ETHNOPHARMACOLOGICAL RELEVANCE: Cancer stands as one of the leading causes of death worldwide according to the World Health Organization (WHO), and it has led to approximately 10 million fatalities in 2020. Medicinal plants are still widely used and accepted form of treatment for most diseases including cancer in Ghana. This review presented Cryptolepis nigrescens (Wennberg) L. Joubert. and Bruyns., Prosopsis africana (Guill. and Perr.) Taub. and Pterygota macrocarpa K. Schum. as medicinal plants that are traditionally used to treat tumour growth, amongst other diseases, in the Ashanti region of Ghana. AIM OF REVIEW: This paper aims to present a comprehensive review on the botanical description, ecological distribution, ethnomedicinal uses, phytochemical composition and ethnopharmacological relevance of C. nigrescens, P. africana and P. macrocarpa. MATERIALS AND METHODS: The review covers works published between 1962 and 2023 from various countries. Published books, thesis, scientific and medical articles on C. nigrescens, P. africana and P. macrocarpa were collected from the following databases: 'Scopus', 'Science Direct', 'Medline', 'PubMed', 'Research Gate' 'Google Scholar, and 'Springer link' using the keywords. RESULTS: Phytochemical analysis of C. nigrescens, P. africana and P. macrocarpa revealed the presence of some prominent bioactive compounds such as convallatoxin, 7,3,4-trihydroxy-3-methoxyflavanone and dioxane, respectively. Plant extracts and isolated compounds of these medicinal plants exhibited a wide range of ethnopharmacological activities including antimicrobial, anti-inflammatory, antioxidant, analgesic, cytotoxic, antimalarial, antipyretic, haematinic, hepato-protective, aphrodisiac and antihypertensive properties. CONCLUSION: The present review on C. nigrescens , P.africana and P. macrocarpa provided a credible summary of the ethnopharmacological research conducted on these medicinal plants till date. The data also highligted the potential therapeutic profiles of these plants in Ghana that could serve as foundation for future studies. Additionally, the information significantly supported the traditional and commercial use of these plants among the people.

Sida cordifolia has been used to treat malaria in Ghana albeit without scientific evidence of antimalarial activity and safety. This work aimed to assess the antimalarial properties and acute toxicity of the aqueous leaf extract of S. cordifolia in murine models. Aqueous extract of the plant was analysed for both suppressive and curative antimalarial properties in chloroquine-sensitive ANKA strains of rodent Plasmodium berghei-infected mice. Acute toxicity evaluation was performed in rats according to the OECD 425 guidelines. The extract displayed antiplasmodial activity in vivo with ED50 of 117.49 ± 15.22 mg/kg and 144.84 ± 18.17 mg/kg in suppressive and curative studies, respectively. The highest % parasitaemia suppression exerted was 76.90 ± 0.64% and 61.50 ± 0.97%, respectively, in the suppressive and curative studies. Survival of infected mice treated with the extract was significantly prolonged. This was dependent on the dose of the extract but imperfectly related to the % parasitaemia suppression. Related antimalarial parameters including percentage hematocrit, changes in body weight, and temperature of experimental mice indicated alleviation of malarial symptoms of treated animals. The extract did not show toxicity in rats. Sida cordifolia L. has antimalarial properties, and was safe. It suppressed parasitaemia in both suppressive and curative studies, was not toxic to animals and prolonged the life of infected animals under treatment. This, therefore, justifies the traditional use of S. cordifolia for the treatment of malaria in Ghana.

