RESUMEN

Chronic kidney disease (CKD) has become a global public health problem with high morbidity and mortality. Renal fibrosis can lead to end-stage renal disease (ESRD). However, there is still no effective treatment to prevent or delay the progression of CKD into ESRD. Therefore, exploring the pathogenesis of CKD is essential for preventing and treating CKD. There are a variety of trace elements in the human body that interact with each other within a complex regulatory network. Iron and copper are both vital trace elements in the body. They are critical for maintaining bodily functions, and the dysregulation of their metabolism can cause many diseases, including kidney disease. Ferroptosis is a new form of cell death characterized by iron accumulation and lipid peroxidation. Studies have shown that ferroptosis is closely related to kidney disease. However, the role of abnormal copper metabolism in kidney disease and its relationship with ferroptosis remains unclear. Here, our current knowledge regarding copper metabolism, its regulatory mechanism, and the role of abnormal copper metabolism in kidney diseases is summarized. In addition, we discuss the relationship between abnormal copper metabolism and ferroptosis to explore the possible pathogenesis and provide a potential therapeutic target for CKD.

RESUMEN

Purpose To analyze the expression of FOXA3 in colorectal cancer(CRC)and its correlation with clinicopatho-logical features. Methods FOXA3 mRNA expression in 31 CRC cancer tissues and their matched normal tissues was detec-ted by real-time quantitative PCR(RT-qPCR).The protein ex-pression of FOXA3 in 120 CRC cancer tissues was detected by immunohistochemical EnVision two-step method, and the clini-copathologic features such as lymph node metastasis and immu-nohistochemical expression were analyzed. Results The mRNA expression level of FOXA3 in colorectal cancer tissues was sig-nificantly higher than that in paired paracancer tissues(t=2.952,P=0.006 1).FOXA3 protein expression level in color-ectal cancer tissues was not significantly correlated with gender, age, site and size of patients, but significantly correlated with the degree of tissue differentiation(P=0.006)and lymph node metastasis(P=0.002).The degree of differentiation was nega-tively correlated with FOXA3 expression, while lymph node me-tastasis was positively correlated with FOXA3 expression.Sur-vival analysis showed that higher FOXA3 expression was associ-ated with worse overall survival(P<0.000 1),and FOXA3 was an independent risk factor for prognosis in patients with colorectal cancer. Conclusion This study suggests that FOXA3 may play a promoting role in the occurrence and development of colorectal cancer, and FOXA3 may be a molecular marker for the diagnosis, metastasis and prognosis of colorectal cancer.

RESUMEN

This paper reports the maternal and fetal outcomes of three twin pregnancies with chronic hypertension and obstructive sleep apnea-hypopnea syndrome (OSAHS) who were treated with continuous positive airway pressure (CPAP). All three women with twin pregnancies were diagnosed with chronic hypertension. Furthermore, symptoms such as snoring and apnea assisted the diagnosis of OSAHS through polysomnography monitoring. Case 1 was treated with CPAP at 28 gestational weeks. The blood pressure increased gradually after the first month of CPAP treatment, with an elevated urine protein concentration. At 34 gestational weeks, the pregnant woman underwent a cesarean section due to the development of hemolysis, elevated liver enzymes, and low platelet syndrome. Case 2 was treated with CPAP at 11 gestational weeks, with stable blood pressure throughout the pregnancy, and was delivered through cesarean section at 37 weeks of pregnancy. Case 3 started CPAP at 13 gestational weeks for four months, and increased blood pressure and urine protein were observed. Medication brought the blood pressure down, and urine protein became negative. At 32 gestational weeks, a cesarean section was performed because of premature rupture of the membrane. Her CPAP treatment continued till delivery with good maternal and infant outcomes. The treatment outcomes of the three cases suggest that CPAP may prolong the time of blood pressure rise among twin pregnancies where chronic hypertension and OSAHS coexist, which potentially reduces the occurrence of adverse maternal and infant outcomes.

