RESUMEN

Pine wood nematode disease is currently the most deadly forest disease in China, and the Monochamus alternatus is its primary vector. Controlling the M. alternatus is crucial for managing pine wood nematode disease. This study, based on the selected HasA (pGHKW4) secretory expression vector, used electroporation to combine the genetically modified high-toxicity toxin Cry3Aa-T with the entomopathogenic bacterium Yersinia entomophaga isolated from the gut of the M. alternatus. The SDS-PAGE and Western blotting techniques were employed to confirm the toxin protein's secretion capability. The engineered bacteria's genetic stability and effectiveness in controlling M. alternatus were assessed for their insecticidal activity. The results of the SDS-PAGE and Western blotting analyses indicate that the HasA system effectively expresses toxin protein secretion, demonstrates certain genetic stability, and exhibits high insecticidal activity against M. alternatus. This study constructed a highly toxic entomopathogenic engineered bacterial strain against M. alternatus larvae, which holds significant implications for controlling M. alternatus, laying the foundation for subsequent research and application of this strain.

To screen out suitable herbicides for peach nurseries, we treated the potted seedlings of the peach rootstock 'Nemaguard' with eleven herbicides under recommended doses to investigate the changes of physiological indices and comprehensively evaluate the safety of different herbicides using principal component analysis (PCA). The results showed that soil application of quizalofop-p exhibited no detectable phytotoxicity on rootstock seedlings, while the remaining herbicides generated multiple symptoms, including green loss, wilting, spot, and withering. Starane caused rapid wilting and death, with a 100.0% phytotoxicity index (PI). Soil application of n-(phosphonomethyl)glycine, glufosinate-ammonium, acetochlor, and MCPA-Na showed a PI>65.0%. As compared with the control, all herbicides inhibited leaf area growth to varying degrees, with a 10.0%-56.2% and 5.8%-44.4% reduction in young leaf area and mature leaf area, respectively. All herbicides, except quizalofop-p, increased the electrolyte permeability of leaf and root tip cells by 21.2%-145.0% and 36.9%-291.4%, respectively, and significantly inhibited root growth. The total root length, root surface area, root volume, and the number of root tips significantly decreased by 37.3%-75.3%, 35.7%-83.0%, 44.3%-89.9%, and 42.6%-73.7%, respectively. Although net photosynthetic rate (Pn) and transpiration rate (Tr) of leaves were not significantly affected by quizalofop-p, mesotrione-atrazine, MCPA-Na·bentazone, bensulfuron-methyl·quinclorac, and bensulfuron-methyl·acetochlor, there was significant reduction of 29.6%, 28.9%, 28.4% and 27.9% in Pn and 21.9%, 29.2%, 26.4%, and 19.7% in Tr post soil application of n-(phosphonomethyl)glycine, glufosinate-ammonium, acetochlor, and MCPA-Na. The overall safety ranking of the 11 examined herbicides is as follows: quizalofop-p>bensulfuron-methyl·acetochlor>bensulfuron-methyl·quinclorac>esotrione·atrazine> auizalofop-p·fluoroglycofen>acetochlor>MCPA-Na·bentazone>MCPA-Na>n-(phosphonomethyl)glycine>glufosinate-ammonium>sterane.

Reverse osmosis (RO) membrane materials play a key role in determining energy consumption. Currently, CTA is regarded as having one of the highest degrees of chlorine resistance among materials in the RO process. The hollow fiber membrane has the advantages of a large membrane surface area and a preparation process without any redundant processes. Herein, response surface methodology with Box-Behnken Design (BBD) was applied for optimizing the preparation conditions of the cellulose triacetate (CTA) hollow fiber RO membrane. There were four preparation parameters, including solid content, spinning temperature, post-treatment temperature, and post-treatment time, which could affect the permeability of the membrane significantly. In this study, the interaction between preparation parameters and permeability (permeate flux and salt rejection) was evaluated by regression equations. Regression equations can be applied to obtain the optimized preparation parameters of hollow fiber RO membranes and reasonably predict and optimize the permeability of the RO membranes. Finally, the optimized preparation conditions were solid content (44%), spinning temperature (167 °C), post-treatment temperature (79 °C), and post-treatment time (23 min), leading to a permeability of 12.029 (L·m-2·h-1) and salt rejection of 90.132%. This study of reinforced that CTA hollow fiber membrane may promote the transformation of the RO membrane industry.

Forest soil CO2 efflux (FCO2 ) is a crucial process in global carbon cycling; however, how FCO2 responds to disturbance regimes in different forest biomes is poorly understood. We quantified the effects of disturbance regimes on FCO2 across boreal, temperate, tropical and Mediterranean forests based on 1240 observations from 380 studies. Globally, climatic perturbations such as elevated CO2 concentration, warming and increased precipitation increase FCO2 by 13% to 25%. FCO2 is increased by forest conversion to grassland and elevated carbon input by forest management practices but reduced by decreased carbon input, fire and acid rain. Disturbance also changes soil temperature and water content, which in turn affect the direction and magnitude of disturbance influences on FCO2 . FCO2 is disturbance- and biome-type dependent and such effects should be incorporated into earth system models to improve the projection of the feedback between the terrestrial C cycle and climate change.

