RESUMEN

BACKGROUND: Botulinum toxin alone is unable to sufficiently reduce the muscle in cases of severe static glabellar lines due to the folded skin and dermal breakdown that frequently accompany these conditions. Augmentation of the soft tissue and removal of folded skin at the same time is the final solution. To simultaneously resolve interbrow skin laxity and replenish tissue volume, we present for the first time the method of glabellar lines excision combined with FDFG. METHODS: This retrospective study involved 23 patients with moderate-to-severe static glabellar lines underwent resection and/or free dermal fat grafting (FDFG) from June 2022 to June 2023. Fifteen of them underwent glabellar lines excision combined with FDFG, and seven were filled only. These patients were followed up at least 6 months to evaluate the effect. We utilized FACE-Q and WSRS for assessment in order to investigate the clinical results. RESULTS: There is no complication such as discoloration, hematoma, infection and palpability in all cases. After 6-15 months of follow-up, all the patients' dynamic and static lines were improved to a certain degree, and the patients were satisfied with the results with the WSRS score decreased from 3.5 ± 0.47 to 1.8 ± 0.62, and FACE-Q assessments in "Line between the eyebrows" decreased from 87 ± 7.39 to 43 ± 10.3. CONCLUSIONS: Resection in conjunction with FDFG is a brief, innovative and effective technique to correct static and dynamic severe glabellar wrinkles and maintain an acceptable outcome over an extended period of time which worthy clinical promotion. Schizochytrium sp. hasreceived much attention for itsability to synthesize and accumulate high-level docosahexaenoic acid (DHA), which can reach nearly 40 % of total fatty acids. In this study, the titer of DHA in Schizochytrium sp. was successfully improved by enhancing DHA storage through overexpressing the diacylglycerol acyltransferase (ScDGAT2C) gene, as well as optimizing the supply of precursors and cofactors required for DHA synthesis by response surface methodology. Notably, malic acid, citric acid, and biotin showed synergistic and time-dependent effects on DHA accumulation. The maximum lipid and DHA titers of the engineered Schizochytrium sp. strain reached 84.28 ± 1.02 g/L and 42.23 ± 0.69 g/L, respectively, with the optimal concentration combination (1.62 g/L malic acid + 0.37 g/L citric acid + 8.28 mg/L biotin) were added 48 h after inoculation. This study provides an effective strategy for improving lipid and DHA production in Schizochytrium sp.

BACKGROUND: Eicosapentaenoic acid (EPA) is widely used in the functional food and nutraceutical industries due to its important benefits to human health. Oleaginous microorganisms are considered a promising alternative resource for the production of EPA lipids. However, the storage of EPA in triglyceride (TG) becomes a key factor limiting its level. RESULTS: This study aimed to incorporate more EPA into TG storage through metabolic engineering. Firstly, key enzymes for TG synthesis, the diacylglycerol acyltransferase (DGAT) and glycerol-3-phosphate acyltransferase (GPAT) genes from Schizochytrium sp. HX-308 were expressed in Yarrowia lipolytica to enhance lipid and EPA accumulation. In addition, engineering the enzyme activity of DGATs through protein engineering was found to be effective in enhancing lipid synthesis by replacing the conserved motifs "HFS" in ScDGAT2A and "FFG" in ScDGAT2B with the motif "YFP". Notably, combined with lipidomic analysis, the expression of ScDGAT2C and GPAT2 enhanced the storage of EPA in TG. Finally, the accumulation of lipid and EPA was further promoted by identifying and continuing to introduce the ScACC, ScACS, ScPDC, and ScG6PD genes from Schizochytrium sp., and the lipid and EPA titer of the final engineered strain reached 2.25 ± 0.03 g/L and 266.44 ± 5.74 mg/L, respectively, which increased by 174.39% (0.82 ± 0.02 g/L) and 282.27% (69.70 ± 0.80 mg/L) compared to the initial strain, respectively. CONCLUSION: This study shows that the expression of lipid synthesis genes from Schizochytrium sp. in Y. lipolytica effectively improves the synthesis of lipids and EPA, which provided a promising target for EPA-enriched microbial oil production.

