While physical activity and zinc supplementation have shown benefits in diabetes management, little is known about their effect on less severe glucose homeostasis disorders, such as impaired glucose tolerance. On the other hand, sirtuins have an important role in glucose metabolism and insulin sensitivity, but to date, there is no information about the impact of zinc supplementation or physical activity on their regulation in individuals with impaired glucose homeostasis. The aim of this study was to assess the effect of supplemental zinc, muscle-resistance training, and their combination on the expression of selected sirtuins in insulin-sensitive tissues of rats with impaired glucose tolerance. Thirty male Wistar rats with impaired glucose tolerance were fed a high-fat diet for 12 weeks while subjected to zinc supplementation, resistance training, both, or none. Morphometric and metabolic evaluations were performed at the end of the experimental period, and gene expression of sirtuins 1, 2, 4, and 7 was assessed in liver, gastrocnemius muscle, and white adipose tissue. Results showed that zinc supplementation and/or resistance training did not improve metabolic parameters of rats with impaired glucose tolerance, nor did they affect the expression of selected sirtuins in any of the tissues evaluated. However, the expression of some sirtuins was associated with metabolic parameters in a tissue-specific manner. Additional studies are needed to evaluate whether zinc supplementation and/or resistance training can improve metabolic status or modulate sirtuins expression in advanced stages of impaired glucose homeostasis.

Voice production can be influenced by interindividual variations related to genetic, physiological, behavioral, and several environmental factors. Here we examined the effect of zygosity on speaking fundamental frequency (F0) statistical descriptors. Our aims were: (1) to determine whether the genetic similarity between monozygotic (MZ) and dizygotic (DZ) twins affects F0 characteristics, and (2) to quantify the contribution of genetic factors to these characteristics. The study involved 79 same-sex twin pairs of Brazilian Portuguese speakers, comprising 65 MZ and 14 DZ twins, aged 18 to 66 years (31.7 ± 11.6 years), with 21 male and 58 female pairs. Participants were recorded while uttering a greeting phrase and the Brazilian Portuguese version of the 'Happy Birthday to You' song. Speech segments were analyzed using Praat free software, and F0 measures were automatically extracted in both Hertz and semitone scales. Statistical descriptors, including centrality, dispersion, and extreme values of F0 were examined, and the ACE model (i.e., total genetic effects, A; shared environmental influences, C; and nonshared environmental influences, E) was employed to estimate the additive effect;ts of monozygosity. As anticipated, we observed a zygosity effect on several F0 parameters, with more similarity between MZ twins compared to DZ twins. We discuss the genetic influences on F0 parameters and the absence of a monozygosity effect in two of them. Additionally, we briefly address potential biases associated with the selected measurement scale for statistical modeling. Finally, we explore the influence of genetic factors on F0 patterns, as well as environmental, life history and linguistic factors, particularly concerning F0 variation in speech.

AIMS: To evaluate the effects of early and late pre-partum shearing of Corriedale ewes carrying single fetuses on placental and lamb development and neonatal lamb behaviour. METHODS: At 70 days of gestation, 37 multiparous Corriedale ewes with known gestation dates were randomly allocated into three groups: S70 (n = 12) and S110 (n = 12), shorn at Day 70 and 110 of gestation, and US (n = 13), that were not shorn pre-partum. Gestation length, lambing duration, placental expulsion time, placental weight, number and weight of cotyledons, and placental efficiency (lamb weight/placental weight) were determined. At birth, body temperature, weight, morphometric measurements, ponderal index (lamb weight/lamb crown-rump length) and behaviour were recorded, with weight remeasured 72 hours later. Four male lambs per group were slaughtered immediately after parturition, and organ weight and perirenal brown fat weight and energy content recorded. RESULTS: Pre-partum shearing, regardless of timing, increased total dried placental weight per ewe. Pre-partum shearing at Day 70 of gestation increased the number and mean weight of the > 2 to ≤ 3-cm cotyledons, while pre-partum shearing at Day 110 of gestation increased only the weight of these cotyledons. At birth and at 72 hours, lambs from ewes shorn either at 70 days of gestation (birth: 5.5 (95% CI = 4.6-6.4) kg, p = 0.001; 72 hours: 6.4 (95% CI = 6.1-6.8) kg, p = 0.002) or at 110 days of gestation (birth: 5.4 (95% CI = 4.4-6.4) kg, p = 0.001; 72 hours: 6.5 (95% CI = 5.9-7.1) kg, p = 0.001) were heavier than lambs from unshorn ewes (birth: 4.0 (95% CI = 3.3-4.8) kg; 72 hours: 4.8 (95% CI = 3.5-6.1) kg). Lambs born to S70 and S110 ewes suckled sooner (31.5 (95% CI = 14.5-48.5) minutes, p = 0.001; and 39.3 (95% CI = 23.7-55.0) minutes, p = 0.001 respectively), than lambs born to the US group (70.3 (95% CI = 38.6-102.1) minutes). There was no evidence for an effect of pre-partum shearing on gestation length, parturition length, time of placental expulsion, placental efficiency, weight and energy of perirenal brown fat, and lamb temperature at birth. CONCLUSIONS AND CLINICAL RELEVANCE: Shearing ewes pre-partum may lead to placental changes affecting lamb development and behaviour and associated with higher survival. The findings suggest pre-partum shearing may improve lamb survival, farm profitability and sustainability.

