RESUMEN

PURPOSE: The study aims to assess the effects of dexmedetomidine (Dex) pretreatment on patients during cardiac valve replacement under cardiopulmonary bypass. METHODS: For patients in the Dex group (n = 52), 0.5 µg/kg Dex was given before anesthesia induction, followed by 0.5 µg/kg/h pumping injection before aortic occlusion. For patients in the control group (n = 52), 0.125 ml/kg normal saline was given instead of Dex. RESULTS: The patients in the Dex group had longer time to first dose of rescue propofol than the control group (P = 0.003). The Dex group required less total dosage of propofol than the control group (P = 0.0001). The levels of cardiac troponin I (cTnI), creatine kinase isoenzyme MB (CK-MB), malondialdehyde (MDA), and tumor necrosis factor-α (TNF-α) were lower in the Dex group than the control group at T4, 8 h after the operation (T5), and 24 h after the operation (T6) (P <0.01). The Dex group required less time for mechanical ventilation than the control group (P = 0.003). CONCLUSION: The study suggests that 0.50 µg/kg Dex pretreatment could reduce propofol use and the duration of mechanical ventilation, and confer myocardial protection without increased adverse events during cardiac valve replacement.

RESUMEN

Chemotherapy-induced neuropathic pain (CINP) is a dose-limiting adverse event affecting 40% of chemotherapy patients. MiRNA-mRNA interaction plays an important role in various processes. However, detailed profiling of miRNA-mRNA interactions in CINP remains unclear. Here, a rat-based CINP model was established using paclitaxel, followed by nociceptive behavioral tests related to mechanical allodynia, thermal hyperalgesia, and cold allodynia. The landscape of miRNA-mRNA interaction in the spinal dorsal horn was investigated through mRNA transcriptomics and small RNA sequencing. Under CINP condition, 86 differentially expressed mRNAs and 56 miRNAs were identified. Gene Set Enrichment Analysis (GSEA), Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses indicated the activity of Odorant binding, postsynaptic specialization and synaptic density, extracellular matrix, mitochondrial matrix, retrograde endocannabinoid signaling, and GTPase activity. Protein-protein interaction (PPI), networks of circRNA-miRNA-mRNA, lncRNA-miRNA-mRNA, and TF-genes were demonstrated. We next explored the immune infiltration microenvironment and found a higher infiltration abundance of Th17 and a lower abundance of MDSC in CINP. RT-qPCR and dual-luciferase assays were used to verify the sequencing results, and single-cell analysis based on the SekSeeq database was conducted. Combined with bioinformatics analyses and experimental validations, Mpz, a protein-coding gene specifically expressed in Schwann cells, was found critical in maintaining CINP under miRNA regulation. Therefore, these data highlight the expression patterns of miRNA-mRNA, and the underlying mechanism in the spinal dorsal horn under CINP condition, and Mpz may serve as a promising therapeutic target for patients with CINP.

Early Cretaceous mafic rocks are first reported in the northern Guangxi region from the western Qin-Hang belt in the interior South China Block. A systematic investigation of zircon U-Pb dating, whole-rock geochemistry, Sm-Nd isotopes and zircon Hf-O isotopes for these mafic rocks reveals their petrogenesis and the mantle composition as well as a new window to reconstruct lithospheric evolution in interior South China Block during Late Mesozoic. Zircon U-Pb dating yielded ages of 131 ± 2 Ma to 136 ± 2 Ma for diabase and gabbro from Baotan area, indicating the first data for Early Cretaceous mafic magmatism in the western Qing-Hang belt. These mafic rocks show calc-alkaline compositions, arc-like trace element distribution patterns, low zircon εHf(t) of - 9.45 to - 6.17 and high δ18O values of + 5.72 to + 8.09‰, as well as low whole-rock εNd(t) values of - 14.27 to - 9.53. These data suggest that the studied mafic rocks are derived from an ancient lithospheric mantle source that was metasomatized during Neoproterozoic subduction. Thus, the occurrence of these mafic rocks indicates a reactivation of Neoproterozoic subducted materials during an extension setting at Late Mesozoic in the western Qin-Hang belt, an old suture zone that amalgamates the Yangtze and Cathaysia blocks.

The understanding of surface chemistry changes on oxygen electrodes is critical for the development of reversible solid oxide fuel cell (RSOFC). Here, we report for the first time that the electrochemical potentials can drastically affect the surface composition and hence the electrochemical activity and stability of PrBaCo2 O5+δ (PBCO) electrodes. Anodic polarization degrades the activity of the PBCO electrode, whereas the cathodic bias could recover its performance. Alternating anodic/cathodic polarization for 180 h confirms this behavior. Microstructure and chemical analysis clearly show that anodic bias leads to the accumulation and segregation of insulating nanosized BaO on the electrode surface, whereas cathodic polarization depletes the surface species. Therefore, a mechanism based on the segregation and incorporation of BaO species under electrochemical potentials is considered to be responsible for the observed deactivation and recovery process, respectively.

We systematically investigated the size- and shape-dependent SERS activities of plasmonic core-shell nanoparticles towards detection of the pesticide thiram. Monodisperse Au@Ag nanocubes (NCs) and Au@Ag nanocuboids (NBs) were synthesized and their Ag shell thickness was precisely adjusted from ∼1 nm to ∼16 nm. All these nanoparticles were used as SERS substrates for thiram detection, and the Raman intensities with three different lasers (514 nm, 633 nm and 782 nm) were recorded and compared. Our results clearly show that: (1) the excitation wavelength discriminated particle shapes regardless of particle sizes, and the maximized Raman enhancement was observed when the excitation wavelength approaches the SERS peak (provided there is significant local electric field confinement on the plasmonic nanostructures at that wavelength); (2) at the optimized laser wavelength, the maximum Raman enhancement was achieved at a certain threshold of particle size (or silver coating thickness). By exciting particles at their optimized sizes with the corresponding optimized laser wavelengths, we achieved a detection limit of roughly around 100 pM and 80 pM for NCs and NBs, respectively.
