RESUMEN

We present the development of an experimental platform that can collect four frames of x-ray diffraction data along a single line of sight during laser-driven, dynamic-compression experiments at the National Ignition Facility. The platform is comprised of a diagnostic imager built around ultrafast sensors with a 2-ns integration time, a custom target assembly that serves also to shield the imager, and a 10-ns duration, quasi-monochromatic x-ray source produced by laser-generated plasma. We demonstrate the performance with diffraction data for Pb ramp compressed to 150 GPa and illuminated by a Ge x-ray source that produces ∼7 × 1011, 10.25-keV photons/ns at the 400 µm diameter sample.

Carbamazepine is a medicine used to manage epilepsy and partial or tonic-clonic seizures. This study aimed at formulating and obtaining carbamazepine orodispersible tablets for paediatric use at a 50 mg dose, with a diameter not greater than 6 mm and a tablet weight of 80 mg, through a direct compression process. The SeDeM pre-formulation/formulation method was used to define the characteristics of both carbamazepine and the selected excipients for direct compression. This study succeeded in formulating and obtaining the proposed tablets. Following the application of the SeDeM method, the tablets met the mass uniformity test and showed appropriate hardness values for orodispersible tablets. The tablets also met the United States Pharmacopeia (USP) test specifications at t = 60 min. The orodispersible tablets obtained may improve compliance with paediatric treatment with carbamazepine, ensuring the safety and effectiveness of the medicine.

In support of future radiation-effects testing, a combined environment source has been developed for the National Ignition Facility (NIF), utilizing both NIF's long-pulse beams, and the Advanced Radiographic Capability (ARC) short pulse lasers. First, ARC was used to illuminate a gold foil at high-intensity, generating a significant x-ray signal >1 MeV. This was followed by NIF 10 ns later to implode an exploding pusher target filled with fusionable gas for neutron generation. The neutron and x-ray bursts were incident onto a retrievable, close-standoff diagnostic snout. With separate control over both neutron and x-ray emission, the platform allows for tailored photon and neutron fluences and timing on a recoverable test sample. The platform exceeded its initial fluence goals, demonstrating a neutron fluence of 2.3 ×1013 n/cm2 and an x-ray dose of 7 krad.

The global coronavirus disease 2019 (COVID-19) pandemic has altered entire nations and their health systems. The greatest impact of the pandemic has been seen among vulnerable populations, such as those with comorbidities like heart diseases, kidney failure, obesity, or those with worse health determinants such as unemployment and poverty. In the current study, we are proposing previous exposure to fine-grained volcanic ashes as a risk factor for developing COVID-19. Based on several previous studies it has been known since the mid 1980s of the past century that volcanic ash is most likely an accelerating factor to suffer from different types of cancer, including lung or thyroid cancer. Our study postulates, that people who are most likely to be infected during a severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) widespread wave will be those with comorbidities that are related to previous exposure to volcanic ashes. We have explored 8703 satellite images from the past 21 y of available data from the National Oceanic and Atmospheric Administration (NOAA) database and correlated them with the data from the national institute of health statistics in Ecuador. Additionally, we provide more realistic numbers of fatalities due to the virus based on excess mortality data of 2020-2021, when compared with previous years. This study would be a very first of its kind combining social and spatial distribution of COVID-19 infections and volcanic ash distribution. The results and implications of our study will also help countries to identify such aforementioned vulnerable parts of the society, if the given geodynamic and volcanic settings are similar.

We present experimental studies of inverted-corona targets as neutron sources at the OMEGA Laser Facility and the National Ignition Facility (NIF). Laser beams are directed onto the inner walls of a capsule via laser-entrance holes (LEHs), heating the target interior to fusion conditions. The fusion fuel is provided either as a wall liner, e.g., deuterated plastic (CD), or as a gas fill, e.g., D2 gas. Such targets are robust to low-mode drive asymmetries, allowing for single-sided laser drive. On OMEGA, 1.8-mm-diameter targets with either a 10-µm CD liner or up to 2 atm of D2-gas fill were driven with up to 18 kJ of laser energy in a 1-ns square pulse. Neutron yields of up to 1.5 × 1010 generally followed expected trends with fill pressure or laser energy, although the data imply some mix of the CH wall into the fusion fuel for either design. Comparable performance was observed with single-sided (1x LEH) or double-sided (2x LEH) drive. NIF experiments tested the platform at scaled up dimensions and energies, combining a 15-µm CD liner and a 3-atm D2-gas fill in a 4.5-mm diameter target, laser-driven with up to 330 kJ. Neutron yields up to 2.6 × 1012 were measured, exceeding the scaled yield expectation from the OMEGA data. The observed energy scaling on the NIF implies that the neutron production is gas dominated, suggesting a performance boost from using deuterium-tritium (DT) gas. We estimate that neutron yields exceeding 1014 should be readily achievable using a modest laser drive of ∼300 kJ with a DT fill.

