RESUMEN

Providing bedding or access to an outdoor run are husbandry aspects intended to improve pig welfare, which is currently financially supported through animal welfare schemes in several European countries. However, they may significantly affect the environment through changes in feed efficiency and manure management. Therefore, the aim of this paper was to compare farms differing in animal welfare relevant husbandry aspects regarding (1) the welfare of growing-finishing pigs and (2) environmental impact categories such as global warming (GW), acidification (AC), and freshwater (FE) and marine eutrophication (ME), by employing an attributional Life Cycle Assessment. We collected data on 50 farms with growing-finishing pigs in seven European countries. Ten animal-based welfare indicators were aggregated into three pig welfare indices using principal component analysis. Cluster analysis of farms based on husbandry aspects resulted in three clusters: NOBED (31 farms without bedding or outdoor run), BED (11 farms with bedding only) and BEDOUT (eight farms with bedding and outdoor run). Pigs on farms with bedding (BED and BEDOUT) manipulated enrichment more often (P < 0.001), pen fixtures less frequently (P = 0.003) and showed fewer oral stereotypies (P < 0.001) than pigs on NOBED farms. There were fewer pigs with a short(er) tail on farms with than without bedding (P < 0.001). Acidification of BEDOUT and BED farms was significantly higher (compared to NOBED farms P = 0.002) due to higher ammonia emissions related to farmyard manure. Also, BEDOUT farms had higher ME than NOBED farms (P = 0.035). There were no significant differences regarding GW and FE between husbandry clusters, due to the large variability within clusters regarding feed composition and conversion. Therefore, both husbandry aspects associated with improved animal welfare have a significant influence on some environmental impacts, such as acidification and marine eutrophication. Nevertheless, the large variation within clusters suggests that trade-offs may be minimised through e.g. AC and ME.

Nitrous oxide emissions can contribute significantly to the carbon footprint of municipal wastewater treatment plants even though emissions from conventional nitrogen removal processes are assumed to be moderate. An increased risk for high emissions can occur in connection with process disturbances and nitrite (NO2-) accumulation. This work describes the findings at a large municipal wastewater treatment plant where the levels of NO2- in the activated sludge process effluent were spontaneously and strongly increased on several activated sludge lines which was suspected to be due to shortcut nitrogen removal that stabilized for several months. The high NO2- levels were linked to a dramatic increase in nitrous oxide (N2O) emissions. As much as over 20% of the daily influent nitrogen load was emitted as N2O. These observations indicate that highly increased NO2- levels can occur in conventional activated sludge processes and result in high nitrous oxide emissions. They also raise questions concerning the risk of increased greenhouse gas (GHG) emissions of the nitritation-denitritation processes - although the uncontrolled nature of the event described here must be taken into consideration - and underline the importance of continuous monitoring and control of N2O emissions.

The aim of this work was to study the operational performance and the microbial community dynamics during the start-up of ANITATMMox technology implemented at full-scale wastewater treatment plant in Finland to treat reject water from anaerobic digesters. The average ammonium removal in the studied setup reached around 90%, withstanding ammonium loads up to 0.13 g N m-2h-1. The nitrite concentration in the effluent did not exceed 10 mg L-1, and there was a slight accumulation of NO3--N during the operation which was controlled. Thus, the result showed a robust success to high ammonium loading in presence of organic matter. The sequencing showed a heterogeneous microbial population where Methanosaeta, WCHA1-57 genus, Sphingobacteriia, Chlorobia and diverse unknown fungi were found as dominant phylotypes. Moreover, members of the Brocadiaceae family were dominant in the adhered biomass, mostly represented by Candidatus Scalindua, rarely reported in WWTPs. Overall, the results demonstrated a drastic effect of region-specific operational conditions on carrier biofilm microbial communities as it was demonstrated by the microbial studies.