The incidence of squamous cell carcinoma (SCC) is on the rise, making it a significant global health concern. Environmental risk factors are crucial to the development of SCC. This study sought to examine comprehensively the impact of these factors on the onset of SCC. We conducted a cross-sectional study involving 480 participants at Beijing tertiary care hospital. Utilizing structured questionnaires, data on demographics, environmental exposures, medical history and clinical characteristics were collected. The cohort was composed of 272 men (56.67%) and 208 women (43.33%). The majority (44.38%) were between ages of 41 and 60, and Type III skin predominated (34.79%). Most of the participants belonged to the middle socioeconomic class (60.83%). 'Vegetarian' dietary habits (46.67%) were prevalent, as was the 'Sedentary' lifestyle (49.79%). Regarding environmental exposures, moderate sun exposure of 3 to 5 h per day (54.58%) and UV protective eyewear (30.83%) were prevalent. The majority (69.58%) of respondents indicated 'Never' exposure to carcinogens. A variety of wound characteristics were observed, with 'non-smokers' (64.17%) dominating. Most SCC lesions were located on the extremities (40.21%), lasted less than 6 months (44.38%) and measured 1-3 cm (39.79%). The majority (54.58%) did not have a history of cutaneous injuries. Our research uncovered substantial relationships between SCC and numerous environmental variables, gender, Fitzpatrick skin type, occupation, duration of sun exposure, exposure to carcinogens, dietary practices, history of skin wounds, wound location, duration, size and depth were significantly associated with the onset of SCC. These results highlighted the complexity of SCC aetiology and need for individualized prevention and treatment strategies.

RESUMEN

Due to the complexity of metabolic and regulatory networks in microorganisms, it is difficult to obtain robust phenotypes through artificial rational design and genetic perturbation. Adaptive laboratory evolution (ALE) engineering plays an important role in the construction of stable microbial cell factories by simulating the natural evolution process and rapidly obtaining strains with stable traits through screening. This review summarizes the application of ALE technology in microbial breeding, describes the commonly used methods for ALE, and highlights the important applications of ALE technology in the production of lipids and terpenoids in yeast and microalgae. Overall, ALE technology provides a powerful tool for the construction of microbial cell factories, and it has been widely used in improving the level of target product synthesis, expanding the range of substrate utilization, and enhancing the tolerance of chassis cells. In addition, in order to improve the production of target compounds, ALE also employs environmental or nutritional stress strategies corresponding to the characteristics of different terpenoids, lipids, and strains.

It is well known that polyunsaturated fatty acids (PUFAs) in Schizochytrium sp. are mainly synthesized via the polyketide synthase (PKS) pathway. However, the specific mechanism of PKS in fatty acid synthesis is still unclear. In this work, the functions of ORFA, ORFB, ORFC, and their individual functional domain genes on fatty acid synthesis were investigated through heterologous expression in Yarrowia lipolytica. The results showed that the expression of ORFA, ORFB, ORFC, and their individual functional domains all led to the increase of the very long-chain PUFA content (mainly eicosapentaenoic acid). Furthermore, the transcriptomic analysis showed that except for the 3-ketoacyl-ACP synthase (KS) domain of ORFB, the expression of an individual functional domain, including malonyl-CoA: ACP acyltransferase, 3-hydroxyacyl-ACP dehydratase (DH), 3-ketoacyl-ACP reductase, and KS domains of ORFA, acyltransferase domains of ORFB, and two DH domains of ORFC resulted in upregulation of the tricarboxylic acid cycle and pentose phosphate pathway, downregulation of the triacylglycerol biosynthesis, fatty acid synthesis pathway, and ß-oxidation in Yarrowia lipolytica. These results provide a theoretical basis for revealing the function of PKS in fatty acid synthesis in Y. lipolytica and elucidate the possible mechanism for PUFA biosynthesis.

Owing to the important physiological functions, polyunsaturated fatty acids (PUFAs) play a vital role in protecting human health, such as preventing cancer, cardiovascular disease, and diabetes. Specifically, Yarrowia lipolytica has been identified as the most popular non-conventional oleaginous yeast, which can accumulate the abundant intracellular lipids, indicating that has great potential as an industrial host for production of PUFAs. Notably, some novel engineering strategies have been applied to endow and improve the abilities of Y. lipolytica to synthesize PUFAs, including construction and optimization of PUFAs biosynthetic pathways, improvement of preucrsors acetyl-coA and NADPH supply, inhibition of competing pathways, knockout of ß-oxidation pathways, regulation of oxidative stress defense pathways, and regulation of genes involved in upstream lipid metabolism. Besides, some bypass approaches, such as strain mating, evolutionary engineering, and computational model based on omics, also have been proposed to improve the performance of engineering strains. Generally, in this review, we summarized the recent advances in engineering strategies and bypass approaches for improving PUFAs production by Y. lipolytica. In addition, we further summarized the latest efforts of CRISPR/Cas genome editing technology in Y. lipolytica, which is aimed to provide its potential applications in PUFAs production.

