RESUMEN

The state of rat kidneys after injection of bone marrow multipotent stromal cells (MSC) labeled with Vybrant CM-Dil into intact or resected liver was studied by fluorescence microscopy. The main structural changes in the kidneys after MSC injection into intact and partially resected liver manifested as granular dystrophy and necrobiotic/necrotic changes in single epithelial cells of the distal tubules and collecting ducts, thrombosis of some vessels, progression of an ascending urinary tract infection (detection of dust-like fluorescent objects), which can be due to the immunomodulating or even immunosuppressive influence of MSC and their detritus. MSC injected into the intact or resected liver, as well as the products of their degradation were not detected in the kidneys at all terms of observation. After injection of MSC into partially resected liver, manifestations of bacterial contamination of the renal medulla appeared later. The injection of MSC into the liver can be complicated by thrombosis of the renal vessels, which should be taken into account when using this administration route in the cell therapy.

Helicobacter pylori induced gastritis accounts for 70% of cases in the structure of this pathology. Features of the long-term inflammatory reaction of the mucous membrane are directly related to the mechanisms of bacterial pathogenicity, and features of immunogenesis within narrow limits of the specific tissue microenvironment of organ structures. Mast cells appear to be one of the key players (promoters) in the regulation of the inflammatory mediator cascade and the formation of cytokine-induced expression. Possessing a wide arsenal of biologically active substances, mast cells are able to participate in the formation of the immune response and resistance of the gastric mucosa, modulating both pro- and anti-inflammatory effects. The antigen-presenting features of mast cells are of interest in terms of interaction with H. pylori and induction of mucosa bacterial colonization. The aim of study was to assess the mast cell tryptase profile of the gastric mucosa in the immunopathogenesis of H. pylori-associated inflammation. Material and methods. The study included 19 biopsies of the gastric mucosa with unknown status of H. pylori infection. Microslides were stained with hematoxylin and eosin, and Giemsa's dye for plain microscopy. H. pylori infection of the gastric mucosa was detected using the immunohistochemical method. Using double immunofluorescent labeling, localization of tryptase-positive mast cells and H. pylori strains was detected. Results. In patients infected with H. pylori (n=12), there was a significant increase in the number of tryptase-positive mast cells (177.99±30.55 vs 88.58±11.49; p<0.05) with activation of secretory pathways and release of protease into the extracellular matrix of the gastric mucosa. The quantitative parameters of mast cells in the group of patients with an undetected pathogen and signs of a chronic inflammation of the gastric mucosa were statistically significantly lower than in the group of infected patients. Co-localization of tryptase-positive mast cells and H. pylori strains (with the formation of areas of large free-lying granule accumulation around the glands with pronounced degree of H. pylori contamination) was detected in gastrobiopsy specimens, the fact evidencing their close involvement in the development of inflammatory reactions of the gastric mucosa. Conclusion. The study demonstrated the features of mast cells and H. pylori interaction revealing previously unknown aspects of gastritis pathophysiology. The data obtained contribute a valuable insight to choose a treatment strategy for H. pylori-associated gastritis.

The effects of inhalation exposure to a mixture containing acetone, acetaldehyde, and ethanol in concentrations typical of closed environment in space vehicles on the structure of mesenteric lymph nodes in F1 male mice were studied by histological technique. The long-term exposure to modeled atmosphere led to pronounced structural changes in these nodes that were clearly seen on day 22 and increased by day 36 of the experiment. The thickness of the capsule and trabeculae of mesenteric lymph nodes as well as diameter of lymphatic sinuses did not differ from the control values up to day 8, but then increased on days 22-70. Starting from day 22, the thickness of the medullary cords decreased and attained the minimum to the end of the experiments, which can indicate depletion of immunocytopoiesis and inhibition of humoral immunity. The present data are important for the space medicine, because they indicate structural changes in the peripheral lymphoid organs, the key elements of immune system.

RESUMEN

The safety of Grippol vaccine was assessed by macro- and microscopic methods in the experiment. The morphological features of group lymphoid nodules of the small intestine were studied on days 4, 7, 14 and 30 after vaccination. The structural morphological dynamics of lymphoid tissue under these conditions is shown. Because of the use of the vaccine, the lymphocytopoietic processes in the body are activated, which indicates the formation of an immune response.

