RESUMEN

The mulberry plant is a member of the Moraceae family and belongs to the Morus genus. Its entire body is a treasure, with mulberries, mulberry leaves, and mulberry branches all suitable for medicinal use. The main active ingredient in mulberries is mulberry polysaccharide. Studies have shown that polysaccharides from different parts of mulberry exhibit antioxidant, antidiabetic, antibacterial, anti-inflammatory, and blood pressure-lowering properties. There are more studies on the biological activities, extraction methods, and structural characterization of polysaccharides from different parts of mulberry. However, the structural characterization of mulberry polysaccharides is mostly confined to the types and proportions of monosaccharides and the molecular weights of polysaccharides, and there are fewer systematic studies on polysaccharides from different parts of mulberry. In order to better understand the bioactive structure of mulberry polysaccharides, this article discusses the recent research progress in the extraction, separation, purification, bioactivity, structural modification, and application of polysaccharides from different parts of mulberry (mulberry leaves, mulberry fruits, and mulberry branches). It also delves into the pharmacological mechanisms of action of mulberry polysaccharides to provide a theoretical basis for further research on mulberry polysaccharides with a view to their deeper application in the fields of feed and nutraceuticals.

Response surface methodology was selected to explore the ultrasonic-assisted cellulase extraction conditions of Garcinia mangostana rind polysaccharides (GMRPs), and the optimum values of each condition were as follows: ratio of raw material to liquid of 1:50 g/mL, ultrasonic time of 40 min, enzyme concentration of 4 %, and ultrasonic power of 179 W. Based on the above conditions, the average extraction rate of GMRPs was 15.56 %. GMRPs were modified by carboxymethylation, and the relationship between the amount of chloroacetic acid and the substitution degree of carboxymethylated derivative was compared. Based on the results of single factor experiment, it was shown that the amount of chloroacetic acid significantly affected the degree of substitution of derivative products. The above research provides some valuable theoretical references for the preparation of GMRPs and its carboxymethylation products.

The shaddock ped polysaccharide (SPP) was extracted by ultrasound-assisted enzyme method. Phosphorylated shaddock ped polysaccharides (P-SPP) and acetylated shaddock ped polysaccharides (Ac-SPP) were obtained by chemical modification of SPP. The characterization methods such as infrared spectroscopy and nuclear magnetism were employed to characterize the structures of the two derivatives. The antioxidant activity of SPP and its derivatives was investigated by measuring their DPPH radical scavenging capacity, hydroxyl radical ion scavenging capacity and superoxide anion scavenging capacity. In comparison, P-SPP showed better antioxidant activity. The results indicated that the antioxidant activity of the polysaccharides varied with different chemical modifications.

In this study, Shatian pomelo peel was used as the raw material for extracting polysaccharides using hot water extraction (HW) and ultrasonic-assisted enzyme (UVE) methods, respectively. The optimal parameters for extractingShatian pomelo peel polysaccharides (StPP) using the ultrasound-assisted enzymatic method were determined using response surface methodology (RSM). The optimal conditions for the extraction of StPP were as follows: ultrasound power 350 W, ultrasound time 50 min, enzymatic digestion time 50 min, compound enzyme addition 1.5%, and enzymatic digestion temperature 55 °C. The yield of StPP was found to be 30.1310% under these conditions. Comparing the physicochemical properties and antioxidant activity of StPP extracted using different methods, it was observed that ultrasound-assisted enzyme extraction resulted in higher yield, sugar content and glucuronic acid content of StPP compared to traditional hot water extraction. Additionally, StPP extracted by ultrasound-assisted enzyme extraction showed better antioxidant activity. These results suggest that ultrasound-assisted enzymatic extraction is an effective method to enhance the activity of natural polysaccharides.

