RESUMEN

This psychophysiological study is the first to examine the relationship between emotional tears and emotional piloerection (i.e., goosebumps). Although both phenomena have been related to peak states of being moved, details about their temporal occurrence and the associated levels of physiological arousal have remained unknown. In our study, we used emotionally powerful film scenes that were self-selected by participants. Our findings show that even within peak moments of emotional arousal, a gradation of intensity is possible. The overlap of tears and goosebumps signifies a maximal climax within peak moments. On the side of the stimulus, we found that displays of prosocial behavior play a crucial role in the elicitation of tears and goosebumps. Finally, based on the results of a formal film analysis of the tears-eliciting clips provided by our participants, as compared to randomly extracted, equally long control clips from the same films, we show how the technical and artistic making of the clips was optimized for the display of social interaction and emotional expressions.

The human spermatozoa membrane is characterized by a unique fatty acyl composition with significant amounts of highly unsaturated fatty acids, particularly docosahexaenoic acid (22:6), whereby phosphatidylcholine (PtdCho) (16:0/22:6) is the most abundant glycerophospholipid. The large amount of highly unsaturated fatty acyl residues is crucial for the fluidity of the membrane and, therefore, the successful fertilization process. Consequently, however, the spermatozoa are very sensitive to reactive oxygen species (ROS) that are generated under conditions of "oxidative stress" and key players in many pathological conditions. Lipid oxidation of the sperm membrane is accompanied by the loss of the oxidatively modified unsaturated residue (normally in the sn-2 position) and the generation of saturated lysophosphatidylcholine (LysoPtdCho). Although other lysolipids are also generated, LysoPtdCho is the "marker" lipid of choice due to the high abundance of PtdCho. In particular, obesity (body mass index >30 kg/m(2)) is characterized by increased ROS generation and negatively affects the reproductive potential. We will show here that the LysoPtdCho/PtdCho ratio can be easily determined by matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). The data found do correlate with clinical markers of sperm quality. A very interesting aspect is that the LysoPtdCho/PtdCho ratios determined in the spermatozoa extracts correlate with the LysoPtdCho/PtdCho values determined in the organic extracts of erythrocytes. Thus, there is no absolute need for a sperm investigation, but an estimation of the fertilizing ability of the corresponding male could be also made directly from the blood which is more readily available than the spermatozoa.

This study investigated the effect of one-week consumption of 165 g/day fresh blue honeysuckle berries (208 mg/day anthocyanins) in 10 healthy volunteers. At the end of intervention, levels of benzoic (median 1782 vs 4156), protocatechuic (709 vs 2417), vanillic (2779 vs 4753), 3-hydroxycinnamic (143 vs 351), p-coumaric (182 vs 271), isoferulic (805 vs 1570), ferulic (1086 vs 2395), and hippuric (194833 vs 398711 µg/mg creatinine) acids by LC/MS were significantly increased in the urine. Clinical chemistry safety markers were not altered. Oxidative stress markers, erythrocyte glutathione peroxidase (0.73 vs 0.88 U/g Hb) and catalase (2.5 vs 2.8 µkat/g Hb) activities, and erythrocyte/plasma thiobarbituric acid reactive substance (522 vs 612/33 vs 38 µmol/g Hb/protein) levels were significantly increased, without change in plasma antioxidant status. Nonsignificant changes of advanced oxidation protein products and oxidized LDL were observed. The results provide a solid base for further study of metabolite excretion and antioxidant parameters after ingestion of anthocyanins.

Effector CD4+ T cell subsets, whose differentiation is facilitated by distinct cytokine cues, amplify the corresponding type of inflammatory response. Regulatory T (Treg) cells integrate environmental cues to suppress particular types of inflammation. In this regard, STAT3, a transcription factor essential for T helper 17 (Th17) cell differentiation, is necessary for Treg cell-mediated control of Th17 cell responses. Here, we showed that anti-inflammatory interleukin-10 (IL-10), and not proinflammatory IL-6 and IL-23 cytokine signaling, endowed Treg cells with the ability to suppress pathogenic Th17 cell responses. Ablation of the IL-10 receptor in Treg cells resulted in selective dysregulation of Th17 cell responses and colitis similar to that observed in mice harboring STAT3-deficient Treg cells. Thus, Treg cells limit Th17 cell inflammation by serving as principal amplifiers of negative regulatory circuits operating in immune effector cells.

IL-10 is a potent regulator of the innate and adaptive immune responses. Several cell types produce IL-10 and its receptor chains and these may regulate different immune responses. Here we report that inactivation of the IL-10 receptor (IL-10R1) gene in mice leads to an increased susceptibility to chemically induced colitis as in the classical IL-10-deficient mutant. To identify the cells regulated by IL-10 in immune responses, we generated several cell type specific IL-10R1-deficient mutants. We show that, in an IL-10-dependent LPS model of endotoxemia, dampening of the immune response requires expression of IL-10R1 in monocytes/macrophages and/or neutrophils but not in T cells nor B cells. As the macrophage and/or neutrophil-specific IL-10-deficient mutants also display the same phenotype, our results suggest that an autocrine loop in monocytes/macrophages is the most probable mechanism for the regulation of an LPS-induced septic shock. In contrast, in an IL-10-regulated T-cell response to Trichuris muris infection, IL-10 acting on T cells or monocytes/macrophages/neutrophils is not critical for the control of the infection.