Eicosapentaenoic Acid (EPA) is an essential ω-3 polyunsaturated fatty acid for human health. Currently, high-quality EPA production is largely dependent on the extraction of fish oil, but this unsustainable approach cannot meet its rising market demand. Biotechnological approaches for EPA production from microorganisms have received increasing attention due to their suitability for large-scale production and independence of the seasonal or climate restrictions. This review summarizes recent research on different microorganisms capable of producing EPA, such as microalgae, bacteria, and fungi, and introduces the different EPA biosynthesis pathways. Notably, some novel engineering strategies have been applied to endow and improve the abilities of microorganisms to synthesize EPA, including the construction and optimization of the EPA biosynthesis pathway, an increase in the acetyl-CoA pool supply, the increase of NADPH and the inhibition of competing pathways. This review aims to provide an updated summary of EPA production.

OBJECTIVES: This study explored and analyzed the application value of informatization-based extended care on children discharged with leukemia. METHODS: 109 children receiving chemotherapy for acute leukemia in our hospital from January 2018 to January 2020 were selected as research subjects. The children were divided into the control group (n=53) and the observation group (n=56) on the basis of admission time point. The control-group children were given conventional nursing measures, and the observation-group children received informatization-based extended nursing care. Patients, self-care ability and their satisfaction with nursing care, and children's quality of life, anxiety and depression between the two groups were compared prior and post intervention. RESULTS: After intervention, the score in each dimension and the total scores of caring abilities in the two groups of patients increased remarkably than those before intervention (P<0.05), and the caring skills, health knowledge and the improvement of total score of nursing ability in observation group was more obvious than which in the control group (P<0.05). Besides, the score in each dimension and the total scores of the living quality in the two groups of children after intervention increased significantly than those before intervention (P<0.05), and the indexes in observation group were remarkably higher than those in control group (P<0.05). The scores of anxiety and depression in both groups after intervention were significantly lower than those before intervention (P<0.05), and the reduction degree in observation group was greater than that in control group (P<0.05). In addition, the satisfaction rate of parents in observation group was notably higher than that in control group (P<0.05). CONCLUSION: The informatization-based extended nursing care can effectively promote the nursing ability of parents on children with acute leukemia, improve the life quality of children and reduce their adverse psychological moods, which is conducive to improving the nursing satisfaction, and is worthy of clinical promotion.

Biofertilizer plays a significant role in crop cultivation that had reduced its inorganic fertilizer use. The effects of inorganic fertilizer reduction combined with Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer on the growth, quality, soil nutrients and diversity of the soil bacterial community in the rhizosphere soil of pakchoi were studied. The experiment composed of 6 treatments, including CK (no fertilization), DL (10% inorganic fertilizer reduction combined with Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer), ZL (25% inorganic fertilizer reduction combined with Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer), SL (50% inorganic fertilizer reduction combined with Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer), FHF (100% inorganic fertilizer) and JZ (100% inorganic fertilizer combined with sterilized Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer). Compared with conventional fertilization, the 25% reduction in chemical fertilizer applied with the Pennisetum giganteum mixed nitrogen-fixing biofertilizer resulted in higher plant height, plant weight, chlorophyll content, soluble protein content, soluble sugar content, vitamin C content, alkali hydrolyzed nitrogen content, available phosphorus content, available potassium content and organic matter content in pakchoi, and these variables increased by 11.81%, 8.54%, 7.37%, 16.88%, 17.05%, 23.70%, 24.24%, 36.56%, 21.09% and 19.72%, respectively. In addition, the 25% reduction in chemical fertilizer applied with the Pennisetum giganteum mixed nitrogen-fixing biofertilizer also had the lowest nitrate content, which was 53.86% lower than that with conventional fertilization. Different fertilizer treatments had a significant effect on the soil bacterial community structure. Compared with conventional fertilization, the coapplication of Pennisetum giganteum z.x.lin mixed nitrogen-fixing biofertilizer and inorganic fertilizer significantly increased the relative abundance of Proteobacteria and Actinobacteria in the soil. The results of the redundancy analysis (RDA) showed that soil organic matter, alkali-hydrolyzed nitrogen, available phosphorus, available potassium, pH and water content had a specific impact on the soil bacterial community. Among the factors, soil water content was the main factor affecting the soil bacterial community, followed by soil organic matter, soil pH, available potassium, soil available phosphorus and soil alkali-hydrolyzed nitrogen.

