RESUMEN

Chronic kidney disease (CKD) has become a global public health problem with high morbidity and mortality. Renal fibrosis can lead to end-stage renal disease (ESRD). However, there is still no effective treatment to prevent or delay the progression of CKD into ESRD. Therefore, exploring the pathogenesis of CKD is essential for preventing and treating CKD. There are a variety of trace elements in the human body that interact with each other within a complex regulatory network. Iron and copper are both vital trace elements in the body. They are critical for maintaining bodily functions, and the dysregulation of their metabolism can cause many diseases, including kidney disease. Ferroptosis is a new form of cell death characterized by iron accumulation and lipid peroxidation. Studies have shown that ferroptosis is closely related to kidney disease. However, the role of abnormal copper metabolism in kidney disease and its relationship with ferroptosis remains unclear. Here, our current knowledge regarding copper metabolism, its regulatory mechanism, and the role of abnormal copper metabolism in kidney diseases is summarized. In addition, we discuss the relationship between abnormal copper metabolism and ferroptosis to explore the possible pathogenesis and provide a potential therapeutic target for CKD.

RESUMEN

Objective To investigate the therapeutic effect of targeting and killing CD248-positive myofibroblasts on bleomycin-induced pulmonary fibrosis in mice. Methods IgG78-DM1, an antibody-maytansine 1 (DM1) conjugate targeting CD248, was prepared. The drug conjugation efficiency was measured and calculated by UV spectrophotometer, and the identification of IgG78-DM1 was performed through SDS-PAGE and Western blot analysis. In vitro, the binding activity of IgG78-DM1 on CD248-positive myofibroblasts was detected by flow cytometry and the cytotoxicity of IgG78-DM1 to CD248-positive myofibroblasts was evaluated by CCK-8 assay. In vivo, C57BL/6 male mice were randomly divided into control group, idiopathic pulmonary fibrosis group, human IgG-DM1 (hIgG-DM1) control group, and IgG78-DM1 treatment group. Then, the mouse models with pulmonary fibrosis induced by bleomycin were constructed. Two weeks later, the animal models were intravenously injected with IgG78-DM1. After the treatment of two weeks, lung tissues were collected for Masson staining and Sirius Red staining to evaluate the degree of pulmonary fibrosis. Real-time fluorescence quantitative PCR was used to measure the expression levels of CD248, as well as markers of fibroblastic activation including alpha-smooth muscle actin (α-SMA) and type I collagen alpha 1 (COL1A1). The safety of IgG78-DM1 was preliminarily assessed by conducting liver and kidney function tests. Results IgG78-DM1 was successfully prepared, and its drug conjugation ratio was 3.2. The antibody structure remained stable after conjugation, allowing effective binding and cytotoxicity against CD248-positive myofibroblasts. After treatment with IgG78-DM1, the degree of pulmonary fibrosis in mice significantly reduced, accompanied by the decrease of the expression of CD248, α-SMA, and COL1A1. The liver and kidney function of the mice remained at normal levels compared to the normal control group. Conclusion IgG78-DM1 effectively inhibits pulmonary fibrosis in mice by targeting and killing CD248-positive myofibroblasts. The safety of this strategy is preliminarily assessed.

RESUMEN

OBJECTIVE To mine and analyze the risk signal of glucagon- like peptide -1 receptor agonists (GLP-1RA)related adverse drug reaction (ADR). METHODS ADR data related to GLP- 1RA from April 1,2005 to October 16,2021 in the openFDA database were collected ,and the Bayesian confidence propagation neoral network (BCPNN)method was used for data mining. ADR were classified and described by using systematic organ classification (SOC)of drug ADR terminology set in 24.0 edition of MedDRA. RESULTS & CONCLUSIONS A total of 175 719 ADR reports related to GLP- 1RA were retrieved ,with 140 ADR positive signals ,involving five drugs such as exenatide (77 027 cases of ADR and 31 ADR positive signals ),dulaglutide (45 329 cases of ADR and 26 ADR positive signals ),liraglutide (42 748 cases of ADR and 32 ADR positive signals ), semaglutide(8 844 cases of ADR and 27 ADR positive signals )and lixisenatide (1 771 cases of ADR and 24 ADR positive signals). According to SOC classification ,GLP-1RA-induced ADR involved gastrointestinal system ,hepatobiliary system ,nervous system, urinary and renal system ,endocrine system ,immune system and administration site. In the gastrointestinal system ,the risk of (acute)pancreatitis was higher ,and the order of risk was liraglutide >exenatide>semaglutide>dulaglutide>lixisenatide; ADR signal of hepatobiliary system was stronger for cholelithiasis ,and the order of risk was liraglutide >semaglutide>exenatide> lixisenatide>dulaglutide. In the nervous system ,the risk of taste disorder was higher ;compared with dulaglutide and lixisenatide , liraglutide, exenatide and semaglutide were more likely to cause headache and dizziness. In urinary and renal system , compared with exena tide, dulaglutide and lixisenatide ,liraglutide and semaglutide were more likely to cause acute renal injury. In the endocrine system ,the risk of hypoglycemia was higher ,and the order of risk was exenatide >liraglutide>lixisenatide> semaglutide>dulaglutide. In the immune system ,lixisenatide was more likely to develop urticaria than other drugs ,dulaglutide and liraglutide did not caused ADR signal. Among the administration sites ,the risk of ADR caused by exenatide and dulaglutide was higher, while the risk of related ADR caused by semaglutide was lower. When using GLP- 1RA clinically ,we should closely monitor the renal function and blood glucose of patients ,and pay attention to patients with sudden upper abdominal pain ;in case of relevant ADR ,timely intervention measures should be taken to ensure the safety and effectiveness of medication.

