivated to seek seclusion at calving, but increased stocking density may impede their ability to do so. Providing additional space and a blind may benefit periparturient dairy cows and heifers by facilitating natural calving behaviors.

Organic dairy producers have distinct perspectives, approaches, challenges, and experiences when managing herd health, but little research has been conducted to develop a deeper understanding of producer decision-making related to disease-prevention and treatment strategies. We used 23 semi-structured interviews with organic dairy producers to better understand their perceived herd health challenges and overall disease-prevention strategies; decision-making and attitudes related to vaccination; and approaches to the treatment of infectious diseases using alternative or conventional antimicrobial therapies. Organic dairy producers reported improvements in overall animal health after they transitioned from a conventional to an organic system. Outdoor access, a balanced forage-based diet, and animal and environmental cleanliness were reported as the predominant disease-prevention measures. Over half of producers vaccinated their animals, and almost all preferred to use vaccines sparingly. Among producers who did not use vaccines, reasons cited were low perceived risk for infectious disease and minor vaccine safety concerns, yet many were not opposed to using vaccines in the future. Non-antimicrobial therapies were viewed as alternative effective treatment strategies for most common infectious diseases. Producers used antimicrobials very rarely and generally reserved them for severe cases when alternative treatments failed. Because US organic standards prohibit selling milk from cows that have ever been treated with antimicrobials, producers described the decision to use antimicrobials as extremely challenging and involved balancing considerations of economics, convenience, and animal welfare. This research fosters a better understanding of organic dairy producers' disease-prevention and treatment strategies and could be used by veterinarians and other animal health specialists to facilitate services for organic clients.

Veal calves are at a high risk of disease early in life, which can lead to poor growth. Research is needed to determine interventions that can reduce disease and promote the growth of veal calves. The aim of this study was to determine the effects of fatty acid supplementation and the provision of a dry teat on the incidence of bovine respiratory disease (BRD), navel inflammation, and diarrhea, as well as calf growth. Upon arrival to a commercial veal facility (d 0), 240 Holstein bull calves from 2 cohorts were randomly assigned to 4 treatments using a 2 × 2 factorial arrangement (n = 60/treatment): (1) milk replacer (MR) without fatty acid supplementation and without access to a dry teat (control), (2) MR with fatty acid supplementation (NeoTec5g, Provimi, Brookville, OH) and without access to a dry teat (FAS), (3) MR without fatty acid supplementation and with access to a dry teat (TT), or (4) MR with fatty acid supplementation and with access to a dry teat (FAS+TT). Calves were housed in individual pens from 0 to 9 wk and then paired by treatment at wk 9. Milk replacer was fed twice daily using a step-up program. Fatty acid supplement was added to milk replacer at a feeding rate of 0.5 g/kg of body weight per head per day for the FAS and FAS+TT groups. Health exams were conducted twice weekly for 6 wk to diagnose BRD, navel inflammation, and diarrhea. Body weight, body condition score (BCS), and structural measurements were recorded at wk 0, 5, and 10. Average daily gain (ADG) was calculated for wk 1 to 5, wk 5 to 10, and wk 1 to 10. Health data were analyzed using logistic regression and are reported as relative risk. Body weight, BCS, and structural measurements were analyzed using repeated-measures ANOVA, and ADG was analyzed using a generalized linear model. There was no effect of FAS, TT, or their interaction on body weight or BCS. There was no effect of FAS on ADG wk 1 to 5, wk 5 to 10, or wk 1 to 10. There was a tendency for TT to decrease ADG from wk 1 to 5 but not from wk 5 to 10 or wk 1 to 10. There was a tendency for the interaction of FAS and TT to decrease ADG for wk 1 to 5 but not for wk 5 to 10 and wk 1 to 10. There was no effect of FAS, TT, or their interaction on the risk of BRD, diarrhea, or navel inflammation. We saw no effect of our interventions on calf health or growth. More research is needed to determine whether other factors, such as failure of passive transfer, poor ventilation, barren housing, and low milk allowance in the first few weeks after arrival, may have affected the efficacy of our interventions.

