Spartina alterniflora has rapidly and extensively encroached on China's coastline over the past decades. Among the coastal areas invaded by S. alterniflora, at most 93% are mudflats. However, the effect of S. alterniflora invasion on soil organic carbon (SOC) stocks of coastal mudflats has not been systematically studied on a national scale. Here, we quantified the nationwide changes in SOC stocks in coastal mudflats associated with S. alterniflora invasion between 1990 and 2020. We found that S. alterniflora invasion significantly enhanced SOC stocks in coastal China. Nonetheless, the benefit of S. alterniflora invasion of coastal SOC stock may be weakened by continuing human intervention. We found that S. alterniflora invading mudflats added 2.3 Tg SOC stocks to China's coastal blue carbon, while 1.78 Tg SOC stocks were lost mainly due to human activities, resulted in a net SOC stock gain of 0.52 Tg C. These findings overturned the traditionally thought that S. alterniflora invasion would reduce ecosystem services by highlighting that the historical invasion of S. alterniflora has broadly and consistently enhanced blue carbon stock in coastal China.

This study aims to determine the molecular mechanisms and analgesic effects of transient receptor potential vanilloid 1 (TRPV1) in the treatments of osteoarthritis (OA) and rheumatoid arthritis (RA). We summarize and analyse current studies regarding the biological functions and mechanisms of TRPV1 in arthritis. We search and analyse the related literature in Google Scholar, Web of Science and PubMed databases from inception to September 2023 through the multi-combination of keywords like 'TRPV1', 'ion channel', 'osteoarthritis', 'rheumatoid arthritis' and 'pain'. TRPV1 plays a crucial role in regulating downstream gene expression and maintaining cellular function and homeostasis, especially in chondrocytes, synovial fibroblasts, macrophages and osteoclasts. In addition, TRPV1 is located in sensory nerve endings and plays an important role in nerve sensitization, defunctionalization or central sensitization. TRPV1 is a non-selective cation channel protein. Extensive evidence in recent years has established the significant involvement of TRPV1 in the development of arthritis pain and inflammation, positioning it as a promising therapeutic target for arthritis. TRPV1 likely represents a feasible therapeutic target for the treatment of OA and RA.

Extracellular enzymes play central roles in the biogeochemical cycles in wetland ecosystems. Their activities are strongly impacted by hydrothermal conditions. Under the ongoing global change, many studies reported the individual effects of flooding and warming on extracellular enzyme activities, however, few researches investigated their interactive effects. Therefore, the current study aims to determine the responses of extracellular enzyme activities to warming in wetland soils under divergent flooding regimes. We investigated the temperature sensitivity of seven extracellular enzymes related to carbon (α-glucosidase, AG; ß-glucosidase, BG; cellobiohydrolase, CBH; ß-xylosidase, XYL), nitrogen (ß-N-acetyl -glucosaminidase, NAG; leucine aminopeptidase, LAP), and phosphorus (Phosphatase, PHOS) cycling along the flooding duration gradient in a lakeshore wetland of Poyang Lake, China. The Q10 value, calculated using a temperature gradient (10, 15, 20, 25, and 30 °C), was adopted to represent the temperature sensitivity. The average Q10 values of AG, BG, CBH, XYL, NAG, LAP, and PHOS in the lakeshore wetland were 2.75 ± 0.76, 2.91 ± 0.69, 3.34 ± 0.75, 3.01 ± 0.69, 3.02 ± 1.11, 2.21 ± 0.39, and 3.33 ± 0.72, respectively. The Q10 values of all the seven soil extracellular enzymes significantly and positively correlated with flooding duration. The Q10 values of NAG, AG and BG were more sensitive to the changes in flooding duration than other enzymes. The Q10 values of the carbon, nitrogen, and phosphorus-related enzymes were mainly determined by flooding duration, pH, clay, and substrate quality. Flooding duration was the most dominant driver for the Q10 of BG, XYL, NAG, LAP, and PHOS. In contrast, the Q10 values of AG and CBH were primarily affected by pH and clay content, respectively. This study indicated that flooding regime was a key factor regulating soil biogeochemical processes of wetland ecosystems under global warming.

