RESUMEN

This study was designed to determine the lead or cadmium exposure of Barki rams and the beneficial role of Nannochlorposis oculata (N. oculata) 4% as a feed supplement, as well as its mitigating role against these elements' impacts concerning performance, biochemical markers of liver enzymes and kidney function, thyroid hormone activity, and oxidative stress markers. Six groups of 36 Barki rams (33.63 ± 1.29 kg) were divided into G1: which served as control; G2: was given 4% dietary N. oculata; G3: was given oral 1 mg/kg cadmium chloride; G4: was given 5 mg/kg/day lead acetate; G5: was given oral 1 mg/kg cadmium chloride and 4% dietary N. oculata, and G6: was given oral 5 mg/kg/day lead acetate and 4% dietary N. oculata; and treatments were continued for 60 days. Cadmium and lead-exposed groups exhibited lower and weaker weight gain as well as feed conversion ratio, respectively, than the control and other groups. Additionally, levels of T3, T4, total proteins, albumin, and glutathione (GSH) were significantly reduced in both G3 and G4 compared to control. However, urea, creatinine, ALT, AST, total cholesterol, triglycerides, protein carbonyl content (PCC), and malondialdehyde (MDA) were significantly increased (P ≤ 0.05) in cadmium and lead-exposed groups. Dietary N. oculata (4%) improves serum proteins, creatinine, urea, T4, and oxidative stress indicators as compared to the control group. Finally, 4% dietary N. oculata greatly enhances the investigated parameters in terms of performance, thyroid hormones, serum biochemical, and antioxidant activity and may assist in reducing the endocrine disrupting effects of Pb and Cd.

BACKGROUND: Natural feed supplements are gaining popularity in the animal production sector due to their safety and potential immunostimulatory properties, as well as the ban of some antibiotics and their negative residual effects. This study was carried out for 1 month to investigate the effect of Nannochloropsis oculata supplementation on growth performance and cell-mediated immunological status of rams assessed by leukogram assessment, lipid oxidation product malondialdehyde (MDA), total antioxidant capacity (TAC), interleukin assay after lymphocyte transformation test (LTT) including interleukin 6 (IL6), tumor necrosis factor-alpha (TNF-α), interleukin 12 (IL12), and gamma interferon (γ-IF), as well as Comet assay (% of DNA damage, tail length (px), % DNA in tail, tail moment and Olive tail moment). METHODS: Eighteen Barki rams (26.21 ± 0.64 kg) were divided into 3 equal treatment groups (6 sheep/each), G1: animals served as the control group that was fed the basal diet only, while the other treated groups (G2 and G3 (Nan 1.5% and Nan 3%) were fed the basal diet supplemented with 1.5% and 3% N. oculata (dry matter basis), respectively. RESULTS: The obtained results revealed that G3 showed a significant (P < 0.05) improvement in performance (body weight and body weight gain), the highest significant count (P < 0.05) in lymphocytes, and the lowest significant (P < 0.05) levels of neutrophils and neutrophils and lymphocytes ratio (N/L) ratio. Meanwhile, both levels of N. oculata significantly (P < 0.05) decreased MDA and increased TAC than control which seemed to be directly correlated with supplemented dose. There was a significant (P < 0.05) enhancement in the lymphocyte transformation assay produced significant (P < 0.05) high cytokines (IL6, γ-IF, IL12, and TNF-α) and the lowest significant (P <0.05) percent of DNA damage. The conducted principal component analysis estimated the inter-relationship between parameters and revealed that microalgae correlated strongly with cytokine assay and TAC, and negatively with Comet assay parameters; MDA, and neutrophils. CONCLUSIONS: It can be noted that dietary addition of N. oculata 3% increased sheep's performance while also producing significant-high cytokines. It also enhanced sheep immunology by considerably enhancing lymphocyte transformation ability. The antioxidant activity of Nannochloropsis appears to influence these findings. It was proposed that the Barki rams' basal diet be supplemented with 3% N. oculata.