RESUMEN

We present a polarization-independent metamaterial design for the construction of electrically tunable terahertz (THz) devices. The implemented structure consists of an array of gold crosses fabricated on top of an n-doped gallium arsenide (GaAs) layer. Utilizing THz time-domain spectroscopy, we show that the electric resonance and thus the transmission properties of the cross structure can be tuned by an externally applied bias voltage. We further demonstrate the fast amplitude modulation of a propagating THz wave for modulation frequencies up to 100 kHz.

A multicompartmental bioreactor was conceived and designed to mimic cross talk between cells in different culture chambers connected only by flow, such that cell-cell interaction is mediated by soluble ligands as occurs in the body. The system was tested with a connected culture of murine hepatocytes and human umbilical vein endothelial cells. Metabolites such as albumin, urea, lactate and viability were monitored during the course of the experiments and compared with monoculture conditions in the bioreactor. When the two cell types are placed in connected culture, there is an increase in endothelial cell viability and hepatic glucose synthesis as well as albumin and urea production, while overall lactate production in the system is downregulated. The results show that the multicompartmental bioreactor enhances cell function, effectively combining both heterotypic interactions with increased nutrient availability.

The fruit of Lonicera caerulea L. (blue honeysuckle; Caprifoliaceae) and its phenolic fraction were analyzed for nutrients and micronutrients. The phenolic fraction was prepared from berries percolated with 0.1% H3PO4 and SPE using Sepabeads SP207. The sugar and lipid content was analyzed by HPLC and GC-MS. The total content of anthocyanins was determined using the pH differential absorbance method and aliphatic acids by capillary electrophoresis. MicroLC-MS/MS was used for determination of cyanidin-3-glucoside (the predominant anthocyanin), 3,5-diglucoside, and 3-rutinoside, paeonidin-3-glucoside, 3,5-diglucoside, and 3-rutinoside, delphinidin-3-glucoside and 3-rutinoside, pelargonidin-3-glucoside, 3,5-diglucoside, and 3-rutinoside, quercetin, its 3-glucoside, and 3-rutinoside, epicatechin, protocatechuic, gentisic, ellagic, ferulic, caffeic, chlorogenic, and coumaric acids. The phenolic fraction displayed Folin-Ciocalteu reagent reducing (335 +/- 15 microg of gallic acid equivalent/mg) and DPPH and superoxide scavenging activity (IC50 12.1 +/- 0.1 and 115.5 +/- 6.4 microg/mL) and inhibited rat liver microsome peroxidation (IC50 160 +/- 20 microg/mL). The freeze-dried fruit and its phenolic fraction reduced the biofilm formation and adhesion to the artificial surface of Candida parapsilosis, Staphylococcus epidermidis, Escherichia coli, Enterococcus faecalis, and Streptococcus mutans.

RESUMEN

Dr. Ada Sue Hinshaw stepped down from her deanship at the University of Michigan School of Nursing, leading to a conference in honor of her legacy held in May 2006. The topic of the conference highlighted Dr. Hinshaw's accomplishments in the area of health policy and patient safety. The proceedings of the afternoon included four presentations. The common themes of the presentations incorporated the importance of public policy for patient safety, the need for health systems reforms, and the use of appropriate evidence-based practice to advance the nursing sciences. These and other perspectives of the speakers are discussed within the context of the broad categories of system reform, contributions from nursing research, and health policy focus. Finally, recommendations were given for the preferred future.

BACKGROUND: Lonicera caerulea L. (blueberry honeysuckle, Caprifoliaceae) is a traditional crop in northern Russia, China, and Japan. Its fruits are little known as edible berries in North America and Europe. This review deals with the botany and chemical composition of L. caerulea and the biological activity of its main constituents, focusing on the potential health benefits of the berries. METHODS AND RESULTS: PubMed, Science Direct and ISI Web of Knowledge databases were used for this paper. Literature sources include the period 1935-2007. L. caerulea berries a are rich source of phenolic compounds such as phenolic acids as well as anthocyanins, proanthocyanidins and other flavonoids, which display potential health promoting effects. Chemopreventive, antimicrobial, anti-adherence and antioxidant benefits, among others are described for these compounds. CONCLUSIONS: The potential of L. caerulea berries to prevent chronic diseases such as diabetes mellitus, cardiovascular diseases and cancer seems to be related above all to their phenolic content.

Lately certain cytotoxicity of quantum dots (QDs) and some deleterious effects of labeling procedure on stem cells differentiation abilities were shown. In the present study we compared cytotoxicity and intracellular processing of two different-sized protein-conjugated QDs after labeling of the human mesenchymal stem cells (hMSC). An asymmetrical intracellular uptake of red (605 nm) and green (525 nm) quantum dots was observed. We describe for the first time a size-dependent activation of autophagy, caused by nanoparticles.