Surface complexation of arsenite (As(III)) on colloidal ferric hydroxide (CFH) plays an important role not only in the adsorptive immobilization of As(III) but also in the subsequent oxidation of As(III) to arsenate (As(V)) through light-induced ligand-to-metal charge transfer (LMCT) in water at near-neutral pH. However, the effects of natural organic matter (NOM), especially humic substances (HSs) and low molecular weight carboxylic acids (CAs), on the photochemistry of the CFH-As(III) system have not been sufficiently understood. In this work, the inhibition of photooxidation of As(III) in terms of the observed apparent rate constant (kobs) by six HSs (below 16 mg L-1) and seven CAs (below 2.5 mM) has been observed in water containing 66 µM Fe(III) and 5 µM As(III) at pH 7 under simulated solar irradiation consisting of UVA (λmax 365 nm) and UVB (λmax 313 nm) lights. Total inhibition factors (T) have been determined from the combined effect of light-screening factor (S) and competitive complexation factor (C), wherein both S and C varied with NOM concentration. S was obtained by determining the absorbance of NOM, and C was obtained by fitting modified Langmuir or Freundlich models to the amount of As(III) desorbed from CFH upon the addition of NOM. Statistical analysis between the experimental Texp and the calculated one according to Tcal = S × C showed that the Freundlich model (RMSE for HS 0.1609 and for CA 0.1771) was better than the Langmuir model and was statistically robust (QLOO2= 0.691 > 0.5). This work provided an estimation method for the effects of NOM on As(III) photooxidation in the presence of CFH as well as a deeper understanding of the transformation of arsenic species in sunlit water.

In the recent decades, the area of seasonal water (SEW) has substantially increased at the global scale. To evaluate nutrient dynamics in aquatic ecosystems, previous studies have analyzed the determining factors of sediment nutrient content and stoichiometry on whole sediment profiles without depth separation on SEW sites. Such a methodology assumes that SEW sediment is a uniform unit and its nutrient dynamics are regulated by the same mechanism at various depths (uniformity assumption). We tested this assumption using sediment samples from six depth increments of 154 sediment profiles (1 m depth) on SEW sites at Shengjin Lake in subtropical China. We measured sediment total nitrogen (STN), total phosphorus (STP), nutrient fractions, and the molar ratio of STN to STP (RSNP), and investigated their determining factors at various depths. STN, STP, and RSNP were averaged at 1.34 g/kg, 0.55 g/kg, and 5.43, respectively, and all gradually decreased with depth. STN was positively affected by moisture and flooding duration in all depth increments. Instead, the major determining factors of STP changed from particle size at 0-20 cm of depth to pH and electrical conductivity at 30-100 cm of depth. These vertical patterns have close connections with sediment nutrient fractions since sediment N fractions did not shift along profile depths (i.e., over 99 % of STN was organic N) but sediment P fractions did (the percentage of Fe-P and Al-P decreased by 6.25 % but those of Ca-P increased by 4.31 % along the sediment depth gradient). The major determining factors of RSNP showed no obvious vertical patterns because they frequently varied along depth gradients. The results demonstrate that SEW sediment is not a uniform unit and the determining factors of nutrient dynamics change with depth. Our study highlights the importance of improved methodological reflection in studies addressing sediment nutrient dynamics on SEW sites.