ETHNOPHARMACOLOGICAL SIGNIFICANCE AND AIM: Hura crepitans is commonly used to treat liver diseases in Nigeria and Ghana. Previous studies have supported its ethnomedicinal use in protecting the liver. The present study aimed at assessing the effect of H. crepitans stem bark on the subacute carbon tetrachloride (CCl4)-induced liver damage in rats. MATERIALS AND METHODS: The protective activities of ethanolic extract of H. crepitans stem bark was evaluated in CCl4-induced subacute liver damage in rats (1:1 v/v in olive oil, intraperitoneally (i.p.), twice weekly for 8 weeks). Blood samples were obtained from the rats and used for some biochemical analysis such as liver function test (Aspartate transaminase, AST; Alanine aminotransferase, ALT; and Alkaline phosphatase, ALP), liver fibrotic indices (Aspartate platelet ratio index, APRI; AST/ALT and AST/PLT ratios) and oxidative stress markers (Malondialdehyde, MDA; Reduced glutathione, GSH; Glutathione S-transferase, GST; Glutathione peroxidase, GPx; and superoxide dismutase, SOD). Histopathological analyses were carried out to determine the expression of pro-inflammatory (NF-κB, COX-2, IL-17 and IL-23) using immunohistochemical techniques. RESULTS: Oral administration of H. crepitans to CCl4-induced hepatic injured rats significantly decreased oxidative stress, increased the levels of SOD, GSH, GST and GPx with reduced MDA levels. The plant also mitigated liver injury as evidenced in the significantly reduced levels of AST, ALT and ALP, while it inhibited the inflammatory process via the inhibition of NF-κB, and consequently down-regulateed the pro-inflammatory cytokines COX-2, IL-17 and IL-23, respectively. Biochemical observations were supported by improvement in liver microarchitecture. CONCLUSION: The Hura crepitans demonstrated antioxidant, antiinflammatory and antifibrotic effect in hepatic injured rats. The study in a way justifies the traditional use of the plant for the treatment of subacute liver diseases in Nigerian Traditional medicine.

The leaf of Theobroma cacao L. is used in traditional medicine in Ghana for the treatment of malaria, yet, with no scientific evidence of its antimalarial property in animals. It was, therefore, studied to validate the antimalarial property in Plasmodium berghei-infected mice. Infected mice were treated with an aqueous extract of T. cacao leaf at different doses of 100, 200, and 400 mg/kg daily for four days. Parasitaemia was determined before treatment and 24 hours following the last dose of extract. The % reduction in parasitaemia and ED50 and ED90 of the extract were determined. Body weight, rectal temperature, and daily mortality of mice were also recorded. The extract had ED50 and ED90 of 242.20 ± 29.38 and 351.00 ± 29.52 mg/kg/day, respectively. Percentage parasitaemia suppression was significant for all doses. The extract at the maximum dose of 400 mg/kg body weight had the highest % parasitaemia suppression of 79.19%; mean survival time of 24.00 ± 2.19 days and median survival of 23 days; body weight increase of 3.82 ± 0.59; and the lowest body temperature reduction of 0.79 ± 0.11°C. T. cacao leaf extract showed an antimalarial property in P. berghei-infected mice. This reinforces the justification for the use of the plant material in treating malaria in Ghana.

Five new cassane-type diterpenoid heterosides, i. e. two cassane-type amides (1-2), two erythrophlamine-type amine esters (3-4) and a non­nitrogenous erythrophlamine analogue (5) were isolated from the root barks (1-2) and the seeds (3-5) of Erythrophleum suaveolens. Their structures were unambiguously established by interpretation of their HRESIMS, 1D and 2D NMR data, and chemical degradation for sugar determination. Compounds 3-5 were evaluated for their cytotoxicity against a panel of three cell lines, revealing modest to strong activities.

The use of herbal products has increased and become more popularized globally; however, limited studies coupled with questions related to the quality and safety of these herbal products have been raised. Herbal products with hope of their nontoxicity may play a role of alternative to overcome the problems of multi-drug resistant pathogens. Medicinal plants used as raw materials for production may have quality and safety issues due to proximity to wastewater application of fungicides and pesticides, which may be directly deposited superficially or absorbed by the plant system. Therefore, possible contamination of some Ghanaian herbal products cannot be ignored, as it may severely affect human life in the process of treatment. Aim. To evaluate the microbial load and the presence of toxic heavy metals in Mist Amen Fevermix and Edhec Malacure, two polyherbal products used in the treatment of uncomplicated malaria in Ghana. Methods. Thermo Elemental M5 Atomic Absorption Spectrophotometer (AAS) fitted with graphite furnace and an auto sampler was used to determine the heavy metal contents of the herbal products. The herbal samples were evaluated for the microbial load by using the appropriate culture media. Results and Analysis. Mist Amen Fevermix and Edhec Malacure complied with the safety limits evaluated for all different microbial counts and contamination. The following heavy metals were present in Mist Amen Fevermix and Edhec Malacure Mixture: Fe, Ni, K, Zn, Hg, Cu, Mn, Cr, Cd, Pb, Fe, Cu, K, and Na. Ni was below detectable limit in Edhec Malacure. Conclusion. Mist Amen Fevermix and Edhec Malacure may be assured of safety. The products contained heavy metals, but all were within acceptable limit established by the FAO/WHO. The levels of microbial contamination were below the maximum acceptable limit.