The crude Solanum tuberdsm polysaccharides (STP) were extracted with hot water. In the process of extraction, proteins, pigments, small molecules and salts in the mixture were removed by Sevage reagent, diatomite and distilled water dialysis, respectively. In addition, the process conditions of protein removal by response surface methodology (RSM) were optimized, and the optimum process conditions of Sevage method were established as follows: ultrasound power 350 W, ultrasound time 20 min, deproteinization twice, volume ratio of polysaccharide solution to Sevage reagent 1:1 (mL/mL). Under these conditions, the protein removal rate was 93.14%.

According to response surface methodology (RSM), the extraction conditions of ultrasound-assisted extraction of polysaccharide from the rinds of Garcinia mangostana L. (GMRP) were optimized and determined. The optimal conditions obtained through optimization were: the liquid to material ratio was 40 mL/g, ultrasonic power was 288 W and extraction time was 65 min. The average extraction rate of GMRP was 14.73%. Ac - GMRP was obtained by acetylation of GMRP, and the antioxidant activities of the two polysaccharides were compared in vitro. The results indicated that the antioxidant capacity of polysaccharide obtained after acetylation was significantly improved compared with that of GMRP. In conclusion, chemical modification of polysaccharide is an effective measure to improve its properties to a certain extent. Meanwhile, it implies that GMRP has great research value and potential.

In order to improve the yield of mung bean peel polysaccharide, on the basis of single-factor experiments, the ultrasonic assisted extraction conditions were optimized by response surface methodology (RSM). The results showed that under the conditions of material-liquid ratio of 1: 40, temperature 77 °C, ultrasonic power 216 W and extraction time 47 min, the extraction rate of mung bean peel polysaccharide was the best, which was 2.55 %. The extracted polysaccharide was phosphorylated and its antioxidant activity in vitro was studied. The results suggested that the modified polysaccharide had a significant scavenging effect on hydroxyl radicals and enhanced the ability of anti-lipid peroxidation, which offered ideas and methods for the development and application of mung bean peel polysaccharide.

Mung bean peel polysaccharides are one of the main active components in mung bean peel. Acetylated mung bean peel polysaccharides were prepared by extracting and acetylating them, and characterized by infrared and ultraviolet methods to preliminarily understand the structural characteristics and activity of acetylated mung bean peel polysaccharides. Acetylation modification can improve the structure of polysaccharides, thereby causing changes in their properties. The product obtained after acetylation modification exhibited new characteristic absorption peaks at 1732 cm-1 , and the scavenging ability of hydroxyl radicals was improved. Therefore, acetylation modification of mung bean peel polysaccharides could enhance the activity by improving the structure, which provided an experimental basis for the application of mung bean peel polysaccharides.

Ipomoea batatas polysaccharides (IBPs) have many important physiological functions. The optimal extraction conditions were extraction time of 40 min, solid-liquid ratio of 1:8 and ultrasonic power of 240 W. 1D/2D nuclear magnetic resonance (1D/2D NMR) analysis showed that the main chain of IBP-1A was mainly composed of →4)-α-D-Glcp-(1→ and →3, 6)-ß-D-Glcp-(1→ residues. In vivo experiments showed that polysaccharide significantly increased the levels of antioxidation-related enzymes and metabolites in older mice. It could significantly relieve oxidative stress injury and delay aging. Therefore, this study provided a new theoretical basis for the development of IBPs as antioxidant food.

Grapefruit peel polysaccharide has antioxidant, antitumor, hypoglycemic and other biological activities, and chemical modification can further improve the properties of the polysaccharide. Acetylation modification of polysaccharides has the advantages of simple operation, low cost and little pollution, and is widely used at present. Different degrees of acetylation modification have different effects on the properties of polysaccharides, so it is necessary to optimize the preparation technology of acetylated grapefruit peel polysaccharides. In this article, acetylated grapefruit peel polysaccharide was prepared by acetic anhydride method. With the degree of acetyl substitution as the evaluation index, combined with the analysis of sugar content and protein content in the polysaccharide before and after modification, the effects of three feeding ratios of 1:0.6, 1 : 1.2 and 1 : 1.8 (polysaccharide: acetic anhydride, mass/volume) on acetylation modification were explored through single factor experiments. The results showed that the optimum ratio of material to liquid for acetylation modification of grapefruit peel polysaccharide was 1:0.6. Under these conditions, the degree of substitution of acetylated grapefruit peel polysaccharide was 0.323, the sugar content was 59.50 % and the protein content was 1.038 %. The results provide some reference for the study of acetylated grapefruit peel polysaccharide.