The organs of the digestive system experience high sensitivity to the orbital flight factors and may limit the implementation of the professional activities of crews on International space station. The connective tissue as a system-forming matrix of the integrative and buffering metabolic environment has a particular importance in the space biomedicine because it provides the inner organ functionality in the conditions of changing level of the gravitational incentive. Aim - to study the adaptive mechanisms of the fibrous component of the extracellular matrix of the connective tissue of stomach and intestines on the effect of prolonged microgravity. MATERIAL AND METHODS: Using histochemical methods the condition of collagen fibers of a specific tissue microenvironment of the membranes of stomach and intestines of C57BL/6N mice (58 males with an initial body weight of 27.1±0.7 g) after a 30-day space flight and the following 7-day land readaptation was studied as well as in the animals representing corresponding control groups. RESULTS AND DISCUSSION: Laboratory animal presence on the biosatellite «BION-M¼ No. 1 has led to the fibrous reduction of extracellular matrix of connective tissue in the studied organs of digestive system structure except for the proper lamina of the gastric mucous membrane. Fibrillogenesis increase in the gastrointestinal tract in comparison with the indicators of space flight animal group has been found after 7 days of the biosatellite landing. The collagen fibers were not characterized by the significance change from the vivarium control group during the experiment with the land modelling of orbital flight conditions. CONCLUSION: The obtained results represent the evidence of fibrous structure gravity sensitivity of extracellular matrix of the connective tissue and show the relevance in the sphere of preventive measure improvement of the digestive system organs in the profession of astronauts in the orbital flight conditions.

Specialized sports nutrition is one of the most important factors in the extension of the functional potential of athletes, providing adaptive resistance to physical stress, which determines the high physical performance and prolongs athletic longevity of the athletes. The study involved 30 skiers-racers (the average age of 19.5±1.8 years). 12 skiers of the main group within 21 days consumed a specialized food product, obtained on the basis of fermented milk whey containing amino acids, several vitamins, minerals and trace elements, live culture of lactic acid bacteria: L. lactis, L. thermophilus, L. bulgaricus (1.2×108 CFU/cm3). The control group consisted of 18 skiers, those taking the placebo (food starch of the same consistency). After a course of product intake, blood level of hemoglobin increased by 6%, of leukocytes - by 10% due to an increase in the number of granulocytes by 32%, and segmented neutrophils by 16% (p<0.05), there was a tendency to increase the number of red blood cells by 7% with a significant decrease in lymphocyte count by 19%. Erythrocyte sedimentation rate in blood of the skiers from the comparison group increased by 41% (p<0.05), while in the athletes of the main group it decreased by 16% (p>0.05). After product intake it has been established by the method of laser Doppler flowmetry that there was a tendency to increase blood perfusion by 15%, a statistically reliable increase in the flux by 53%, which is based on the improvement of the internal mechanisms of microcirculation regulation. According to the mathematical analysis of cardiac rhythm, centralization of regulation decreased while the activity of an autonomous mechanism for controlling the work of the heart increased. The revealed functional changes ensured an increase of absolute (by 31%, p<0.05) and relative (by 33%, p<0.05) physical performance and aerobic endurance of skiers, contributed to the improvement of short-term memory. The conclusion is made about the expediency of the intake of the specialized food product to enhance the adaptive capacity of athletes under the influence of systematic physical loads.

We studied the reactions of spinal ganglion neurons accompanying regeneration of the skin wound complicated by S. aureus sp. infection. The appearance of neuron groups with different morphofunctional changes was noted: reversible dystrophic and compensatory reactions and irreversible degenerative changes. The proportion of small B neurons was higher than the proportion of large type A neurons at all terms of the experiment. Application of plateletrich plasma to the wound after preliminary hydroimpulsive treatment considerably reduced cell destruction and stimulated regenerative effects in neurons starting from day 7 of the experiment. Application of platelet-rich plasma without hydroimpulsive treatment decelerated wound regeneration and increased the number of destructively changed neurons even in comparison with spontaneous wound healing. Close correlations between protein synthesis markers and stages of the wound process and its characteristics were revealed.

We performed a complex morphological analysis of atherosclerotic plaques obtained from 68 men with coronary atherosclerosis during coronary bypass surgery with endarterectomy. The expression of MMP-2 and MMP-9, collagen IV, CD31, CD34, factor VIII, and of smooth muscle cell actin was measured in the samples by morphometric and immunohistochemical methods. The expression of MMP-2 and MMP-9 as well as the intensity of neoangiogenesis estimated by the expression of CD31, CD34, and factor VIII in unstable plaques was significantly higher than in stable ones. Immunohistochemical analysis showed more intensive collagen IV expression in stable plaques. The observed differences in immunohistochemical phenotypes of stable and unstable atherosclerotic plaques reflect peculiarities of morphogenesis of atherosclerotic foci in the coronary arteries determining their further development.

Number and size of aggregated lymphoid nodules in the small intestine of mice exposed to radiation using a γ-irradiation unit GOBO-60 with 137Cs source were measured using the methods of visual microscopy and morphometry. Morphological status of the nodules was estimated on days 4, 28, 60, and 90 after the exposure. Significant changes were observed in the lymphoid system of the small intestine after exposure to low-intensity radiation. The size of aggregated lymphoid nodules decreased and the relative number of cells with degenerative changes increased. Regenerative processes were observed as soon as on day 4 of the rehabilitation period. However, the number and sizes of aggregated lymphoid nodules (length, width, and section area), and the ratio of lymphoid nodules with germinal center were still lower than in the control. The number of cells forming lymphoid interstitial plaques was close to the control on days 28-90 of the rehabilitation process.

