RESUMEN

Diagnosis and treatment of heart diseases due to changes in the coronary vascularization need a detailed knowledge on the morphology and possible variations of the aortic valves and coronary ostia. This study was performed to clarify details on morphology of these structures in domestic cats. The tricuspid aortic valve was examined in 65 domestic shorthair cats. The location of coronary ostia was determined either inferior to (26 and 20%, left and right coronary ostium - LCO and RCO), at (65 and 66%) or superior to the intercommissural line (9 and 14%). In 13 cats (20%), accessory ostia were found either for left, right or both coronary arteries (LCA and RCA). Their position varied between specimens. They were located beyond the main ostium, at its edge, or inside just below the edge. In one cat, no main trunk of the LCA was found. In one cat, two accessory ostia next to the RCO were observed. Coronary ostia in cats show anatomical variants and morphological anomalies. This study provides basic data useful for, for example, angiography performed for diagnosis of cardiac diseases and as a basis for surgical interventions.

Salmonella Typhimurium is one of the main pathogens compromising porcine and human health as well as food safety, because it is a prevailing source of foodborne infections due to contaminated pork. A prominent problem in the management of this bacteriosis is the number of subclinically infected carrier pigs. As very little is known concerning the mechanisms allowing Salmonella to persist in pigs, the objective of this study was to develop an immunohistochemical approach for the detection of salmonellae in tissue of pigs experimentally infected with Salmonella Typhimurium. Samples were obtained from a challenge trial in which piglets of the German Landrace were intragastrically infected with Salmonella enterica serovar Typhimurium DT104 (1.4-2.1x1010 CFU). Piglets were sacrificed on days 2 and 28 post infection. Tissue samples of jejunum, ileum, colon, ileocecal mesenteric lymph nodes (Lnn. ileocolici), and tonsils (Tonsilla veli palatini) were fixed in Zamboni's fixative and paraffin-embedded. Different immunohistochemical staining protocols were evaluated. Salmonella was detected in varying amounts in the tissues. Brown iron-containing pigments in the lymph nodes interfered with the identification of Salmonella if DAB was used as a staining reagent. Detergents like Triton X-100 or Saponin enhanced the sensitivity. It seems advisable not to use a detection system with brown staining for bacteria in an experimental setup involving intestinal damage including haemorrhage. The use of detergents appears to result in a higher sensitivity in the immunohistochemical detection of salmonellae.

Zinc oxide (ZnO) used in high ('pharmacological') levels to prevent diarrhoea in pigs is assumed to reduce copper (Cu) in tissues and inhibits large intestinal microbial fermentation. To test it, German Landrace pigs were weaned on d28 of age and fed diets containing either 100 (LowZinc, LZn, n = 10) or 3100 mg ZnO/kg (HighZinc, HZn, n = 10). The mixed feed (13.0 MJ ME, 18.5% crude protein) was based on wheat, barley, soya bean meal and maize. After 4 weeks, the HZn group was further fed 100 mg ZnO/kg for another 2 weeks. Caecal contents, faeces and tissues were collected after 4 weeks (n = 5 and n = 10 respectively) and 6 weeks (n = 5 and n = 5 respectively). Faeces and caecal content were analysed for dry matter (DM), pH, ammonia, lactic acid (LA) and short-chain fatty acids (SCFA) on native water basis. anova was performed to elucidate significant differences at p < 0.05. No diarrhoea occurred. After 4 weeks, the caecal contents' pH increased (p < 0.001) and butyric (p < 0.05) and valeric acid (p < 0.01) decreased in the HZn group in comparison with LZn. In faeces, a decrease of acetic (p = 0.009), butyric (p = 0.007) and valeric acid (p = 0.046), as well as reduced acetic:propionic acid (A:P) ratio (p = 0.025) was observed in the HZn group in comparison with LZn. Faecal ammonia decreased in HZn (p = 0.018). No differences (p > 0.05) were recorded in caecal contents after 6 weeks. In faeces, acetic acid remained lower in the HZn group in comparison with LZn (p = 0.006), as did the A:P ratio (p = 0.004). Zn concentration in liver, kidneys and ribs, and Cu concentrations in kidneys increased in HZn. Withdrawal of ZnO resulted in reversibility of the changes. The effect on butyric acid should be discussed critically regarding the energetic support for the enterocytes. High Zn and Cu tissue concentrations should be considered by pet food producers.