Diarrhea is one of the most common causes of antimicrobial use and mortality in young calves. To reduce antimicrobial use and resistance on dairy farms, research on alternative therapies for calf diarrhea is necessary. Our laboratory previously conducted a randomized clinical trial investigating the effectiveness of lactoferrin, an iron-binding protein found in colostrum, as a treatment for calf diarrhea. The trial showed significantly reduced calf mortality in diarrheic calves that were administered lactoferrin. Thus, the objective of this study was to corroborate the results of our prior clinical trial across multiple farms and to investigate the effect of lactoferrin on the morbidity and mortality of preweaned calves with naturally occurring cases of diarrhea. This randomized field trial was conducted on 5 commercial dairy farms in Ohio. In total, 485 calves (≤21 d of age) were enrolled at first diarrhea diagnosis (fecal score ≥2 defined as loose to watery) and randomly assigned to receive an oral dose of lactoferrin (3 g of lactoferrin powder dissolved in 30 mL of water) or 30 mL of water (control) once daily for 3 consecutive days. Health assessments were conducted on the day of diarrhea diagnosis (d 0) and 1, 2, 3, 7, 14, 21, 28, and 35 d following diagnosis. Producer records of disease treatment and mortality were collected 120 d following diagnosis. A Poisson regression model was used to test differences between treatments in disease frequency through 35 d post-diarrhea diagnosis and the incidence risk of treatment and mortality risk 120 d post-diarrhea diagnosis; the model controlled for calf age at enrollment, farm, and treatment. Median calf age at enrollment was 11 d and ranged from 1 to 26 d of age. At study enrollment, 51.3% (123/240) and 52.2% (128/245) of calves in the control and lactoferrin treatment groups, respectively, were diagnosed with severe diarrhea (fecal score = 3). The frequency of disease (diarrhea, dehydration, depression, signs of respiratory disease) through 35 d following diarrhea diagnosis was not significantly different for calves in the lactoferrin and control groups. Overall mortality risk for enrolled calves was 9.9%, and 10.7% (22/243) and 9.1% (26/242) of calves in the lactoferrin and control groups, respectively, died or were culled in the 120 d following diarrhea diagnosis. The relative risk of death or culling did not differ between treatment groups, however. Therefore, as performed in this study, lactoferrin as a treatment for calf diarrhea was not beneficial.

Every year, multiple outbreaks of salmonellosis in humans are linked to contact with mail-order chicks and ducks. The objective of this study was to describe the temporal changes in the prevalence of serovars, genotypes and antimicrobial resistance (AMR) phenotypes of non-typhoidal Salmonella (NTS) recovered from shipped boxes of mail-order hatchling poultry in the United States during 2013 to 2015. In each year, a sample of feed stores belonging to a single national chain participated in the study. The store employees submitted swabs or hatchling pads from hatchling boxes and shipment tracking information of the arriving boxes to the investigators. NTS was cultured from the samples and isolates were sent to the National Veterinary Services Laboratories (Ames, IA) for serotyping, pulsed-field gel electrophoresis (PFGE) and AMR phenotyping. The PFGE patterns of Salmonella serovars isolated from hatchling boxes were compared with those from human outbreaks of salmonellosis linked to live poultry contact. The box-level prevalence of NTS was significantly higher in 2015 compared to 2014. Also, the population of Salmonella serovars recovered in 2015 was more diverse and substantially different from those recovered in the previous two years. Of PFGE patterns recovered from hatchling boxes, seven distinct patterns in 2015, three in 2014 and four in 2013 were indistinguishable from the PFGE patterns of human outbreaks-associated strains in the respective years. Importantly, a significant positive correlation was found between the box-level prevalence of PFGE patterns and the number of human illnesses associated with the same patterns. Also, the proportion of multidrug-resistant isolates was higher in 2014 and 2015 compared to that in 2013. The results demonstrate that shipments of mail-order hatchling poultry are frequently contaminated with Salmonella genotypes indistinguishable from human outbreaks-associated strains each year, and control efforts at hatchery level are likely to have an important public health impact.