RESUMEN

Ethical aspects of training of physicians are considered. The author highlights the importance of teaching the basic disciplines during the training of the neurologist. An analysis of most frequent mistakes that arise in practical activity as a consequence of overestimation of paraclinical methods of investigation, underestimation of a role of treatment that the patient receives due to other indications is presented. The formation of life attitude of the physician is the extremely important component of teaching process. One should not forget the lessons of history shown examples of physicians that gave their lives to save patients. Also, there are examples of physicians who had violated the fundamental humanitarian principles.

The nutrition factor is one of the most important in the achievement of high sport results and in maintaining of athlete health along with the methodological and psychological aspects of training. Improving the methods of training in different sports leads to a significant increase in the level of energy expenditure. In order to provide athletes with a large amount of energy, it is advisable to use specialized products with increased nutritive and biological value. Specialists should pay special attention to the scientific justification of composition of special food products (SPP) with consideration of age and gender of athlete, type of sport, the stages of sports activities, training regimes, the qualifications of the athlete and individual level of metabolism. An analytical review of literature on the appropriateness of the application of SPP for athlete nutrition in the preparatory period was conducted to justify the need for further research in this field.

The basal metabolism rate (BMR) is of great importance in the assessment of daily energy requirements and physical activity level of a person. Article reviews the factors influencing the BMR. The BMR significantly correlates with weight, height, age, sex. Prediction equations based on these factors account for approximately 70% of the variability of the basal metabolism. However, these equations do not take into account the effect of body composition. The BMR is associated with lean body mass. Among body composition indicators lean body mass is the strongest determinant of BMR. The rate of basal metabolism, predicted on the basis of the relationship with the lean body mass covers the same 70% of the variability that are determined by gender and age differences, as most of the variability due to gender and age differences. In addition, the role of physical activity and nutrition as factors that affect the value of the BMR is discussed. The conclusion is made that lean body mass is the strongest predictor of BMR. Body fat mass also affects the basal metabolic rate as well as physical activity level and nutrition.

The article presents the anthropometric indicators of 210 old age men (aged 60-74 years), 108 elderly men (aged 75-90 years) and 125 long-liver men (aged 90-98 years) of the Slavic ethnic group, living in Moscow and Moscow region. Significant differences in basic anthropometric parameters have been established in body weight and body mass index between the three age groups. Average values of body height in elderly men were lower by 0.6% (p>0.05), and in long-liver men were significantly lower by 4.6% than in old age men. Diameters did not have statistically significant differences in all age groups. Averages values of all circumferences of available limbs significantly differed in three age groups, averages values of waist and thigh circumference significantly differed only in the group of long-liver men and old age men. Averages values of breast circumference were not statistically different in three age groups. Average values of all eight skin-fat folds were significantly lower in long-liver men compared with old age men, whereas in elderly men only skin-fat folds of shoulder front and back, forearms, chest and lower leg were significantly less than in old age men. Somatotypological analysis revealed a different frequency of somatotype occurrence and the prevalence of 3 main types among men of older age groups (old age men, elderly men, long-liver men) - chest (18.5, 26.2 and 28.4%), abdominal-muscle (20.1, 15.4 and 16.8%) and abdominal (20.8, 22.2 and 19.4% respectively). Also the peculiarities of the component body composition of men of older age groups were revealed. Body composition analysis revealed a decrease in the average indicators of the absolute content of bone, fat and muscular body components from the old age to the period of longevity (long-liver men). Absolute content of bone tissue in elderly men (7.9±0.3 kg) didn't differ and in the period of longevity (6.8±0.2 kg) was less by 1.18 fold (p<0.05) than in old age men (8.0±0.3 kg). Absolute amount of body fat tissue in elderly men (16.1±1.2 kg) was less by 1.09 fold (p<0.05) and in long-liver men (12.5±1.0 kg) was less by 1.41 fold (p<0.05) than in old age men (17.6±1.4 kg). Absolute amount of body muscle tissue in elderly men (18.2±0.3 kg) was less by 1.17 fold (p<0.05) and in the period of longevity (16.3±0.2 kg) was less by 1.31 fold (p<0.05) than in old age men (21.3±0.2 kg). Average relative indicators of the major body components with aging undergo a number of changes, which are reflected in the reduction of quantity of bone and muscle tissue and increasing rates of adipose tissue.