Liver disease is a major health problem and its treatment is costly in most developing countries with attendant adverse effects. This study aimed at determining the acute hepatoprotective efficacy of Duranta erecta hydroethanolic extracts of leaves, ripe and unripe fruits against CCl4-, and acetaminophen-induced hepatotoxicity in animals. Materials and Methods. CCl4 (1 mL/kg body weight in olive oil) and acetaminophen (500 mg/kg b.wt) were used to induce hepatotoxicity in the animals. Animals were treated with extracts at 250 mg/kg b.wt and standard drug, silymarin (100 mg/kg), for 7 days. Hepatoprotective efficacy was assessed by assaying serum biochemical markers such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyl transferase (γGT), bilirubin (Bil), antioxidative biomarkers including reduced glutathione (GSH), glutathione peroxidase (GPx), glutathione transferase (GST), superoxide dismutase (SOD), malondialdehyde (MDA), hydrogen peroxidase (H202), and nitric oxide (NO), as well as histological observations. Results. Exposure of the animals to CCl4 and acetaminophen resulted in liver injury as evidenced by elevated ALT, AST, ALP, γGT, Bil, MDA, H2O2, and NO levels with resultant derangement in liver microarchitecture. Pretreatment with hydroethanolic extracts, particularly ripe fruits of Duranta erecta, led to a reduction in these indicators and an increase in GSH, GPx, GST, and SOD levels. Biochemical data were supported by improvement in liver structure. Conclusion. The findings suggest that hydroethanolic Duranta erecta ripe fruits extract possesses hepatoprotective and antioxidative activities against CCl4- and acetaminophen-induced toxicity and could be developed as a potent agent for drug-induced liver diseases.

The use of plant-based medicine is popular amongst individuals and communities in developing countries. Duranta erecta has been used in Africa and Asia to treat a wide range of diseases. This study evaluated the phytochemical profile and antioxidant and antimicrobial activities of D. erecta to ascertain its health benefits in traditional medicine. Phytochemical constituents and antimicrobial effect of the hydroethanolic extract of D. erecta leaves (DRL), unripe fruits (DRU), and ripe fruits (DRR) were investigated by standard methods. Elemental analyses were carried out by atomic absorption spectroscopy (AAS) on the raw sample and extract. FTIR and UV-VIS spectroscopy were used to identify functional groups. Extracts were screened for their possible antioxidant activities by three tests. The total phenolic and total tannin contents were evaluated by using the Folin-Ciocalteu method. Total flavonoid content was determined by the aluminium chloride colorimetric assay method. The antioxidant activities were evaluated using the DPPH scavenging activity. The results of phytochemical screening showed the presence of triterpenoids, sterols, alkaloids, flavonoids, saponins, glycosides, and tannins. FTIR analysis revealed the presence of alcohols, phenols, alkanes, aldehydes, ketones, aromatics, aliphatic amines, aromatic amines, amides, carboxylic acids, esters, nitro compounds, alkynes, primary and secondary amines, and alkyl halides. Iron, zinc, and copper were also detected. Total phenolic and tannin contents ranged from 2.20 ± 0.15 to 14.54 ± 0.29 mg gallic acid equivalent (GAE)/100 g and 3.55 ± 0.07 to 13.82 ± 0.04 mg GAE/100 g, respectively. Total flavonoid content varied from 41.76 ± 0.96 to 343.49 ± 3.45 µg quercetin equivalent (QE)/100 g. The highest DPPH scavenging activity was recorded in the methanolic fraction of the leaves. The antimicrobial assay of the extract or fractions recorded no activity against the test organisms. The outcome of this study affirmed that D. erecta contains phytochemicals and bioactive compounds that could be of health benefit.