In the US, nontyphoidal Salmonellae are a common foodborne zoonotic pathogen causing gastroenteritis. Invasive Salmonella infections caused by extended-spectrum cephalosporin resistant (ESCR) phenotypes are more likely to result in treatment failure and adverse health outcomes, especially in severe pediatric Salmonella infections where the extended-spectrum ß-lactams are the therapy of choice. To examine the genetic and epidemiologic characteristics of ESCR Salmonellae which may enter the food chain, we characterized 44 ceftiofur-resistant Salmonella isolates from the National Animal Health Monitoring System (NAHMS) 2011 beef cattle feedlot health and management study. As part of the NAHMS Feedlot 2011 study, 5050 individual fecal samples from 68 large (1000+ head capacity) feedlots were cultured for Salmonella spp. The resulting 460 positive samples yielded 571 Salmonella isolates with 44 (8%) expressing an AmpC ß-lactamase phenotype. These phenotypic blaCMY-2Salmonella isolates represented 8 serotypes, most commonly S. Newport (n=14, 32%), S. Typhimurium (n=13, 30%), and S. Reading (n=5, 11%), followed by S. Dublin, S. Infantis, S. Montevideo, S. Rough O:i;v:1;7, and S. Uganda. Carriage of the blaCMY-2 gene was confirmed for all isolates expressing an AmpC ß-lactamase phenotype by PCR. Additionally, all 44 isolates were shown to carry the blaCMY-2 gene on a large IncA/C plasmid, a gene/plasmid combination which has been previously reported in multiple species. Other plasmids, including IncN, FIC, and FIIA, were also detected in some isolates. Cattle fed chlortetracycline were less likely to be positive for a blaCMY-2Salmonella isolate in their enteric flora compared to those not receiving chlortetracycline during the feeding period. Carriage of blaCMY-2 was more prevalent in Salmonella isolates originating from lighter weight cattle, cattle fed tylosin and dairy breeds. Our characterization of the NAHMS Feedlot 2011 study Salmonella isolates with ESCR phenotype shows that while other cephalosporin resistance mechanisms have been reported in US cattle, specific serotypes harboring blaCMY-2 on IncA/C plasmids may be the dominant resistance genotype.

Dairy calves are at high risk for morbidity and mortality early in life. Understanding producer attitudes is important for implementation of best management practices to improve calf health. The objectives of this study were to evaluate usage frequency and producer attitudes on key calf management practices between conventional and organic dairy operations. A cross-sectional survey was mailed to conventional and organic dairy producers in Ohio and Michigan that included questions on cow-calf separation, colostrum management, and vaccination use. The overall survey response rate was 49% (727/1,488); 449 and 172 conventional and organic producer respondents, respectively, were included in the final analysis. Binary, cumulative, and multinomial logistic regression models were used to test differences within and between herd types for management practices and producer attitudes. The majority of conventional (64%, 279/439) producers reported separating the calf from the dam 30 min to 6 h after birth. More organic (34%, 56/166) than conventional (18%, 80/439) producers reported separation 6 to 12 h after birth, and organic producers were more likely to agree time before separation is beneficial. Few conventional (10%, 44/448) and organic (3%, 5/171) producers reported measuring colostrum quality. Most conventional producers (68%, 304/448) hand-fed the first feeding of colostrum, whereas the majority of organic producers (38%, 69/171) allowed calves to nurse colostrum. Last, 44% (188/430) of conventional producers reported vaccinating their calves for respiratory disease, compared with 14% (22/162) of organic producers; organic producers were more likely to perceive vaccines as ineffective and harmful to calf health. Thus, the usage frequency and perceived risks and benefits of calf management practices vary considerably between conventional and organic dairy producers. These findings provide helpful information to understand decision making at the herd level regarding key calf management and health practices, regardless of production systems.