Group housing of gestating sows benefits their welfare by allowing them freedom of movement and the opportunity for social interaction. However, social life could also bring disadvantages for individuals who receive direct aggression or are displaced from the feeder. The aim of this study was to investigate associations between social behaviour, body condition and live weight. Gestating sows (n=298) were investigated on a commercial farm. Sows were housed in mixed parity groups where two single space, ad libitum trough feeders served 12 animals. Sows were weighed, body condition scored and had their back fat layer measured at mixing, 4 weeks after insemination and again before farrowing. Social status was estimated based on the numbers of won and lost agonistic interactions at mixing and at the end of gestation. In addition, tear staining was scored before the farrowing and reproductive performance data were collected. With the aid of video recordings, 100 to 150 interactions per group were observed. Winning percentage at mixing and at the end of gestation were associated (P<0.05) and appeared relatively stable within individuals. Tear staining scores and litter sizes were not associated with winning percentage at the end of gestation. However, live weight, relative weight, body condition and back fat thickness were associated with winning percentage (P<0.05), giving heavier animals an advantage. Low winning percentage related to lower live weight gain, probably due to poorer success in competition for feed. Live weight within a mixed parity group could be used as a proxy measure for social status. Sows with low body condition score and submissive sows might need special attention with regard to group dynamics and housing to alleviate the effects of competition in group housing.

Tail biting is detrimental to animal welfare and has negative consequences for producer economy. Poor health is one of the risk factors for tail biting. The first aim of this study was therefore to test for links between health status and behavior related to tail biting at the individual level. The second aim of this study was to test whether variation in cytokines was related to variation in social behavior. These small molecules produced upon immune activation are known to influence behavior both in the direction of withdrawal and increased aggression. This could potentially increase non-functional social behavior and thereby the risk of a tail biting outbreak. To investigate this, we collected behavioral data, health data, feeding data and blood samples from undocked boars at a test station farm in Norway. We compared groups with three different diagnoses: osteochondrosis diagnosed by computer tomography scanning (OCSAN), osteochondrosis diagnosed by clinical examination (OCCLIN) and respiratory tract disease (RESP), with healthy controls (CTR). We tested whether the diagnoses were associated with feeding and growth, social behavior and cytokine levels. We then tested whether there were correlations between cytokine levels and social behavior. We also provide raw data on cytokine levels in the extended sample (N=305) as there are few publications on cytokine levels measured in pigs living under commercial conditions. OCCLIN pigs visited the feeder less, and fed longer compared to CTR pigs. Pigs diagnosed with RESP showed a large drop in growth the first week after filming, which corresponds to the week they were likely to have been diagnosed with illness, and a tendency to compensatory increase in the week after that. Social behavior differed between experimental groups with OCSCAN pigs receiving more social behavior (both aggressive and non-aggressive) compared to CTR, and RESP pigs tending to perform more ear- and tail-biting than controls. There were no differences in absolute levels of cytokines between categories. However IL1-ra and IL-12 showed correlations with several behaviors that have been shown by others to be associated with current or future tail biting activity. To our knowledge, this is the first published study indicating a role for illness in non-functional social behavior in pigs and the first showing a correlation between cytokine levels and social behavior.

AIMS: Toxigenic strains of Paenibacillus polymyxa were isolated from buildings connected with the symptoms of ill health. Our aim was to identify the toxic compounds of Paenibacillus polymyxa and to describe their toxic actions. METHODS AND RESULTS: The toxins of Paenibacillus polymyxa were purified and analysed by HPLC and mass spectrometry. Toxic fusaricidins A and B, and LI-F05a with mass ions at m/z 883·7, 897·6 and 897·6, respectively, were found. The cytotoxicity of purified fusaricidins A and B was measured using boar sperm, porcine tubular kidney epithelial cells and murine fibroblasts. The ion channel forming properties of fusaricidins were studied using the black lipid membrane (BLM) technique. Fusaricidins A and B depolarized the mitochondria of boar sperm, porcine tubular kidney epithelial cells and murine fibroblasts at concentrations of 0·5-1 µg ml-1 and caused nuclear fragmentation and induced apoptosis at concentrations of 2·5-5 µg ml-1 . Furthermore, fusaricidins A and B induced K+ permeating single channels. CONCLUSIONS: It was concluded that fusaricidins were toxic to mitochondria and induced apoptosis in mammalian cells. It was proposed that the observed toxicity of fusaricidins is due their ion channel forming properties. SIGNIFICANCE AND IMPACT OF THE STUDY: This paper revealed, for the first time, the mode of action of Paenibacillus polymyxa fusaricidins toxins towards mammalian cells. Fusaricidins, due to their potassium ionophoricity and mitochondria depolarizing impacts, may have contributed to the health damage observed at sites where the producer strains were isolated at high density.