The use of medicinal plants for the treatment of diseases including malaria is commonplace in Ghanaian traditional medicine, though the therapeutic claims for most plants remain unvalidated. Antiplasmodial activity of the aqueous extracts and successively obtained petroleum ether, ethyl acetate and methanol fractions of the whole Phyllanthus fraternus plant, the leaves of Tectona grandis, Terminalia ivorensis and Bambusa vulgaris, and roots of Senna siamea were studied against Plasmodium falciparum chloroquine-sensitive 3D7 and chloroquine-resistant W2 strains. The aqueous extracts were assessed against human umbilical vein endothelial cells (HUVECs) for cytotoxicity, and the organic solvent fractions against human O(+) erythrocytes for haemolytic effect. Both extracts and fractions demonstrated antiplasmodial activity to varied extents. The aqueous extract of T. ivorensis was the most active (3D7, IC50 0.64 ± 0.14; and W2, IC50 10.52 ± 3.55 µg/mL), and together with P. fraternus displayed cytotoxicity (CC50 6.25 ± 0.40 and 31.11 ± 3.31 µg/mL, respectively). The aqueous extracts were generally selective for 3D7 strain of P. falciparum (selectivity indexes (SIs) ≥3.48) but only that of S. siamea was selective for the W2 strain (SI > 2.1). The organic solvent fractions also displayed antiplasmodial activity with the methanol fractions of P. fraternus and T. grandis, and the fractions of B. vulgaris showing activity with IC50 below 1 µg/mL against P. falciparum 3D7 strain; some fractions showed haemolytic effect but with low to high selectivity indexes (SI ≥ 4). The results while justifying the traditional use of the plant materials in the treatment of malaria, however, suggest their cautious use.

ETHNOPHARMACOLOGICAL RELEVANCE: Ethnobotanical survey was performed to document medicinal plants employed in the management of malaria in the Bosomtwe and Sekyere East Districts of the Ashanti Region (Ghana), in comparison with the plant ingredients in herbal antimalarial remedies registered by the Ghana Food and Drug Administration. MATERIALS AND METHODS: Two hundred and three (203) herbalists from 33 communities within the two districts were interviewed on medicinal plants they use to manage malaria. A literature search was made to determine already documented plants. In addition, 23 finished marketed herbal products indicated for the management of malaria were identified and their labels examined to find out which of the plants mentioned in our survey were listed as ingredients and whether these products are in anyway regulated. RESULTS: Ninety-eight (98) species of plants were cited for the management of malaria. In comparison with literature citations, 12 (12.2%) species were reported for the management of malaria for the first time and 20 (20.4%) others for the first time in Ghana. Twenty-three (23) finished marketed herbal antimalarial products examined contained aerial or underground parts of 29 of the plants cited in our survey as ingredients. Twenty-two (22) of these products have been registered by the Ghana Food and Drugs Authority, four (4) of which were included in the recommended herbal medicine list for treating malaria in Ghana. CONCLUSION: This study provides new additions to the inventory of medicinal plants used for the management of malaria and reports the commercial availability and regulation of finished marketed labelled herbal products intended for the treatment of malaria in Ghana.

BACKGROUND: Hoslundia opposita Vahl. (Lamiaceae), a common local shrub in Ghana, is traditionally known not only for its pharmacological benefits but also for its insecticidal properties. Its acaricidal property, however, has not been investigated. OBJECTIVE: To test the acaricidal effects of the crude extract and fractions of H. opposita leaves as well as to isolate and characterize the acaricidal principles. MATERIALS AND METHODS: The crude methanolic extract, pet. ether, ethyl acetate and aqueous fractions of the leaves of H. opposita were tested against the larvae of the cattle tick, Amblyomma variegatum, using the Larval Packet Test. A bioassay-guided isolation was carried out to identify the acaricidal principle obtained from the ethyl acetate fraction. RESULTS: The active principle was characterised as ursolic acid, a triterpene previously isolated from the leaves of the same plant. The extract and fractions were less potent than the control, malathion (LC(50) 1.14 × 10(-4) mg/ml). Among the plant samples however the crude methanolic extract exhibited the highest effect against the larvae (LC(50) 5.74 × 10(-2) mg/ml), followed by the ethyl acetate fraction (LC(50) 8.10 × 10(-2) mg/ml). Ursolic acid, pet. ether and aqueous fractions however showed weak acaricidal effects with LC(50) values of 1.13 mg/ml, 8.96 × 10(-1) mg/ml and 1.44 mg/ml, respectively. CONCLUSION: Ursolic acid was not as potent as the crude methanolic extract and the ethyl acetate fraction from which it was isolated. The overall acaricidal effect of H. opposita may have been due to synergy with other principles having acaricidal properties.