RESUMEN

Objective Explore differences in the cognitive abilities of socially different Labradors. Methods The dog mentality assessment(DMA)test created by the Swedish Working Dog Association was modified to employ 12 behavioral variables from five subtests of the DMA test, social contact, play Ⅰ,distance-play,ghosts and play Ⅱ,to assess sociability of the dogs.In accordance with the scoring criteria,49 labradors provided by the China Guide Dog Training Centre in Dalian were scored on the social behavioral variables and classified into high(n=15)and low(n=34)sociability groups by cluster analysis.A new system to test canine cognitive ability was developed using the dog cognitive development battery, which tests various domains of cognitive ability such as social cue use, unsolvable task, inhibitory control, cognitive flexibility, working memory, and multistep problem solving task. The dogs'behavioral performance and duration of the test were also recorded.Statistical analysis was performed to determine assess differences in the cognitive abilities of socially diverse dogs. Results Dogs in the high and low social subgroups differed significantly in behavioral variables of the unsolvable task, inhibitory control test, and multistep problem solving task.In the unsolvable task, dogs in the high social group looked at people for significantly longer than dogs in the low social grouping(P=0.008)and looked at people with significantly less latency time than dogs in the low social group(P=0.0001).In the inhibitory control, dogs in the high social group chose significantly more correctly than dogs in the low social group(P=0.034)and chose for significantly less time than dogs in the low social group(P=0.039).In the multistep problem solving task, for dogs in the high social group.successfully completed number of stakes was significantly higher than for dogs in the low social group(P=0.044).The percentage of operation pale time was significantly lower than for dogs in the low social group(P=0.05).The average latency time to solve the bone task was significantly higher than for dogs in the low social group(P=0.037).Moreover,the percentage of operation bone time was significantly lower than for dogs in the low social group(P=0.038).In tests involving a manipulable apparatus, dogs in the high social group spent more time looking at people than dogs in the low social group and less time manipulating the apparatus than dogs in the low subgroup, but no statistically significant differences were observed(P>0.05).Conclusions Highly sociable labradors have a greater cognitive ability, they are more able to suppress impulses during tests, more able to complete the multistep problem solving task, and more inclined to change strategies to seek new cues from people rather than obsessing over manipulating the apparatus when they are unable to solve a problem.

RESUMEN

Objective To explore the regulatory effect of miR-26b-5p on PI3K/PIP3 signaling path-way and its effect on L-type calcium channel of atrial cardiomyocytes in rats with ischemic ar-rhythmia.Methods A total of 72 SD rats were randomly divided into sham operation group,mod-el group,LY294002 group, negative control group, overexpression group and combination group, with 12 rats in each group. In 24 h after corresponding intervention, rat model of ischemic arrhyth-mia was established in the other groups except the sham operation group(only thoracotomy but without ligation).During the modeling process, arrhythmia indexes were detected in each group, and Curtis-Walker scoring was used for arrhythmia score.Fluorescence quantitative PCR was em-ployed to detect the expression of miR-26b-5p in atrial myocardiocytes,whole-cell patch-clamp technique was used to record the ICa-L of atrial myocardiocytes, and Western blotting was applied to measure the expression of CACNA1C,calmodulin and PI3K/PIP3 pathway related proteins.Re-sults Compared with the model group, the number of left ventricular preventricular contrac-tions,duration of ventricular tachycardia, duration of ventricular fibrillation, arrhythmia score,ab-solute value of ICa-L peak density of atrial myocardiocytes, and expression levels of CACNA1C and calmodulin were significantly increased, and the levels of miR-26b-5p,p-PI3K,PIP3 and p-Akt were obviously decreased in the LY294002 group(P<0.05).While the overexpression group had decreased number of left ventricular preventricular contractions, shorter duration of ventricular tachycardia, shorter duration of ventricular fibrillation, lower arrhythmia score, reduced absolute value of ICa-L peak density, and lower expression levels of CACNA1C and calmodulin, and in-creased levels of miR-26b-5p,p-PI3K,PIP3 and p-Akt than the model group(P<0.05).The number of left ventricular preventricular contractions, duration of ventricular tachycardia,dura-tion of ventricular fibrillation, arrhythmia score, absolute value of ICa-L peak density and expres-sion levels of CACNA1C and calmodulin were significantly increased, and the expression of p-PI3K,PIP3 and p-Akt were obviously decreased(0.82±0.08 vs 1.09±0.11,0.91±0.09 vs 1.17± 0.11,0.94±0.09 vs 1.20±0.12,P<0.05)in the combination group than the overexpression group. Conclusion Overexpression of miR-26b-5p may block the L-type calcium channel in atrial myocardiocytes in ischemic arrhythmia rats by activating PI3K/PIP3 related signaling pathway, and thus improve the performance of arrhythmia in rats.