Multistate outbreaks of salmonellosis associated with live poultry contact have been occurring with increasing frequency. In 2013, multistate outbreaks of salmonellosis were traced back to exposure to live poultry, some of which were purchased at a national chain of farm stores (Farm store chain Y). This study was conducted at 36 stores of Farm store chain Y and was concurrent with the timing of exposure for the human outbreaks of salmonellosis in 2013. We used environmental swabs of arriving shipment boxes of hatchling poultry and shipment tracking information to examine the distribution, diversity and anti-microbial resistance of non-typhoidal Salmonella (NTS) across farm stores and hatcheries. Isolates recovered from shipment boxes underwent serotyping, anti-microbial resistance (AMR) testing and pulsed-field gel electrophoresis (PFGE). Postal service tracking codes from the shipment boxes were used to determine the hatchery of origin. The PFGE patterns were compared with the PFGE patterns of NTS causing outbreaks of salmonellosis in 2013. A total of 219 hatchling boxes from 36 stores in 13 states were swabbed between 15 March 2013 and 18 April 2013. NTS were recovered from 59 (27%) of 219 hatchling boxes. Recovery was not significantly associated with species of hatchlings, number of birds in the shipment box, or the presence of dead, injured or sick birds. Four of the 23 PFGE patterns and 23 of 50 isolates were indistinguishable from strains causing human outbreaks in 2013. For serotypes associated with human illnesses, PFGE patterns most frequently recovered from shipment boxes were also more frequent causes of human illness. Boxes positive for the same PFGE pattern most frequently originated from the same mail-order hatchery. Only one of 59 isolates was resistant to anti-microbials used to treat Salmonella infections in people. This study provides critical information to address recurrent human outbreaks of salmonellosis associated with mail-order hatchling poultry.

RESUMEN

Causes of poor fertility after insemination of frozen-thawed (FT) sperm include reduced sperm viability at thawing and a shorter longevity of surviving sperm in the female genital tract due to sub-lethal damage. The present studies examined the effect of incubating thawed boar sperm in seminal plasma (SP) on sperm membrane integrity (viability), and motility in vitro (experiment 1), and fertility in vivo (experiment 2). For experiment 1, FT sperm from five individual boars and a sperm pool from these boars were thawed and incubated for 4 h in media containing 0%, 10%, or 50% autologous seminal plasma (individual boars) or pooled seminal plasma (sperm pool). At approximately 10 min (0 h) and again at 1 h, 2 h, 3 h, and 4 h, sperm populations were examined for percentage sperm viability and percent sperm motility. Each variable progressively decreased during the incubation period. Incubation in 50% SP increased percentages of live sperm (P < 0.0001) and percent sperm motility (P < 0.01) at all time points compared to incubation in either 0% or 10% SP. For experiment 2, multiparous Large white x Landrace sows (n = 82) each received 900 IU eCG at weaning and 750 IU hCG 80 h later to control time of ovulation. Sows were assigned on the basis of parity to be inseminated with pooled semen with or without SP from the boars used in experiment 1. Sows received 3 x 10(9) live fresh-extended sperm (n = 30) or FT sperm thawed in 80 mL BTS extender (n = 26) or 3 x 10(9) live FT sperm thawed in 80 mL BTS containing 50% SP (FT-SP; n = 26). Sows were inseminated at 36 h, and 42 h after hCG injection. Compared to sows receiving fresh semen, the pregnancy rate of FT inseminated sows tended (P = 0.06) to be lower with the FT-SP group being intermediate. Farrowing rates were not different (83.3%, 69.2%, and 65.4% for fresh, FT, and FT-SP, respectively). Inseminations with FT sperm were associated with a reduction in litter size (P < 0.05), which was not evident in the FT-SP group. Taken together, these data confirm an adverse effect of inseminating FT sperm on sperm quality and sow fertility but suggest that thawing FT sperm in 50% SP may partially alleviate these adverse effects.