The natural products have a new chemical structure and biological active diversity, so they are favored by scientific researchers. Gout is a high incidence and high-risk disease, and the current treatments are not satisfactory. Xanthine oxidase (XO) is a key enzyme responsible for the development and progression of various metabolic and oxidative stress-related diseases. Excessive activity of XO leads to elevated serum urate levels, which in turn leads to the development of hyperuricemia. This review mainly introduces the recent progress of the new research on the anti-gout activity of natural products that could provide new treatment methods for gout and information for the discovery and development of new anti-gout drugs.

Pueraria lobata polysaccharides (PLPs) were obtained by a hot water extraction method. Starting from the single factor experiment, the extraction was optimized by response surface methodology, and the following optimal extraction parameters were obtained: the extraction temperature was 84 °C, the liquid-solid ratio was 11 mL/g, the extraction time was 73 min, and the extraction rate of polysaccharides was 8.59%. The Sevag method was used to remove the protein soluble in water and H2O2 was used to remove the pigment; then PLPs were precipitated with three times of anhydrous ethanol, soluble salts and other small molecules were removed by dialysis, and finally refined PLPs were obtained by freeze-drying.

Antitumor, antioxidant, hypoglycemic, and immunomodulatory properties are all exhibited by maize polysaccharides. With the increasing sophistication of maize polysaccharide extraction methods, enzymatic method is no longer limited to a single enzyme to extract polysaccharides, and is more often used in combination with ultrasound or microwave, or combination with different enzymes. Ultrasound has a good cell wall-breaking effect, making it easier to dislodge lignin and hemicellulose from the cellulose surface of the maize husk. The "water extraction and alcohol precipitation" method is the simplest but most resource- and time-consuming process. However, the "ultrasound-assisted extraction" and "microwave-assisted extraction" methods not only compensate for the shortcoming, but also increase the extraction rate. Herein, the preparation, structural analysis, and activities of maize polysaccharides were analyzed and discussed.

The maidenhairtree polysaccharides (MTPs) have important application prospects. So, the extraction, purification, structure, derivatization and biological activities of polysaccharides from leaves, fruits, and testae of maidenhairtree were disscussed. Polysaccharides were extracted by collaborative extraction methods such as ultrasound-assisted extraction and microwave-assisted extraction. The ultrasound-assisted extraction had higher content and higher efficiency. The structural characteristics and structure-activity relationship of maidenhairtree polysaccharides were studied in order to provide theoretical basis and technical support for the further development and utilization of maidenhairtree polysaccharides.

Pueraria lobata polysaccharides (PLPs) were obtained by hot water extraction using Pueraria lobata as raw material. Structural analysis revealed that PLPs may have a repetitive backbone units of →4) -α-D-Glcp (1→4-α-D-Glcp (1→. Phosphorylated Pueraria lobata polysaccharides (P-PLPs), carboxymethylated Pueraria lobata polysaccharides (CM-PLPs) and acetylated Pueraria lobata polysaccharides (Ac-PLPs) were obtained by chemical modifications of PLPs, respectively. The physicochemical properties and antioxidant activities of these four Pueraria lobata polysaccharides were studied in comparison. In particular, the clearance rate of P-PLPs exceeded 80 %, and was expected to achieve the same effect as Vc . The results showed that the effects of different chemical modifications on the antioxidant activity of PLPs varied greatly.