RESUMEN

Extracellular vesicles (EV) are lipid bilayer vesicles with a diameter of 30-1000 nm secreted by cells or microorganisms, which are abundant in proteins, nucleic acids, lipids and other biological information molecules. Therefore, EV can be used as a carrier, transferring materials between cells. At present, in infectious diseases, EV derived from pathogenic microorganisms could be considered as a double-edged sword, which means it can not only play a negative role in the host′s infection immunity, bacteria′s pathogenicity and antibiotic resistance transmission, but also manifest the advantages in clinical diagnosis and treatment.

RESUMEN

Jiaji points have been widely used in the treatment of cerebral palsy, spinal cord injury, stroke, cervical spondylosis, lumbar disc herniation, ankylosing spondylitis, herpes zoster and other diseases, and shown good curative effect. The treatment methods for Jiaji points are rich, including acupuncture, electroacupuncture, moxibustion, percussion, fire needle, acupoint injection, warm acupuncture, and bloodletting. And acupuncture and electroacupuncture are the most application treatment methods. But the treatment shows narrow, scope of diseases, like internal medicine, pediatrics, and gynecology. Such treatment of Jiaji points are seldomly combined with other acupoints. Though Jiaji point treatment has good curative effect for insomnia, depression, uterine fibroids and other diseases, clinical application is few.

RESUMEN

The inclusive wealth approach is increasingly common to measure the sustainable development of the countries. It comprised the natural, human and produced capital of nations to measure social wellbeing. We measure the inclusive wealth of the provinces in China from 2000 to 2015 and reports the sustainable use of the resources. We identify that three types of capital have increased to varying degrees, with produced capital increasing by 615.6%, natural capital increasing by 33.8%, and human capital increased by 337.0%. The total amount of inclusive wealth has increased by 300.4% in the past 15 years. However, the provinces in China are still facing unbalanced development across the country compared to developed nations. The use of the natural capital, more specifically now-renewable resources, has been restricting the wealth growth in some provinces. Although ecological services account for a small proportion of the total inclusive wealth, more attention is essential for sustainable development. Meanwhile, the rapid growth of carbon damages posed threat to future wealth accumulation. Innovative, coordinated, green, open and shared development are the goals of China 13th and 14th five-year plan and our inclusive wealth of China will be key measurement tool of this achievement.

RESUMEN

Objective:To compare the curative effect of hematoma removal and minimally invasive puncture drainage in hypertensive intracerebral hemorrhage (HICH) under neuroendoscope. Methods:The clinical data of 118 patients with HICH admitted to our hospital from February 2017 to November 2019 were retrospectively analyzed. According to the different surgical methods, the patients were divided into group A ( n=59, minimally invasive puncture and drainage) and group B ( n=59, neuroendoscopic hematoma removal). The perioperative indexes, complications and neurological function indexes [neuron specific enolase (NSE), neurotrophic factor (NGF), substance P (SP)], inflammatory factor index [interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α)] and related scale scores [National Institutes of Health Stroke Scale (NIHSS), modified Barthel Index (mBI)] were compared between the two groups. Results:The operation time, cerebrospinal fluid recovery time, hematoma removal time and hospitalization time of group B were shorter than those of group A ( P<0.05). On the 7th day after operation, the NSE, SP, IL-6, TNF-α in the two groups were decreased, and the levels of NSE, SP, IL-6 and TNF-α in group B were lower than those in group A ( P<0.05); on the 7th day after operation, the levels of NGF in two groups were increased, and those in group B were higher than those in group A ( P<0.05). 6 months after operation, the NIHSS decreased and mBI increased in both groups ( P<0.05), and NIHSS in group B was lower than that in group A, but mBI was higher than that in group A ( P<0.05). There was no difference in the incidence of complications between the two groups ( P>0.05). Conclusions:Compared with minimally invasive puncture and drainage, neuroendoscope hematoma removal for HICH patients can effectively improve perioperative indicators, reduce neurological damage and body inflammatory stimulation, and improve the self-care ability of patients, which is safe and reliable.