Osteoarthritis (OA) is the most common degenerative joint disease that causes painful swelling and permanent damage to the joints in the body. The molecular mechanisms of OA are currently unknown. OA is a heterogeneous disease that affects the entire joint, and multiple tissues are altered during OA development. To better understand the pathological mechanisms of OA, new approaches, methods, and techniques need to be used to understand OA pathogenesis. In this review, we first focus on the epigenetic regulation of OA, with a particular focus on DNA methylation, histone modification, and microRNA regulation, followed by a summary of several key mediators in OA-associated pain. We then introduce several innovative techniques that have been and will continue to be used in the fields of OA and OA-associated pain, such as CRISPR, scRNA sequencing, and lineage tracing. Next, we discuss the timely updates concerning cell death regulation in OA pathology, including pyroptosis, ferroptosis, and autophagy, as well as their individual roles in OA and potential molecular targets in treating OA. Finally, our review highlights new directions on the role of the synovial lymphatic system in OA. An improved understanding of OA pathogenesis will aid in the development of more specific and effective therapeutic interventions for OA.

Understanding the influencing factors of cyberbullying is key to effectively curbing cyberbullying. Among the various factors, this study focused on the personal level of individual students and categorized the influencing factors of cyberbullying among college students into five sublevels, i.e., background, Internet use and social network habits, personality, emotion, and literacy related to digital citizenship. Then a questionnaire survey was applied to 947 Chinese college students. The results show that cyberbullying among Chinese college students are generally at a low level. There are many factors influence cyberbullying. Specifically, at the personal background level, gender has a significant impact on cyberbullying and being cyberbullied. In terms of personal Internet use and social network habits, students' average daily online time has no significant correlation with cyberbullying and being cyberbullied; however, the proportion of online non-learning time has a significantly positive correlation with cyberbullying, and the proportion of online learning/work time has a significant impact on being cyberbullied. At the personality level, the Big Five personality traits have varying degrees of correlation with and influence on cyberbullying and being cyberbullied. At the personal emotions level, students' life satisfaction has a significantly negative correlation with cyberbullying and being cyberbullied while it only has a significant impact on being cyberbullied; the personal stress and empathetic concern aspects of empathy have a significantly positive correlation with cyberbullying and being cyberbullied among female students. At the literacy related to digital citizenship level, students' understanding of and compliance with Internet etiquette have significantly negative impacts on cyberbullying; the ability to communicate and collaborate online and Internet addiction have significantly positive impacts on cyberbullying and being cyberbullied; the understanding of and compliance with relevant digital laws and regulations have significantly negative correlations with cyberbullying and being cyberbullied. Overall, college students' digital citizenship level has a significantly negative correlation with cyberbullying but no significant correlation with being cyberbullied. Finally, analysis and suggestions were provided according to these statistical results and the effects of these factors on cyberbullying and being cyberbullied among college students, so as to help solve this problem and provide a new perspective for research in this field.

Iron species have essential influence on the environmental/geochemical behaviors of arsenic species in water and soil. Colloidal ferric hydroxide (CFH) induces photooxidation of arsenite (As(III)) to arsenate (As(V)) in water at neutral pH through surface complexation and ligand-to-metal charge transfer (LMCT). However, the effect of the co-existing natural organic matter (NOM) on the complexation-photolysis in this process has remained unclear. In the present work, the photooxidation of As(III) induced by CFH was investigated in the presence of various carboxylic acids and polyphenols as simple model compounds of NOM. Two different light sources of ultraviolet A (UVA) (λmax = 365 nm) and ultraviolet B (UVB) (λmax = 313 nm) were used for photooxidation treatment of the experimental ternary system and the control binary system respectively. The obtained results demonstrated that all investigated NOM inhibited the photooxidation of As(III) in the As(III)/CFH system at pH 7. Moreover, the correlation analysis between the pseudo-first order rate constant kobs and various property parameters of NOM showed that the stable constant for the complexation between Fe(III) and NOM (logKFe-NOM) as well as the molecular weight of NOM and the percentages of total acidity of NOM exhibited significant correlations. A simple quantitative structure-activity relationship (QSAR) model was established between kobs and these three parameters utilizing a multiple linear regression method, which can be employed to estimate the photooxidation efficiency of As(III) in the presence of ferric iron and NOM. Thus, the present work contributes to the understanding of the environmental interactions between NOM and iron.

