RESUMEN

The aim of this study was the development of a methodology for the integral study of the antagonistic activity of normal human microbiota against Staphylococcus aureus to enable direct selection (without prior isolation of pure cultures) of potentially highly efficient probiotic preparations. The selection of bacterial representatives of normal human nasal microbiota capable of antagonizing S. aureus was carried out using two complimentary methods: replica-plating and deferred antagonism procedures. The material of the anterior nares from healthy human subjects was plated onto the surface of different nutrient media agar plates followed by incubation under appropriate conditions. The grown bacterial colonies were replica-plated to Petri dishes with nutrient agar overlayed with a thin layer of a soft agar which contained the culture of an indicator S. aureus strain. This agar also supported the growth of potential probiotic strains. The potential probiotic strains were selected by their ability to suppress the growth of S. aureus around their colonies. Most active strains-inhibitors may be used to develop probiotic preparations with targeted activity against S. aureus.

RESUMEN

Two clinical cases of perforation of a previously undiagnosed colon diverticulum in patients with coronavirus infection caused by the SARS-CoV-2 virus treated at the Hospital №1 of Nalchik. Both patients were elderly, overweight, had a lot of chronic concomitant diseases. Patients received hormone therapy and were targeted: the first patient twice (tocilizumab on the first day of hospitalization and olokizumab on the 7th day of inpatient treatment). The second patient received levilimab on the 3rd day of his stay in the hospital. A short time after targeting, both patients developed acute diffuse abdominal pain, the patients were transferred to the surgical department and operated on. During the operation, both patients were found to have previously undiagnosed diverticular disease, complicated by diverticular perforation and peritonitis on the background of immunosuppression. Both patients died. Thus, when using targeted therapy for patients with COVID-19, it is necessary to take into account that they may have previously undiagnosed chronic diseases that can cause fatal complications against the background of immunosuppression.

The concentrations of ATP, IL-6, and IL-10 were measured in extracts of plucked hair follicles from healthy volunteers (normal values) and patients with androgenetic alopecia and then, ATP, IL-6, and IL-10 content was calculated for each follicle. The resulting values were directly proportional to hair follicle length, except for IL-6. The concentration of extracted ATP correlated with lactate dehydrogenase activity indicating cell damage. In patients with androgenetic alopecia, IL-10 content exceeded the normal values in follicles with a length <1 mm and ATP content surpassed the normal in follicles >2 mm long. The content of IL-6 and IL-10 measured by ELISA was comparable with results of mRNA expression assayed by RT-PCR, which attested to moderate level of gene expression. The content of ATP and IL- 10, but not IL-6 depended on the length of plucked hair follicle and on pathogenetic factors affecting hair growth.

RESUMEN

The studied levels of cytokines (levels of Pro-inflammatory cytokine - interleukin-1ß, anti-inflammatory cytokine - interleukin-10, CD40 ligand (CD40L)) in patients with arterial hypertension and ischemic heart disease. Indicators of Pro-inflammatory cytokine - interleukin-1ß and anti-inflammatory cytokine - interleukin-10 were significantly increased in patients with arterial hypertension and ischemic heart disease compared with values in healthy individuals. Elevated interleukin-1ß and CD40L in patients with arterial hypertension and ischemic heart disease compared with values in healthy individuals indicate inflammatory process in the endothelium. An increase in the IL-10 may indicate the presence of chronic systemic inflammation, in which vascular endothelium is likely to take an active part. Long-term high IL-1ß rates lead to increased activity of cells of species immunity, whose metabolic products are involved in the activation of the adrenal glands. Thus, the cytokine profile disorder can be considered as an additional laboratory indicator of atherosclerotic vascular damage.

Levels of IL-1beta, IL-6, TNFalpha, IL-10 and TGF-beta in blood and wound of 56 patients with odontogenic phlegmons in dynamic of infection process were expected. In accordance with known facts production of proinflammatory cytokines (IL-1beta, IL-6, TNFalpha in blood in acute phase of the inflammation process was much more intensive than in normal conditions. Increased proinflammatory cytokines generation led to significant raising of IL-10 concentration, but elevation of its level in blood was only at the 10-14 the days of therapy. The dynamic of these cytokines concentrations in wound were similar. On other side the negative role in regeneration of wound plays TGFbeta. Minimal concentration of this cytokine in blood was found at 1-3 from treatment beginning. On the background of the intensive therapy the level of TGFbeta raised fastly and at 10-14 days was much more than in control conditions. Wound level of TGFbeta was more than in normal conditions from the third day. These changes may be one of the reason of hyper proliferation of keratinocytes, that led rough scars.

RESUMEN

Treatment with the phytopreparation from papaya accelerated wound healing and reduced the severity of local inflammation in rats with burn wounds. The effect of this phytopreparation can be related to an increase in the effectiveness of intracellular bacterial killing by tissue phagocytes due to the inhibition of bacterial catalase. Antioxidant activity of the preparation decreases the risk of oxidative damage to tissues.