Temporomandibular disorders (TMD) can be treated by promoting cartilage regeneration with biomaterials. However, there are deficiencies in the infiltration function of bone filler biological materials. In this study, stems cells were loaded onto gelatin methacryloyl (GelMA) hydrogel microspheres endowed with superwettable properties and TGF-ß sustained-release function, which can quickly infiltrate the irregular surface of the temporomandibular joint (TMJ) bone defect area and accelerate cartilage healing. First, to improve cell adhesion and spreading function, the BMSCs-coated GelMA microspheres were endowed with superwetting property. At the same time, the swelling adsorption characteristics of gelatin microspheres could be used to load recombinant TGF-ß within the microspheres, which could in turn promote the chondrogenic differentiation of multi-potent bone marrow mesenchymal stem cells. The SEM imaging demonstrated that BMSCs-coated GelMA microsphere has superwettable and superhydrophilic property, which enabled rapid adaptation to the bone defect surface morphology, which is conducive to tissue repair. Furthermore, the cartilage defect model showed that rBMSCs-coated GelMA microspheres promote temporomandibular joint arthritis repair. In conclusion, our study established that BMSC-coated GelMA microspheres endowed with superwetting properties, can colonize the bone defect repair site better with sustained release of growth factors, thus providing an innovative strategy for promoting cartilage regeneration.

The early diagnosis of recurrence and metastasis is critically important for decreasing the morbidity and mortality associated with oral cancers. Although liquid biopsy methods hold great promise that provide a successive "time-slice" profile of primary and metastatic oral cancer, the development of non-invasive, rapid, simple, and cost-effective liquid biopsy techniques remains challenging. In this study, an ultrasensitive and selective electrochemical liquid biopsy is developed for oral cancer screening based on tracking trace amounts of cancer biomarker by functionalized asymmetric nano-channels. Detection via antigen-antibody reactions is assayed by evaluating changes in ionic current. Upon the recognition of cancer biomarker antigens in bio-fluids, the inner wall of nano-channel immobilized with the corresponding antibodies undergoes molecular conformation transformation and surface physicochemical changes, which significantly regulate the ion transport through the nano-channel and help achieve sensitivity with a detection limit of 10-12 g mL-1 . Furthermore, owing to the specificity of the monoclonal antibody for the antigen, the nano-channel exhibits high selectivity for the biomarker than for structurally similar biological molecules present in bio-fluids. The effectiveness of this technique is confirmed through the diagnosis of clinical cases of oral squamous cell carcinoma. This study presents a novel diagnostic tool for oral cancer detection in bio-fluids.

RESUMEN

In this study, the first complete mitochondrial genome of Chrysolina aeruginosa Fald was assembled and analyzed. The total length of this mitochondrial genome is 16,335 base pairs. It consists of 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and an AT-rich region. Phylogenomic analysis indicated that C. aeruginosa Fald is sister to Chrysodinopsis sp. This study provides new molecular data for the further taxonomic and phylogenetic studies of the Chrysomelidae of Coleoptera.

OBJECTIVE: To investigate the correlations of hepatitis B virus markers and hepatitis B virus -DNA vectors in blood between women in perinatal period and cord blood, and to assess the risk of HBV infections status in pregnant women to intrauterine fetal infective. METHODS: We selected 612 pregnant women who decided to delivery in hospital, in compliance with the principles of informed consent. According the difference of hepatitis virus serological markers existing in pregnant women, samples were divided into six groups. We used enzyme-linked immunosorbent assay to detect hepatitis virus serological markers, existing in serum of mother and cord blood. Real-time fluorescence quantitative PCR was supplied to test HBV-DNA load levels in these two kinds of biological specimen. RESULTS: In group A, hepatitis B virus "big 3 positives" or 1,3 positive 149 lying-in woman examples, two positive rates of HBV-DNA about pregnant women and cord blood are 99.33% and 32.21%; in group B, positive rates of HBV-DNA in two kinds of specimen are 20.00% and 3.08%; in group C and D, two positive rates are and the average contents of HBV-DNA, the results as mentioned in each group respectively are 65.52%, 12.07% and 13.56%, 1.69% respectively. Control group is group E, 86 lying-in woman examples and the detecting results orderly are 1.16%, 0. There was a significant difference in positive rate of HBV-DNA in cord blood between group A and group B subgroups (chi2 = 54.09, P < 0.01). There is significant positive correlation between HBV-DNA vectors existing in mother's serum and the positive rate of HBV-DNA in cord blood. Hepatitis B virus the mother blood " big 3 positives " is the umbilicus blood HBV-DNA 6345 times that carries quantity in average. CONCLUSION: (1) During the perinatal period, along with the HBV-DNA load levels arising of pregnant women, the risk of HBV infections status in pregnant women to intrauterine fetal infective increased. (2) Suggested to develop the compound pattern human hepatitis B immunoglobulin: Increase the composition of efficient price HBeAb-can be combinated HBeAg, HBsAb can be combinated HBsAg, strengthen the hinderance and break hepatitis B virus disseminate. (3) Our government should strengthen the propaganda of hepatitis B virus education. Establish and perfect to surround and produce the system of health protection.