Experiments on the National Ignition Facility (NIF) to study hohlraums lined with a 20-mg/cc 400-µm-thick Ta_{2}O_{5} aerogel at full scale (hohlraum diameter = 6.72 mm) are reported. Driven with a 1.6-MJ, 450-TW laser pulse, the performance of the foam liner is diagnosed using implosion hot-spot symmetry measurements of the high-density carbon (HDC) capsule and measurement of inner beam propagation through a thin-wall 8-µm Au window in the hohlraum. Results show an improved capsule performance due to laser energy deposition further inside the hohlraum, leading to a modest increase in x-ray drive and reduced preheat due to changes in the x-ray spectrum when the foam liner is included. In addition, the outer cone bubble uniformity is improved, but the predicted improvement in inner beam propagation to improve symmetry control is not realized for this foam thickness and density.

Pexophagy is a peroxisome degradation process. The last two steps of penicillin biosynthesis in Penicillium rubens are carried out in peroxisomes. These organelles proliferate in large numbers during this process, so that after the penicillin secretion, their removal is essential as a regulatory mechanism. In this work, two pexophagy modes are described for the high-penicillin producing strain P. rubens P2-32-T, by transmission electron microscopy (TEM) on 24- and 48-h cultures (when maximum penicillin production is achieved). The obtained images show peroxisome phagocytosis by vacuoles in three different ways: macropexophagy, micropexophagy, and a new proposed model: unipexophagy.

Prolactin (PRL) is a pleiotropic hormone secreted by several cells and tissues in the body, such as mammary glands, T-lymphocytes, hypothalamus, among others. This hormone possess neuroprotective properties against glutamate-excitotoxicity through the activation of NF-kB, suggesting it could exert an antioxidant action. However, the role of PRL on the antioxidant defense during glutamate-induced excitotoxicity is not clear to date. Therefore, in the present study, we have evaluated the effect of PRL on SOD activity and protein content of both of its isoforms (Mn2+-SOD and Cu2+/Zn2+-SOD), as well as, its action on mitochondrial activity in primary culture of hippocampal neurons of rats. Additionally, we have evaluated the possible antioxidant effect of PRL through the determination of lipid peroxidation products (LPO), measured as malondialdehyde (MDA). Results show that PRL enhances the activity and the protein content of Mn2+-SOD and Cu2+/Zn2+-SOD in neurons exposed to glutamate-induced excitotoxicity. Moreover, our results demonstrate that PRL prevents mitochondrial dysfunction induced by glutamate and significantly decreases the levels of LPO products. To our knowledge, this is the first time that a potential antioxidant effect of PRL has been described in hippocampal neurons exposed to glutamate excitotoxicity, opening questions of its potentiality for therapeutics.

An experimental Taenia crassiceps mouse model was used to assess the role of Taenia solium metacestode factor (Fac) in human neurocysticercosis. Intraperitoneal infection with T. crassiceps metacestodes or subcutaneous inoculation with a T. crassiceps metacestode factor (Fac) produced significant impairment of performance (learning) in the Barnes maze and induced bilateral hippocampal sclerosis in mice. Several staining techniques revealed important cell dispersion, extensive apoptosis and cell loss in the dentate gyrus, hilus and CA1-CA3 regions of both hippocampi, as well as intense deterioration of the adjacent cortex. An outstanding disruption of its histoarchitecture in the surrounding tissue of all these regions and apoptosis of the endothelial cells were also observed.

INTRODUCTION AND OBJECTIVES: Pediatric Systemic Lupus Erythematosus (pSLE) is an autoimmune disorder of children. Early disease onset raises the probability of genetic etiology and it is more severe than adult SLE. PATIENTS AND METHODS: Herein an eight-year-old girl with pSLE from consanguineous parents is reported. RESULTS: Although she was diagnosed as pSLE since the age of two years, Whole Exome Sequencing (WES) revealed a rare stop-gained C>T mutation in C1QA gene. The variant was validated and segregated in patient and the family. Furthermore, serum levels of the C1q protein were measured and found to be much lower than normal ranges. CONCLUSIONS: This study indicated that C1Q deficiency should be considered as a differential diagnosis of pSLE. Therefore, measurement of C1q should be recommended in all cases with pSLE