A field survey was conducted on five fish farms to trace glyphosate and malathion pollution with some physicochemical parameters. A precise half-life time, LC50-96h, of these agrochemicals on Oreochromis niloticus, as well as chronic exposure with organic selenium (OS) supplementation, were experimentally investigated. Oreochromis niloticus was subjected to the following: (negative control); (2 mg L-1 glyphosate); (0.5 mg L-1 malathion); (glyphosate 1.6 mg L-1 and 0.3 mg L-1 malathion); (glyphosate 2 mg L-1 and OS 0.8 g kg-1 diet); (malathion 0.5 mg L-1 and OS 0.8 g kg-1 diet) and (glyphosate 1.6 mg L-1; malathion 0.3 mg L-1 and OS 0.8 g kg-1 diet). Furthermore, data from the analyzed pond revealed a medium risk quotient (RQ) for both agrochemicals. The detected agrochemicals were related to their application, and vegetation type surrounding the farms, also their biodegradation was correlated to water pH, temperature, and salinity. Glyphosate and malathion had half-lives of 2.8 and 2.3 days and LC50-96h of 2.331 and 0.738 mg L-1, respectively. The severest nervous symptoms; increased oxidative stress markers, as well as high bacterial count in the livers and kidneys of fish challenged with Aeromonas hydrophila, were observed in the combined exposure, followed by a single exposure to malathion and then glyphosate. Organic selenium mitigated these impacts.

BACKGROUND: Pesticide exposure is thought to be a major contributor to living organism health deterioration, as evidenced by its impact on both cultured fish species and human health. Commercial fish diets are typically deficient in selenium (Se); hence, supplementation may be necessary to meet requirements during stress. Therefore, this study was conducted to investigate the protective role of selenium yeast (SY) supplementation for 60 days against the deleterious effects of glyphosate and or malathion chronic toxicity at sublethal concentrations in Oreochromis niloticus . METHODS: Two hundred and ten fish were divided into seven groups (n = 30/group) as follows: G1 (negative control); G2 (2 mg L- 1 glyphosate); G3 (0.5 mg L- 1 malathion); G4 (glyphosate 1.6 mg L- 1 and malathion 0.3 mg L- 1); G5 (glyphosate 2 mg L- 1 and SY 3.3 mg kg- 1); G6 (malathion 0.5 mg L- 1 and SY 3.3 mg kg- 1); and G7 (glyphosate 1.6 mg L- 1; malathion 0.3 mg L- 1 and SY 3.3 mg kg- 1). RESULTS: Results revealed significant alteration in growth performance parameters including feed intake (FI), body weight (BW), body weight gain (BWG), specific growth rate (SGR), feed conversion ratio (FCR), and protein efficiency ratio (PER). G4 has the highest documented cumulative mortalities (40%), followed by G3 (30%). Additionally, the greatest impact was documented in G4, followed by G3 and then G2 as severe anemia with significant thrombocytopenia; leukocytosis; hypoproteinemia; increased Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST), urea, and creatinine, as well as malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GPx). Considering the previously mentioned parameters, selenium yeast (Saccharomyces cerevisiae) (3.3 mg kg- 1 available selenium) mitigated the negative impact of both the agrochemicals, whether exposed singly or in combination, in addition to their antioxidative action. CONCLUSIONS: In conclusion, our study found that organophosphorus agrochemicals, single or combined, had negative impacts on Oreochromis niloticus regarding growth performance, biochemical and hematological changes in the serum, as well as induced oxidative damage in liver and kidney tissues. Supplementation of SY at the rate of 3.3 mg kg- 1 diet (2.36 mg kg- 1 selenomethionine and 0.94 mg organic selenium) ameliorated the fish performance and health status adversely affected by organophosphorus agrochemical intoxication.

The effects of two probiotics on NH3 degradation, as well as the magnetic field (21.56 m tesla) on the germination and proliferation of Bacillus spores, were studied in-vitro. Additionally, the effect of these probiotics on water quality maintenance in Litopenaeus vannamei holding ponds was investigated. For 180 min, NH3 degradation was assessed as follows: Set 1: ammonia-free tap water with NH3; Probiotic A (5 × 1010 viable Bacillus spores/g) with NH3; Probiotic B (multi spp. 2 × 109 CFU/g) with NH3; and Set 2: same as set 1 with 30 mg L-1 OM. The magnetic field was tested on Probiotic A (3.5 × 107 CFU) for 36 h in triplicate. In the presence of organic matter, both probiotics degrade NH3. The viable Bacillus count increased within 6 h of being exposed to the magnetic field, reaching its peak after 36 h. Firstly, fifteen ponds (250,000 PL/acre) were investigated, then 360 water samples were collected from the same corresponding pond for 8 weeks, and subjected to T1: control; T2: Probiotic A (0.007 g/m3/2 weeks); T3: Probiotic B (0.03 g/m3/2 weeks). Both probiotics with TVC and NH3 demonstrated a negative correlation, on the other hand, they showed a significant (P ≤ 0.01) improvement in DO and pH. Overall, both probiotics were able to degrade NH3 and the magnetic field (21.56 m tesla) was efficient to improve the germination and proliferation of Bacillus spores in-vitro. Probiotics were also effective for reducing TVC and NH3 levels by increasing dissolved oxygen and pH in pond water.