The article presents the anthropometric parameters of 251 elderly women (75-90 years) and 125 long-liver women (90-98 years) of the Slavic ethnic group, living in Moscow and Moscow region. Significant differences in basic anthropometric characteristics between two age groups have been demonstrated. Average values of body weight and height, circumferences and quantities of skin-fat folds were significantly lower in long-liver women in compare with representatives of the elderly, whereas diameters had no statistical significant differences. Somatotypological analysis revealed a frequency of occurrence of different somatotypes and prevalence of the three main types among elderly and long-liver women - asthenic (32.2-34.0%), pyknic (29.3-30.0%) and europlastic (20.0-21.2%) somatotype. Some features of body composition characteristics of elderly and long-livers women have been demonstrated as well. Estimated absolute amount of bone compartment did not differ in two women groups, while relative amount of bone compartment in elderly women (15.30±0.21%) was lower by 1.11 fold (p<0.05) than in long-liver women (17.05+±0.17%). The content of fat and muscular body compartment was significantly (p<0.05) lower in long-liver women as compared with the elderly women. The absolute amount of fat body compartment in long-liver women was 9.15±1.22 vs 13.13±0.49 kg in elderly women, the relative amount of fat body compartment - 14.39±0.26 vs 18.04±0.05%; the absolute amount of muscular body compartment - 23.04±0.26 vs 28.06±0.47 kg, the relative amount of fat body compartment - 36.22±0.15 vs 38.54±0.16%.

A comprehensive anthropometric and bioelectrical study of 651 women of mature age (relative norm) aged 22 to 55 years living in the Moscow region, was conducted. The somatotyping was performed according to I.B. Galant-V.P. Chtetzov-B.A. Ni kitiuk scheme. The quantitative distribution of women according to their somatotypological characteristics was obtained, the anthropometric peculiarities and the specific features of body composition in women belonging to different somatotypes were determined. It was found that among the women of mature age studied, mesoplastic (26.9%) and stenoplastici (16.7%) somatotypes were dominant. The least frequently encountered were the representatives of asthenic (1.7%) and athletic (5.2%) somatic types. Women of different somatotypes vary significantly in height and weight and other anthropometric parameters.

The physical state of body construction of 651 practically health women 20-55 years old were studied by using a complex of anthropometric and bioelectric methods (conditional norm, slavonians, inhabitants of Moscow region). The leptosome constitutional group was established in 152 cases (23.3%), mesosome group was exposes in 259 cases (39.8%), megalosome group was revealed in 189 cases (29.0%) and indeterminate group was fixed in 78 cases (8.0%). Women of mesoplastic somatotype were established in 26.9%, sthenoplastic somatotype--in 16.7%, picnic somatotype--in 13.7%, subathletic somatorype--in 11.5%, euriplastic somatotype--in 12.3%, athletic somatotype--in 5.2% and asthenic somatotype--in 1.7%. Sizes and weight parameters were depending on type of women body construction. Norm of body mass index for asthenic women was fixed as 16.9; for sthenoplastic somatotype--20.3; for athletic somatotype--23.1; for subathletic somatotype--24.2; for mesoplastic somatotype--24.6; for euriplastic somatotype--24.7; for picnic somatotype--26.2. The length of women body was varied from 158.2 +/- 0.5 (152.0-165.0) cm for sthenoplastic somatotype and 159.6 +/- 0.4 (152.0- 164.8) cm for mesoplastic somatotype to 169.4 +/- 1.7 (162.2-187.4) cm for athletic somatotype. Body weight was also in correspondence with type of constitution. It was 47.8 +/- 0.6 (44.2-54.6) kg for asthenic somatotype, 50.6 +/- 0.4 (42.6-54.2) kg for sthenoplastic somatotype, 62.2 +/- 0.2 (56.3-64.9) kg for mesoplastic somatotype, 68.6 +/- 0.7 (64.5-82.7) kg for picnic somatotype, 66.2 +/- 1.0 (62.0-76.5) kg for athletic somatotype, 68.2 +/- 1.0 (63.4-76.5) kg for subathletic somatotype and 75.0 +/- 1.0 (68.4-92.5) kg for euriplastic somatotype. Mass of bone component was minimal for women of picnic somatotype (6.4 +/- 0.1 kg) as well as asthenic somatotype (7.3 +/- 0.3 kg) and maximal for subathletic (9.3 +/- 0.1 kg) and euriplastic (9.4 +/- 0.1 kg) somatotypes. Skeletal muscles mass was varied from 18.7 +/- 0.4 kg (asthenic somatotype) to 30.2 +/- 0.4 kg (athletic somatotype). Fat tissue mass was varied from 7.0 +/- 0.2 kg (asthenic somatotype) to 32.2 +/- 0.2 kg (euriplastic somatotype). There were established intercommunications between women constitutional characteristics and pecualiarities of their body construction.