RESUMEN

The occurrence and development of malignant tumors is closely related to the tumor microenvironment. Tumor-associated neutrophils (TAN) are important inflammatory cells in the tumor microenvironment. Studies have confirmed that TAN plays an important role in tumor growth, invasion, metastasis, pathological angiogenesis, and immunity escapes. TAN can be activated as an anti-tumor N1 type and a tumor-promoting N2 type. This article reviews the biological characteristics of TAN and the role of TAN in the malignant tumors and its mechanism.

RESUMEN

Objective To explore the changes in inflammatory reactions in tail-suspension mice infected by Klebsiella pneumoniae from spaceflight. Methods Tail suspension was used to simulate the physiological effects of microgravity.C57BL/6 mice were randomly divided into control (Con),control+K.pneumoniae T16-169 (Con+T16-169),tail suspension (TS) and tail suspension+K.pneumoniae T16-169 (TS+T16-169) groups. The level of inflammatory cytokines TNF-α,IL-6 and IL-1β mRNA in lung tissue and the plasma cytokine concentration were detected by RT-qPCR and xMAP technology, and HE staining was used to represent the morphological changes in lung tissue. Results Compared with the control group, the expression of inflammatory cytokines in lung tissue and plasma concentrations of all experimental groups were increased, and the difference in TS+T16-169 group was the most significant (P<0.01 or P<0.001).HE staining showed that the lung tissues in Con+T16-169 and TS+T16-169 groups were damaged in different degrees, and the damage of TS+T16-169 group was the most serious. Conclusion The K.pneumoniae from spaceflight significantly increases the expression of inflammatory cytokines in lung tissue and plasma concentrations after infecting tail-suspension mice, and induces more serious damages to the lung tissue, which suggests that inflammatory reactions can be increased in tail suspension mice infected by K.pneumoniae from spaceflight.

RESUMEN

BACKGROUND: Anterior cervical decompression and fusion has been widely used in the treatment of multi-level cervical spondylotic myelopathy, but accompanied with lots of complications.OBJECTIVE: To investigate the efficacy of zero-profile interboby fixation system for multi-level cervical spondylotic myelopathy.METHODS: Seventy-one patients with multi-level cervical spondylotic myelopathy were randomly divided into two groups, and the patients in group A accepted zero-profile interboby fixation system, and group B accepted cage interboby fixation system. The Japanese Orthopaedic Association score, fusion rate, as well as the incidence of dysphagia and esophageal fistula were detected to compare the efficacy between two groups.RESULTS AND CONCLUSION: (1) All cases were followed-up for 3-34 months, average of 17.5 months. The excellent and good rate at the last follow-up showed no significant difference between two groups. (2) The final fusion rate did not differ significantly between two groups, but the fusion rate in the group A was significantly higher than that in the group B at 6 and 9 months postoperatively (P < 0.05). (3) There was one patient with mild dysphagia in the group A (3%), three mild, five medium, and two severe dysphagia in the group B (29%), which showed significant difference between two groups (P < 0.05). No internal fixation loosening occurred in the group A, but three cases in the group B. The blood loss, operation time and radiology times in the group A were significantly lower than those in the group B (P < 0.05). (4) These results suggest that the effect of these two surgical methods in promoting functional recovery of spinal cord and final fusion rate show no significant differences; however, the zero-profile interboby fixation system exhibits better postoperative stability and interim fusion rate, with lower incidence of dysphagia.