Water in its three ambient phases plays the central thermodynamic role in the terrestrial climate system. Clouds control Earth's radiation balance, atmospheric water vapour is the strongest "greenhouse" gas, and non-equilibrium relative humidity at the air-sea interface drives evaporation and latent heat export from the ocean. In this paper, we examine the climatologically relevant atmospheric relative humidity, noting fundamental deficiencies in the definition of this key observable. The metrological history of this quantity is reviewed, problems with its current definition and measurement practice are analysed, and options for future improvements are discussed in conjunction with the recent seawater standard TEOS-10. It is concluded that the International Bureau of Weights and Measures, (BIPM), in cooperation with the International Association for the Properties of Water and Steam, IAPWS, along with other international organisations and institutions, can make significant contributions by developing and recommending state-of-the-art solutions for this long standing metrological problem, such as are suggested here.

Clouds control Earth's radiation balance, atmospheric water vapour is the strongest "greenhouse" gas, and non-equilibrium relative humidity at the air-sea interface drives evaporation and latent heat export from the ocean. On climatic time scales, melting ice caps and regional deviations of the hydrological cycle result in changes of seawater salinity, which in turn may modify the global circulation of the oceans and their ability to store heat and to buffer anthropogenically produced carbon dioxide. In this paper, together with three companion articles, we examine the climatologically relevant quantities ocean salinity, seawater pH and atmospheric relative humidity, noting fundamental deficiencies in the definitions of those key observables, and their lack of secure foundation on the International System of Units, the SI. The metrological histories of those three quantities are reviewed, problems with their current definitions and measurement practices are analysed, and options for future improvements are discussed in conjunction with the recent seawater standard TEOS-10. It is concluded that the International Bureau of Weights and Measures, BIPM, in cooperation with the International Association for the Properties of Water and Steam, IAPWS, along with other international organisations and institutions, can make significant contributions by developing and recommending state-of-the-art solutions for these long standing metrological problems in climatology.

The increasing incidence of type 1 diabetes observed in the past 60 years has spawned massive efforts in multiple research fields to elucidate the aetiology of this disease. While GWAS studies provide a good genetic basis for the current knowledge, it is clear that environmental triggers and their influence in disease prevalence and origin are highly important. The realization of disease heterogeneity has created a requirement for better biomarkers to complement the known autoantibody markers and to more successfully predict the severity and onset time of the disease. Such biomarkers would be needed both for prevention as well as for monitoring disease activity and response to preventive and therapeutic measures. Systematic holistic approaches concentrating on the triggering molecular mechanisms, pancreatic beta cells, immune response, as well as the influence of diet and environment, are necessary to understand the disease pathogenesis and find a cure. The current genomic knowledge is being broadened with accompanying studies in epigenetics and transcriptomic regulation, metabolomics, proteomics and lipidomics, covering the whole system from beta cells, the profile and cellular balance of the infiltrating lymphocytes, to gut microbiota and viral infections. Here we highlight interesting recent findings in type 1 diabetes research.

Hepatitis E virus (HEV) is a zoonotic agent that causes acute hepatitis in humans, and infects several animal species, most importantly swine. In the current study, that presents the first evidence of HEV infections in pigs in Finland, genetic divergence and transmission of HEV was investigated among pigs at a swine test station at two occasions. In 2007, HEV RNA was found in 25% of pens, and 35% of 2-3 month-old pigs at the station. Three different isolates, comprising 13 sequences of HEV genotype 3 e that were imported from different farms were detected. In 2010, 39% of pigs were HEV RNA positive on weeks 1, 3, or 5 of a 3-month follow-up, and 11 sequences, all representing one of the isolates that was also present in 2007, were detected. The isolate was considered to be either re-introduced to, or to persist at the station, and it was transmitted between the pigs. The study sheds light on the rate and time of HEV transmission in swine, and describes the epidemiologic variability of HEV isolates over time.