The corrosion inhibition action of Phalaris canariensis extract on 316 L stainless steel in the H2O-35 (wt%) LiCl mixture at different temperatures has been evaluated with the aid of weight loss, potentiodynamic polarization curves, linear polarization resistance and electrochemical impedance spectroscopy tests. These studies were complemented by Fourier Transformed Infrared spectroscopy, FTIR, gas/mass chromatography analytical techniques and detailed scanning electronic microscopy studies. Results have indicated that Phalaris canariensis extract is an efficient inhibitor, with an efficiency that increases with its concentration, but it decreases as the temperature increases. Phalaris canariensis extract is physically adsorbed onto stainless steel according to a Temkin type of adsorption isotherm. Phalaris canariensis extract affected both anodic and cathodic electrochemical reactions with a stronger effect on the anodic ones, acting, thus, as a mixed type of inhibitor. Main compounds contained in the Phalaris canariensis extract were palmitic and oleic acids, responsible for its inhibitory properties.

Background: Campylobacter is the leading cause of bacterial gastroenteritis worldwide. It is generally associated with an acute gastrointestinal infection causing a self-limiting diarrheal episode. However, there is evidence that persistent/recurrent carriage of Campylobacter also occurs. In hyperendemic settings the epidemiology and consequences of persistent Campylobacter enteric infections is poorly studied. Methods: Risk factors for and growth consequences of persistent Campylobacter infections detected by polymerase chain reaction (qPCR) were evaluated with data from the MAL-ED birth cohort study in children 0-24 months of age between November 2009 and February 2012. A persistent Campylobacter infection was defined as three or more consecutive Campylobacter positive monthly stools. Findings: Across all study sites, 45.5% (781/1715) of children experienced at least one persistent Campylobacter episode. The average cumulative duration of days in which children with persistent Campylobacter were positive for Campylobacter spp. was 150 days (inter-quartile range: 28-236 days). Children who experienced a persistent Campylobacter episode had an attained 24-month length-for-age (LAZ) score that was 0.23 (95% (CI): -0.31, -0.15) less than children without a persistent Campylobacter episode. Among children who had at least one episode of Campylobacter over a 3-month or 9-month window, persistent episodes were not significantly associated with poorer 3-month weight gain (-28.7 g, 95% CI: -63.4 g, 6.0 g) but were associated with poorer 9-month linear growth (-0.134 cm 95% CI: -0.246, -0.022) compared to children with an episode that resolved within 31 days. Interpretation: Persistent/recurrent Campylobacter infection is common among children and has a measurable negative impact on linear growth in early childhood. Yerba mate (Ilex paraguariensis) represents a culture of economic, social, and ecological importance for the cultivation regions. Due to the chemical, physical, and biological variations that occur in the different soils where yerba mate is economically exploited, the symbiotic associations with arbuscular mycorrhizal fungi (AMF) guarantee the plant's ability to absorb nutrients. The purpose of this study was to identify and quantify the occurrence of arbuscular mycorrhizal fungi in different environments of yerba mate cultivation. The research was performed in four areas located in the rural area of the municipality of Seberi/RS: Environment with production of yerba mate in the conventional system, silvopastoral system, organic system, and native forest. The normality of residuals and homogeneity of variances assumptions were verified using the Lilliefors and Chi-square tests and the averages compared by the Tukey's test at 5% probability of error. In addition to calculations of diversity, equivalent species, and evenness indices. The presence of AMF spores showed a direct relationship with the phosphorus (P) availability in each treatment, with a count reduction in the organic system, with P content lower than 3 mg kg-1 of soil. The species with the highest predominance were the Acaulosporaceae (Acaulospora colombiana, A. delicata, and A. tuberculata), followed by the Glomaceae (Glomus ambisporum and Glomus pansihalos) in the conventional and silvopastoral systems. The silvopastoral and conventional systems showed the highest levels of Shannon-Weaver diversity (H') and Pielou's evenness, demonstrating greater diversity and consequently greater richness and uniformity.