Salmonellae are among the most widespread sources of foodborne infections and Salmonella Typhimurium, in particular, is correlated with human disease caused by the consumption of contaminated pork. Intraepithelial lymphocytes (IEL) have early contact with intestinal antigens and play an important role in the detection of pathogenic bacteria. The objective of this study was to determine whether a presumed probiotic Enterococcus faecium strain could improve histomorphological and immune system-related parameters of gut function after a Salmonella challenge in weaned pigs. In particular the morphological parameters villus length and width, crypt depth and width as well as the actual enlargement of the intestinal epithelial surface were calculated and the number of IEL was evaluated in sections of the porcine gut. Weaned piglets were challenged with Salmonella enterica serovar Typhimurium DT 104, and half of them also received Enterococcus faecium NCIMB 10415 in the diet. Animals were sacrificed at days post infection (DPI) 2 and 28. The effect of the factors "time post-infection/age" and "probiotic treatment" on jejunal morphology and IEL numbers and distribution was evaluated by light microscopy. The time post-infection had significant effects in both feeding groups. Animals sacrificed at DPI 28 had longer and wider villi, deeper and wider crypts, a higher villus enlargement factor, a higher ratio between villus and crypt enlargement factors as well as more IEL. Probiotic treatment resulted in longer villi, a higher ratio of villus surface/crypt circumference enlargement factors and significantly more IEL. The larger total number of IEL displayed by the probiotic group resulted from significantly higher numbers of IEL at the nuclear and apical levels of the intraepithelial compartment but not from the number of IEL situated at the basement membrane. The probiotic effects were only measurable 28 DPI. It is proposed that Enterococcus faecium NCIMB 10415 exerts an immune modulatory effect by increasing the numbers of intraepithelial lymphocytes.

The aim of the present study was to test the effect of Enterococcus faecium NCIMB 10415 (E. faecium) on CD4+ T helper immune cell subpopulations and CD25+ cells in ileal lymphatic tissue after challenge with Salmonella (S.) Typhimurium DT 104. German Landrace piglets treated with E. faecium (n=16) as a feed additive and untreated controls (n=16) were challenged with S. Typhimurium 10 days after weaning. The expression of lineage specific T helper cell subtype master transcription factors on mRNA level was measured in the whole tissue of the gut associated lymphoid tissues (ileocecal mesenteric lymph node, ileum with Peyer's patches and papilla ilealis) and in magnetically sorted T helper cells from blood and ileocecal mesenteric lymph nodes at two and 28 days post infection. CD25 protein expression of T helper cells was studied by flow cytometry in ileal Peyer's patches, lymph nodes and blood. Distribution and morphology of CD25+ cells was demonstrated in situ by immunohistochemistry in paraffin embedded specimens of the ileum and the ileocecal mesenteric lymph nodes. The data provide evidence for a higher T helper 2 cell driven immune response in the control group compared to the E. faecium treated group (P<0.05) in CD4+ magnetically sorted lymphocytes from the ileocecal mesenteric lymph nodes at two and 28 days post infection. We did not observe differences for CD25+ cells in immunohistochemistry and flow cytometry between E. faecium fed pigs and the control group, but provided a detailed description of the occurrence and morphology of these cells in the gut associate lymphoid tissues of piglets. In conclusion we suggest that (i) prolonged feeding with E. faecium can result in changes of the T helper cell response leading to a stronger infection with S. Typhimurium and (ii) that it is important to examine purified immune cells to be able to detect effects on T helper cell subpopulations.

AIMS: This study investigated the impact of zinc oxide (ZnO) on Campylobacter coli by in vivo and in vitro assays. METHODS AND RESULTS: By in vitro growth inhibition assays, a high susceptibility of Camp. coli against ZnO could be observed. At concentrations ≥ 2.6 mmol l(-1) ZnO, a decline in cell numbers occurred. Quantitative real-time PCR assays demonstrated an up-regulation of the main oxidative stress gene (katA) in response to ZnO treatment. The expression level of katA was increased by fivefold after ZnO treatment. An experiment was carried out in pigs to elucidate the impact of ZnO as feed supplement on Camp. coli faecal excretion. Feeding a high-dosage ZnO concentration (3100 mg kg(-1) ) to piglets significantly reduced the faecal excretion of Camp. coli by up to 1 log CFU g(-1) as compared to animals receiving a low (40 mg kg(-1) ) or medium (100 mg kg(-1) ) ZnO diet. CONCLUSION: In vitro assays showed a high susceptibility of Camp. coli against ZnO. Adding high levels of ZnO to the diet of weaned piglets reduced Camp. coli excretion significantly. There is evidence for the induction of an oxidative stress response by ZnO supplementation in Camp. coli. SIGNIFICANCE AND IMPACT OF THE STUDY: Supplementation of a high-dosage ZnO diet to piglets can reduce the Camp. coli load, potentially leading to a lower contamination risk of meat during slaughter.