Both the presence of owned dogs and stray dogs allows the spread of Toxocara, a parasite whose eggs can be found in soil, water and food. Animals, including horses, serve as definitive and paratenic hosts. In México, where consumption of horse meat is common, Toxocara is a zoonotic parasite. The aim of this study was to identify the presence of anti-Toxocara antibodies in work horses and horses intended for human consumption by ELISA. ELISA was chosen for analysis as paratenic hosts do not shed Toxocara eggs in their feces. Blood samples were collected from a total of 188 horses, 94 of which were work horses and 94 horses from the slaughter house. Samples were analyzed by ELISA, and the general equine seroprevalence was found to be 44.6% (n = 188). Adult horses for slaughter had a 61.7% greater presence of anti-Toxocara antibodies (p = 0.006). Toxocara IgG antibodies were found in horses, confirming that horses are paratenic hosts and possible sources of infection for other animals and people.(AU)


Tanto a presença de cães com dono quanto de cães vadios permitem a disseminação de Toxocara, e o parasita está presente no solo, na água e nos alimentos. Animais, incluindo cavalos, apresentam-se como hospedeiros definitivos e paratênicos. No México, o consumo de carne de cavalo é comum, e Toxocara é um parasita zoonótico. ELISA foi escolhido para análise, já que hospedeiros paratênicos não jogam ovos de Toxocara em suas fezes. O objetivo deste estudo foi identificar a presença de anticorpos anti-Toxocara por ELISA, em cavalos de trabalho e em cavalos para o consumo humano. As amostras de sangue foram retiradas de 188 cavalos: 94 cavalos de trabalho e 94 cavalos de trabalho do matadouro. Soros dos animais foram analisados por ELISA e 44,6% dos equinos apresentaram anticorpos anti-Toxocara. Cavalos adultos para abate têm 61,7% mais elevada a presença de anticorpos anti-Toxocara (P = 0,006). Anticorpos IgG Toxocara foram encontrados em cavalos, confirmando cavalos paratênicos como hospedeiros e possíveis fontes de infecção para outros animais e pessoas.(AU)

BACKGROUND: Drug reaction with eosinophilia and systemic symptoms (DRESS) is a severe delayed hypersensitivity reaction. The determination of drug causality is complex. The lymphocyte transformation test (LTT) has been reported positive in more than 50% of DRESS cases. Nevertheless, the sensitivity and specificity of LTT specifically in DRESS have not yet been established. Rechallenge with the culprit drug is contraindicated and cannot be used as gold standard for sensitivity and specificity determination. OBJECTIVE: To estimate the sensitivity and specificity of LTT in a clinically defined series of patients with DRESS. METHODS: Some 41 patients diagnosed with DRESS were included in the study. The results of the algorithm of the Spanish Pharmacovigilance System were used as the standard for a correct diagnosis of drug causality. A standard LTT was performed with involved drugs in acute or recovery samples. A stimulation index (SI) ≥2 in at least one concentration except for beta-lactams (SI ≥3) and contrast media (SI ≥4) was considered positive. Contingency tables and ROC curves were used for analysis. RESULTS: Sensitivity and specificity of LTT in the recovery phase of DRESS were 73% and 82%, respectively, whereas in the acute phase, they were only 40% and 30%, respectively. Comparison of skin tests and LTT confirmed a higher sensitivity and specificity of LTT in DRESS. LTT showed high sensitivity (S) and specificity (Sp) for anticonvulsants (S 100%, Sp 100%; P = .008), anti-TB drugs (S 87.5%, Sp 100%; P = .004), and beta-lactams (S 73%, Sp 100%; P = .001). ROC curves revealed that the best criteria for LTT positivity for all drugs are SI ≥2 in at least one concentration, increasing overall sensitivity to 80%, and for beta-lactams from 73% to 92%. CONCLUSIONS AND CLINICAL RELEVANCE: LTT is a good diagnostic tool for drug causality in DRESS, mainly when performed in the recovery phase.

AIMS: To establish the role of cardiolipin (CL) of the membrane in response to the presence of tetradecyltrimethylammonium in Pseudomonas putida A (ATCC 12633). METHODS AND RESULTS: Two ORFs of Ps. putida A (ATCC 12633), which in Ps. putida KT2440 encode the putative CL synthase genes cls and cls2, were cloned, sequenced and mutated. Only the double mutant lacking cls and cls2 showed a reduction of the CL content, 83% lower than the amount produced by the wild-type. Accompanying this change was a 40% decrease in the content of unsaturated fatty acid. Consequently, the membrane of the mutant was more rigid than the one of the parental strain, as observed using fluorescence polarization techniques. The mutant strain showed reduced viability in the presence of tetradecyltrimethylammonium. The incorporation of exogenous CL into its membrane relieved sensitivity to the cationic detergent. CONCLUSIONS: Pseudomonas Putida cells with low levels of CL die in the presence of tetradecyltrimethylammonium, because they cannot counter the fluidizing effect of the cationic surfactant. SIGNIFICANCE AND IMPACT OF THE STUDY: The modification in the membrane phospholipids composition allows knowing the adaptation strategy of Ps. putida when these bacteria are exposed to cationic surfactant.