RESUMEN

Objective To investigate the therapeutic effect of targeting and killing CD248-positive myofibroblasts on bleomycin-induced pulmonary fibrosis in mice. Methods IgG78-DM1, an antibody-maytansine 1 (DM1) conjugate targeting CD248, was prepared. The drug conjugation efficiency was measured and calculated by UV spectrophotometer, and the identification of IgG78-DM1 was performed through SDS-PAGE and Western blot analysis. In vitro, the binding activity of IgG78-DM1 on CD248-positive myofibroblasts was detected by flow cytometry and the cytotoxicity of IgG78-DM1 to CD248-positive myofibroblasts was evaluated by CCK-8 assay. In vivo, C57BL/6 male mice were randomly divided into control group, idiopathic pulmonary fibrosis group, human IgG-DM1 (hIgG-DM1) control group, and IgG78-DM1 treatment group. Then, the mouse models with pulmonary fibrosis induced by bleomycin were constructed. Two weeks later, the animal models were intravenously injected with IgG78-DM1. After the treatment of two weeks, lung tissues were collected for Masson staining and Sirius Red staining to evaluate the degree of pulmonary fibrosis. Real-time fluorescence quantitative PCR was used to measure the expression levels of CD248, as well as markers of fibroblastic activation including alpha-smooth muscle actin (α-SMA) and type I collagen alpha 1 (COL1A1). The safety of IgG78-DM1 was preliminarily assessed by conducting liver and kidney function tests. Results IgG78-DM1 was successfully prepared, and its drug conjugation ratio was 3.2. The antibody structure remained stable after conjugation, allowing effective binding and cytotoxicity against CD248-positive myofibroblasts. After treatment with IgG78-DM1, the degree of pulmonary fibrosis in mice significantly reduced, accompanied by the decrease of the expression of CD248, α-SMA, and COL1A1. The liver and kidney function of the mice remained at normal levels compared to the normal control group. Conclusion IgG78-DM1 effectively inhibits pulmonary fibrosis in mice by targeting and killing CD248-positive myofibroblasts. The safety of this strategy is preliminarily assessed.

RESUMEN

OBJECTIVE To mine and analyze the risk signal of glucagon- like peptide -1 receptor agonists (GLP-1RA)related adverse drug reaction (ADR). METHODS ADR data related to GLP- 1RA from April 1,2005 to October 16,2021 in the openFDA database were collected ,and the Bayesian confidence propagation neoral network (BCPNN)method was used for data mining. ADR were classified and described by using systematic organ classification (SOC)of drug ADR terminology set in 24.0 edition of MedDRA. RESULTS & CONCLUSIONS A total of 175 719 ADR reports related to GLP- 1RA were retrieved ,with 140 ADR positive signals ,involving five drugs such as exenatide (77 027 cases of ADR and 31 ADR positive signals ),dulaglutide (45 329 cases of ADR and 26 ADR positive signals ),liraglutide (42 748 cases of ADR and 32 ADR positive signals ), semaglutide(8 844 cases of ADR and 27 ADR positive signals )and lixisenatide (1 771 cases of ADR and 24 ADR positive signals). According to SOC classification ,GLP-1RA-induced ADR involved gastrointestinal system ,hepatobiliary system ,nervous system, urinary and renal system ,endocrine system ,immune system and administration site. In the gastrointestinal system ,the risk of (acute)pancreatitis was higher ,and the order of risk was liraglutide >exenatide>semaglutide>dulaglutide>lixisenatide; ADR signal of hepatobiliary system was stronger for cholelithiasis ,and the order of risk was liraglutide >semaglutide>exenatide> lixisenatide>dulaglutide. In the nervous system ,the risk of taste disorder was higher ;compared with dulaglutide and lixisenatide , liraglutide, exenatide and semaglutide were more likely to cause headache and dizziness. In urinary and renal system , compared with exena tide, dulaglutide and lixisenatide ,liraglutide and semaglutide were more likely to cause acute renal injury. In the endocrine system ,the risk of hypoglycemia was higher ,and the order of risk was exenatide >liraglutide>lixisenatide> semaglutide>dulaglutide. In the immune system ,lixisenatide was more likely to develop urticaria than other drugs ,dulaglutide and liraglutide did not caused ADR signal. Among the administration sites ,the risk of ADR caused by exenatide and dulaglutide was higher, while the risk of related ADR caused by semaglutide was lower. When using GLP- 1RA clinically ,we should closely monitor the renal function and blood glucose of patients ,and pay attention to patients with sudden upper abdominal pain ;in case of relevant ADR ,timely intervention measures should be taken to ensure the safety and effectiveness of medication.

RESUMEN

Extracellular vesicles (EV) are lipid bilayer vesicles with a diameter of 30-1000 nm secreted by cells or microorganisms, which are abundant in proteins, nucleic acids, lipids and other biological information molecules. Therefore, EV can be used as a carrier, transferring materials between cells. At present, in infectious diseases, EV derived from pathogenic microorganisms could be considered as a double-edged sword, which means it can not only play a negative role in the host′s infection immunity, bacteria′s pathogenicity and antibiotic resistance transmission, but also manifest the advantages in clinical diagnosis and treatment.