RESUMEN

Researchers in the Global South (GS, developing countries) make valuable contributions to the field of comparative physiology, but face economic and scientific disparities and several unique challenges compared with colleagues in the Global North (developed countries). This Perspective highlights some of the challenges, knowledge gaps and disparities in opportunity faced by GS researchers, especially those at early-career stages. We propose collaborative solutions to help address these issues, and advocate for promoting investment and cultural and societal change for a more inclusive research community. Additionally, we highlight the role of GS researchers in contributing expert knowledge on local biodiversity and the environment; this knowledge can help to shape the future of comparative physiology, allowing us to achieve a better understanding of the evolution of physiological mechanisms and to develop innovative solutions to environmental and biomedical challenges. With this Perspective, we hope to highlight the need to foster a more diverse, equitable and inclusive research landscape in comparative physiology; one that empowers GS scientists to address the global challenges associated with biodiversity loss, climate change and environmental pollution.

Edwardsiella ictaluri and Flavobacterium covae are two bacteria species that cause diseases in farm-raised channel catfish (Ictalurus punctatus) that cause heavy economic damage to the aquaculture industry, particularly to the channel catfish farming. In search for environmentally benign antibacterial compounds active against E. ictaluri and F. covae, we investigated the constituents isolated from Brazilian red, brown and green propolis. We have also synthetically modified active constituents to see if lipophilicity plays a role in enhancing antibacterial activities. Vestitol, neovestitol and methylvestitol were found to be the active constituents with minimum inhibitory concentration (MIC) relative to drug control florfenicol (RDCF) values (MIC-RDCF) of 7.6, 7.6 and 7.9 mg/L, respectively, against F. covae. The activity against E. ictaluri was not significant.

Understanding the role of open vegetation, particularly in white-sand ecosystems (WSE) and savannas, is crucial for elucidating their role in Amazonian biotic diversification. These ecosystems predominantly develop on sandy terrains, suggesting that the geological substrate significantly influences the vegetation upon it. Therefore, the interaction between landscape changes and biotic diversification is closely tied to the dynamics and resilience of these sandy substrates. Current WSE and savannas in lowland Amazonia colonized fluvial sediments deposited during the past 120 ka, with marked synchronicity over the last 23 ka, as shown by optically stimulated luminescence (OSL) and radiocarbon ages of such sandy substrates. In contrast, sandy substrates supporting open vegetation in highland areas, unsuitable for Quaternary sand accumulation, would have persisted beyond the Quaternary, as ancient sedimentary rocks in these areas are prone to developing sandy soils. The current distribution of open vegetation ecosystems in lowland Amazonia is coupled with the deposition and erosion of sandy sediments by Quaternary fluvial systems, while weathering sandy substrates in highland areas serve as long-term and resilient refugia beyond the Quaternary. The contrasting spatiotemporal dynamics of landscape changes in lowland and highland areas has implications for biodiversification or extinction events leading to current biogeography patterns in Amazonia.

PURPOSES: To measure the test-retest reliability and the clinical usefulness of the Work Limitation Questionnaire, and to compare the in-person with the telephone application. METHODS: Cross-sectional, exploratory study. The Work Limitation Questionnaire was answered three times: twice in person, to measure test-retest reliability and clinical usefulness, and once, by telephone, to measure the validity of the telephone application. RESULTS: Fifty-six individuals (32 men) with mild to moderate disabilities after stroke were included. Test-retest reliability was very high (ICC 0.96; 95% CI 0.94 to 0.98; p < 0.01), the clinical usefulness was high (9 out of 12 points), and the correlation between in-person and telephone applications was high (ρ = 0.7; 95% CI 0.5 to 0.9; p < 0.01). The average productivity loss was 4% (SD 5, min-max 0 to 15%). CONCLUSIONS: The Work Limitation Questionnaire showed adequate test-retest reliability and clinical usefulness in individuals with stroke. The telephone application produced comparable results to in-person applications. The participants reported low productivity loss, which may be related to the mild impairments of the included sample.