Municipal wastewater treatment plant (WWTP) influent is typically dependent on diurnal variation of urban production of liquid waste, infiltration of stormwater runoff and groundwater infiltration. During wet weather conditions the infiltration phenomenon typically increases the risk of overflows in the sewer system as well as the risk of having to bypass the WWTP. Combined sewer infrastructure multiplies the role of rainwater runoff in the total influent. Due to climate change, rain intensity and magnitude is tending to rise as well, which can already be observed in the normal operation of WWTPs. Bypass control can be improved if the WWTP is prepared for the increase of influent, especially if there is some storage capacity prior to the treatment plant. One option for this bypass control is utilisation of on-line weather-radar-based forecast data of rainfall as an input for the on-line influent model. This paper reports the Viikinmäki WWTP wet weather influent modelling project results where gridded exceedance probabilities of hourly rainfall accumulations for the next 3 h from the Finnish Meteorological Institute are utilised as on-line input data for the influent model.

Tail biting has several identified feeding-related risk factors. Tail biters are often said to be lighter and thinner than other pigs in the pen, possibly because of nutrition-related problems such as reduced feed intake or inability to use nutrients efficiently. This can lead to an increase in foraging behavior and tail biting. In this study, a total of 55 pigs of different ages were selected according to their tail-biting behavior (bouts/hour) and pen-feeding system to form eight experimental groups: tail-biting pigs (TB), victim pigs (V) and control pigs from a tail-biting pen (Ctb) and control pen (Cno) having either free access to feed with limited feeding space or meal feeding from a long trough. After euthanasia, a segment of jejunal cell wall was cut from 50 cm (S50) and 100 cm (S100) posterior to the bile duct. Villus height, crypt depth and villus : crypt ratio (V : C) were measured morphometrically. Blood serum concentration of minerals and plasma concentration of amino acids (AA) was determined. Villus height was greater in Cno than Ctb pigs in the proximal and mid-jejunum (P < 0.05), indicative of better ability to absorb nutrients, and increased with age in the proximal jejunum (P < 0.001). Serum mineral concentration of inorganic phosphate (Pi) and calcium (Ca) was lower in Ctb compared with Cno pigs, and that of Pi in V compared with all the other pigs. Many non-essential AA were lower in pigs from tail-biting pens, and particularly in victim pigs. Free access feeding with shared feeding space was associated with lower levels of essential AA in blood than meal feeding with simultaneous feeding space. Our data suggest that being a pig in a tail-biting pen is associated with decreased jejunal villus height and blood AA levels, possibly because of depressed absorption capacity, feeding behavior or environmental stress associated with tail biting. Victim pigs had lower concentrations of AA and Pi in plasma, possibly as a consequence of being bitten.

AIMS: To determine whether therapeutic treatment of pigs with chlortetracycline affects the susceptibility of their Campylobacter isolates for tetracycline, ciprofloxacin and erythromycin. METHODS AND RESULTS: Minimum inhibitory concentrations (MICs) and presence of a tetracycline resistance gene tet(O) were studied in Campylobacter collected before, during and after chlortetracycline treatment. Tetracycline MICs and the presence of tet(O) for additional Campylobacter coli isolates collected previously from seven farrowing farms were also determined. Isolates with ciprofloxacin MICs above the epidemiological cut-off value (ECOFF) were subtyped by flaA restriction fragment length polymorphism (RFLP). Tetracycline MICs of 221 Camp. coli isolates remained under the ECOFF at all sampling stages as well as the MICs for 63 isolates from the other farms. The ciprofloxacin MIC was above the ECOFF for 22% of the isolates, and one Camp. coli isolate had an erythromycin MIC above the ECOFF. None of the studied 300 Campylobacter isolates from nine herds carried tet(O). flaA-RFLP typing revealed the heterogeneity of Camp. coli isolates with high ciprofloxacin MICs. CONCLUSION: Use of chlortetracycline did not increase the MIC values for the antimicrobials studied. SIGNIFICANCE AND IMPACT OF THE STUDY: This study demonstrated that susceptibility of Camp. coli isolates is not affected by chlortetracycline therapy if tet(O) is not present in Camp. coli population.