RESUMEN

The inclusive wealth approach is increasingly common to measure the sustainable development of the countries. It comprised the natural, human and produced capital of nations to measure social wellbeing. We measure the inclusive wealth of the provinces in China from 2000 to 2015 and reports the sustainable use of the resources. We identify that three types of capital have increased to varying degrees, with produced capital increasing by 615.6%, natural capital increasing by 33.8%, and human capital increased by 337.0%. The total amount of inclusive wealth has increased by 300.4% in the past 15 years. However, the provinces in China are still facing unbalanced development across the country compared to developed nations. The use of the natural capital, more specifically now-renewable resources, has been restricting the wealth growth in some provinces. Although ecological services account for a small proportion of the total inclusive wealth, more attention is essential for sustainable development. Meanwhile, the rapid growth of carbon damages posed threat to future wealth accumulation. Innovative, coordinated, green, open and shared development are the goals of China 13th and 14th five-year plan and our inclusive wealth of China will be key measurement tool of this achievement.

RESUMEN

Objective:To compare the curative effect of hematoma removal and minimally invasive puncture drainage in hypertensive intracerebral hemorrhage (HICH) under neuroendoscope. Methods:The clinical data of 118 patients with HICH admitted to our hospital from February 2017 to November 2019 were retrospectively analyzed. According to the different surgical methods, the patients were divided into group A ( n=59, minimally invasive puncture and drainage) and group B ( n=59, neuroendoscopic hematoma removal). The perioperative indexes, complications and neurological function indexes [neuron specific enolase (NSE), neurotrophic factor (NGF), substance P (SP)], inflammatory factor index [interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α)] and related scale scores [National Institutes of Health Stroke Scale (NIHSS), modified Barthel Index (mBI)] were compared between the two groups. Results:The operation time, cerebrospinal fluid recovery time, hematoma removal time and hospitalization time of group B were shorter than those of group A ( P<0.05). On the 7th day after operation, the NSE, SP, IL-6, TNF-α in the two groups were decreased, and the levels of NSE, SP, IL-6 and TNF-α in group B were lower than those in group A ( P<0.05); on the 7th day after operation, the levels of NGF in two groups were increased, and those in group B were higher than those in group A ( P<0.05). 6 months after operation, the NIHSS decreased and mBI increased in both groups ( P<0.05), and NIHSS in group B was lower than that in group A, but mBI was higher than that in group A ( P<0.05). There was no difference in the incidence of complications between the two groups ( P>0.05). Conclusions:Compared with minimally invasive puncture and drainage, neuroendoscope hematoma removal for HICH patients can effectively improve perioperative indicators, reduce neurological damage and body inflammatory stimulation, and improve the self-care ability of patients, which is safe and reliable.

RESUMEN

The occurrence and development of malignant tumors is closely related to the tumor microenvironment. Tumor-associated neutrophils (TAN) are important inflammatory cells in the tumor microenvironment. Studies have confirmed that TAN plays an important role in tumor growth, invasion, metastasis, pathological angiogenesis, and immunity escapes. TAN can be activated as an anti-tumor N1 type and a tumor-promoting N2 type. This article reviews the biological characteristics of TAN and the role of TAN in the malignant tumors and its mechanism.

RESUMEN

BACKGROUND: Anterior cervical decompression and fusion has been widely used in the treatment of multi-level cervical spondylotic myelopathy, but accompanied with lots of complications.OBJECTIVE: To investigate the efficacy of zero-profile interboby fixation system for multi-level cervical spondylotic myelopathy.METHODS: Seventy-one patients with multi-level cervical spondylotic myelopathy were randomly divided into two groups, and the patients in group A accepted zero-profile interboby fixation system, and group B accepted cage interboby fixation system. The Japanese Orthopaedic Association score, fusion rate, as well as the incidence of dysphagia and esophageal fistula were detected to compare the efficacy between two groups.RESULTS AND CONCLUSION: (1) All cases were followed-up for 3-34 months, average of 17.5 months. The excellent and good rate at the last follow-up showed no significant difference between two groups. (2) The final fusion rate did not differ significantly between two groups, but the fusion rate in the group A was significantly higher than that in the group B at 6 and 9 months postoperatively (P < 0.05). (3) There was one patient with mild dysphagia in the group A (3%), three mild, five medium, and two severe dysphagia in the group B (29%), which showed significant difference between two groups (P < 0.05). No internal fixation loosening occurred in the group A, but three cases in the group B. The blood loss, operation time and radiology times in the group A were significantly lower than those in the group B (P < 0.05). (4) These results suggest that the effect of these two surgical methods in promoting functional recovery of spinal cord and final fusion rate show no significant differences; however, the zero-profile interboby fixation system exhibits better postoperative stability and interim fusion rate, with lower incidence of dysphagia.