BACKGROUND: Dapagliflozin prevents myocardial dysfunction in chronic kidney disease patients regardless of residual kidney function. We hypothesized that this effect is extensible also to patients on dialysis. RESEARCH DESIGN AND METHODS: The DARE-ESKD-2 is an ongoing, single-center, open-label randomized clinical trial designed to determine the effects of adding dapagliflozin to standard treatment on myocardial function and structure. Eligible patients were adults on a regular dialysis scheme for more than 3 months. Pregnancy, liver failure, allergy to the investigational drug, and prior use of SGLT2i were exclusion criteria. Participants were randomized in a 1:1 ratio to dapagliflozin or standard treatment groups for 24-weeks. The primary goal is to compare the change in NT-proBNP levels between study arms, and secondary goals include comparing the between-group difference in left ventricle global longitudinal strain, indexed mass, ejection fraction, and E/e` ratio, and on symptoms scale and 6-minute walk test distance. An exploratory analysis will evaluate changes in body composition and bone densitometry. RESULTS: The trial has finished the enrollment of 80 patients, who are currently being followed-up. CONCLUSIONS: This trial will provide novel data on myocardial effects of SGLT2i in dialysis recipients. Results from this study may provide evidence to support SGLT2i use in ESKD.

BACKGROUND: Access to essential diagnostics is crucial for primary healthcare (PHC) in low-and-middle income countries (LMICs). Many LMICs have invested in equipping PHC with point-of-care (PoC) diagnostics for infectious diseases, however there has been no similar investment to improve PHC capacities for clinical chemistry. The biochemistry gap is among the deterrents to universal health coverage. METHODS: A social sciences project was conducted with the aim to understand the key PHC stakeholders' insights on the pertinence of PoC biochemistry for PHC in LMICs. Data generation was conducted between July-November 2023 in Mongolia, Nigeria and Peru. Decision-makers in healthcare delivery, healthcare professionals, and patient and community advocates were engaged using a combination of sampling techniques. Unstructured individual and group conversations, and non-participant observation were conducted. Analysis involved an inductive line-by-line coding on printed transcripts, followed by a deductive coding and theme-by-theme analysis on digitized transcripts. RESULTS: Fifteen, 51 and 20 informants from Mongolia, Nigeria and Peru, respectively, participated. Fifty-five of the 94 informants were female. Most informants considered that PoC biochemistry in PHC would be pertinent, from a clinical and a resources-saving perspective. Those households that currently bear the burden of referrals (i.e., the poor, the bedridden, the older adults) would benefit the most from the deployment of PoC biochemistry for essential biochemistry parameters. Improved access to PoC glycated hemoglobin (HbA1c), lipid, liver and kidney profile was perceived as helpful to inform clinicians' decision-taking. The value of PoC biochemistry for the management of noncommunicable diseases (diabetes, hypertension) and infectious conditions (dengue, malaria, tuberculosis), to improve child health outcomes (severe dehydration in children with diarrhea and/or malnutrition) and to reduce preventable causes of death (dengue-related renal failure) was highlighted. CONCLUSIONS: PoC biochemistry holds potential to revert the impact that the biochemistry gap has for patient care in some LMICs' PHC settings. PoC equipment for parameters such as HbA1c, urea, creatinine or electrolytes could enhance community-level management of preventable causes of mortality, improve service delivery for patients affected by locally-prevalent infectious conditions, and improve the psychosocial and economic wellbeing of patients facing the burden of referrals to remote biochemistry-equipped centers. Volcanic eruptions can release large amounts of tephra, lava, and gases, drawing attention due to their magnitude, energy, and impact on life and the environment. Among the most documented and sometimes dramatic effects of volcanic ashes are those linked to the input of diverse elements in the environment, which are released as a consequence of ash weathering. Laboratory studies have been conducted to investigate and predict the environmental input of chemical elements from volcanic ashes. This research paper describes the optimization of batch leaching tests used to investigate the release of ions from ashes collected in the Andes Cordillera after the eruption of the Puyehue volcano in 2011. Chemometric multivariate strategies were employed to evaluate the influence of variables affecting the leaching of volcanic ash. The effects of the main variables, namely contact time, the acidity of the leaching agent, the solid/liquid ratio, the particle size, and the stirring speed, were studied in leaching tests. To determine the optimal conditions for selected metal determinations, we employ Darringer's desirability function, which allows for the simultaneous optimization of the selected responses (element concentrations during the leaching process). Multielemental analysis (Na, Mg, Al, Si, P, Cl, K, Ca, V, Cr, Mn, Fe, Ni, Cu, Zn, Sr, Cd, Hg, Tl, and Pb) was quantified by ICP-MS (inductively coupled plasma-mass spectrometry) following adequate dilution of test leaching. These results established the optimal experimental conditions for leaching volcanic ash. The most significant variables were the solid/liquid ratio and the stirring speed, resulting in two groups of elements with an adequate global desirability function (D) value.

