RESUMEN

Tissue engineering combines suitable cells, engineering methods, and proper biochemical factors to develop functional and biological tissues and repair damaged tissues. In this study, we focused on synthesizing and characterizing a nanocomposite scaffold based on glycerol and azelaic acid (Gl-Az) combined with copper chromite (CuCr2O4) nanoparticles in order to increase the osteogenic differentiation efficiency of human adipose-derived stem cells (hADSCs) on fabricated scaffolds. The degradability and hydrophobicity properties as well as mechanical and thermal behaviors of nanocomposite scaffolds were investigated. Next, the cell toxicity of glycerol, azelaic acid and CuCr2O4 nanoparticles was studied by MTT assay test and acridine orange staining. Finally, the osteogenic differentiation of hADSCs on Gl-Az-CuCr2O4 scaffolds was examined using alkaline phosphatase activity (ALP) and calcium content. The obtained results demonstrated that Gl-Az-1%CuCr2O4 not only showed appropriate mechanical strength, biocompatibility and degradability but also influenced the capability of hADSCs to differentiate into osteogenic lineages. The hADSCs culture in Gl-Az-1%CuCr2O4 showed a significant increase in ALP activity levels and calcium biomineralization after 14 days of osteogenic differentiation. In conclusion, the Gl-Az-1%CuCr2O4 nanocomposite could be used as a biocompatible and degradable scaffold to induce the bone differentiation of hADSCs and it could be a promising scaffold in bone regenerative medicine.

Background: Previous animal studies have shown a protective effect of 5-phosphodiesterase inhibitors on cancer therapeutics-related cardiac dysfunction (CTRCD) of anthracyclines. Aim: The aim of this study was to evaluate the clinical effect of sildenafil on the primary prevention of CTRCD in human. Materials and Methods: In this randomized double-blind clinical trial, the primary end point was efficacy in preventing the reduction of left ventricular ejection fraction (LVEF). The intervention group patients received sildenafil at a dose of 25 milligrams twice a day before starting the chemotherapeutic regimen, and the control group received placebo. All the patients at baseline and after the 6-month follow-up underwent 4D and speckle-tracking echocardiography and cardiac MRI, accompanied by hs-troponin I and NT-Pro-BNP measurement. Results: Sixty patients were enrolled in this study, and data from 52 patients (24 patients in the intervention group and 28 patients in the control group) were used in the final analysis. Our findings showed that in the intervention and control groups, LVEF was dropped from 61.28 ± 7.36 to 51.57 ± 7.67 (difference (D) = -9.71 ± 11.95, p=0.003) and from 57.9 ± 7.29 to 50.2 ± 7.02% (D = -7.7 ± 5.93; p=0.001), respectively (between-group difference = -2.01%, p=0.26). CTRCD was detected in 11 patients in the control group (42.8%) and 10 in the intervention group (41.6%, p=0.51). Conclusion: Consumption of sildenafil for primary prevention of anthracycline-induced cardiac toxicity seems to be unbeneficial. This trial is registered with IRCT20180506039554N1.

Tetanus is an acute, often fatal, infectious neuromuscular disease in all farmed mammals caused by Clostridium tetani. The disease is sporadic but outbreaks of tetanus have been described, as a result of wound contaminated with spores of C. tetani, which sporulates to the vegetative form and produce toxins. The present study reports an outbreak of tetanus in a sheep flock, shortly after ear tagging. Three sheep from a large flock (with a population of 1000 sheep) were presented with signs of: convulsion, limb stiffness, incoordination and trismus ("lock jaw"). There were wounds and scabs in most livestock where ear tags had been attached 1 week prior. Clinical examination revealed tachycardia, dyspnoea with dilated nostrils, mild fever, erected ear pinnae, teeth grinding, mild bloat, muscles rigidity, prolapse of third eyelid and anxiety. According to the history stated by the owner, the case fatality rate of the disease from the beginning was 50% during the outbreak. Necropsy did not reveal any significant finding. Gram-positive bacilli with terminal spores representing C. tetani were isolated in anaerobic cultures which were taken from ear wounds. Procaine penicillin G was administrated at 20 000 IU/kg BW for 5 days, but antiglobulin was not available to treat affected animals. Mortality significantly declined one day after onset of treatment. In this report, the organism was probably introduced by contaminated instruments which were used for ear tagging of sheep. Wound exudation and adhesion following rubbing, created a favourable anaerobic condition for the spores to germinate with production of neurotoxin. Vaccination can protect animals against tetanus, but it does not preclude the need to apply standard hygienic principles when performing management procedures causing wounds. In pasture holding system, many pathogens are present in environment, so tetanus should be considered important in farm animals, because of its high fatality rate and the long course of convalescence.
