RESUMEN

An analytic design of a prototype filter for M-channel maximally decimated cosine-modulated Near Perfect Reconstruction (NPR) filter banks is proposed in this work. The prototype filter is created using the least-square (CLS) method with weighted constraints, which is one-dimensional and requires single-parameter optimization. Compared to existing approaches, this suggested method achieves rapid convergence by analytically determining the optimal step size, ensuring the 3 dB cutoff frequency at π/2 M. The simulation results for design examples outperform the techniques in the available literature in terms of amplitude and aliasing distortion, reaching distortion around 2.4489 × 10-4 and 3.4907 × 10-9, respectively. This optimization algorithm's usefulness is further demonstrated with the sub-band coding of ECG signals. Implementing optimal prototype filters has tangible real-world effects, especially in critical sectors like healthcare and communications, improving diagnostics accuracy, data transmission efficiency, and overall performance.

Adaptability to intracellular or extracellular cues is essential for maintaining cellular homeostasis. Metabolic signals intricately control the morphology and functions of mitochondria by regulating bioenergetics and metabolism. Here, we describe the involvement of PHLPP1, a Ser/Thr phosphatase, in mitochondrial homeostasis. Microscopic analysis showed the enhanced globular structure of mitochondria in PHLPP1-depleted HEK 293T and C2C12 cells, while forced expression of PHLPP1 promoted mitochondrial tubularity. We show that PHLPP1 promoted pro-fusion markers MFN2 and p-DRP1Ser637 levels using over-expression and knockdown strategies. Contrastingly, PHLPP1 induced mitochondrial fragmentation by augmenting pro-fission markers, t-DRP1 and pDrp1Ser616 upon mitochondrial stress. At the molecular level, PHLPP1 interacted with and caused dephosphorylation of calcineurin, a p-DRP1Ser637 phosphatase, under basal conditions. Likewise, PHLPP1 dimerized with PINK1 under basal conditions. However, the interaction of PHLPP1 with both calcineurin and PINK1 was impaired upon CCCP and oligomycin-induced mitochondrial stress. Interestingly, upon mitochondrial membrane depolarization, PHLPP1 promoted PINK1 stabilization and parkin recruitment to mitochondria, and thereby activated the mitophagy machinery providing a molecular explanation for the dual effects of PHLPP1 on mitochondria under different conditions. Consistent with our in-vitro findings, depletion of phlp-2, ortholog of PHLPP1 in C. elegans, led to mitochondrial fission under basal conditions, extended the lifespan of the worms, and enhanced survival of worms subjected to paraquat-induced oxidative stress.

Glass ionomer cement (GIC) is a material used for restoration in dentistry. The stress in a material right before it yields in a flexure test is referred to as flexural strength. The aim of this study was to evaluate the flexural strength of two different glass ionomers after immersion in fruit juices. Five samples of D-Tech and Shofu GIC were prepared from each for the in vitro study. The eight samples were subjected to a flexural strength process and two were left as control which was followed by the determination of maximum force and displacement, then the collected data were used to determine flexural strength and analyzed using SPSS software (IBM SPSS Statistics for Windows, Version 22.0. Armonk, NY: IBM Corp). The mean value of flexural strength of the D-Tech GIC immersed in fruit juices is 4.4375 and the mean value of Shofu GIC immersed in fruit juices was 8.4975. Paired t-test was done between the two groups and the P value was 0.00, which is considered highly statistically significant. From this study, it is evident that the GIC brand Shofu has higher flexural strength compared to D-Tech. Flexural strength was decreased for both the GIC groups when immersed in grape juice.

Cyclic 3',5' adenosine monophosphate (cAMP) is a ubiquitous signaling molecule, but its role in higher plants was in doubt due to its very low concentration. In this study we wanted to look at the flux of cAMP in response to light in algae, considered to be the more primitive form of photosynthetic organisms. While it did not fluctuate very much in the tested green algae, in the cyanobacterium Arthrospira platensis its level was closely linked to exposure to light. The expression from cyaC, the major isoform of adenylate cyclase was strongly influenced by exposure of the cells to light. There was about 300 fold enhancement of cyaC transcripts in cells exposed to light compared to the transcripts in cells in the dark. Although post-translational regulation of adenylate cyclase activity has been widely known, our studies suggest that transcriptional control could also be an important aspect of its regulation in A. platensis.