The study was performed to investigate and to describe features of gross and microscopic morphology of craniocervical junction (CCJ) in dogs. Seventy mature dogs (38 females, 32 males) of different body weight, representing small, medium and large breeds of dolicho-, mesati-, and brachycephalic morphotype were dissected. Morphological details were localised using an operating microscope with integrated video channel. Occurrence and distribution of fibrocartilage in the ligaments from 10 dogs was analysed histologically. Three new pairs of ligaments were described and named: dorsal ligaments of atlas, cranial internal collateral ligaments of atlas, and caudal internal collateral ligaments of atlas. Several new findings in the course of the known ligaments were found relating to breed and body weight. For the first time enthesis fibrocartilage was identified in ligaments of CCJ in dogs. Sesamoidal fibrocartilage was identified in the transversal ligament of atlas in large dogs. The findings are discussed for clinical importance.

Essential oils (EO) are being considered as possible alternatives to in-feed antibiotic growth promoters in pig nutrition. The effects of an EO mixture consisting of limonene, eugenol and pinene (10.0, 2.0, and 4.8 mg/kg diet, resp.) on gut physiology and ecology were studied in piglets. The experiment was conducted at low (commercial farm) and high hygienic conditions (experimental farm), to elucidate interactions between EO supplementation and husbandry methods. Piglets were weaned at 28 days of age, when they were offered either a control diet (C) or C with EO. Four piglets were sacrificed in each group on day 29, 30, 33 and 39. Digesta from the third distal part of the small intestine and from the colon were sampled and analysed for pH, dry matter, lactic acid, short chain fatty acids and ammonia concentrations. Enterobacteria, enterococci, lactobacilli and yeast counts were obtained by plating. Genomic DNA was extracted from digesta and polymerase chain reaction-denaturing gradient gel electrophoresis was performed. Individual microbial communities were identified at each farm. Age affected the intestinal parameters. No effects of the EO with exception for a significant reduction in colon bacterial diversity at 39 days of age could be recorded at experimental farm.

It is generally accepted that the intestinal microbiota plays an important role in sustaining health and productivity of animals. Chlorella vulgaris, a naturally occurring green microalga, is believed to influence performance and health, including bird reproduction and egg quality. The nutritive value of open or indoor cultured C. vulgaris depends upon the technological process used to treat the algal mass. In the present paper, it is presented and discussed how 2 differentially processed C. vulgaris powders (spray-dried: SD-CV; bullet-milled and spray-dried: BMSD-CV) affected crop and cecal microbiota in laying hens. Polymerase chain reaction-denaturing gradient gel electrophoresis (DGGE) of bacterial 16S rRNA gene fragments was applied. The diversity of the crop universal bacterial DGGE fingerprints was not affected (6.4 +/- 1.65, 5.4 +/- 1.19, and 5.5 +/- 1.35 in the control, SD-CV, and BMSD-CV, respectively). Most of the bands from the corresponding positions in the gels were closely related to Lactobacillus sp. The DGGE fingerprints of V2-V3 fragments of 16S rRNA of crop lactobacilli had lower diversity in the control hens (8.7 +/- 1.22) than in the SD-CV (9.2 +/- 1.77) and BMSD-CV (9.9 +/- 1.88); thus, feeding C. vulgaris resulted in increased lactobacilli diversity in crop. A band closely related to Lactobacillus ingluviei was present in 9 out of 12 hens in the control group but in only 1 bird in the SD-CV and in 5 out of 11 birds in the BMSD-CV, suggesting a negative effect of C. vulgaris on this lactobacillus. Feeding C. vulgaris to laying hens also resulted in increased bacterial community diversity in the ceca. No effect of the technological processing of the microalgae on the microbial diversity could be observed. The diversity of the ceca universal bacterial DGGE fingerprints was lower in the control group than in the SD-CV and BMSD-CV (5.6 +/- 1.72 vs. 9.16 +/- 2.64 and 9.31 +/- 2.41, respectively). Most of the sequences retrieved from the DGGE bands formed ceca that were closely related to Ruminococcaceae, Lachnospiraceae, and lactobacilli, giving further insight into still poorly discovered intestinal microbiota of laying hens.