BACKGROUND: This study aimed to determine the association between sleep quality and oral lichen planus (OLP). MATERIAL AND METHODS: The protocol was registered in PROSPERO. The inclusion criteria used in the literature search consisted of studies that investigated sleep quality in patients with OLP, without language or publication time restrictions. The outcome measures included sleep quality, expressed by the Pittsburgh Sleep Quality Index, Epworth Sleepiness Scale and General Health Questionnaire scores. A search was performed in PubMed, Embase, Web of Science, LILACS and Scopus. Another search in Google Scholar and manual search through the references as well were performed up to January 2024. Bias risk was determined using the JBI Critical Appraisal Checklist. RESULTS: Four observational case-control studies with a total of 1125 participants were identified. Mean age ranged between 40 and 65.2 years, and all participants were over 18 years old. In all studies, OLP patients had higher sleep disturbance scores than control groups (p<.05). Patients with erosive OLP type had higher mean scores for insomnia. CONCLUSIONS: Even though the results disclose an association between OLP and sleep disturbances, the evidence for such association is limited, because of the small number of studies published and their heterogeneity.

Utilizing coastal land for agriculture presents challenges such as low water content, high soil salinity, and low organic compound content. To support plant growth under these conditions, biofertilizers composed of plant growth promoting Rhizobacteria (PGPR), especially those inhabiting coastal areas, are needed. The Parangkusumo sand dunes on the southern coast of Java, Indonesia, is a unique coastal ecosystem characterized by arid conditions, high temperatures, and high soil salinity. To date, no studies have reported the isolation of PGPR from this ecosystem. This study is the first to isolate and identify PGPR associated with Spinifex littoreus, a dominant plant species in the Parangkusumo sand dunes, which are adapted to the harsh condition of Parangkusumo sand dunes. Ten rhizobacterial isolates were obtained, with five identified as members of the Bacillaceae family. All isolates demonstrated phosphate solubilization activity, while seven exhibited cellulolytic activity. One isolate, Priestia aryabhattai strain 2, notably showed phosphate solubilization and nitrogen fixation activities. The findings of this PGPR activity screening offer valuable insights for developing biofertilizers tailored for coastal agricultural applications.

Background: Infections caused by antibiotic-resistant bacteria pose a major challenge to modern healthcare. This systematic review evaluates the efficacy of machine learning (ML) approaches in predicting antimicrobial resistance (AMR) in critical pathogens (CP), considering Whole Genome Sequencing (WGS) and antimicrobial susceptibility testing (AST). Methods: The search covered databases including PubMed/MEDLINE, EMBASE, Web of Science, SCOPUS, and SCIELO, from their inception until June 2024. The review protocol was officially registered on PROSPERO (CRD42024543099). Results: The review included 26 papers, analyzing data from 104,141 microbial samples. Random Forest (RF), XGBoost, and logistic regression (LR) emerged as the top-performing models, with mean Area Under the Receiver Operating Characteristic (AUC) values of 0.89, 0.87, and 0.87, respectively. RF showed superior performance with AUC values ranging from 0.66 to 0.97, while XGBoost and LR showed similar performance with AUC values ranging from 0.83 to 0.91 and 0.76 to 0.96, respectively. Most studies indicate that integrating WGS and AST data into ML models enhances predictive performance, improves antibiotic stewardship, and provides valuable clinical decision support. ML shows significant promise for predicting AMR by integrating WGS and AST data in CP. Standardized guidelines are needed to ensure consistency in future research.

RESUMEN

Dacryocystitis, inflammation and infection of the lacrimal sac, is most commonly caused by infection from Staphylococcus and Streptococcus species. This report highlights a rare case of chronic dacryocystitis due to the atypical pathogen Mycobacterium abscessus. A 62-year-old woman presented with several months of left medial canthal pain, tenderness, and discharge. Exam demonstrated a left tender medial nodule, and imaging showed left lacrimal sac dilation and fluid collection consistent with dacryocystitis. She underwent external dacryocystorhinostomy with drainage and culture of the abscess, which was positive for M. abscessus. Her post-surgical treatment required an extended course of antibiotics, including omadacycline and azithromycin, with slow but progressive symptomatic improvement. This case is only the second reported case of dacryocystitis due to M. abscessus and suggests a role for culturing lacrimal sac abscesses intraoperatively due to the need for extended antibiotic therapy for atypical infections that may have high antibiotic resistance.