BACKGROUND: Mast cells are known to affect the primary and secondary immune responses against parasites, and this effect is partially mediated through the release of pro-angiogenic mediators. The aim of this study was to explore the effect of the mast cell stabilizer (MCS), ketotifen, with and without albendazole, an anti-parasitic prescription medicine, on the inflammatory response against Trichinella spiralis, with the overall aim to investigate its effect on angiogenesis accompanying nurse cell formation. METHODS: The effect of ketotifen and albendazole was explored in eight groups of female BALB/c mice. Four groups were sensitized with a small dose of T. spiralis larvae. The drug regimen was then applied to both sensitized (challenged) and non-sensitized mice. The parasite load was assessed by histopathological examination of the small intestine and muscle tissue, and angiogenesis was assessed by immunohistochemistry to determine the expression of vascular endothelial growth factor (VEGF). RESULTS: Sensitized mice showed a significantly lower parasite load and a more pronounced inflammatory response than mice receiving a single infective dose of T. spiralis larvae. All treated groups showed a significant reduction in parasite count compared to the control groups (groups IAa and IBa), reaching approximately an 98.8% reduction in adult parasite count in the sensitized group treated with albendazole (groups IIAb and IIBb). MCS significantly decreased the parasite count during both the intestinal or muscular phases, reduced tissue inflammation, and decreased local VEGF expression, both in the non-sensitized and sensitized groups. CONCLUSION: Sensitization with a low dose of T. spiralis larvae was found to confer a partial protective immunity against re-infection and to positively affect the study outcomes, thus underlining the importance of vaccination, but after extensive studies. The anti-angiogenic effect of MCS protects against larval encystation during the muscle phase. The anti-angiogenic potential of albendazole suggests that the action of this anti-helminthic during trichinellosis is not confined to structural damage to the parasite cuticle but includes an effect on host immunopathological response.

The association of Helicobacter pylori (H. pylori) infection and parasitic infections including Giardia lamblia (G. lamblia), especially in childhood, is widely recognized to be high in developing communities. We aimed to study the impact of concomitant intestinal parasitic and H. pylori infections on the different clinical presentation of infected children and whether this coinfection could in turn cause any alteration in the clinical manifestations of each other. This cross-sectional study included 150 children of both sexes with their age ranging from 1 to 15 years, having gastrointestinal complaints, throughout 8 months duration. All cases were subjected to full history taking, clinical examination and stool analysis by direct wet smear and formalin-ethyl acetate concentration technique, permanent staining with cold acid fast stain in addition to H. pylori coproantigen detection in stool. Parasitic infection was recorded in 58.6% of patients, with G. lamblia the most detected parasite (35.2%). Cases infected with H. pylori were 63 cases (42%) of which 61.9% of cases showed associated parasitic infection. Diarrhea was the most frequent complaint (63.2%) in cases infected with intestinal parasites, while it was less frequently recorded in co-infected cases (35.8%) and in cases with H. pylori infection only (29.1%) (P value 0.0008). On the other hand, vomiting was less recorded in coinfected cases than cases with H. pylori infection. Coinfection with intestinal parasites (including G. lamblia) and H. pylori could modulate the clinical manifestation of each other especially diarrhea in parasitic infections and vomiting in H. pylori infection.