Response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction conditions of Ginkgo biloba leaves polysaccharide (GBLP). The optimum extraction conditions for the ultrasound-assisted extraction of GBLP were obtained as liquid to material ratio of 30 mL/g, ultrasonic power of 340 W, and extraction time of 50 min. Under these conditions, the yield of GBLP was 5.37 %. Two chemically modified polysaccharides, CM-GBLP and Ac-GBLP, were obtained by carboxymethylation and acetylation of GBLP. The physicochemical properties of these three polysaccharides were comparatively studied and their in vitro antioxidant activities were evaluated comprehensively. The results showed that the solubility of the chemically modified polysaccharides was significantly enhanced and the in vitro antioxidant activity was somewhat improved. This suggests that carboxymethylation and acetylation are effective methods to enhance polysaccharide properties, but the results exhibited some uncontrollability. At the same time, GBLP has also shown high potential for research and application.

We introduced a strategy for preparing a carbohydrate microarray and demonstrated its utility for characterizing carbohydrate binding and activities. We isolated the lipopolysaccharide (LPS) components from different bacteria and explored the possibility of immobilizing these glycoconjugates on a high-binding polystyrene plate. Carbohydrate-specific combination was examined by observing the binding of the blood group B analogic LPS O-polysaccharide from Escherichia coli on the high-binding polystyrene plate and anti-B from a broad spectra antibody of human blood serum. Strong binding of antibodies was screened, as it was evident that relative response value is two times higher than control. The hybridization results indicated that this method is a reliable technique for the detection of human intestinal bacteria and is expected to be applied in diagnostics and seroepidemiology.

Rheumatoid arthritis (RA) is a chronic and systemic disease of inflammatory synovitis with unknown etiology. In previous studies, we found that the double-ring conjugated enone structure has anti-rheumatoid arthritis activity and could effectively inhibit the proliferation of rat synovial cells in vitro and has good anti-inflammatory activity in vivo. Herein, we further modified the structure, which was a novel double-ring conjugated enone, to study its anti-rheumatoid arthritis activity. Results showed that the most potent compound 32 could effectively inhibit the proliferation of rat synovial cells in vitro and has better anti-inflammatory activity compared with that of the positive control methotrexate, as shown by in vivo activity evaluation. More interestingly, compound 32 could effectively inhibit the increase of TNF-α, IL-1ß, and IL-6 induced by LPS and regulate the expression of TLR4, MyD88, NF-κB, and IκB in the signaling pathway of TLR4/NF-κB. Our results provided a promising starting point for the development of highly effective small molecules for the treatment of RA.

Fermentum is a common unicellular fungus with many biological activities attributed to ß-polysaccharides. Different in vivo and in vivo experimental studies have long proven that fermentum ß-polysaccharides have antioxidant, anti-tumor, and fungal toxin adsorption properties. However, there are many uncertainties regarding the relationship between the structure and biological activity of fermentum ß-polysaccharides, and a systematic summary of fermentum ß-polysaccharides is still lacking. Herein, we reviewed the research progress about the extraction, structure and modification, structure-activity relationship, activity and application of fermentum ß-polysaccharides, compared the extraction methods of fermentum ß-polysaccharide, and paid special attention to the structure-activity relationship and application of fermentum ß-polysaccharide, which provided a strong basis for the development and application of fermentum ß-polysaccharide.

Taro, a staple food for residents in Africa and parts of Asia, is an important source of carbohydrate. China has abundant taro resources. Taro contains polysaccharide, vitamins, minerals and other substances. Taro polysaccharides, as a significant active ingredient in taro, are mainly composed of monosaccharide units such as glucose, galactose, arabinose, mannose, and so on. Taro polysaccharides have antioxidant, lipid-lowering, and immunomodulatory effects. In today's world, people are interested in food containing natural ingredients, which stimulates the potential of taro polysaccharides in the food, pharmaceutical, medical, and other fields. Herein, the extraction and purification, structural characterization, functional activity, and application of taro polysaccharides are reviewed to strengthen the cognition of taro polysaccharides.