Teaching morphology, a fundamental part of medicine curricula is traditionally based on lectures and practical trainings. We introduced peer-assisted learning (PAL) and student expert teams to the courses to give the students the possibility to improve their free speech and self-confidence. We involved students in active preparation of online materials such as labelled e-slides and e-pics. We offered online digital microscopy (Zoomify) and dissection (CyberPrep) allowing repeating the learned material and studying veterinary morphology outside the dissection theatre. Over 60% of first and third semester students profited from being a peer or being taught by a peer and 50% said the expert teams were an excellent method to learn the topographic anatomy. Almost all students applied Zoomify and CyberPrep and 75% of them found the digital microscopy and dissection to be a helpful or very helpful learning tool. In face of reduced contact hours, these forms of education compensated in part the lost teaching time. We observed improvement of rhetoric and presentation skills and self-confidence. The approaches should therefore find their constant place in the veterinary medicine curricula.

The aim of this study was to evaluate whether a single administration of two strains of Lactobacillus plantarum (DSMZ 8862 and 8866) either before or at the time point of weaning can influence the intestinal microbiota of piglets. A total of 176 piglets were allocated into five groups: control (LP0), administration of 5 x 10(9) or 5x10(10) cfu at day 25 of life (LP1, LP2) and administration of 5 x 10(9) or 5 x 10(10) cfu at day 28 of life (LP3, LP4). Piglets were weaned on day 28 of life. On day 25 (LP1, LP2), 28 (LP0, LP3, LP4), 33 (all groups) and 39 (all groups) of life, 10-13 animals of each group were killed and genomic DNA was extracted from small and large intestinal contents. Denaturing gradient gel electrophoresis demonstrated that administration of L. plantarum had a significant effect in GIT microbial communities as revealed by the Simpson's index of diversity and cluster analysis based on the Dice similarity index; this effect was more pronounced in groups LP3 and LP4. A treatment dependent presence of Clostridium glycolicum-like, Lactobacillus sobrius-like, Eubacterium rectale-like and Roseburia faecalis-like phylotypes was observed. The results show that the administration of L. plantarum at the point of weaning can influence gastrointestinal microbiota in weaning piglets which may have positive results on gastrointestinal health.

The EU ban on in-feed antibiotics has stimulated research on weaning diets as a way of reducing post-weaning gut disorders and growth check in pigs. Many bioactive components have been investigated but only few have shown to be effective. Amongst these, organic acids (OA) have been shown to exert a bactericidal action mediated by non-dissociated OA, by lowering gastric pH, increasing gut and pancreas enzyme secretion and improving gut wall morphology. It has been postulated that they may also enhance non-specific immune responses and improve disease resistance. In contrast, relatively little attention has been paid to the impact of OA on the stomach but recent data show they can differently affect gastric histology, acid secretion and gastric emptying. Butyrate and precursors of butyric acid have received special attention and although promising results have been obtained, their effects are dependent upon the dose, treatment duration, initial age of piglets, gastrointestinal site and other factors. The amino acids (AA) like glutamine, tryptophan and arginine are supportive in improving digestion, absorption and retention of nutrients by affecting tissue anabolism, stress and (or) immunity. Glutamine, cysteine and threonine are important for maintaining mucin and permeability of intestinal barrier function. Spray-dried plasma (SDP) positively affects gut morphology, inflammation and reduces acquired specific immune responses via specific and a-specific influences of immunoglobulins and other bioactive components. Effects are more pronounced in early-weaned piglets and under poorer health conditions. Little interaction between plasma protein and antibiotics has been found, suggesting distinct modes of action and additive effects. Bovine colostrum may act more or less similarly to SDP. The composition of essential oils is highly variable, depending on environmental and climatic conditions and distillation methods. These oils differ widely in their antimicrobial activity in vitro and some components of weaning diets may decrease their activity. Results in young pigs are highly variable depending upon the product and doses used. These studies suggest that relatively high concentrations of essential oils are needed for beneficial effects to be observed and it has been assumed that these plant extracts mimic most of the effects of antibiotics active on gut physiology, microbiology and immunology. Often, bioactive substances protective to the gut also stimulate feed intake and growth performance. New insights on the effects of selected OA and AA, protein sources (especially SDP, bovine colostrum) and plant extracts with anti-bacterial activities on the gut are reported in this review.