Health in relation to tail-biting behaviour was investigated on a problem farm. Quartets (n = 16) of age- and gender-matched fattening pigs including a tail biter (TB, n = 16), a victim (V, n = 16), a control in the same pen (Ctb, n = 10) and a control in a pen where no tail biting was observed (Cno, n = 14) were chosen by direct behavioural observation. Haematological and clinicochemical analyses, autopsy and histological examination of 16 different tissues were carried out. Tail lesion severity was evaluated both macroscopically, on the basis of inspection, and histologically, in the sagittally cut tail. Category effects were tested using Friedman's ANOVA by Ranks, Cochran's Q or a repeated-measure GLM and, if significant, pair-wise tests were conducted using Wilcoxon Signed Ranks or McNemar's Test. The number of received tail bites correlated better with histological than with macroscopic tail lesion scoring because of deep inflammation beneath healthy skin in some cases. Most individuals had mild inflammatory lesions in internal organs suggestive of generalized activation of the immune system, and 30% of the animals were anaemic, possibly because of systemic spread of infectious agents. V had more severe respiratory organ lesions and higher serum protein concentrations than all other categories of pigs. Liver- and muscle-specific enzymes (alanine aminotransferase, alkaline phosphatase and creatine kinase) differed between categories. In conclusion, most animals had signs of generalized activation of the immune system, possibly because of systemic spread of infectious agents. V pigs suffered from more severe inflammatory lesions than TB, Ctb or Cno. Deep infections may exist under healthy skin in the tail of bitten pigs.

In order to determine the effective dose, the effects of orally administered ketoprofen were evaluated in pigs following intravenous challenge with Escherichia coli endotoxin. One hour after the challenge, five groups of pigs were treated with either tap water or ketoprofen (0.5 mg/kg, 1 mg/kg, 2 mg/kg or 4 mg/kg). The body temperature was measured and a total clinical score was calculated after assessing the general behaviour, respiratory rate and locomotion of the pigs. Thromboxane B(2) and ketoprofen concentrations were analysed from blood samples. Ketoprofen treatment significantly reduced the rectal temperature and total clinical scores, and lowered blood thromboxane B(2) concentrations when compared with the control group. Ketoprofen plasma concentrations were lower than previously reported in healthy pigs after similar doses. The appropriate dose of orally administered ketoprofen in pigs in this model is 2 mg/kg, as the higher dose of 4 mg/kg failed to provide an additional benefit.

RESUMEN

In this work we present and discuss the design of an array of soft-sensors to estimate the nitrate concentration in the denitrifying post-filtration unit at the Viikinmäki wastewater treatment plant in Helsinki (Finland). The developed sensors aim at supporting the existing hardware analyzers by providing a reliable back-up system in case of malfunction of the instruments. In the attempt to design easy to implement and interpretable sensors, computationally light linear models have been considered. However, due to the intrinsic nonlinearity of the process, also nonlinear but still computationally affordable models have been considered for comparison. The experimental results demonstrate the potential of the developed soft-sensors and the possibility for an on-line implementation in the plant's control system as alternative monitoring devices.

Effects of environmental enrichment at different stages of life on stress physiology of pigs were investigated in a trial with 63 groups, each of four siblings. In each of the three growing phases (suckling 0 to 4 weeks of age, nursery 5 to 9 weeks, fattening 10 to 24 weeks) pens either were (=E) or were not (=0) enriched. Accordingly, the treatments were (i) 000, (ii) E00, (iii) EE0, (iv) 00E, (v) 0EE and (vi) EEE. The enrichment material, renewed twice daily to leave a thin layer, consisted of wood shavings and chopped straw. Salivary cortisol was sampled hourly from 0700 to 1900 h at the age of 9 and 21 weeks. The presence of a circadian secretion rhythm was evaluated by an intra-assay coefficient of variation-based method. An adrenocorticotropic hormone test was performed at 21 weeks. Treatment effects on the odds of a physiological cortisol rhythm were assessed by logistic regression, and effects on cortisol concentrations with a repeated measures GLM. Substrate-enrichment from 0 to 9 weeks of age increased the odds of a rhythm as compared to barren housing (odds ratio (OR) = 30.0, P < 0.01). A flat cortisol secretion pattern may indicate chronic stress and/or delayed maturation of the rhythm. Barren as compared to enriched rearing (0 to 4 weeks of age) seemed to cause a blunted secretion rhythm at 21 weeks of age. Although behavioural and tail lesion observations provided support to the assumption that a blunted rhythm indicates chronic stress, the biological significance of these cortisol results needs confirmation in future studies.