A robust digital holography recording design is presented to complete the work of switching between in-line and off-axis recording methods. We precisely supervise the off-axis angle in off-axis holography, so that the original reconstructed image can be separated by a minimum off-axis angle. In the design, we can also monitor and remove the negative effects of the reference tilt error in phase-shifting digital holography. Compared with the conventional digital holographic recording configuration, a supervising unit is introduced to control and to monitor the angle between the reference beam and object beam. By the Fourier analysis on the interferograms recorded by the supervising unit and using the corresponding equations, the off-axis angle, which is crucial to reset the object image in holographic reconstruction, can be calculated accurately and then chosen for the best recording angle. For in-line holography, the error affecting the slight tilt of the reference beam on the retrieved object wavefront can be eliminated completely because the tilt angle is detected by another independent device. Furthermore, by using this advanced design, the experimental arrangement can be transformed flexibly from the in-line recording state to the off-axis state or from the latter one to the former without rebuilding the experimental setup. The availability and effectiveness of this design are verified by a series of experiments.

We report on the formation and stability of induced solitons in parity-time (PT) symmetric periodic systems with the logarithmically saturable nonlinearity. Both on-site and off-site lattice solitons exist for the self-focusing nonlinearity. The most intriguing result is that the above solitons can also be realized inside the several higher-order bands of the band structure, due to the change of nonlinear type with the soliton power. Stability analysis shows that on-site solitons are linearly stably, and off-site solitons are unstable in their existence domain.

A simple and convenient method, without the need for any additional optical devices and measurements, is suggested to improve the quality of the reconstructed object wavefront in two-step phase-shifting digital holography by decreasing the errors caused by reference beam tilt, which often occurs in practice and subsequently introduces phase distortion in the reconstructed wave. The effects of reference beam tilt in two-step generalized interferometry is analyzed theoretically, showing that this tilt incurs no error either on the extracted phase shift or on the retrieved real object wave amplitude on the recording plane, but causes great deformation of the recovered object wavefront. A corresponding error detection and correction approach is proposed, and the formulas to calculate the tilt angle and correct the wavefront are deduced. A series of computer simulations to find and remove the wavefront errors caused by reference beam tilt demonstrate the effectiveness of this method.

Two families of gap and twisted surface lattice solitons in diffusive nonlinear periodic media with spatially modulated nonlinearity are reported. It is shown that the existence and stability of such solitons are extremely spatially modulated nonlinearity sensitive. For self-focusing nonlinearity, gap surface solitons belonging to the semi-infinite gap are stable in whole existence domain, twisted surface solitons are also linearly stable in low modulated strength region and a very narrow unstable region near the upper cutoff appears in high modulated strength region. In the self-defocusing case, surface gap solitons belonging to the first gap can propagate stably in whole existence domain except for an extremely narrow region close to the Bloch band, twisted solitons belonging to this gap are unstable in the entire existence domain.

A stable diode pumped Kerr-lens mode-locked (KLM) Yb:LuYSiO5 (Yb:LYSO) laser of generating 61 fs pulses at a central wavelength of 1055.4 nm is experimentally demonstrated. This is, to the best of our knowledge, the first demonstration of femtosecond KLM operation in Yb:LYSO laser, and it is believed that 61 fs is the shortest pulse duration ever produced from an Yb-doped orthosilicate laser. The average output power of the mode-locked laser is 40 mW and the repetition rate is 113 MHz.

A low-threshold self-starting Kerr-lens mode-locked Ti:sapphire laser is demonstrated that uses a tight-focusing cavity design in conjunction with a semiconductor saturable-absorber mirror (SESAM). With 3% and 12% output couplers, we achieve mode-locking thresholds as low as 390 and 600 mW, respectively. Stable femtosecond laser pulses with average power of 114 mW are generated at a pump power of 1.2 W, which corresponds to a typical duration of 17 fs and bandwidth of 47 nm. Mode-locking operation is achieved in a pump power range of 600 mW to 4.8 W at an output coupling of 12%; the advantages of using a SESAM for low-power mode-locking operation are demonstrated.