RESUMEN

Objective To construct the eukaryotic expression vector pEGFP-N1/IL-37b and analyze the expression of IL-37 gene in THP-1 cells. Methods Total RNA was extracted from human peripheral blood mononuclear cells ( PB-MCs) and the coding region of IL-37b gene was amplified by RT-qPCR. Then, the gene was cloned into pEGFP-N1 eu-karyotic expression vector. After transfected the recombinant plasmid into THP-1 cells, the expression of IL-37 was detec-ted by RT-qPCR and Western blot. Results Double restriction enzyme digestion and gene sequencing showed that IL-37b gene was correctly inserted into the eukaryotic expression vector pEGFP-N1. RT-qPCR and Western blot showed that the IL-37 expression level was increased significantly (P<0. 01) after transfection in THP-1 cells. Conclusions We successful-ly constructed a novel anti-inflammatory cytokine IL-37 eukaryotic expression vector pEGFP-N1/IL-37b, which lays a foun-dation for further study on IL-37 functions and its association with related diseases.

RESUMEN

[Summary] A total of 128 individuals with type 2 diabetes underwent continuous glucose monitoring for 3 consecutive days. The dawn phenomenon was defined by three different parameters according to the previous research:(1)the absolute increase of glucose level from nocturnal nadir to prebreakfast value(?G) above 20 mg/dl;(2)?G above 10 mg/dl;( 3 ) insulin requirement increased at least 20%.The participants were secondarily separated by presence/absence of a dawn phenomenon based on the definitions above. The impact on blood glucose fluctuation of different groups was assessed according to the standard deviation of blood glucose( SDBG) , the area under curve above 10 mmol/L ( AUC ) , and the mean amplitude of glycemic excursions ( MAGE ) , etc.The frequencies of dawn phenomenon were 64.8%(?G≥20mg/dl), 85.2%(?G≥10 mg/dl), and 59.4%(rise in insulin requirement≥20%)respectively.The impacts on SDBG, AUC, MAGE, and MODD were without statistical difference(P>0.05) between the presence and absence of the dawn phenomenon patients when?G≥10 mg/dl.However, the differences reached statistical significance(P<0.05) when ?G≥20 mg/dl and the increase in insulin requirement≥20%. Besides, the incidence of dawn phenomenon was positively correlated with HOMA-IR, HbA1C , and free C-peptide.Dawn phenomenon is a very frequent event in type 2 diabetes and not only impacts the overall glycemic control but also exaggerates glucose fluctuation. To be clinically relevant, ?G≥20mg/dl should be taken as the quantitative criterion of the dawn phenomenon.

Objective To study the effect of dexmedetomidine complex with remifentanil in elderly patients with laparoscopic colon cancer resection. Methods 80 cases of elderly patients with laparoscopic colon cancer resec-tion were selected and divided into the study group and the control group with 40 patients in each group according to the random number table method. Two groups were induced conventionally, and intraoperative anesthesia maintained by remifentanil and sevoflurane. The study group were pumped dexmedetomidine 0.8μg/kg 10min before induction, then the doses reduced to 0.2 μg·kg -1 ·h -1 for continuous pumping until to the end of surgery, while the control group was pumped the same dose 0.9% sodium chloride injection, then the heart rate(HR)and mean arterial pres-sure(MAP)of the two groups were compared when entered into surgery room(T0 ),administered 5min(T1 ),after intubation(T2 ),surgical incision(T3 )and after extubation(T4 ),and the compose score, recovery time, and agitation score of two groups were compared. Results HR and MAP of the study group at T1 ,T2 ,T3 and T4 were (62.2 ± 1.1)times/min,(67.3 ±3.2)times/min,(69.2 ±2.3)times/min,(70.2 ±2.5)beats/min and(68.3 ±0.5)mmHg,(70.1 ±0.9)mmHg,(71.5 ±0.9)mmHg,(68.2 ±1.9)mmHg significantly lower than which of the control group (t =9.289,10.293,8.928,9.039,9.278,10.789,9.322,8.399,all P <0.05),and HR and MAP of the control group at T1 ,T2 ,T3 and T4 were significantly higher than T0,the differences were statistically significant(t =8.928, 9.892,8.387,8.927,9,783,9,073,9.039,7.938,all P <0.05);Compose score(4.2 ±1.3)points of the study group was significantly higher than (1.2 ±0.2)points of the control group, but the agitation score of the study group was(1.2 ±0.7)points,which was significantly lower than (2.9 ±1.5)points of the control group, the differences were statistically significant(t =9.38,8.379,all P <0.05).Conclusion Dexmedetomidine complex with remifen-tanil has a anesthetic good effect in elderly patients with laparoscopic colon cancer resection, and which can maintain the more stable hemodynamics.