Genetic selection and better control of the environment of the pigs have resulted in increased production levels concerning both reproduction and growth. Such high performances imply high physiological demands that may deteriorate health and welfare. The aims of this paper are to review the physiological challenges that pigs are facing, to identify possible consequences on health and welfare, to propose ways of detecting and correcting problems whenever possible. At weaning, piglets are submitted to abrupt changes in food supply, housing and social environment. Behavioural changes and efficient adaptations of the digestive tract are critical for their health and welfare. Physiological demands to support these adaptations and risks of failure are inversely related to the age of the pigs. During fattening, modern pigs have high daily weight gain especially of lean tissue as well as elevated feed conversion rate. These high growth performances are suspected to favour stress and disease susceptibility, undesirable behaviours as well as leg weakness, but further experimental data are necessary to validate these effects and find their origin. In reproductive females, high prolificacy generates elevated foetal demands for nutrients and space that are not fully met as shown by an increased number of light piglets having difficulties to adapt successfully to the neonatal life. During lactation, sows with high milk production have high nutrient requirements leading to intense catabolism that may affect their health, welfare and future reproductive abilities.

The effect of ad libitum (ADLIB) feeding strategy on the fertility of the group housed sow was studied in a prospective field trial during 1.5 years. All study animals farrowed under standard farrowing circumstances in crates, and they were provided with an ad libitum feeding throughout the 30-day lactation. After weaning, animals were randomly allocated to one of the two dry sow feeding strategies (AD LIB or CONT). After oestrus detection in groups, they were artificially inseminated and moved into pregnancy pens with partially slatted floor, in groups of 40 sows each. The ADLIB sows (n = 447) were loose housed and provided with ad libitum access to 7.7 MJ/kg feed high in fibre from two feeders per group. The control sows (n = 479; CONT) were also loose housed and given a standard dry sow feed in feeding stalls once a day (2.5 kg/day. The energy content of the feed was 9.3 MJ/kg NE). The feeding strategy (ADLIB vs CONT) had no effect on pregnancy rate (85.8 vs 90.9, p > 0.05), weaning to oestrus interval (7.7 vs 7.3 days, p > 0.05), piglets born alive (11.5 ± 3.5 vs 11.6 ± 3.3, p > 0.05), stillborn piglets (1.2 ± 1.8 vs 0.9 ± 1.5, p > 0.05) nor on progesterone concentration (p > 0.05). CONT sows weaned more piglets (9.7 ± 2.2 vs 9.4 ± 2.0, p < 0.01), whereas the piglets of AD LIB sows were heavier at weaning (8.8 ± 0.9 vs 8.0 ± 1.3 kg, p < 0.05). In conclusion, ad libitum feeding with a high in fibre diet during pregnancy did not affect the reproductive performance.

The objective of this retrospective cohort study was to clarify the effect of re-modelling the breeding unit on farrowing rate. The original study population included 63 sow herds that participated in the Finnish herd surveillance system. In these herds, the breeding units were re-modelled between 1995 and 2002. Information about the production system and other herd data for the last year before and the second year after re-modelling were available for 47 herds. The herds had an average of 68.5 (SD 45) sows per herd. Data were collected during one farm visit per herd. Initially, all herds had individual cages in their breeding units. The piggeries had either re-modelled their cages or switched to a group housing system with deep-litter, solid or slatted (partly or totally) floors. Farrowing rates were gathered from the national database, and data were tested with a linear regression model. Re-modelling did not have an effect on farrowing rate if initial reproductive performance of the herd was neglected. However, when farms were stratified according to performance prior to the beginning of study, namely into 'farms with initially high farrowing rate' (HF) and 'farms with initially low farrowing rate' (LF), the re-modelling had a significant effect on farrowing rate. In the HF farms, re-modelling caused a decrease in farrowing rate. Correspondingly, farrowing rate increased after remodelling in the LF farms. Farrowing rate was also influenced by mean parity of sows and by proportion of sows culled because of leg problems. This study could not show a profound impact of the design of the breeding unit on the fertility of the sow in the modern commercial production environment if the farm's reproductive performance before re-modelling was neglected. However, when initial performance was taken into account, re-modelling revealed significant, although varying effects on farrowing rate.