RESUMEN

Objective To explore the changes in inflammatory reactions in tail-suspension mice infected by Klebsiella pneumoniae from spaceflight. Methods Tail suspension was used to simulate the physiological effects of microgravity.C57BL/6 mice were randomly divided into control (Con),control+K.pneumoniae T16-169 (Con+T16-169),tail suspension (TS) and tail suspension+K.pneumoniae T16-169 (TS+T16-169) groups. The level of inflammatory cytokines TNF-α,IL-6 and IL-1β mRNA in lung tissue and the plasma cytokine concentration were detected by RT-qPCR and xMAP technology, and HE staining was used to represent the morphological changes in lung tissue. Results Compared with the control group, the expression of inflammatory cytokines in lung tissue and plasma concentrations of all experimental groups were increased, and the difference in TS+T16-169 group was the most significant (P<0.01 or P<0.001).HE staining showed that the lung tissues in Con+T16-169 and TS+T16-169 groups were damaged in different degrees, and the damage of TS+T16-169 group was the most serious. Conclusion The K.pneumoniae from spaceflight significantly increases the expression of inflammatory cytokines in lung tissue and plasma concentrations after infecting tail-suspension mice, and induces more serious damages to the lung tissue, which suggests that inflammatory reactions can be increased in tail suspension mice infected by K.pneumoniae from spaceflight.

Objective Invasive candidiasis is associated with a significant mortality clinically. The purpose of this study was to observe the T cells immune response to fructose bisphosphate aldolase (Fba), an immunodominant antigen of Candida albicans, and determine whether the antigen has the possibility of priming cellular immune protection in invasive candiasis. Methods Using ELISPOT assay, we determined the frequencies of positive spot-forming cells (SFCs) of Fba antigen-specific T cells secreting IFN-γ, IL-4 and IL-17A in the peripheral blood mononuclear cells (PBMCs) of 26 healthy individuals. Results After Fba stimulation, the frequencies of positive SFCs of IFN-γ, IL-4 and IL-17A in the 26 healthy subjects were 23 (9.75, 42.50), 0 (0, 0.25) and 1.5 (0.75, 8.25), respectively, with statistically significant differences among the three (P<0.01).The response rates of IFN-γ (100% [26/26]) and IL-17A (76.92% [20/26]) were significantly higher than that of IL-4 (15.38% [4/26]) (P<0.01).Fba-induced strong response (SCFs ≥20) for IFN-γ was observed in 57.69% (15/26) of the healthy individuals, that for IL-17A in only 1, while that for IL-4 in none.Responses of both Th1 and Th17 cells to Fba were found in 65.38% (17/26) of the subjects, that of Th1 cells in 19.23% (5/26), but that of Th2 cells in none. Conclusion Fba of Candida albicans can induce immunodominant responses of Th1 and Th17 cells and is a potential vaccine against invasive candiasis.

RESUMEN

Objective To analyze the expression and clinical significance of tumor suppressor phosphatase -tension protein gene(PTEN)in differentiated thyroid carcinoma and the relationship with the V600E gene site muta-tion of murine sarcoma viral oncogene homolog B1(BRAF).Methods The expression of PTEN was assayed by im-munohistochemical staining in differentiated thyroid carcinoma and adjacent tissues. The differences were compared between clinical pathological features. The BRAF V600E gene mutation was detected by fluorescence quantitative pol-ymerase chain reaction(PCR)and to investigate the correlation between the expression of PTEN and BRAF V600E mutation. Results The negative rate of PTEN was 78.75%(63 /80)in differentiated thyroid carcinoma,13.75%(11 /80)in adjacent tissues(χ2 =27.236,P =0.000).There were significant differences in the expression of PTEN between I -II and III -IV in TNMstage, grade I and II,and lymph node metastasis(χ2 =10.395,6.948,9.263,P =0.000,0.006,0.000).The differences were not significant in the expression of PTEN between different gender,≤45 years and >45 years different pathological types and tumor diameter <2cm and ≥2cm(χ2 =1.113,0.941, 2.301,1.567,P =0.185,0.213,0.087,0.181).There was correlation between the expression of PTEN and BRAF V600E mutations(r =0.301,P =0.004).Conclusion The expression of PTEN is involved in the occurrence and development of differentiated thyroid cancer and is associated with BRAF V600E.The PTEN has the predictive value of prognosis in tumors.
