RESUMEN

Growth factors are commonly added to cell culture media in cellular agriculture to mimic the endogenous process of proliferation and differentiation of cells. Many of these growth factors are endogenous to humans and known to be present in the edible tissues and milk of food animals. However, there is little or no information on the use of growth factors intentionally added in food production before the advent of cultivated meat. Ten commonly used growth factors have been reviewed to include information on their mode of action, bioavailability, occurrence in food and food animals, endogenous levels in humans, as well as exposure and toxicological information drawn from relevant animal studies and human clinical trials with a focus on oral exposure. In addition, a comparison of homology of growth factors was done to compare the sequence homology of growth factors from humans and domestic animal species commonly consumed as food, such as bovine, porcine, and poultry. This information has been gathered as the starting point to determine the safety of use of growth factors in cultivated meat meant for human consumption. The change in levels of growth factors measured in human milk and bovine milk after pasteurization and high-temperature treatment is discussed to give an indication of how commercial food processing can affect the levels of growth factors in food. The concept of substantial equivalence is also discussed together with a conservative exposure estimation. More work on how to integrate in silico assessments into the routine safety assessment of growth factors is needed.

Bisphenol F (BPF, 4-[(4-hydroxyphenyl)methyl]phenol) is a bisphenol that is structurally similar to bisphenol A (BPA). In response to consumer concern towards BPA, industry has started to substitute BPA for BPF and other bisphenol analogues in the production of epoxy resins and coatings for various applications. In 2016, it was reported that commercially sold mustard contained naturally occurring BPF. Here, the existing literature was reviewed to investigate whether other natural sources of BPF among edible plants exist, including their impact on human exposure to BPF. Coeloglossum viride var. bracteatum (rhizome), Galeola faberi (rhizome), Gastrodia elata (rhizome), Xanthium strumarium (seeds) and Tropidia curculioides (root) were found to contain naturally occurring BPF. Botanical extracts from these plants are used in traditional Chinese medicine. The highest values of BPF were recorded for G. elata and T. curculioides. Information on precise doses of the plant extracts used is scarce; however, for G. elata, also known as Tian Ma and available in powder form, a daily exposure of BPF from this source could theoretically amount up to 4.5 µg/kg body weight per day (based on a 70 kg body weight). Therefore, herbal products used in traditional Chinese medicine should be considered as a potential source contributing to the overall human exposure when assessing endocrine-active bisphenolic compounds.

Brominated and phosphorous flame retardants (BFRs, PFRs) are added to household and consumer products to reduce their flammability. Some FRs are persistent in the environment and may have adverse health effects. As exposure indoors contributes significantly to total exposure, we wanted to estimate the exposure of children (3 years of age) through dust ingestion, inhalation, and dermal absorption. We measured 17 BFRs and 10 PFRs in indoor dust, predicted their respective concentrations in the indoor air and assessed children's exposure. Among the BFRs, decabromodiphenyl ether (BDE-209) had highest median level in the dust (411 ng/g) followed by decabromodiphenyl ethane (DBDPE, 119 ng/g) and bis-ethylhexyl tetrabromophthalate (BEH-TEBP, 106 ng/g). Among the PFRs, trisbutoxyethyl phosphate (TBOEP) had the highest concentration (11100 ng/g) followed by tris(2-chloroisopropyl) phosphate (TCIPP, 1870 ng/g) and triphenyl phosphate (TPHP, 773 ng/g). FR concentration in air predicted from dust concentrations were within the interquartile range of experimental data for 10/13 of BFRs and 4/8 of PFRs compared. Dust ingestion was the major route of exposure (75-99%) for higher molecular weight BFRs, TBOEP and phenyl based PFRs (73-77%). Inhalation was important for volatile BFRs like pentabromobenzene (PBB 71%) and pentabromotoluene (PBT 52%) and dermal exposure for volatile chlorinated PFRs (TCEP 84%, TCIPP 77%). Margins of Exposure (MoE) were calculated as the ratio of total exposure to oral Reference Dose (RfD). MoEs were lowest for TCEP (220), TBOEP (240) and TCIPP (830), and > 1000 for all other FRs. These MoEs imply no risk for Finnish children by the studied FRs.