Patients with hereditary tyrosinemia type I (HT1) present acute and irreversible liver and kidney damage during infancy. CRISPR-Cas9-mediated gene correction during infancy may provide a promising approach to treat patients with HT1. However, all previous studies were performed on adult HT1 rodent models, which cannot authentically recapitulate some symptoms of human patients. The efficacy and safety should be verified in large animals to translate precise gene therapy to clinical practice. Here, we delivered CRISPR-Cas9 and donor templates via adeno-associated virus to newborn HT1 rabbits. The lethal phenotypes could be rescued, and notably, these HT1 rabbits reached adulthood normally without 2-(2-nitro-4-trifluoromethylbenzyol)-1,3 cyclohexanedione administration and even gave birth to offspring. Adeno-associated virus (AAV)-treated HT1 rabbits displayed normal liver and kidney structures and functions. Homology-directed repair-mediated precise gene corrections and non-homologous end joining-mediated out-of-frame to in-frame corrections in the livers were observed with efficiencies of 0.90%-3.71% and 2.39%-6.35%, respectively, which appeared to be sufficient to recover liver function and decrease liver and kidney damage. This study provides useful large-animal preclinical data for rescuing hepatocyte-related monogenetic metabolic disorders with precise gene therapy.

BACKGROUND: Many favorable traits of crops and livestock and human genetic diseases arise from multiple single nucleotide polymorphisms or multiple point mutations with heterogeneous base substitutions at the same locus. Current cytosine or adenine base editors can only accomplish C-to-T (G-to-A) or A-to-G (T-to-C) substitutions in the windows of target genomic sites of organisms; therefore, there is a need to develop base editors that can simultaneously achieve C-to-T and A-to-G substitutions at the targeting site. RESULTS: In this study, a novel fusion adenine and cytosine base editor (ACBE) was generated by fusing a heterodimer of TadA (ecTadAWT/*) and an activation-induced cytidine deaminase (AID) to the N- and C-terminals of Cas9 nickase (nCas9), respectively. ACBE could simultaneously induce C-to-T and A-to-G base editing at the same target site, which were verified in HEK293-EGFP reporter cell line and 45 endogenous gene loci of HEK293 cells. Moreover, the ACBE could accomplish simultaneous point mutations of C-to-T and A-to-G in primary somatic cells (mouse embryonic fibroblasts and porcine fetal fibroblasts) in an applicable efficiency. Furthermore, the spacer length of sgRNA and the length of linker could influence the dual base editing activity, which provided a direction to optimize the ACBE system. CONCLUSION: The newly developed ACBE would expand base editor toolkits and should promote the generation of animals and the gene therapy of genetic diseases with heterogeneous point mutations.

Kumari et al. investigated the arsenic resistance and arsenic biotransforming ability in three bacterial species, mainly Rhodococcus sp.. But in the use of response surface methodology to optimize key parameters that affect the removal of arsenic, the effects of dissolved oxygen and the effect of HA addition on the adsorption or oxidation of arsenic by bacteria were ignored. We therefore recommend taking the impacts of the above-mentioned factors on the removal of arsenic into examination.

Cytosine base editors (CBEs) enable programmable C-to-T conversion without DNA double-stranded breaks and homology-directed repair in a variety of organisms, which exhibit great potential for agricultural and biomedical applications. However, all reported cases only involved C-to-T substitution at a single targeted genomic site. Whether C-to-T substitution is effective in multiple sites/loci has not been verified in large animals. Here, by using pigs, an important animal for agriculture and biomedicine, as the subjective animal, we showed that CBEs could efficiently induce C-to-T conversions at multiple sites/loci with the combination of three genes, including DMD, TYR, and LMNA, or RAG1, RAG2, and IL2RG, simultaneously, at the embryonic and cellular levels. CBEs also could disrupt genes (pol gene of porcine endogenous retrovirus) with dozens of copies by introducing multiple premature stop codons. With the CBEs, pigs carrying single gene or multiple gene point mutations were generated through embryo injection or nuclear transfer approach.