BACKGROUND: Diagnosis of intestinal capillariasis depending on microscopic detection of parasitic stages is of low sensitivity, especially in cases with low worm burden. There is a necessity to develop sensitive and specific diagnostic tools of capillariasis for early treatment to avoid complications. Western blot (WB) technique showed promising results for antigen recognition patterns in several parasitic infections. AIM OF THE STUDY: This study is directed to identify and evaluate relevant proteins of intestinal capillariasis crude worm antigens using WB immunodiagnosis. MATERIALS AND METHODS: Capillaria crude worm antigens were extracted and analyzed using SDS-PAGE. Sixty serum samples belonging to 3 groups (20 individuals each) were included; Group I (shedding Capillaria in feces), Group II (infected with other parasites) and Group III (negative parasitological results). Reactivity of the resulting bands of Capillaria with serum samples was analyzed using WB technique. RESULTS: Thirty-two immunoreactive bands were detected in WB analysis representing recognition of proteins with molecular weights (MW) varying from 19 to 110 kDa. Immunodominant proteins of 23.5, 31, 36.5, 40.5 and 44 kDa were recognized, respectively, in 35%, 30%, 85%, 95% and 75% of sera from patients with confirmed capillariasis and in 30%, 25%, 35%, 25%, and 20% of sera from those infected with other parasitic infections. One serum sample from group III gave reaction with 31 kDa band. CONCLUSION: Immunodiagnosis of intestinal capillariasis using WB proved that 23.5, 31, 36.5, 40.5 and 44 kDa bands could be considered useful tools for diagnosis of capillariasis.

BACKGROUND: Children with underlying malignancies and those on chemotherapy are at risk for having intestinal parasitic infections, which can lead to a severe course and death. This cross-sectional study was done to assess the copro-parasitological and copro-molecular prevalence of entero-parasites in children with malignancies and those on chemotherapy. PROCEDURE: Stool samples were collected from 137 Egyptian hospitalized cancerous children with different malignancies in the National Cancer Institute, and receiving chemotherapy.Faecal samples were examined microscopically. Genomic copro-DNA was extracted from fecal samples and amplified by 3 separate nPCR assays targeting Cryptosporidium, G. intestinalis and Entamoeba histolytica complex. RESULT: The overall prevalence of enteroparasites was 6.6 % (9 cases). Only Giardia copro-DNA was encountered in 2 (1.4%) faecal samples of patients. Coproscopy detected parasites in 7 cases: Blastocystis spp. in 5 cases (3.6%), Hymenolepis nana in 1 case (0.7%) and Ascaris lumbericoides in 1 case (0.7%). CONCLUSION: Low prevalence may be due to patient's use of prophylactic anti-parasitic and anti-fungal drugs, a standard protocol, basic hygienic practices and good nursing all of which are preventive against enteroparasites transmission. Among studied variables only diarrhoeic individuals who had a solid tumor, and soft/liquid stool with mucus and blood were predictors of intestinal parasitism.

Egypt's population is growing with the biggest hurdle facing the Government is to secure animal protein. Broilers provide quality protein of reasonable price. This study was conducted to investigate the outcomes of dietary organic betaine (betafin S4) on productive, epigenetic make up of lipoprotein lipase gene (LPL) promoter, some blood biochemical, and economic parameters in male broilers at finishing period. Eighty one commercial Arbor Acre Plus males, 21 days old, were randomly allocated to three groups, with three replicates each in battery cages under thermo-neutral environment till 42 days. The examined groups received yellow corn-soy basal diet, supplemented with 0 (G1), 1.5 (G2) and 3.0 g (G3) betaine/kg diet, respectively. The mRNA expression levels of LPL gene were analyzed by real-time quantitative PCR. Methylation pattern on LPL gene promoter was determined by bisulfite sequencing. Doses of betaine statistically (P ≤ .05) improved tested performance parameters; while carcass yield % and abdominal fat deposition did not achieve significant changes. The expression of LPL mRNA showed an inverse relationship with betaine dose, which illustrated as a trend toward increase in G2 and decrease in G3. Regarding serum biochemistry, both treated groups when compared to control group revealed a significant improvement (P ≤ .01) in albumin level, simultaneously, a significant increase (P ≤ .05) was recorded in uric acid and triglyceride levels, additionally, strong positive (P ≤ .01) correlation between betaine dose and previously mentioned parameters was reported. Betaine is recommended in finishing male broilers as production costs were reduced by 3.97%-4.37% per kg, respectively. In conclusion, incorporation of 0.15-0.30% organic betaine to male broilers diets during finishing period improves the growth performances.