Several biological processes are involved in the oxidative stress present in diabetes mellitus; among them we can find glucose autooxidation, proteins glycation and decreased antioxidant defenses. Free radicals yielded at mitochondrial level could be a trigger to unchain the vicious circle of the oxidative stress in diabetes mellitus. Aims: to determine antioxidant system’s alterations and indicators of oxidative damage on lipids and proteins in patients with type II Diabetes and a control group. Materials and Methods: It was analyzed 120 serum samples; 60 from patients that suffer type II diabetic from endocrinology surgery belonging “Casa de atención al paciente diabético”in Santa Clara, Villa Clara and 60 samples from healthy individuals used as control group. Spectrofotometric techniques were used to assess levels of Superoxide dismutase and Catalase activity as well as concentrations of reduced glutathione, malonildialdehide and advanced products of proteins Oxidation. Results were compared using the statistical software SPSS. Results: Diabetes type 2 patients showed decreased of Superoxide Dismutase and Catalase enzymatic activity (p = 0,003) and (p = 0,013) respectively as well as and reduced Glutathione levels (p = 0,038). Malondialdehide and Advanced Products of Proteins Oxidation were increased (p = 0,000) in diabetics patients compared with control group. Conclusions: it was found redox alterations in patients that suffer type 2 Diabetes. These alterations are evidenced by a reduced antioxidant enzymatic system and damage on macromolecules such as lipids and proteins...

Introducción: Desde la infancia y la adolescencia pueden aparecer alteraciones en el metabolismo de los lípidos e hidratos de carbono junto con alteraciones en la nutrición, y servir como marcadores cuya identificación en etapas tempranas de la vida puede menguar el desarrollo de episodios patológicos (diabetes mellitus tipo 2, cardiopatías, nefropatías). Sonora (México) es uno de los Estados con mayor prevalencia nacional de obesidad en edades tempranas y de enfermedades crónico-degenerativas y sus complicaciones. Por tal motivo nos propusimos determinar el perfil metabólico y el estado de nutrición en adolescentes de 15-18 años de una comunidad de Sonora. Pacientes y métodos: Estudio transversal analítico, de 56 adolescentes de 15-18 años de edad, en quienes se determinó el peso, la talla, el índice de masa corporal, la glucemia, el perfil lipídico, los valores de insulina y el índice de resistencia a la insulina (HOMA-IR), tomando como punto de corte 3,16 para considerar resistencia a la insulina (RI). Resultados: De los 56 adolescentes, 25 (44,6%) eran de sexo masculino y 31 (55,4%) de sexo femenino. La prevalencia de sobrepeso-obesidad era del 32,1%. Un 17,9% presentaba RI, un 33,3% un índice cintura de cadera elevado, un 8,9% colesterol elevado, un 16,1% triglicéridos elevados, un 25% una disminución de HDL, un 12,3% insulina elevada y solamente el 1,6% glucosa elevada. El 44,4% de los pacientes con sobrepeso-obesidad presentó RI. Conclusión: Uno de cada 4 adolescentes presenta, por lo menos, alguna alteración en el perfil metabólico, y 1 de cada 3 en el estado de nutrición (AU)


Introduction: Alterations in lipid and carbohydrate metabolism coupled with changes in nutrition, may appear in childhood and adolescence and which serve as markers to identify early in life may reduce the development of pathological events (type 2 diabetes, heart disease, nephropathy). Sonora, Mexico, is one of the states with the highest national prevalence of obesity at an early age, and chronic degenerative diseases and their complications. Therefore we decided to determine the metabolic profile and nutritional status in adolescents 15 to 18 years in a community of Sonora, Mexico. Patients and methods: A cross-sectional survey of 56 adolescents aged 15 to 18 years who were weight, height, body mass index, glycemia, lipid profile, insulin, HOMA-IR (resistance index insulin) taking as cutoff consider 3.16 for IR (insulin resistance). Results: Of the 56 teenagers got 25 (44.6%) males and 31 (55.4%) female; prevalence of overweight-obesity 32.1%, 17.9% had IR; 33.3% waist hip ratio high, 8.9% high cholesterol, 16.1% triglycerides, 25% decrease in HDL, high insulin 12.3% and only 1.6% high glucose. The 44.4% of patients with overweight-obesity presents IR. Conclusion: 1 in 4 teens has at least some alteration in the metabolic profile and 1 in 3 in the state of nutrition (AU)