AIM: The growth promoting effect of the blue-green filamentous alga Spirulina platensis (SP) was observed on meat type Japanese quail with antibiotic growth promoter alternative and immune enhancing power. MATERIALS AND METHODS: This study was conducted on 180 Japanese quail chicks for 4 weeks to find out the effect of diet type (vegetarian protein diet [VPD] and fish meal protein diet [FMPD])- Spirulina dose interaction (1 or 2 g/kg diet) on growth performance, gut microbiota, and sensory meat quality of growing Japanese quails (1-5 weeks old). RESULTS: Data revealed improvement (p<0.05) of weight gain, feed conversion ratio and European efficiency index due to 1, 2 g (SP)/kg VPD, and 2 g (SP)/kg FMPD, respectively. There was a significant decrease of ileum mean pH value by 1 g (SP)/kg VPD. Concerning gut microbiota, there was a trend toward an increase in Lactobacilli count in both 1; 2 g (SP)/kg VPD and 2 g (SP)/kg FMPD. It was concluded that 1 or 2 g (SP)/kg vegetarian diet may enhance parameters of performance without obvious effect on both meat quality and gut microbiota. Moreover, 1 and/or 2 g (SP) may not be invited to share fish meal based diet for growing Japanese quails. CONCLUSION: Using of SP will support the profitable production of Japanese quails fed vegetable protein diet.

Limited data is available on the epidemiologic status of schistosomiasis mansoni in Egypt. The present work aimed to explore the seroepidemiological status of Schistosoma inansoni infection in Egypt by screening inhabitants of different Egyptian govemorates and its correla- tion with morbid symptoms and risk factors. Health questionnaires and indirect haemagglutination test (IHAT) were performed upon a cross-sectional study of 1788 individuals from 22 govemorates. Socio-demographic varjables included sex, age, residence and.canal water contact. A multivariate regressi6n model was used to assess associations betWeen S.mansoni infection and socio-demographic variables; S.mansoni significant titre ≥ 1:160 was detected in 43% of samples. S. mansoni showed the highest prevalence in Al-Fayoum (15.2%), Kafr EI-Sheikh (11.2%) then Assiut (10.9%) while the least positiveresults were from Matrouh (0.2%). This may be the first indication to emerging foci in Cairo, Luxor, Aswan, Suez, Port Said and the Red Sea Govemorates. Anti-S.mansoni antibodies were least de- tected at 1 1-2Oys while they were the highest at 41-SOys, the highest titres (1/1280) were achieved by the age group 31 -4Oys.Male gender was a risk factor as 48.2% of males were IHAT +ve. Contacting canal water tends to be advantageous for schistosomiasis mansoni as 72.6% had a histoxy of canal contact and 96.7% of them achieved the highest titre. The alteration in the actual prevalence of schistosomiasis mansoni in Egypt with emergence of new foci including Cairo, Luxor, Aswan, Mersa-Matrouh and the north-eastern province alongside Suez Canal that may be explained by the associated socioepidemiologic risk factors.

Blastocystis hominis provides major challenges for laboratory diagnosis due to its polymorphic nature in wet mounts which can result in confusion with other protozoa, yeast or even fat globules. Studies revealed that simple smears were less sensitive than in vitro cultivation using different media for the detection of B. hominis in stool specimens. Cultures of B. hominis are usually enriched by different types of sera to enhance growth and multiplication of the parasite. The aim of this study is to assess the use of two sera types other than horse serum that is commonly used in culture media for the growth, multiplication and detection of B. hominis in examined stool samples and comparing the results with those obtained using horse serum. Fifty stool samples were collected from patients suffering from different colonic manifestations attending Cairo University Hospitals. The samples were freshly cultured in three different culture media using horse serum (in Jones' medium), donkey serum (as a modification ii Jones' medium) and human plasma (in modified Pavlova's medium) in adequate preparations. Cultures were then left for incubation and examined by direct microscopy to detect Blastocystis hominis. The result showed of 50 stool samples studied,. 18 samples (36%) were positive results for B. hominis. The number of positive results obtained by horse serum, donkey serum and human plasma were 13,18and 11 respectively. Paired comparisons were made between each 2 cultures with each culture set as a reference once to detect the most appropriate one for diagnosis When horse was set as the reference method, donkey serum showed a sensitivity of 100% and specificity of 86.5% with a 90% agreement between the 2 methods. While human plasma showed a sensitivity of 46.2% and specificity of 86.5% with an agreement of 76%. In addition, the vacuolar form was the commonest pattern observed in this study throughout all the three cultures.
